Molecular Monitoring Of Methicillin-Resistant Staphylococcus Aureus (Mrsa) In A Hospital Setting, Jessica Ammon May 2018

Molecular Monitoring Of Methicillin-Resistant Staphylococcus Aureus (Mrsa) In A Hospital Setting, Jessica Ammon

Honors Theses

Methicillin resistant Staphylococcus aureus (MRSA) is a potentially pathogenic bacterium that poses a serious risk in healthcare settings. MRSA can be characterized by a genetic element, known as the staphylococcal cassette chromosome, which harbors the gene responsible for methicillin resistance, mecA. MRSA can be classified into two categories: community acquired (CA) and hospital acquired (HA). S. aureus strains represent a major health concern due to their prevalence in healthcare facilities and their rapidly evolving antibiotic resistance. The current study investigated the association between MRSA isolates obtained from patients and from the intensive care units in a local hospital. Among the …

Antibiotic Resistant Bacteria In Water Environments In Louisville, Kentucky., Amy Priest May 2018

Antibiotic Resistant Bacteria In Water Environments In Louisville, Kentucky., Amy Priest

College of Arts & Sciences Senior Theses

Antibiotic resistant bacterial strains are an increasing problem, particularly in clinical health care settings. As a result, bacterial infections are becoming increasingly challenging to treat with more cases becoming life threatening. Aquatic environments facilitate microbial diversity and the transfer of genetic elements and thus may serve as a reservoir for antibiotic resistant microbes. Human misuse of antibiotics may further facilitate the spread of resistance in water environments. With little known about the bacteria communities in local water environments, this study aimed to learn more about these populations through the following aims: 1) identify the microbial community composition from water environments …

Most Effective Interventions For Disaster Relief Preparation, Elizabeth Keating, Faith Edwards, Cheryl Swanson Phd, Rn May 2018

Most Effective Interventions For Disaster Relief Preparation, Elizabeth Keating, Faith Edwards, Cheryl Swanson Phd, Rn

Anna Vaughn School of Nursing Undergraduate Work

Because disasters are sometimes unpredictable and difficult to measure, it is exceedingly important to have properly trained healthcare professionals that are prepared for a variety of natural and manmade crises. Victims affected by disasters may be numerous and have severe injuries, but nurses, as first-responders, with proper education and experience can drastically reduce the amount of casualties in a particular disaster. However, it is clear that nurses currently feel unprepared in the event of a natural or manmade disaster, and therefore need more training and preparation for such occurrences. This systematic research review set out to find the most effective …

An Analysis Of Chinese Trade And Fta Using Gravity Model, Minyu Liu May 2018

An Analysis Of Chinese Trade And Fta Using Gravity Model, Minyu Liu

Applied Economics Theses

This paper assesses the impacts of the Free Trade Agreement on the volume of both the import and export of China and FTA countries or districts. We used a gravity model with dummy variables for China and each Chinese FTA partner country to account for the specificities of particular trade relations. By using the relevant theories and methods for the trade gravity model, we estimate a pooled cross-section model with data for 33 countries or districts over 15 years. The result indicates that the GDP, per capita GDP, distance, FTA, and language are the main factors that influence the volume …

The Prevention And Management Of Hospital Acquired Infections, Sofia Rahmanzai May 2018

The Prevention And Management Of Hospital Acquired Infections, Sofia Rahmanzai

Theses/Capstones/Creative Projects

Hospital acquired infections, or HAIs, appear in patients during treatment and are unrelated to the original illness. These infections occur at least 48 hours after hospital admission or 30 days after discharge. With over 700,000 individuals directly affected by HAIs, more attention needs to go into prevention and surveillance. It has been reported that some health care facilities have reduced HAIs by up to 70%. However, this will take a conscious effort from all healthcare facilities, care teams, physicians and nurses. Some possible solutions include infection prevention, increasing surveillance, and improving staff education and accountability. Despite these efforts, no institution …

