Shigella are bacteria that are responsible for millions of infections and hundreds of thousands of deaths every year. The emergence of antibiotic resistant Shigella adds to the potentially devastating effect that these bacteria can have on human health. Shigella flexneri utilize specialized molecular machinery called the Type III secretion system to infect humans and cause disease. Research of this machinery promises to provide the knowledge, tools, and direction for the development of new avenues to combat shigellosis. This research focuses on the understanding and engineering of nonribosomal peptide biosynthetic pathways in Streptomyces coelicolor CH999, Escherichia coli BAP1 and Saccharomyces cerevisiae BJ5464-NpgA. The biosynthetic systems of indigoidine from bacteria and beauvericin/bassianolide from fungi were studied in this research. The production of these valuble products was significantly increased by enhancing their synthetic pathway with metabolic engineering approaches.
Indigoidine is a bacterial natural product with antioxidant and antimicrobial activities. Its bright blue color resembles the industrial dye indigo, thus representing a new natural blue dye that may find uses in industry. Indigo is a dark blue crystalline powder and has …

The emergence of the porcine epidemic diarrhea virus (PEDv) in the United States in 2013 resulted in billions of dollars in annual losses for the U.S. swine industry. Infection with PEDv causes severe diarrhea and vomiting in pigs, spreads rapidly through ingestion of infected manure, and produces nearly 100% mortality in pre-weaned piglets. Because swine manure slurry is a valuable crop nutrient source, concerns about virus persistence in stored manure remain a major barrier to proper manure management. There are two main methods that are used to model the spread of an infectious disease: agent-based modelling and equation based modelling. In this paper, we compare the results from an example implementation of each method, and show that although the agent-based model takes longer to setup and run, it provides additional information that is not available when using an equation based model. Specifically, the ability of the agent-based model to capture heterogeneous mixing and agent interactions enables it to give a better overall view of an outbreak. A parent's death it is one of the most stressful and traumatic events in a child's life (Guldin et al., 2015; Worden, 1996, 2008). In particular, when bereavement is linked to a parent's suicide, children face unique challenges and are more vulnerable to potentially negative outcomes (Brent, Melhem, Donohoe, & Walker, 2009; Haine, Ayers, Sandler, & Wolchik, 2008; Pitman, Osborn, King, & Erlangsen, 2014; Young et al., 2012). We model the nonresponse probabilities as logistic functions ofthe outcome variable and other covariates in the survey sampling study withcallback. The identification aspect of this callback model is investigated. Semiparametricmaximum likelihood estimators of the parameters in the responseprobabilities are proposed and studied. As a result, an efficient estimator ofthe mean of the outcome variable is constructed using the estimated responseprobabilities. Expansive soils usually recognized as swell-shrink soils have been a problem for civil infrastructure for a long time. It has been a very common practice to use chemical stabilizers including cement and lime to stabilize expansive soils, especially for lightly loaded structures. However, due to the the detrimental effects of these stabilizers on the environment and several occurrences of premature failures after stabilizing with chemical additives, engineers are in search of sustainable stabilization alternatives. Sources Of Health Financing And Health Outcomes: A Panel Data Analysis, Tomoki Fujii

We study the differential impacts of public and private sources of health spending on health outcomes using a triple difference approach. We find that private health spending has on average a higher health-promoting effect than public health spending. This result is robust with respect to the choice of outcome measure and covariates in the regression and driven primarily by the countries with ineffective governments. Once we restrict our sample to countries with effective governments, private health spending is no better than public health spending for improving the health outcome.

The purpose of this study was to explore attitudes and perception of stakeholders (teachers, principals, nutrition service directors, nutrition service supervisors, school nurses, nutrition specialists and administrators) toward the implementation of the local school wellness policy in public schools within Kent County, Michigan. The subjects of interest were stakeholders from more than 300 schools and 20 public school districts within the Kent Intermediate School District. Invasive species represent major threats to biodiversity, global economies, and human health. Consequently, extensive research has been directed towards improving methods that restrict and contain them. Yet, control measures can also act as agents of selection by significantly impacting the reproductive capacity of invasives (in the context of “eco-evo” dynamics). The end result is that control measures subsequently alter the fitness landscape of an invasive over ecologically-relevant time, and lose their efficacy by so doing. The purpose of this study was to measure hormonal and metabolic markers in humans to see if simulating dehydration would emulate measures that are seen in prediabetic humans. 60 volunteers were divided into equal categories by sex and body mass index of normal or overweight. Participants completed two oral glucose tolerance tests (OGTT) after being intravenously infused with a isotonic or hypertonic saline solution for 120 minutes in a counter balanced order. Samples collected at five different large bird poultry processing facilities over a period of 7 months from prescald to post debone locations were enumerated for Enterobacteriaceae, Salmonella spp., and Campylobacter spp. and the results were used to create Quantitative Microbial Risk Analyses (QMRA) models for parts, ground, and mechanically separated chicken (MSC) products. Sensitivity analyses indicated the points in the process at which reductions would be most advantageous to the endpoint and simulation models were run to test reductions required to meet the current USDA performance standards.

