Effect Of Non-Alcoholic Fatty Liver Disease (Nafld) On Hepatic Drug Metabolism Enzymes In Human, Rohitash Jamwal Jan 2018

Effect Of Non-Alcoholic Fatty Liver Disease (Nafld) On Hepatic Drug Metabolism Enzymes In Human, Rohitash Jamwal

Open Access Dissertations

Significant changes in dietary habits have led to a rampant increase in metabolic disorders. Non-alcoholic fatty liver disease (NAFLD) is one such disorder characterized by the excess buildup of fat in hepatocytes of people who drink little or no alcohol. If not managed, NAFL (simple steatosis) progress into nonalcoholic steatohepatitis (NASH) and further deteriorate to cirrhosis leading to severe illness or even death. Drug disposition proteins (enzymes and transporters) in liver control the systemic exposure of drugs and xenobiotics in human and drive the efficacy as well as adverse events in the body. Therefore, it is critical to address the …

Vascular Cognitive Impairment And Dementia: The Importance Of Mixed Pathologies From Mouse Models To Humans, Alex Marian Helman Jan 2018

Vascular Cognitive Impairment And Dementia: The Importance Of Mixed Pathologies From Mouse Models To Humans, Alex Marian Helman

Theses and Dissertations--Molecular and Cellular Biochemistry

Age-related neurologic disease is a significant and growing burden on our society. Although the largest share of research effort has typically been devoted to the common neurodegenerative illnesses (such as Alzheimer’s disease, or AD), the reality is that nearly all cases of neurodegenerative disease possess elements of mixed pathology. Vascular contributions to cognitive impairment and dementia (VCID) is a complex form of dementia, combining aspects of vascular disease and other forms of dementia, such as Alzheimer’s disease. This pathology is heterogeneous and can include cerebral amyloid angiopathy (CAA), hemorrhages, white matter infarcts, and changes to the neurovascular unit. Given the …

Controlling Platelet Secretion To Modulate Hemostasis And Thrombosis, Smita Joshi Jan 2018

Controlling Platelet Secretion To Modulate Hemostasis And Thrombosis, Smita Joshi

Theses and Dissertations--Molecular and Cellular Biochemistry

Upon vascular injury, activated blood platelets fuse their granules to the plasma membrane and release cargo to regulate the vascular microenvironment, a dynamic process central to platelet function in many critical processes including hemostasis, thrombosis, immunity, wound healing, angiogenesis etc. This granule- plasma membrane fusion is mediated by a family of membrane proteins- Soluble N-ethyl maleimide Attachment Receptor Proteins(SNAREs). SNAREs that reside on vesicle (v-SNAREs) /Vesicle-Associated Membrane Proteins(VAMPs) interact with target/t-SNAREs forming a trans-bilayer complex that facilitates granule fusion. Though many components of exocytic machinery are identified, it is still not clear how it could be manipulated to prevent …

Minority Stress And Mental Health Among Transgender Persons, Elizabeth Alice Sapareto Jan 2018

Minority Stress And Mental Health Among Transgender Persons, Elizabeth Alice Sapareto

Walden Dissertations and Doctoral Studies

Transgender people, a minority population, are at increased risk for negative health and mental health consequences. Profound societal discrimination and stigmatization lead to systemic institutional barriers and lack of access to health care services. Research with lesbian, gay, and bisexual populations shows a strong association between minority stress and mental health; however, there is a gap in research for the transgender population. This study, based on theories of minority stress, positive psychology, the biopsychosocial model, and the transgender model, was conducted to clarify this relationship for the transgender population. Four research questions were proposed. A final sample of N = …

Leadership, Organizational Turnarounds, And Gil Hodges's 1969 New York Mets, John Rebecchi