The Subcloning And Expression Of Munc18a In Escherichia Coli For Antibody Production And Analysis In Mast Cell Degranulation Reactions, Brandi Goble May 2018

The Subcloning And Expression Of Munc18a In Escherichia Coli For Antibody Production And Analysis In Mast Cell Degranulation Reactions, Brandi Goble

Honors Theses

Mast cells are secretory cells responsible for fighting off infection through the early recognition of pathogens. This process is completed through the secretion of proinflammatory mediators that are stored in secretory granules within the cytoplasm of the cell. The degranulation secretion process relies on regulated fusion of secretory granules to the cell membrane via membrane-bound SNARE proteins that bridges the two opposed membranes. The intricate regulation of SNARE-mediated mast cell degranulation is not well understood. However, Sec1/Munc18 (SM) proteins, specifically the Munc18 isoforms, are known to play a critical role in the process (Brochetta, et. al., 2014). The Xu lab …

Defending Wild Dogs: Population Dynamics And Disease In Endangered African Wild Dogs, Elizabeth Claire Arredondo

Graduate Theses and Dissertations

African wild dogs (Lycaon pictus) are endangered carnivores whose population is decreasing from habitat loss and fragmentation, interspecific competition, and disease. Survival rates are especially low in Kruger National Park (KNP), though it is unclear why. I estimated the abundance in KNP and survival rates over different time spans, six years and nine months, using public photographic survey data. In 2015, there were 298 (SE=12.1) individuals. Using a mark-recapture analysis in program R, I found that the survival rate between 2009-2015 was only 3.2%, and within the 9-month survey period (September 2014 – June 2015), monthly survival rates for the …

Spatial Ecology Of The Western Massasauga (Sisturus Tergeminus) In A Large Interior Wetland, Joshua Mead

Master's Theses or Doctor of Nursing Practice

Insight into the spatial ecology of a population of animals provides information valuable to the management and conservation of a species. Reptiles are facing global declines, with 1 in 5 species currently threatened with extinction. For cryptic taxa such as snakes, radio-telemetry allows for individuals to be reliably located on a consistent basis. I used radio-telemetry to investigate the spatial ecology of the Western Massasauga (Sistrurus tergeminus) at Cheyenne Bottoms in Barton County, Kansas. Eighteen individuals (12 male and 6 female) were implanted with very high frequency (VHF) radio transmitters during 2016-2017 and tracked twice weekly from July to November …

T Socio-Ecology Of Managed Honeybees (Apis Mellifera) In The Louisville Metro Area., Haileigh M. Arnold

Electronic Theses and Dissertations

Humans have a long history of the practice of beekeeping to harness the power of pollination. This managed pollinator system consists of beekeepers, honeybees, and their environment. However, recent disease, pesticide use, and land use factors honeybee threaten this relationship. In the face of such concerns it is important to examine the factors that impact and can help sustain our managed pollinator systems. In this thesis, the national and Kentucky state-level policies that affect managed pollinator systems were examined and socio-ecological factors that may contribute to honeybee hive growth and losses were assessed along an urban development gradient in Louisville, …

Malaria In The Prehistoric Caribbean : The Hunt For Hemozoin., Mallory D. Cox May 2018

Malaria In The Prehistoric Caribbean : The Hunt For Hemozoin., Mallory D. Cox

Electronic Theses and Dissertations

With the increase in resistance to anti-malarials and global warming trends expanding the habitation range of the mosquito vector, research highlighting the biogeographical contexts of infected populations is critical to understanding epidemiological patterns. A bioarchaeological approach to epidemiology can shed light on previous disease patterns and aid in the prediction of future outbreaks of diseases like malaria. Currently, there is no direct evidence of malaria in the Americas prior to European contact; however, skeletal, archaeological, paleoenvironmental, historic, and ethnohistorical evidence strongly suggest the presence of Plasmodium spp. malaria in indigenous Caribbean skeletal remains held in the Yale Peabody Museum of …

