Direct And Indirect Effects Of Invasive Cirsium Arvense On Pollination In Southern Appalachian Floral Communities, Jesse Daniels Dec 2018

Direct And Indirect Effects Of Invasive Cirsium Arvense On Pollination In Southern Appalachian Floral Communities, Jesse Daniels

Electronic Theses and Dissertations

Invasive plants can alter pollination dynamics in invaded communities by disrupting patterns of pollinator visitation, pollen transfer dynamics (conspecific [CP] and heterospecific [HP]), and reproductive success. The direction of invasive effects (competitive, neutral, and facilitative) may be partially determined by spatial scale and species’ floral traits. Here, we investigated pollinator visitation, CP and HP receipt, and pollen tube growth for species in a C. arvense present community and non-present community at two scales. At the community-level, the effect of C. arvense on pollinator visitation varied among species. Floral symmetry seemed to explain this variation. At the floral neighborhood-level, we found …

Localization Of Sip470, A Plant Lipid Transfer Protein In Nicotiana Tabacum, Shantaya Andrews Dec 2018

Localization Of Sip470, A Plant Lipid Transfer Protein In Nicotiana Tabacum, Shantaya Andrews

Electronic Theses and Dissertations

SABP2-interacting protein 470 (SIP470), a non-specific lipid transfer protein (nsLTP), was discovered in a yeast two-hybrid screening using SABP2 as bait and tobacco leaf proteins as prey. SABP2 is an important enzyme in systemic acquired resistance that converts salicylic acid to methyl salicylate. Localization studies are an important aspect to understanding the biological function of proteins. nsLTPs are generally considered apoplastic proteins and has been localized intracellularly and extracellularly. Transient expression shows highest expression of SIP470-eGFP at 2 days post infiltration into Nicotiana benthamiana. Confocal microscopy showed localization near the periphery of the cell. Subcellular localization using differential centrifugation showed …

Gut Microbiota Regulates The Interplay Between Diet And Genetics To Influence Insulin Resistance, Jeralyn Jones Franson Dec 2018

Gut Microbiota Regulates The Interplay Between Diet And Genetics To Influence Insulin Resistance, Jeralyn Jones Franson

Theses and Dissertations

Insulin resistance and obesity are major public health concerns. The impact of diet and genetics on insulin resistance and obesity is well accepted. Additionally, the gut microbiota has been shown to influence obesity and metabolic disorders. However, much remains to be understood about the role of gut microbiota in the development of insulin resistance and obesity. We utilized a mouse model lacking PAS kinase, a protein involved in cellular metabolism, in order to better understand the relationship between diet, genetics and the gut microbiota. Previous research has shown that mice lacking PAS kinase were protected from the effects of a …

Engineering Cell-Free Biosystems For On-Site Production And Rapid Design Of Next-Generation Therapeutics, Kristen Michelle Wilding

Theses and Dissertations

While protein therapeutics are indispensable in the treatment of a variety of diseases, including cancer, rheumatoid arthritis, and diabetes, key limitations including short half-lives, high immunogenicity, protein instability, and centralized production complicate long-term use and on-demand production. Site-specific polymer conjugation provides a method for mitigating these challenges while minimizing negative impacts on protein activity. However, the location-dependent effects of polymer conjugation are not well understood. Cell-free protein synthesis provides direct access to the synthesis environment and rapid synthesis times, enabling rapid evaluation of multiple conjugation sites on a target protein. Here, work is presented towards developing cell-free protein synthesis as …

Microbial And Environmental Drivers Of Soil Respiration Differ Along Montane To Urban Transitions, Kerri Ann Russell Dec 2018

Microbial And Environmental Drivers Of Soil Respiration Differ Along Montane To Urban Transitions, Kerri Ann Russell

