Compartmentalization Of Dna-Based Molecular Computing Elements Using Lipid Bilayers, Aurora Fabry-Wood
Compartmentalization Of Dna-Based Molecular Computing Elements Using Lipid Bilayers, Aurora Fabry-Wood
Biomedical Engineering ETDs
This dissertation will present a progression from the detection of double-stranded DNA using a combination of toehold-mediated strand displacement and DNAzyme reactions in dilute saline solutions, to the generation of separate compartments to allow standardization of DNA computing elements, by protecting from complementary strands. In well-mixed solutions complementary regions cause spurious interactions. Importantly, these compartments also provide protection from nucleases. Along the way we will also explore the use of silica microsphere supported lipid bilayers to run compartmentalized DNA reactions on a fluid surface and the design of a molecule capable of DNA-based transmembrane signal transduction.

Huntington's disease (HD) is a rare autosomal dominant neurodegenerative disorder caused by a cytosine-adenine-guanine (CAG) trinucleotide repeat (TNR) expansion within the HTT gene. The mechanisms underlying HD-associated cellular dysfunction in pluripotency and neurodevelopment are poorly understood. We had previously identified downregulation of selected DNA repair genes in HD fibroblasts relative to wild-type fibroblasts, as a result of promoter hypermethylation. Here, we tested the hypothesis that hypomethylation during cellular reprogramming to the induced pluripotent stem cell (iPSC) state leads to upregulation of DNA repair genes and stabilization of TNRs in HD cells. We sought to determine how the HD TNR region …

An Evaluation Of Unmanned Aerial System Multispectral And Thermal Infrared Data As Information For Agricultural Crop And Irrigation Management, Mitch Maguire
Department of Agricultural and Biological Systems Engineering: Dissertations, Theses, and Student Research
Spatial irrigation management has been steadily advancing over the last several years. A current issue with managing irrigation spatially on sub-field scale is the inability to readily collect the spatial field data necessary to properly manage irrigation. Multispectral and thermal infrared imagery used in informing irrigation management decisions was previously collected by satellite and manned aircraft remote sensing platforms. These remote sensing platforms pose issues concerning economic feasibility, revisit intervals, and weather factors that inhibit the collection of data. Recent developments in unmanned aerial systems, which provide an additional means of collecting multispectral and thermal infrared data, have the potential …

The Impact Of Violence On The Emerging Development Of Emotion Regulation: The Role Of The Caregiver, Christina Caiozzo
The Impact Of Violence On The Emerging Development Of Emotion Regulation: The Role Of The Caregiver, Christina Caiozzo
Dissertations (1934 -)
Caregivers teach children how to navigate an emotional world (Thompson, 1994), and children’s ability to manage emotional reactions underlies their mental health as well as their social and academic performance (Zeman, Cassano, Perry-Parish, & Stegall, 2006). However, children who experience adverse life events are at risk for impaired development of emotion regulation (Maughan & Cicchetti, 2002). Little is known about how parents of at-risk youth can continue to foster healthy development of emotion regulation for their children. Therefore, the current study aims to identify specific parenting practices that promote adaptive emotion regulation in at-risk preschoolers. Multi-method, multi-informant data were collected …

Design, Synthesis And Study Of Redox And Optoelectronic Properties Of Aromatic Oxidants And Polycyclic Aromatic Hydrocarbons, Mohammad Mosharraf Hossain
Dissertations (1934 -)
Organic materials play a significant role for the next generation photovoltaic devices that convert solar energy into usable forms of energy. In this regard, polycyclic aromatic hydrocarbons (PAHs) are fundamental tools in the developing area of molecular electronics and photovoltaics as they show excellent optical/electronic properties and are well-suited for applications in such developing areas as flexible display devices, field effect transistors and solar cell panels. Design and synthesis of novel materials for photovoltaics applications would require the proper understanding the mechanism of charge transport and identification of the structural features necessary in a particular molecular wire or PAH. Inference And Analysis Of Multilayered Mirna-Mediated Networks In Cancer, Duc Do
Inference And Analysis Of Multilayered Mirna-Mediated Networks In Cancer, Duc Do
Dissertations (1934 -)
MicroRNAs (miRNAs) are small noncoding transcripts that can regulate gene expression, thereby controlling diverse biological processes. Aberrant disruptions of miRNA expression and their interactions with other biological agents (e.g., coding and noncoding transcripts) have been associated with several types of cancer. The goal of this dissertation is to use multidimensional genomic data to model two different gene regulation mechanisms by miRNAs in cancer. This dissertation results from two research projects. The first project investigates a miRNA-mediated gene regulation mechanism called competing endogenous RNA (ceRNA) interactions, which suggests that some transcripts can indirectly regulate one another's activity through their interactions with …
Understanding Relationships Between Early Life Toxic Stress, Childhood Socioeconomic Disadvantage, And Allostatic Load In Adolescence, Amanda King
Dissertations (1934 -)
Chronic disease prevalence among children and adolescents is rising, which is thought to result in part from elevations in allostatic load (AL). AL is the cumulative physiological dysregulation that results from exposure to biological, social and environmental stressors over time. Socioeconomic disparities in chronic disease and AL have been well-documented in adult populations, including links between childhood socioeconomic disadvantage (CSD) and AL, yet little is known as to whether CSD may begin to impact AL earlier in life. Differential exposure and vulnerability to stress among racial/ethnic minorities may increase risk for elevated AL among those experiencing CSD. Framed by the …
The Forbidden Zone Writers: Femininity And Anglophone Women War Writers Of The Great War, Sareene Proodian
The Forbidden Zone Writers: Femininity And Anglophone Women War Writers Of The Great War, Sareene Proodian
Dissertations (1934 -)
This dissertation examines the texts of Anglophone women writers from the First World War. Women’s roles in the war—volunteer nurses, ambulance driver, munitions workers, and land girls—gave them the opportunity to leave the protection of their homes and enter the masculine dominated public sphere. In this dissertation, I examine different genres of women’s writing from the war and trace three aspects of simultaneity as these writings explore the new freedoms, and new and old constraints, that the war brought to women. The three principles of simultaneity explain the conflicting emotions women feel over what the war means for them in …

