The overarching goals of the current work are to fill key gaps in the current understanding of alcohol consumption and the risk of metastasis to the liver. Considering the evidence this research group has compiled confirming that the hepatic matrisome responds dynamically to injury, an altered extracellular matrix (ECM) profile appears to be a key feature of pre-fibrotic inflammatory injury in the liver. This group has demonstrated that the hepatic ECM responds dynamically to alcohol exposure, in particular, sensitizing the liver to LPS-induced inflammatory damage. Microbotryum lychnidis-dioicae causes anther smut fungus in its host plant, Silene latifolia. The goal of this work is to identify and characterize the virulence determinants of this pathogen to better understand the molecular basis behind this host-pathogen interaction. This work studied for the first time the key effectors in the mechanism of infection by this fungal species. Using, yeast two-hybrid screens, I have identified the host plant interaction partners for the effector, MVLG_01732. A second effector MVLG_05720, interacts with other fungal proteins that appear to facilitate the fungal establishment and colonization during the infection. Our findings indicate that a …

The aim of this dissertation study was to examine the impact of the implementation of a new Compassionate Care (CC) curriculum on the quality of care provided by Certified Nursing Assistants (CNAs) to residents with Alzheimer’s disease (AD). More specifically, this dissertation used Kirkpatrick’s model of evaluation to assess the reactions, learning, and behavior change of the CNAs exposed to the curriculum, and ultimately the impact of the curriculum on the stress levels of residents with AD. Familial hypercholesterolemia (FH) is a common genetic disease and has been studied with the aim of finding a curative measure for decades. FH is caused by mutations in the low-density lipoprotein receptor (LDLR) resulting in defects in LDL-cholesterol (LDL-C)-receptor mediated endocytosis and development of premature cardiovascular disease (CVD). Here I describe the use of a corrected and non-corrected LDLR FH cell model to investigate receptor-mediated endocytosis and statin effects. For these studies, we reprogrammed FH fibroblast cells to induced pluripotent stem cells (iPSC) and confirmed their pluripotency and ability to differentiate to hepatocyte-like cells (HLC). A recent study demonstrated an association between the Porphyromonas gingivalis (Pg)infection and the progression of ESCC (Gao et al., 2016). However, how Pg infection affects the nature of ESCC remains unknown. We examined the effects of Pg on the proliferation and chemotherapy drug-induced apoptosis of ESCC cells and elucidated the signaling molecules involved in these processes. Utilizing a holistic approach that includes local voices may help a program’s effectiveness. Scholars are searching for a framework flexible enough to accommodate the varied nature of programs. This dissertation examined the contributing factors to the success of high achieving African American male high school students who attend an all-male, predominantly Black, college-preparatory academy. The study took place in an urban, Midwestern school district and highlighted the lived experience and perspectives of students who have at least a 3.0 grade point average. This research builds upon the framework of the Health Traditions Model (HTM) as described in 1994 by Rachel Spector, a scholar and practitioner of a cultural care model of health among multicultural populations. I integrate this model into a study of Syrian refugees’ resettlement experience in Louisville, Kentucky. Syrian refugees’ voices and narratives of their corresponding Arab-Islamic and folk approaches to health, illness, and healing are the launch site for this project. Accurate disulfide bond formation is important for proper folding, stability and function of exported proteins. The process of disulfide bond formation, termed oxidative protein folding, is catalyzed by thiol-disulfide oxidoreductase enzymes. Oxidative protein folding pathways influence processes essential for bacterial physiology and pathogenicity. In the Gram-positive actinobacterial pathogens Actinomyces oris and Corynebacterium diphtheriae oxidative protein folding is catalyzed by the primary thiol-disulfide oxidoreductase MdbA. MdbA is required for assembly of adhesive pilus, which mediate receptor-dependent bacterial interactions, or coaggregation, in A. oris. Prostate apoptosis response-4 (Par-4) is a 38 kDa largely intrinsically disordered tumor suppressor protein that functions in cancer cell apoptosis. Par-4 down-regulation is often observed in cancer while up-regulation is characteristic of neurodegenerative conditions such as Alzheimer’s disease. Cleavage of Par-4 by caspase-3 activates tumor suppression via formation of an approximately 25 kDa fragment (cl-Par-4) that enters the nucleus and inhibits Bcl-2 and NF-ƙB, which function in pro-survival pathways. Here, we have investigated the structure of cl-Par-4 using biophysical techniques including circular dichroism (CD) spectroscopy, dynamic light scattering (DLS), and intrinsic tyrosine fluorescence. Microalgae are one of the most promising feedstocks for biodiesel production due to their high lipid content and easy farming. However, the extraction of lipids from microalgae is energy intensive and costly and involves the use of toxic organic solvents. Compared with organic solvent extraction, supercritical CO2 (SCCO2) has demonstrated advantages through lower toxicity and no solvent-liquid separation. Ultrahigh stocking density (a.k.a., mob grazing) is proposed as a management tool that results in greater harvest efficiency, animal performance, aboveground plant production, species richness, and soil carbon content. The study objective was to determine grazing treatment, haying, or non-defoliated control effects on forage utilization, aboveground production, animal performance, and soil properties. Recently, imaged-based high-throughput phenotyping methods have gained popularity in plant phenotyping. Imaging projects the 3D space into a 2D grid causing the loss of depth information and thus causes the retrieval of plant morphological traits challenging. Biological diversity, or biodiversity, is a multi-dimensional concept that can be decomposed to measure information about taxonomic, phylogenetic, and functional variation within communities. Although the dimensions of biodiversity are interrelated, the assumption that measuring one dimension of diversity can inform about patterns in another dimension does not necessarily follow from theory or empirical study. Infectious disease assays can be imperfect. When estimating disease prevalence, these imperfections are accounted for by incorporating assay sensitivity and specificity into point and variance estimates. Unfortunately, these accuracy measures are often treated as fixed constants, rather than acknowledging that they are estimates from an assay validation process. The purpose of this study is to show the detrimental effect of not taking into account this sampling variability when samples are obtained through group testing (aka, pooled testing). Real time spatial information about water balance components is important for designing VRI prescription maps. Brown adipose tissue (BAT) is a crucial regulator in energy expenditure by dissipating energy in the form of heat through mitochondrial uncoupling protein 1 (UCP1). Previously, we demonstrated that n-3 PUFA (Omega-3 Polyunsaturated Fatty Acids) increase brown adipogenesis by miRNA-mediated epigenetic mechanisms. Given that BAT is formed at the late stage of pregnancy, we hypothesized prenatal exposure of n-3 PUFA through maternal nutrition potentiates BAT development. Primary objectives of the present study include exploring the role of both A1C values and the anti-diabetic medication, metformin, on cognitive decline through measured by a brief neuropsychological exam (BNE) and the Montreal Cognitive Assessment (MoCA). Method: Eleven years of archival cognitive testing and medical information from a total of 479 East Central Florida Memory Disorder Clinic patients (52.8 % female, 88.5% Caucasian, Mage = 78.79 years) was utilized for the present study.