Walden Dissertations and Doctoral Studies

The impact of the 2008 global financial crisis, shifting market demands, and prolonged underperformance has forced organizations to devise and implement turnaround strategies or risk business failure. Researchers have pointed to the importance of leadership in the turnaround process, yet there are a limited number of research studies identifying characteristics of successful turnaround leaders. Using the full range leadership model, the purpose of this nonexperimental, ex post facto study was to examine the leadership style of Gil Hodges, manager during the 1969 New York Mets successful turnaround season and explore the organization's culture and climate. Data were collected using the …

Marital Status And Burdensomeness As Risk Factors Of Suicide Ideation In Poststroke Patients, Froso Andreou Jan 2018

Marital Status And Burdensomeness As Risk Factors Of Suicide Ideation In Poststroke Patients, Froso Andreou

Walden Dissertations and Doctoral Studies

Suicide ideation, suicide attempts, and suicide (SISAS) are increased in poststroke patients, yet not everyone who has suffered a stroke is at risk for SISAS. Two risk factors for SISAS, marital status and burdensomeness, may be of particular relevance to poststroke patients. The majority of poststroke patients have a disability that may require help from a family member with basic functions such as dressing and bathing. It was not known if being married decreases risk of SISAS for stoke victims as shown in studies with nonpoststroke subjects or increases risk for SISAS due to its influence on feelings of burdensomeness. …

Theses and Dissertations

I wish I could believe in something. Having grown up in a religious household, I have continually teetered between faith and doubt. Landscapes seen and unseen are my last source of awe; here my doubt is suspended – for a moment. Using unfired clay, I create alternate landscapes inspired by sublime philosophy. The sublime experience is born in a sense of amazement linked to fear of something beyond our understanding or control. The amazing intricacy of microbiology, a whole universe existing alongside and inside us, fascinates me. The abundance of unfamiliar life in my work triggers a cautious curiosity. My …

Human onchocerciasis - commonly known as river blindness - is one of the most devastating yet neglected tropical diseases, leaving many millions in sub-Saharan Africa blind and/or with chronic disabilities. Attempts to eliminate onchocerciasis, primarily through the mass drug administration of ivermectin, remains challenging and has been heightened by the recent news that drug-resistant parasites are developing in some populations after years of drug treatment. Needed, and needed now, in the fight to eliminate onchocerciasis are new tools, such as preventive and therapeutic vaccines. This review summarizes the progress made to advance the onchocerciasis vaccine from the research laboratory into …

The Relationships Between Older, Physically Impaired Men And Their Pets., Jennifer Hughes Williams Jan 2018

The Relationships Between Older, Physically Impaired Men And Their Pets., Jennifer Hughes Williams

Walden Dissertations and Doctoral Studies

The present study examined older male adults' lived experiences with pet ownership. There is robust literature on older adults and the human-animal bond, but presently there are no qualitative phenomenological studies that focus solely on older physically impaired males who live alone and the relationships they have with their pets. With the guiding framework of biopsychosocial theory and using intensive interviews, 10 older males (65+) who suffered from some physical impairment discussed the relationships they have with their pets, and how their companion animals affect their physical, emotional, and social health. All interviews were transcribed, coded, and analyzed for themes. …

Impaired Cardiorespiratory Fitness Following Thoracic Radiotherapy, Justin M. Canada Jan 2018

Impaired Cardiorespiratory Fitness Following Thoracic Radiotherapy, Justin M. Canada

Theses and Dissertations

Cancer (CA) is the second leading cause of death in the United States preceded only by cardiovascular disease (CVD). Over the past 30 years, the 5-year survival rate for all cancers combined has increased by more than 20%. This improved survival rate is due to early diagnosis and advances in treatment involving a multimodality treatment approach that includes radiotherapy [RT] with about half of all CA patients receiving some type of RT sometime during the course of their treatment. Cardiotoxicity is one of the most important adverse reactions of RT and leads to a meaningful risk of CVD-related morbidity and …

Utilizing Voltage-Gated Calcium Channels To Assess The Activity Of Cathinone Derivatives At Human Monoamine Transporters, Brian A. Ruiz Jan 2018

Utilizing Voltage-Gated Calcium Channels To Assess The Activity Of Cathinone Derivatives At Human Monoamine Transporters, Brian A. Ruiz