Development Of Blended Polymeric Fibers To Alter The Release Of Peptide Targeting Porphyromonas Gingivalis., Sonali Suresh Sapare

Electronic Theses and Dissertations

Periodontitis is a chronic inflammatory disease that infects the tissues of the periodontium. It is estimated that 47.2% or 67.2 million American adults suffer from mild, moderate and severe periodontitis. Globally, 30-50% of the adult population is afflicted with periodontal disease, making it one of the most prevalent infectious diseases in the world. Therapeutics targeting P. gingivalis may be effective to alter periodontitis progression. However, the current treatment modalities that target critical pathogens to maintain host-biofilm homeostasis are limited, urging the development of specifically targeted therapeutics to limit P. gingivalis recolonization of the oral cavity after periodontal treatment and healing. …

Electronic Theses and Dissertations

Legionella pneumophila is an aquatic organism capable of intracellular replication within a wide range of protozoan hosts and within human macrophages where it causes Legionnaires’ Disease. L. pneumophila manipulates a variety of host cell processes by translocating, into the host cell cytosol, more than 300 effector proteins via the type IVB secretion system (T4SS). The AnkB effector from the AA100/130b strain (AnkB-AA100) contains a C-terminal CaaX motif that rapidly becomes farnesylated upon translocation into the host cell and anchored to the cytosolic face of the Legionella-containing vacuole (LCV) membrane, which is essential for intracellular replication. We show that a …

Reforming Regenerative Medicine Regulation, Sarah Duranske May 2018

Reforming Regenerative Medicine Regulation, Sarah Duranske

Georgia State University Law Review

Regenerative medicine is defined as the branch of medicine that develops methods to regrow, repair, or replace damaged or diseased cells or tissues. It includes a variety of approaches, such as transplanting cells to promote healing, editing genes in cells to attack cancer, and even building organs from biological materials. Regulating regenerative medicine therapies is no easy task. Finding a balance between competing interests–enabling timely access for needy patients while simultaneously ensuring a positive benefit/risk profile and promoting the development of beneficial innovations–is hard enough at any given point in time. But add in constantly advancing scientific knowledge and increasing …

Enhanced Production Of Pro-Il-1Βeta Contributes To Immunopathology During The Coinfection Of Influenza A Virus And Streptococcus Pneumoniae, Angeline E. Rodriguez May 2018

Enhanced Production Of Pro-Il-1Βeta Contributes To Immunopathology During The Coinfection Of Influenza A Virus And Streptococcus Pneumoniae, Angeline E. Rodriguez

Graduate Theses/Dissertations

Viral bacterial coinfections are known to cause severe pneumonia, especially in the elderly and in pediatric patients. Antibiotics like β-Lactams kill the bacteria but fail to improve symptoms suggesting a faulty immune system may play an important role in the disease. Interleukin-1β (IL-1β) is an important immune signaling cytokine responsible for inflammation. It exists as an inactive precursor that can be activated by caspase-1 containing inflammasomes (multi-protein complex). Influenza A virus (IAV) and Streptococcus pneumoniae (S. pneumoniae) activate the inflammasome through the NOD-like receptor protein NLRP3 and/or AIM2. Previous reports in mice indicate that IL-1β levels are dramatically …

The Role Of Freshwater Drum As A Host Of Freshwater Mussels, Unionidae, Michael S. Martin May 2018

The Role Of Freshwater Drum As A Host Of Freshwater Mussels, Unionidae, Michael S. Martin

Graduate Theses/Dissertations

The Freshwater Drum, Aplodinotus grunniens, serves as fish host to multiple mussel species. Some mussel species are “drum specialists” in the sense that Freshwater Drum is apparently the only host that they utilize. Freshwater Drum diet consists partly of mussels, which it crushes with specially adapted pharyngeal teeth. The Freshwater Drum’s habit of feeding on mussels should favor adaptations to use the species as a host. Mussels that use Freshwater Drum as host could adapt to attract the fish. I predicted that many other species will have some ability to use Freshwater Drum as host because of its habit of …

Investigation Of Bradyzoite Differentiation Initiation In Toxoplasma Gondii, Harim I. Won, Paul H. Davis Ph.D. May 2018

Investigation Of Bradyzoite Differentiation Initiation In Toxoplasma Gondii, Harim I. Won, Paul H. Davis Ph.D.