Theses and Dissertations

In natural ecosystems, like deciduous and coniferous forests, soil CO2 flux or soil respiration is highly variable and influenced by multiple factors including temperature, precipitation, dissolved soil organic carbon (DOC), dissolved organic matter (DOM), and bacterial and fungal biomass and diversity. However, as the human population continues to grow rapidly, so too do urbanized landscapes with unknown consequences to soil respiration. To determine the extent urbanization influences seasonal shifts in microorganisms and environmental drivers alter soil respiration, we evaluated bacterial and fungal communities, soil physiochemical characteristics, and respiration in forested and urbanizing ecosystems in three watersheds across northern Utah, USA. …

Repopulation And Stimulation Of Porcine Cardiac Extracellular Matrix To Create Engineered Heart Patches, Silvia Juliana Moncada Diaz Dec 2018

Repopulation And Stimulation Of Porcine Cardiac Extracellular Matrix To Create Engineered Heart Patches, Silvia Juliana Moncada Diaz

Theses and Dissertations

Heart failure is the main cause of death for both men and women in the United States. The only proven treatment for patients with heart failure is heart transplantation. The goal of this research is to create patches of tissue that could mimic the function of the native heart to repair the damaged portions of the heart. In this study, whole porcine hearts were decellularized to create a 3D construct that was recellularized with cardiomyocytes (CM) differentiated from human induced pluripotent stem (IPS) cells. At day 4 of differentiation, IPS-derived CMs were implanted onto cardiac extracellular matrix (cECM) and ten …

Characterization Of Pyrolysis Products From Fast Pyrolysis Of Live And Dead Vegetation, Mohammad Saeed Safdari Dec 2018

Characterization Of Pyrolysis Products From Fast Pyrolysis Of Live And Dead Vegetation, Mohammad Saeed Safdari

Theses and Dissertations

Wildland fire, which includes both planned (prescribed fire) and unplanned (wildfire) fires, is an important component of many ecosystems. Prescribed burning (controlled burning) is used as an effective tool in managing a variety of ecosystems in the United States to reduce accumulation of hazardous fuels, manage wildlife habitats, mimic natural fire occurrence, manage traditional native foods, and provide other ecological and societal benefits. During wildland fires, both live and dead (biomass) plants undergo a two-step thermal degradation process (pyrolysis and combustion) when exposed to high temperatures. Pyrolysis is the thermal decomposition of organic material, which does not require the presence …

In amyotrophic lateral sclerosis (ALS), mitochondrial dysfunction and oxidative stress form a vicious cycle that promotes neurodegeneration and muscle wasting. To quantify the disease-stage-dependent changes of mitochondrial function and their relationship to the generation of reactive oxygen species (ROS), we generated double transgenic mice (G93A/cpYFP) that carry human ALS mutation SOD1G93A and mt-cpYFP transgenes, in which mt-cpYFP detects dynamic changes of ROS-related mitoflash events at individual mitochondria level. Compared with wild type mice, mitoflash activity in the SOD1G93A (G93A) mouse muscle showed an increased flashing frequency prior to the onset of ALS symptom (at the age of 2 months), whereas …

Theses and Dissertations

The Intermountain West is comprised of impressive land formations, numerous ecoregions, and a unique biota. The area has many flora and fauna that have been investigated, but the region is generally considered undersampled when it comes to insects. However, I propose the matter to be a lack of shared experience in identifying key insect species and the underutilization of professional and personal collections. These impediments are highlighted by two insect groups in the Intermountain West: spongillaflies and tiger moths.Spongillaflies can be difficult to recognize for the general entomologist and have rarely been recorded in the Intermountain West. My colleagues and …

The Design, Construction, And Thermal Diffusivity Measurements Of The Fluorescent Scanning Thermal Microscope (Fstm), Samuel Hunter Hayden

Theses and Dissertations

Over the life of nuclear fuel, inhomogeneous structures develop, negatively impacting thermal properties. New fuels are under development, but require more accurate knowledge of how the properties change to model performance and determine safe operational conditions. Measurement systems capable of small–scale, pointwise thermal property measurements and low cost are necessary to measure these properties and integrate into hot cells where electronics are likely to fail during fuel investigation. This project develops a cheaper, smaller, and easily replaceable Fluorescent Scanning Thermal Microscope (FSTM) using the blue laser and focusing circuitry from an Xbox HD-DVD player. The FSTM also incorporates novel fluorescent …