Theses and Dissertations

Cathinones are psychostimulant compounds heavily implicated as drugs of abuse. They exert their physiological actions at the monoamine transporters, which are responsible for maintaining synaptic neurotransmitter homeostasis. Monoamine transporters produce currents during transport and have been shown to depolarize cell membranes and activate voltage-gated calcium channels in mammalian expression systems. This phenomenon is harnessed in an assay which measures these induced calcium transients, allowing for quantification of pharmacodynamic effects of compounds at monoamine transporters. It is unknown if this electrical coupling occurs in neurons, but the implications if it does are significant. In the current work, fluorescent resonance energy transfer …

Impact Of Phosphoinositides On Regulation Of K-Atp By Atp And Hydrogen Sulfide, Tyler Hendon Jan 2018

Impact Of Phosphoinositides On Regulation Of K-Atp By Atp And Hydrogen Sulfide, Tyler Hendon

Theses and Dissertations

Hydrogen sulfide (H2S) reduces ischemia reperfusion (IR) injury by stimulating adenosine triphosphate (ATP) sensitive potassium channels (KATP) [1-5]. Demonstrating H2S stimulation is unique to KATP, as other inwardly rectifying potassium (Kir) channels demonstrate inhibition or are unaffected [6]. We recently showed that H2S inhibits Kir2 and Kir3 by decreasing channel sensitivity to phosphatidylinositol 4,5-bisphosphate (PI(4,5)P2 or PIP2) [6]. Here, we test the hypothesis that H2S regulation of Kir6.2, a pore-forming subunit of the KATP channel, is also dependent on PIP2. Using whole-cell patch-clamp we show that H2S increases the activity of Kir6.2 channels expressed in HEK-293 cells. To study the …

Aerosolized Surfactants: Formulation Development And Evaluation Of Aerosol Drug Delivery To The Lungs Of Infants, Susan Boc Jan 2018

Theses and Dissertations

The overall aim of this research project was to develop surfactant dry powder formulations and devices for efficient delivery of aerosol formulations to infants using the excipient enhanced growth (EEG) approach. Use of novel formulations and inline delivery devices would allow for more efficient treatment of infants suffering from neonatal respiratory distress syndrome and bronchiolitis.

A dry powder aerosol formulation has been developed using the commercial product, Survanta ® (beractant) and EEG technology to produce micrometer-sized hygroscopic particles. Spray drying and formulation parameters were initially determined with dipalmitoylphosphatidylcholine (DPPC, the dominant phospholipid in pulmonary surfactant), which produced primary particles 1 …

Teachers' Lived Experiences Of A Positive Intervention On Emotional Behavior Disorder Students, Leslie Deann Williams-Britt Jan 2018

Teachers' Lived Experiences Of A Positive Intervention On Emotional Behavior Disorder Students, Leslie Deann Williams-Britt

Walden Dissertations and Doctoral Studies

Champions for Change Mentorship Program (CFCMP) was implemented to address the academic and social struggles of emotional behavior disorder (EBD) students. The problem was that although the passing of the California Assembly Bill 1729, called the Other Means of Correction law, had prompted a greater need for positive behavior intervention support at Tier 2 and Tier 3 levels, implementation of the CFCMP continued to generate serious concerns involving students with behavior issues. The conceptual framework for this study is Bandura's social learning theory which guided CFCMP to incorporate cognitive and observational learning to develop academically and socially sound students with …

Effects Of Occupational Stress On Decision-Making Style In Law Enforcement Officers, Christine M. Crippen Jan 2018

Effects Of Occupational Stress On Decision-Making Style In Law Enforcement Officers, Christine M. Crippen

Walden Dissertations and Doctoral Studies

Police officers deal with a variety of stresses from different sources. Organizational stresses have the most effect on police officers, often more than stressful critical incidents. Previous research has indicated that over time, the mundane organizational and operational stresses of the job can result in a variety of effects from psychological to physiological, and this stress can impact police performance and public safety. However, there remains an important gap in the current literature regarding the impact of stress on the decision-making style of police officers. Police officers often make split-second decisions that can affect their life, the public, and other …

