Microvesicle particles (MVPs) are produced from cellular membranes and are thought to mediate cell-cell communication, including in response to stressors such as UVB radiation and thermal burn injury. Previous studies have shown that stress-induced MVP release requires the platelet-activating factor (PAF) receptor in human keratinocytes and that pharmacological inhibition of acid sphingomyelinase (ASM) blocked this release. To validate a genetic role for ASM in MVP release, we used CRISPR-Cas9 gene silencing in human keratinocytes and primary fibroblasts derived from ASM-knockout mice. Modulation of macrophage polarization is required for effective tissue repair and regenerative therapies. Conversion of macrophages from inflammatory M1 to fibrotic M2 phenotype could help in diseases such as chronic wound which are stuck in inflammatory state. During the inflammatory phase, macrophages are of the inflammatory phenotype (M1) and distribute pro-inflammatory cytokines including TNF-a and IL1[beta] which are microbicidal and recruit/activate cells. In normal wound healing macrophages then switch to a fibrotic phenotype (M2) promoting wound closure by angiogenesis, and matrix deposition. The use of the standard two dimensional (2D) cell culture has laid down the fundamentals of molecular and cell biology. However, recent advances in cell-based regenerative medicine raises the concern on deconstructing cellular behaviors in more physiologically relevant three dimensional (3D) microenvironments. Differences in cell response in 2D versus 3D systems arise from the perturbations in gene expression patterns that stem from how cells sense their underlying 2D or surrounding 3D matrices and adjust their phenotypes accordingly. The invasion of novel habitats by non-native plant species is a worldwide problem with serious economical and ecological implications. Broad biotic and abiotic filters contribute to the overall invasibility of non-native species. Invasive plants have the ability to rapidly establish themselves and outcompete their native counterparts as result of their relationship with herbivorous arthropods. Because invasive plants displace native plants, reduce overall plant diversity, and alter vegetation structure, this can have cascading effects on insect herbivores, particularly those that rely on one or a few food plants, and arthropod predators. Mesenchymal stem cells (MSCs) are adult multipotent stem cell that can differentiate into mesodermal lineages such osteoblast, adipocytes, and chondrocytes, or can be transdifferentiated into clinically relevant lineages such as cardiac or neural cells using in vitro reprogramming techniques. Chemical/biological sensors serve many purposes in protecting machinery, the environment, and human life/wellness. Graphene, a two-dimensional (2-D) material made up of carbon atoms in a honeycomb-like lattice, is promising for applications to chemical/biological sensing due to its unique properties. Functionalization of graphene by surface decorating with nanoparticles and increasing interior adsorption sites can tailor its catalytic activity and electrical properties, and hence, important for detecting and distinguishing trace hazardous gases. Dementia caregiver burnout is detrimental to both the familial caregiver and their loved ones with dementia. As the population of older adults increases, both the number of individuals with dementia and their corresponding caregivers increase as well. Thus, we are interested in developing a potential tool to non-invasively detect signs of caregiver burnout using a mobile application combined with machine learning. Huntington’s disease (HD) is a degenerative disorder caused by expanded CAG repeats in the Huntingtin gene, which results in cognitive problems as well as muscle weakness, chorea, rigidity, and dystonia. Most research in HD has focused on neurodegeneration, but recent studies have found peripheral defects that may help explain the debilitating motor symptoms of HD. We have shown that skeletal muscle from the R6/2 transgenic mouse model of HD is hyperexcitable due to decreases in resting chloride and potassium currents. Gametes of gray treefrogs, Dryophytes chrysoscelis, are deposited into freshwater ponds. Sperm undergo spermatogenesis and maturation beginning in the seminiferous tubules and migrating to the lumen. In mammals and fishes, these cells are immotile within the isosmotic fluid of the testes and have motility activated by exposure to a hyper- or hypoosmotic medium. Water flows into or out of the sperm cell, altering intracellular ionic concentrations, and ultimately stimulates flagellar movement. Anthropogenic alterations to nitrogen, carbon, and phosphorus bioavailability have increased the flux of these resources into the biosphere and altered stream ecosystem function. Streams modify the transport of these resources to receiving ecosystems through uptake, transformation, and mineralization. Understanding how streams process carbon, nitrogen, and phosphorus can provide insight about how stream ecosystems function in landscapes where human modification is inescapable. I investigated how land use in agricultural regions affect resource availability to primary producers and consumers and the subsequent impact on stream processes. Very Long Engagements: The Persistent Authority Of Bridewealth In A Post-Apartheid South African Community, Michael W. Yarbrough