Journal Articles

This Article predicts trademark law’s impending neural turn. A growing legal literature debates the proper role of neuroscientific evidence. Yet outside of criminal law, analysis of neuroscientific evidence in the courtroom has been lacking. This is a mistake given that most of the applied research into brain function focuses on building better brands, not studies of criminal defendants’ grey matter. Judges have long searched for a way to measure advertising’s psychological hold over consumers. Advertisers already use brain imaging to analyze a trademark’s ability to stimulate consumer attention, emotion, and memory. In the near future, businesses will offer a neural …

Superelastic And Ph-Responsive Degradable Dendrimer Cryogels Prepared By Cryo-Aza-Michael Addition Reaction, Juan Wang, Hu Yang

Chemical and Biochemical Engineering Faculty Research & Creative Works

Dendrimers exhibit super atomistic features by virtue of their well-defined discrete quantized nanoscale structures. Here, we show that hyperbranched amine-terminated polyamidoamine (PAMAM) dendrimer G4.0 reacts with linear polyethylene glycol (PEG) diacrylate (575 g/mol) via the aza-Michael addition reaction at a subzero temperature (-20 °C), namely cryo-aza-Michael addition, to form a macroporous superelastic network, i.e., dendrimer cryogel. Dendrimer cryogels exhibit biologically relevant Young's modulus, high compression elasticity and super resilience at ambient temperature. Furthermore, the dendrimer cryogels exhibit excellent rebound performance and do not show significant stress relaxation under cyclic deformation over a wide temperature range (-80 to 100 °C). The …

Nkx2-1-As1 Negatively Regulates Cd274/Pd-L1, Cell-Cell Interaction Genes, And Limits Human Lung Carcinoma Cell Migration., Hasmeena Kathuria, Guetchyn Millien, Liam Mcnally, Adam C. Gower, Jean-Bosco Tagne, Yuxia Cao, Maria I. Ramirez

Center for Translational Medicine Faculty Papers

The function of most long noncoding RNAs (lncRNAs) is unknown. However, recent studies reveal important roles of lncRNAs in regulating cancer-related pathways. Human antisense lncRNA-NKX2-1-AS1 partially overlaps the NKX2-1/TTF1 gene within chromosomal region 14q13.3. Amplification of this region and/or differential expression of genes therein are associated with cancer progression. Herein we show higher levels of NKX2-AS1 and NKX2-1 in lung adenocarcinomas relative to non-tumor controls but no correlation between NKX2-1-AS1 and NKX2-1 levels across specimens, or with amplification of the 14q13.3 region, suggesting that NKX2-1-AS1 and NKX2-1 are independently regulated. Loss-and-gain of function experiments showed that NKX2-1-AS1 does not regulate …

Evaluation Of Alternative Valorization Options For Institutional And Industrial Food Wastes, Swati Hegde Dec 2018

Evaluation Of Alternative Valorization Options For Institutional And Industrial Food Wastes, Swati Hegde

Theses

In recent years many countries and several regions in the United States have passed legislation on banning landfilling of organic waste. It is a well-known fact that organic wastes, including food scraps, generate methane under anaerobic landfill environments and contribute significantly to global warming. As more regions are mandated to follow the landfill ban, there is an increasing demand for alternative technologies for the disposal, treatment and upcycling of food wastes. Disposal and treatment may not result in any value-added products and hence are not the most efficient ways to manage food waste. As food waste has a high energy …

Case Study: Environmental Requirements And Its Impacts Upon The Conservation Center For Wildlife Care, Chase Flemmer Dec 2018

Case Study: Environmental Requirements And Its Impacts Upon The Conservation Center For Wildlife Care, Chase Flemmer