Pre-Death Grief, Resourcefulness And Perceived Stress Among Care Givers Of Partners With Young Onset Dementia, Karie Kobiske
Dissertations (1934 -)
Over 200,000 Americans are diagnosed with young onset dementia (YOD). YOD is the dementia diagnosed prior to the age of 65. Most persons of YOD are cared for by their partners. Caregiving for a partner diagnosed with YOD has unique challenges including multiple losses resulting from the functional, cognitive, and behavioral declines which can be demanding and stressful. These losses experienced by the caregiver of a partner diagnosed with YOD have been termed pre-death grief. Caregivers of partners with YOD often report high levels of burden and stress resulting in depression, anxiety, hopelessness, as well as increase morbidity and mortality. …

Global warming, as a consequence of excessive CO2 production mainly due to anthropogenic actions, is one of the main concerns of society due to the effects it can cause in the survival of humans, plants and animals. Several climatic consequences have already been reported, such as warming the oceans and changing biodiversity in various regions of the planet. One of the greenhouse gases responsible for global warming, which causes a lot of concern, is methane gas from digestion of food by ruminants. Besides that, emissions of greenhouse gases are represented also by waste management, rice cultivation, burning of residues from …

Climatic Drivers Of Ponderosa Pine Growth In Central Idaho, Joseph L. Pettit, R. Justin Derose, James N. Long
Wildland Resources Faculty Publications
Despite the widespread use of ponderosa pine as an important hydroclimate proxy, we actually understand very little about its climate response in the Northern Rockies. Here, we analyze two new ponderosa pine chronologies to investigate how climate influences annual growth. Despite differences in precipitation amount and timing and large elevation differences (1820 m versus 1060 m), ring width at both sites was strongly driven by water availability. The mid-elevation, water-limited site responded well to previous fall precipitation whereas the wetter, high-elevation site responded to growing season precipitation and temperature. When precipitation and temperature were simultaneously accounted for using the standardized …
Reducing Urban Pollution From Coastal Settlements With Green Roofs, Alexander Phillips
Reducing Urban Pollution From Coastal Settlements With Green Roofs, Alexander Phillips
The Plymouth Student Scientist
It was identified that traditional infrastructural methods for solving coastal pollution were becoming too costly and insufficient in combatting the effects of climate change. This report investigates the sustainable drainage method of green roofing as a possible approach to reducing coastal pollution. A generic computational hydraulic model was created, using Plymouth as a design template, to represent 3 different scenarios in which a combined sewage system would require overflow relief. Storm simulations were run through the scenarios to determine a set of baseline sewage spilling results. The simulations were rerun after allowances for green roofs, with substrates of 50mm and …
How Does Individual Quality And Mate Attractiveness Affect Courtship And Mating Behaviour?, Laura Cook
How Does Individual Quality And Mate Attractiveness Affect Courtship And Mating Behaviour?, Laura Cook
The Plymouth Student Scientist
Courtship and mating are costly and as a result, organisms are faced with trade-off decisions between the costs and benefits which arise from them. Individuals use information on their own quality and the attractiveness of their potential mate to determine whether to court and mate, and if so how much energy to invest in these activities. An individual’s quality can affect its ability to perform sexual activities and determine the energy the individual is able to invest in such activities. Organisms assess one another on their relative attractiveness, which allows them to determine the relative benefits they will receive from …

Trichomonas Vaginalis Cytoadherence Is Modulated By Oxidative Stress, Magali Ayala
Trichomonas Vaginalis Cytoadherence Is Modulated By Oxidative Stress, Magali Ayala
Theses and Dissertations
The adherence of Trichomonas vaginalis to the human urogenital tract is a prerequisite for the pathogenesis of the infection. This multifactorial process is modulated by environmental factors such as iron, estrogen, temperature and pH and specific receptor/ligand interactions. During natural infections, the anaerobe T. vaginalis is exposed to oxygen resulting in oxidative stress. This environmental change may alter trichomonad cytoadherence to the ectocervical tissue. Therefore, it was hypothesized that changes in the redox status of T. vaginalis may modulate cytoadherence levels. Results showed that oxidative stress reduces levels of trichomonad cytoadherence to HeLa cells by > 50% when exposed to 150 …
