Kinetics Of Dextromethorphan-O-Demethylase Activity And Distribution Of Cyp2d In Four Commonly-Used Subcellular Fractions Of Rat Brain, Barent N. Dubois, Farideh Amirrad, Reza Mehvar

Pharmacy Faculty Articles and Research

The purpose of this study was to compare the enzymatic kinetics and distribution of cytochrome P450 2D (CYP2D) among different rat brain subcellular fractions.

Rat brains were used to prepare total membrane, crude mitochondrial, purified mitochondrial, and microsomal fractions, in addition to total homogenate. Michaelis–Menten kinetics of the brain CYP2D activity was estimated based on the conversion of dextromethorphan (DXM) to dextrorphan using UPLC-MS/MS. Protein levels of CYP2D and subcellular markers were determined by Western blot.

Microsomal CYP2D exhibited high affinity and low capacity, compared with the mitochondrial CYP2D that had a much lower (∼50-fold) affinity but a higher (∼six-fold) …

Selenate Prevents Adipogenesis Through Induction Of Selenoprotein S And Attenuation Of Endoplasmic Reticulum Stress, Choon Young Kim, Kee Hong Kim

Department of Food Science Faculty Publications

The conversion of preadipocytes to adipocytes (adipogenesis) is a potential target to treat or prevent obesity. Selenate, an inorganic form of selenium, elicits diverse health benefits, mainly through its incorporation into selenoproteins. The individual roles of selenium and certain selenoproteins have been reported. However, the effects of selenate treatment on selenoproteins in adipocytes are unclear. In this study, the effects of selenate pretreatment on selenoprotein and endoplasmic reticulum (ER) stress during adipogenesis were examined in vitro. The selenate pretreatment dose-dependently suppressed the adipogenesis of 3T3-L1 preadipocytes. The selenate pretreatment at 50 µM for 24 h almost completely suppressed adipogenesis without …

Prostate cancer is a leading cause of cancer death in men over 50 years of age, and there is a characteristic marked decrease in Zn content in the malignant prostate cells. The cause and consequences of this loss have thus far been unknown. We found that in middle-aged rats a Zn-deficient diet reduces prostatic Zn levels (P = 0.025), increases cellular proliferation, and induces an inflammatory phenotype with COX-2 overexpression. This hyperplastic/inflammatory prostate has a human prostate cancer-like microRNA profile, with up-regulation of the Zn-homeostasis-regulating miR-183-96-182 cluster (fold change = 1.41-2.38; P = 0.029-0.0003) and down-regulation of the Zn …

Action potentials (APs) are the functional units of fast electrical signaling in excitable cells. The upstroke and downstroke of an AP is generated by the competing and asynchronous action of Na+- and K+-selective voltage-gated conductances. Although a mixture of voltage-gated channels has been long recognized to contribute to the generation and temporal characteristics of the AP, understanding how each of these proteins function and are regulated during electrical signaling remains the subject of intense research. AP properties vary among different cellular types because of the expression diversity, subcellular location, and modulation of ion channels. These complexities, in addition to the …

The Chemotactic Role Of Metallothionein In The Extracellular Environment, Sadikshya Bhandari Nov 2018

The Chemotactic Role Of Metallothionein In The Extracellular Environment, Sadikshya Bhandari

Doctoral Dissertations

Metallothionein (MT) is a cysteine rich stress response protein that lacks a signal peptide and thus has historically been considered an intracellular protein. However, the presence of MT in a variety of extracellular fluids and spaces suggests an additional extracellular role for this protein. We have shown that MT released in the extracellular environment can bind to the surface of lymphocytes and is a chemoattractant for Jurkat T cells among other cells. Additionally, when Jurkat T cells are pre-incubated with MT, there is a decline in the subsequent chemotactic response to SDF-1 α. Jurkat T cells pre-incubation with SDF-1 α …

Climate changes are predicted to drive changes in plant species composition and vegetation cover around the world. Preserved specimens and other botanical information that we gather today—a period future practitioners may look back on as an early stage of modern anthropogenic climate change—will be of value to conservation managers and conservation biologists in the decades and centuries ahead. Here, we present suggestions for the systematic collection, long-term curation (in museums, herbaria, and other research institutions), and maintenance of plant specimens, along with associated data and analyses on the plants and vegetation present today and in the past. The primary aim …

Dairy Calves' Personality Traits Predict Social Proximity And Response To An Emotional Challenge, B Lecorps, S Kappel, Dm Weary, Mag Von Keyserlingk

School of Biological and Marine Sciences

The assessment of individual traits requires that tests are reliable (i.e. consistency over time) and externally valid, meaning that they predict future responses in similar contexts (i.e. convergent validity) but do not predict responses to unrelated situations (i.e. discriminant validity). The aim of this study was to determine if dairy calf personality traits (Fearfulness, Sociability and Pessimism), derived from behaviours expressed in standardized tests, predict individuals’ responses in related situations. The first experiment tested if the trait ‘Sociability’ was related to the expression of social behaviour in the home-pen, with calves assigned individual proximity scores (based on proximity to other …

Microbial Diversity In Urban Environments: Concern For Antibiotic Resistance, Ality Aghedo, Mangala Tawde, Nazrul I. Khandaker

Publications and Research

We are almost oblivious to the presence of microorganisms in our daily lives, but they exist and come into contact with us all the time. While some bacteria are harmless, and even beneficial, other bacteria can cause infections. A common treatment to bacterial infections is antibiotics and the success of an antibiotic depends on the resistance of the bacteria to the antibiotic. We conducted experiments to identify the types of bacteria that can be found on surfaces or within soil environments that come into contact with a vast number of people on a daily basis and how this can affect …