This article examines the persistent authority of the customary practice for forming recognized marriages in many South African communities, centered on bridewealth and called “lobola.” Marriage rates have sharply fallen in South Africa, and many South Africans blame this on the difficulty of completing lobola amid intense economic strife. Using in-depth qualitative research from a village in KwaZulu-Natal, where lobola demands are the country’s highest and marriage rates its lowest, I argue that lobola’s authority survives because lay actors, and especially women, have innovated new repertoires of lobola behavior that allow them to pursue emerging needs and desires for marriage …

Deregulation of genome duplication is a common theme in cancer cells. Although multitude of pathways has been discovered and implicated in the ability of oncogenes or loss of function tumor suppressors to induce oncogenesis, most of the pathways ultimately converge in deregulated genome duplication or uncoupling of DNA replication and segregation in cancer cells. It is important to determine how known oncogenes deregulate genome duplication, and if prevention of deregulated genome duplication could be a feasible target to thwart uncontrolled proliferation of cancer cells. This study investigated wild caught striped mullet (Mugil cephalus) at Merritt Island National Wildlife Refuge (MINWR) for levels of 15 perfluoroalkyl acids (PFAA) in tandem with individual fecundity measurements (Oocyte sub-stage 2 late, n=42) and oocyte reproductive stages (Stages 1–5, n=128). PFAAmeasurementswere quantified in stripedmullet liver (n=128),muscle (n=49), and gonad (n=10). No significant negative impacts of liver PFAA burden on wild-caught,mullet fecundity endpoints were observed in this study; however, changes in PFAAwere observed in the liver asmullet progressed through different sub-stages of oocyte development. This Article explores the intersection of race and disability in the context of employment discrimination, arguing that people of color with disabilities can and should obtain more robust relief for their harms by asserting intersectional discrimination claims. Professor Kimberlé Crenshaw first articulated the intersectionality framework by explaining that Black women can experience a form of discrimination distinct from that experienced by White women or Black men, that is, they may face discrimination as Black women due to the intersection of their race and gender. Objective: Fecal contamination of potable water leads to unsafe water supply. Although many urban areas of large metropolitan cities receive safe water, peri-urban areas are often not monitored by public health authorities and water supply and quality remain unknown. We assessed microbiological quality and rates of antimicrobial resistance in viable indicator bacteria in two peri-urban communities of Karachi, Pakistan. Many jobs in today’s society require sitting at a desk with little physical activity. Individuals who engage in ten hours of sedentary behavior per day double their CVD risk. Standing desks are thought to decrease sedentary time and improve cardiovascular health. Acute use of standing desks is shown to lower PWV. However, chronic effects remain unknown. In Sub-Saharan African countries, soil fertility is low due to unsustainable agricultural practices and little to no access to chemical fertilizer. Poor yields have resulted in many of these countries struggling to produce enough food to feed their growing populations. Human urine has been used as an effective, low-cost alternative to chemical fertilizers in greenhouse and plot trials on a variety of vegetables, but its effects on elemental grain composition in cereal crops are largely unknown. As digital technologies have expanded, so have the literacy sponsors that support and shape how those technologies are used. This project focuses on one of these growing sites of sponsorship surrounding a specific health-tracking technology: wearable Fitbit devices. While much of the work on literacy sponsorship has focused on institutional sponsors as agents, I argue that the picture becomes more complicated and interesting when we place our focus on how users—often considered the sponsored—can become agents in a system that may have marginalized, excluded, or used them.

This research investigates the ecology of: ringed seals (Pusa hispida), moose (Alces alces), and bowhead whales (Balaena mysticetus). Each of these species draws upon specialized strategies to meet the physical demands of life in their extreme environments. Water security has become a large issue with populations rising and weather patterns changing to drier and hotter conditions in southern Africa. Lesotho, a mountainous country landlocked by South Africa, is responsible for a large amount of the rainfall in southern Africa. As such, Lesotho provides water to South Africa in exchange for national economic growth. Lesotho, however, is also prone to droughts and with water reserves being sold to South Africa, water security has become a national issue. Through the continued breeding of the red fox in captivity, there has been an explosion of variations in color, size, and temperament. This thesis investigated the restoration of mauri (life-force) to Ōkahu Bay, Auckland New Zealand. Ōkahu Bay is part of the land and waters of Ngāti Whātua Ōrākei, a Māori hapū (sub-tribe). Ngāti Whātua Ōrākei has been driving the restoration, restoring Ōkahu Bay based on their worldview, visions, and concerns. This vision and control of the restoration process allows them to bring in the hapū in sustainable engagement and have the long-term vision and commitment necessary for self-determination. This thesis involves examining the influence of feminism, platform and community on the fandom of Sarah J Maas (SJM) novels on the social network site Tumblr. The goals are to show how a shift to an open social network site has changed the practice of fandom and how various feminisms are applied to popular culture and learned through Tumblr. An ethnographic study and textual analysis on SJM fandom posts and interactions on Tumblr were utilized in order to reach these goals.