Campylobacter jejuni, a leading cause of bacterial gastroenteritis worldwide, is strongly associated with the consumption and/or mishandling of raw contaminated poultry products. Thus, interventions aiming to reduce C. jejuni counts on poultry products could greatly reduce the incidence of human campylobacteriosis. In the first study, the efficacy of a generally recongnized as safe (GRAS) compound, carvacrol (CR; derived from oregano oil), as an antimicrobial wash treatment to reduce C. jejuni on chicken skin was evaluated. The false alarm of a Hawaiian nuclear attack in January 2018 is an example of the lack of U.S. preparedness for attacks using nuclear and other weapons of mass destruction. To address such threats, this Article proposes the establishment of a nationwide integrated defense of health countermeasures initiative (DHCI). DHCI is a multi-prong program to create a defensive triad comprising government, private industry, and academia to develop countermeasures for health threats posed by chemical, biological, radiological, and nuclear (CBRN) attacks. Study Design: Systematic literature review.
Objectives: The aims of this study were to (1) describe the clinical features, disabilities, and incidence of neurologic deficits of pyogenic spondylodiscitis prior to treatment and (2) compare the functional outcomes between patients who underwent medical treatment alone or in combination with surgery for pyogenic spondylodiscitis.

Social, health, and environmental policies are critical tools for providing the conditions needed for healthy populations. However, current policy analyses fall short of capturing their full potential impacts across the life course and from generation to generation. We argue that the field of Developmental Origins of Health and Disease (DOHaD), a conceptual and research framework positing that early life experiences significantly affect health trajectories across the lifespan and into future generations, provides an important lens through which to analyze social policies. After the emergence of H5N1 avian influenza in 1997 Hong Kong implemented a sophisticated system to regulate live poultry markets. While this system is well implemented and thorough, it also has limitations. The rise of H7N9 avian influenza (which is typically acquired through contact with poultry, including in live markets) makes this an appropriate time to revisit the ethical and practical issues related to this trade. Workers on prison farms in the United States commonly grow produce and row crops and raise livestock. Teaching good agricultural practices to prisoners and prison staff increases the safety of the food produced and reduces the likelihood of an illness outbreak. We developed a food safety curriculum for use on prison farms in Iowa. In developing the curriculum, we encountered many challenges that resulted in modifications in the content and delivery method. We present the content of the curriculum and provide educators with a better understanding of challenges and potential solutions related to working with prison farms.

Extension educators have been enlisted to assist farmers in meeting requirements of the Food Safety Modernization Act Produce Safety Rule (PSR). Although food safety is a familiar topic for Extension educators, helping farmers learn how to prepare for PSR regulations is new. In this article, we describe a needs assessment conducted in the north central United States according to a modified Delphi approach. Results revealed unique characteristics of farmers in the region, least understood components of the PSR, preferences regarding educational tools, and the need for materials for varied audiences. The human papillomavirus (HPV) is the most common sexually transmitted infection (STI) among sexually active men and women in the U.S. HPV can lead to several types of cancers (e.g., cervical, penile, and anal cancers) that affect the Hispanic community disproportionately. In the U.S., Hispanic women have the highest rate of HPV incidence and the second highest death rate due to HPV-related cervical cancer, and Hispanic men are most affected by HPV-related penile cancers. This study aimed to determine relation between family perceptions towards health role in filariasis countermeasures using the Health Belief Model. An analytical survey was applied with a cross-sectional study approach. The study population was families at three villages that are Lambaro Bileu, Lambaet, and Cot Preh of Kuta Baro Primary Health Care. β-Glucan derived from cell walls of Candida albicans is a potent immune modulator. It has been shown to induce trained immunity in monocytes via epigenetic and metabolic reprogramming and to protect from lethal sepsis if applied prior to infection. Since β-glucan-trained monocytes have not been classified within the system of mononuclear phagocytes we analyzed these cells metabolically, phenotypically and functionally with a focus on monocyte-to-macrophage differentiation and compared them with naïve monocytes and other types of monocyte-derived cells such as classically (M1) or alternatively (M2) activated macrophages and monocyte-derived dendritic cells (moDCs). Deeper exploration of uncharacterized Gcn5-related N-acetyltransferases has the potential to expand our knowledge of the types of molecules that can be acylated by this important superfamily of enzymes and may offer new opportunities for biotechnological applications. While determining native or biologically relevant in vivo functions of uncharacterized proteins is ideal, their alternative or promiscuous in vitro capabilities provide insight into key active site interactions. Additionally, this knowledge can be exploited to selectively modify complex molecules and reduce byproducts when synthetic routes become challenging.