Reporting From The City: Vignettes From Constantinople In The Dispatches Of Lord Strangford During The Eastern Crisis Of The 1820s, Theophilus C. Prousis

History Faculty Research and Scholarship

The tangled web of the Eastern Question became the single most explosive force in European great power politics during the nineteenth and early twentieth centuries, and Constantinople became the epicenter of this contentious dispute in Ottoman-European relations. Eyewitness commentaries by diplomats, travelers, residents, and others who visited this fabled city conveyed images and episodes about various topics, including European interactions with the Ottoman Empire, European designs on contested lands, and Ottoman politics and policy. These scenes and stories not only shed light on the geopolitical heart of the Eastern Question but also reinforce the centrality of this volatile issue in …

Mathematical models are key tools for the development of surveillance, preparedness and response plans for the potential events of emerging and introduced foreign animal diseases. Creating these types of plans requires data; when data are incomplete, mathematical models can help fill in missing information, provided they are informed by the data that are available. In the United States, the most complete national-scale data available on cattle shipments are based on Interstate Certificates of Veterinary Inspection, which track the shipment of cattle between states; data on intrastate cattle shipments are lacking. Here we develop four new datasets on intrastate cattle shipments …

Mapping Colonial Interdependencies In Dutch Brazil: European Linen & Brasilianen Identity, Carrie Anderson Nov 2018

Mapping Colonial Interdependencies In Dutch Brazil: European Linen & Brasilianen Identity, Carrie Anderson

Artl@s Bulletin

In Dutch Brazil, the Brasilianen were essential allies to the West India Company. To maintain this critical alliance, the Dutch presented them with gifts of linen, a fabric in high demand. Representations of Brasilianen wearing linen garments were pervasive and include an image on Joan Blaeu’s 1647 map of the Brazilian Captaincies of Rio Grande and Paraíba. Traditional interpretations of these linen-clad Brasilianen prioritize a center/periphery model; in contrast, I argue that these pictured linens document the interdependencies between the WIC and the Brasilianen, a position supported by digital maps plotting Dutch/indigenous exchanges.

Victor’s Dual Diagnosis: An Exploration of Mental Illness in Frankensteinian Times

Before the advances of modern psychology, treatment of the mentally insane consisted of cruel and torturous methods that involved beating, starving, or bleeding patients often until the point of death. It was not until the late eighteenth century that a revolutionary kind of moral treatment was introduced by William Tuke, an English Quaker and founder of The Friends’ Retreat. Founded in 1879, the small retreat in York set the precedent for future asylums with their meticulous record keeping that included their own standardized diagnoses and symptoms of mental illnesses. …

The Manuscript Map Of The Dagua River. A Rare Look At A Remote Region In The Spanish Colonial Americas, Juliet Wiersema Nov 2018

The Manuscript Map Of The Dagua River. A Rare Look At A Remote Region In The Spanish Colonial Americas, Juliet Wiersema

Artl@s Bulletin

The Manuscript Map of the Dagua River Region (1764) is a hand-drawn map produced in the Spanish Viceroyalty of Nueva Granada. While created as visual testimony for a land dispute, I argue that a careful art historical reading of the Dagua River Map, considered in conjunction with eighteenth-century archival documents, nineteenth-century explorers’ accounts, and surviving historical maps, reveals other narratives about ethnicity, industry, and society in a remote region of a peripheral Spanish viceroyalty. The Dagua River map highlights the incontrovertible place that geography held for those—namely enslaved and freed Africans—who came to control trade and transport in the region, …

Cartographic Beasts At The Tail-End Of The Long Renaissance: Style And Sources For The Tabula Geographica Regni Chile (1646), Catherine Burdick Nov 2018

Cartographic Beasts At The Tail-End Of The Long Renaissance: Style And Sources For The Tabula Geographica Regni Chile (1646), Catherine Burdick

Artl@s Bulletin

In mid-seventeenth century Europe, as “plain” Enlightenment mapping conventions displaced Renaissance pictorialism, cartographic beasts passed into disuse. In this paper I analyze a final retreat for mapped fauna, Alonso de Ovalle’s Tabula geographica regni Chile (Rome, 1646), as a tail-end of the pictorial style which characterized the cartography of the long Renaissance. Yet we shall see that this map’s outmoded pictorialism was appropriate to its function as an early conveyance of Chile in Spain, and that Ovalle’s South American critters, adapted from exploration documents, evoked the authority of great voyages before Philip IV.

This dissertation assesses Kenya’s capacity to effectively prepare and respond to oil spill incidents and compares it to recommended international practice. The study is based on analysis of available data and questionnaires administered to personnel involved in oil spill preparedness and response in Kenya as well as International Organizations involved in oil spill response and training. Globally sea borne trade has been increasing and this has reflected in Kenya where the country has embarked on expanding its shipping industry. This exposes the country to the risk of oil spill and further compounded by busy tanker route through the territorial sea …

Background We sought to determine if the prevalence of antibiotic-resistant Escherichia coli differed across retail poultry products and among major production categories, including organic, “raised without antibiotics”, and conventional. Results We collected all available brands of retail chicken and turkey—including conventional, “raised without antibiotic”, and organic products—every two weeks from January to December 2012. In total, E. coli was recovered from 91% of 546 turkey products tested and 88% of 1367 chicken products tested. The proportion of samples contaminated with E. coli was similar across all three production categories. Resistance prevalence varied by meat type and was highest among E. …