Sites contaminated with chlorinated hydrocarbons are frequent and widespread, and with the rising use of insensitive high explosive (IHE) compounds, more widespread contamination is inevitable. In the cases of both classes of organic contaminants, natural attenuation is a critical component of our understanding of the environmental fate of these compounds. This dissertation is intended to expand the knowledge of potential abiotic natural attenuation mechanisms and, in the case of the study of chlorinated hydrocarbons, to examine degradation under variable pH conditions in the hopes of helping to develop minimally invasive remediation techniques. The results indicated that precipitated hydrolyzed Fe(II) species …

Social media data promise to inform the disaster response community, but effective mining remains elusive. To assist in the analysis of community reports on disaster from social media, I draw on an integrated model of psycholinguistic theory to investigate the patterns by which language use changes as a function of environmental influence. Using social media corpora from several disasters and non-disasters, I examine variations in patterns of lexical choice between domain independent paired antonyms with respect to an Internet-specific base rate to determine generic sentinels of breach of canonicity. I examine social media content with respect to disaster proximity and …

Mapping Hybrid Lethal Genes On The X Chromosome Of C. Briggsae, Blaine E. Bittorf Jan 2018

Mapping Hybrid Lethal Genes On The X Chromosome Of C. Briggsae, Blaine E. Bittorf

Browse all Theses and Dissertations

In the cross of C. nigoni males to C. briggsae hermaphrodites, all F1 males arrest during embryogenesis. However in the reciprocal cross there are some viable F1 male progeny. This unidirectional male-specific lethality in the F1 hybrids has been attributed to a hybrid lethal gene in a 500 Kb region of the X chromosome of C. briggsae. Cbr-him-8 is a recessive maternal suppressor of the male-specific lethal phenotype, due to the requirement of the him-8 protein for proper X chromosome pairing. Without proper pairing of any one of the chromosomes in the Caenorhabditis genome, genes present on the unpaired chromosome …

It can be improved by continuously monitoring patients and their environment using the Internet of Things (IoT) based devices. These sensor data streams so obtained are essential to comprehend multiple factors triggering asthma symptoms. In order to support physicians in exploring causal associations and finding actionable insights, a visualization system with a scalable cloud infrastructure that can process multimodal sensor data and Patient Generated Health Data (PGHD) is necessary. In this thesis, we describe a cloud-based asthma management and visualization platform that …

Modeling Of Soil Formation On The Basis Of Chemical Weathering: Applications From Percolation Theory, Fang Yu Jan 2018

Modeling Of Soil Formation On The Basis Of Chemical Weathering: Applications From Percolation Theory, Fang Yu

Browse all Theses and Dissertations

The concept of soil formation has been studied since the beginning of 19th century. However, until recently, there has been little concrete progress on developing an analytical result to relate soil depth or quality to measurable variables that represent the five soil-forming factors including time, parent material, topography, climate, and organisms. It has become increasingly clear that soil formation rates are closely related to chemical weathering rates. In this dissertation, we propose a theoretical approach to model soil formation process within the theoretical framework of percolation theory, which has been shown to successfully predict solute transport in heterogeneous media. From …

Multiple myeloma is a hematological malignancy characterized by clonal proliferation of plasma cells generally caused by chromosomal abnormalities. It occurs in the bone marrow, which is the microenvironment of multiple myeloma. Exosomes (EXs) are 30-100 nm membrane-derived micro-vesicles containing various of bioactive molecules, such as microRNAs, to mediate the cell-cell interaction. Numerous studies reported that exosomes play a significant role in tumor microenvironment. Angiogenesis has the important implication in tumor exacerbation to supply nutrients to promote the progression of cancer cells through endothelial cells (ECs). Some studies demonstrated that microRNA-29b (miR-29b) can suppress tumor development and inhibit angiogenesis. Therefore, in …
