We report the optimization of detergent-mediated reconstitution of an integral membrane-bound protein, full-length influenza M2 protein, by direct insertion into detergent-saturated liposomes. Detergent-mediated reconstitution is an important method for preparing proteoliposomes for studying membrane proteins, and must be optimized for each combination of protein and membrane constituents used. The purpose of the reconstitution was to prepare samples for site-directed spin-labeling electron paramagnetic resonance (SDSL-EPR) studies. Our goals in optimizing the protocol were to minimize the amount of detergent used, reduce overall proteoliposome preparation time, and confirm the removal of all detergent. The liposomes were comprised of (1-palmitoyl-2-oleyl-sn-glycero-phosphocholine (POPC) and 1-palmitoyl-2-oleyl-sn-glycero-3-[phospho-rac-(1-glycerol)] …

Infertility affects 6.7 million women in the United States (Chandra, Copen, & Stephen, 2013). Women’s experiences with infertility are not only influenced by biological health factors, but also by social, cultural, and personal variables. Given the prevalence and complexity of infertility, additional research is needed to further examine the nuances of women’s experiences. The purpose of this multicase study, as informed by four individual cases, was to explore how women construct their infertility narratives. Review of reflective journals found five common elements: (1) Emotional Rollercoaster, (2) Mind-Body (Dis)Connection, (3) Secret Identity, (4) Supportive vs. Constrained Communication Patterns, and (5) Fatalistic- …

Patient safety is a key aspect for good consumer care. When an individual is hospitalized or receives medication the family wants the patient safety to be above all factors. For instance, a drug can do both either cure the disease or perhaps, give rise to an adverse event. A drug administered for an indicated condition has substantial power to reduce or cure a disease and further to prevent it from happening again in the future but at the risk of side effects. At present, there are several methods in patient safety and in particular in the area of signal detection …

Towards End-To-End Semi-Supervised Deep Learning For Drug Discovery, Xiaoyu Zhang

Computer Science and Engineering Theses - Archive

Observing the recent progress in Deep Learning, the employment of AI is surging to accelerate drug discovery and cut R&D costs in the last few years. However, the success of deep learning is attributed to large-scale clean high-quality labeled data, which is generally unavailable in drug discovery practices. In this thesis, we address this issue by proposing an end-to-end deep learning framework in a semi supervised learning fashion. That is said, the proposed deep learning approach can utilize both labeled and unlabeled data. While labeled data is of very limited availability, the amount of available unlabeled data is generally huge. …

Planning algorithms have attained omnipresent successes in several fields including robotics, animation, manufacturing, drug design, computational biology and aerospace applications. Path Planning is an essential component for autonomous robots. The problem involves searching the configuration space and constructing a desired collision-free path that connects two states (the start and the goal) for a robot to gradually navigate from one state to another. In global path planners, the complete path is computed prior to the robot set off. Sampling based planning like Rapidly Expanding Random Trees (RRT) and Probabilistic Road Maps (PRM) used for single or multi-query planning has gained popularity …

Truth And Strength In Vulnerability: Using Topoanalysis To Reveal The Need For Expanding The Modernist Literary Canon, Misty Dawnmarie Falkenstein

English Theses - Archive

In the late nineteenth and early twentieth centuries, the public role of women shifted dramatically. Women asserted themselves in politics, education, and work in a way foreign to their Victorian predecessors. Although these “New Women” altered the gender landscape and set in motion a new path for women that would continue even into the twenty-first century, their writing still goes largely unnoticed in the current study of the Modernist literary canon. This project makes a case for expanding the current Modernist literary canon to include more of these women, especially women of disenfranchised racial, ethnic, cultural, and economic groups. Writers …

Methanogens, Plausible Extraterrestrial Life Forms On Mars, And Their Tolerance To Increasing Concentrations Of Illite Clay, Chandler Kern

Biological Sciences Undergraduate Honors Theses

Methanogens, some of Earth’s most primitive prokaryotic organisms, are candidates for possible life forms capable of inhabiting Mars. Specifically, four different species (Methanobacterium formicicum, Methanococcus maripaludis, Methanosarcina barkeri, Methanothermobacter wolfeii) were analyzed for their tolerance to the presence of illite clay. Illite is a crystalline mineral that has been identified from regions of Mars’s surface. Results indicated that all four species grew with some success in the illite at different concentrations. This experimentation with methanogens’ abilities to survive and reproduce in the presence of illite allows for a more accurate understanding of the potential capability of microbial …

Synergistic Negative Effects Of Thermal Stress And Altered Food Resources On Echinoid Larvae, Colette Feehan, Zoe Ludwig, Suzannah Yu, Diane K. Adams

Department of Biology Faculty Scholarship and Creative Works

Multiple changes to the marine environment under climate change can have additive or interactive (antagonistic or synergistic) effects on marine organisms. Prompted by observations of anomalously warm sea temperatures and low chlorophyll concentrations during the 2013–2016 warm “Blob” event in the Northeast Pacific Ocean, we examined the combined effects of thermal stress and a shift in food resources on the development of a larval echinoid (Strongylocentrotus droebachiensis) in the laboratory. A high concentration of phytoplankton yielded faster echinus rudiment development at warm versus historical temperature, indicating a mitigating effect of abundant food on thermal stress; however, low phytoplankton concentration or …