Construction Management

As the population of California continues to grow, urban spaces are rapidly being developed into areas considered to be environmentally sensitive. In order to prevent extensive damage to our state’s delicate ecosystems, environmental regulations are often enforced by local organizations during the course of impactful operations such as construction projects. These regulations will often vary from one site to another, depending on a given site’s location and proximity to the environmentally sensitive areas. No two jobs will posses the exact same specifications for dealing with this aspect of construction, and as such some will require more intense regulations than others. …

Noise Induced Hearing Loss Accountability On Jobsites Within The Construction Industry, Joseph O. Lamphere Dec 2018

Noise Induced Hearing Loss Accountability On Jobsites Within The Construction Industry, Joseph O. Lamphere

Construction Management

Noise induced hearing loss is a major issue on jobsites within the construction industry. Every day workers are exposed to hazardous levels of noise, but they are not being properly managed to mitigate this issue. Workers have misconceptions about the topic, but more surprisingly are sometimes indifferent to the effects; believing hearing loss is inevitable, or it can be cured with a hearing aide. Construction has inherent dangers that workers have to deal with every day. Hearing loss might not be on the top of their lists, but the effects are permanent, and they cannot be cured with a hearing …

Heteroleptic Nickel Complexes Of A Bulky Bis(Carbene)Borate Ligand, Jorge L. Martinez, Wei-Tsung Less, Maren Pink, Chun-Hsing Chen, Jeremy M. Smith Dec 2018

Heteroleptic Nickel Complexes Of A Bulky Bis(Carbene)Borate Ligand, Jorge L. Martinez, Wei-Tsung Less, Maren Pink, Chun-Hsing Chen, Jeremy M. Smith

Chemistry: Faculty Publications and Other Works

Bis(carbene)borate ligand transfer to the nickel nitrosyl synthon Ni(NO)(PPh3)2Br provides the new nickel nitrosyl complex Ph2B(t BuIm)2Ni(NO)(PPh3). The solid state structure reveals a trigonal pyramidal nickel ion with a very long bond to the apical PPh3 ligand. The complex reversibly dissociates PPh3 in solution to afford three‐coordinate Ph2B(t BuIm)2Ni(NO), with NMR data providing evidence for PPh3 binding at low temperatures. Ligand transfer to Ni(PMe3)2Cl2 provides the square planar complex, Ph2B(t BuIm)2Ni(PMe3)Cl, which shows no evidence for rearranging to the form a homoleptic complex with two bis(carbene)borate ligands. This complex is a suitable synthon for the [Ph2B(t BuIm)2Ni]+ fragment, as demonstrated …

‘Presume Not That I Am The Thing I Was’: Altering Perceptions Of The Disabled Via The Staging Of Disability In Early Modern England, William Nyfeler

All NMU Master's Theses

Attitudes toward people with physical or mental disabilities have varied throughout history. Each society collectively defines what is considered normal and abnormal, and those values change over time. Many cultural factors impact how much these views change, including the dominant social philosophies and religions of an era. In Early Modern England, the rise of large public theaters and an increasingly permissive society contributed to the development of plays becoming a powerful tool for swaying public opinion.

Using this new pulpit, Shakespeare and his contemporaries staged plays that often depicted disability and deformity in negative ways, including the implications that a …

Limited By Design: Expanding The Boundaries Of Schooling Policies From Demanded Passivity To Transformative Practice, Natalia T. Carvalho-Pinto

Masters Theses

Schooling is often imagined as the great equalizer and the primary venue towards personal and collective liberation. However, hegemonic ideals and practices have limited the ability for educators to craft and utilize policy in a way that best benefits their students. In this thesis, I utilize the state of Michigan’s Common Core curriculum to examine the limits beyond policy and practice. Specifically, I argue that the demands for student passivity and institutional measurable as signs of success, prohibit student growth. Throughout the document, I describe hope in the ability to instill a variety of transformative educational theories and practices that …