Theses/Capstones/Creative Projects

Like other eukaryotic organisms, Toxoplasma gondii promoters feature both constitutive and life-stage regulated cis-elements. Using a transcriptomic microarray approach, a cluster of transcripts upregulated early during bradyzoite differentiation was identified. Computational analysis of the promoter regions of these “up-early” transcripts identified a shared upstream consensus motif, a putative transcription factor binding site. Using a dual luciferase assay adapted for recombinational cloning and reporter gene quantification by qPCR, we demonstrate developmental stage-specific expression of the luciferase reporter gene inserted downstream of the transcription factor binding site. The shared consensus motif was found to be an autonomous cis-element by conversion …

Investigation Of Catalytic Activity Of Stable Β-Ketoacyl-Acp Substrate Analogs In Quorum Sensing Signal Synthesis, Nhu Lam May 2018

Investigation Of Catalytic Activity Of Stable Β-Ketoacyl-Acp Substrate Analogs In Quorum Sensing Signal Synthesis, Nhu Lam

Boise State University Theses and Dissertations

Quorum sensing is an intercellular mechanism used by many bacterial pathogens to regulate behaviors, such as motility, virulence, and antibiotic production. Disruption of quorum sensing is shown to be deleterious to bacterial pathogenicity, without causing lethality. In Gram-negative bacteria, acyl-homoserine lactone (AHL or acyl-HSL) signal molecules are synthesized by AHL synthase enzymes using acylated-acyl carrier protein (acyl-ACP) and S-adenosyl-L-methionine (SAM) as their substrates. Pantoea stewartii EsaI, which causes Stewart’s wilt in corn, and Yersinia pestis YspI, which causes bubonic plague are AHL synthases that use 3-oxohexanoyl-ACP and 3-oxooctanoyl-ACP as the acyl-substrate to synthesize 3-oxohexanoyl-HSL and 3-oxooctanoyl-HSL signals, respectively. Unfortunately, the …

Can Alcaligenes Be A New Treatment For Staphylococcus Species?, Devin Narwani

Undergraduate Honors Theses

Members of the Staphylococcus genus are known to cause serious human infections in the clinical environment. Many species of this genus are typically found among the normal flora of the skin, nasal cavities, and the linings of the throat; however, the bacterium becomes an opportunistic infection when there is a break in the physical barriers or when a contaminated medical device is implanted in the human body. As more resistant strains are beginning to appear it is becoming drastically harder to treat patients infected with this bacterium. One such strain is Methicillin Resistant Staphylococcus aureus (MRSA). MRSA is a major …

The opiate epidemic has had an impact on the lives of millions of people in this county. As we explore ways to address this issue, it is important to consider the long-term consequences that the epidemic will have. One long-term consequence is the spread of the hepatitis C virus (or HCV). Approximately 3.2 million people in the United States are chronically infected with this virus. Without consideration about ways to treat this virus and prevent its transmission, there will be large numbers of people who will die from complications of HCV in the future. These losses will contribute to the …

University Faculty Publications and Creative Works

Thirteen percent of pregnancies result in preterm birth or stillbirth, accounting for fifteen million preterm births and three and a half million deaths annually. A significant cause of these adverse pregnancy outcomes is in utero infection by vaginal microorganisms. To establish an in utero infection, vaginal microbes enter the uterus by ascending infection; however, the mechanisms by which this occurs are unknown. Using both in vitro and murine models of vaginal colonization and ascending infection, we demonstrate how a vaginal microbe, group B streptococcus (GBS), which is frequently associated with adverse pregnancy outcomes, uses vaginal exfoliation for ascending infection. GBS …
