PVSRIPO, a variant of the poliovirus, is a revolutionary virus that demonstrates key characteristics for oncolytic virotherapy due to its ability to attack glioblastomas. The first criteria that PVSRIPO demonstrates is tumor-targeting tropism. PVSRIPO binds to nectin-like molecule 5 (Necl-5), a poliovirus receptor (PVR), found on different types of cancers such as lung cancer, colorectal cancer, and glioblastomas. This allows the virus to have a high binding affinity to tumor cells. Secondly, after modifying the poliovirus by substituting its internal ribosome entry site (IRES) with the Human Rhinovirus 2's (HRV2) IRES, the poliovirus becomes PVSRIPO, an attenuated version of the …

Breast tumors reprogram their cellular metabolism, nutrient uptake, and utilization-associated biochemical processes. These processes become further transformed as genetically predisposed metastatic breast tumor cells colonize specific organs. Breast tumor cells often metastasize to the brain, bone, lung and liver. Massagué and colleagues isolated organotropic subclones and established organ-specific gene signatures associated with lung-, bone-, and brain-specific metastatic triple-negative breast cancer (TNBC) MDA-MB-231 cells. Using these genetically characterized metastatic subclones specific to lung (LM4175), bone (BoM1833), and brain (BrM-2a), we evaluated marine natural products for the ability to differentially suppress metastatic breast cancer cells in a target organ-dependent manner. Surface tension plays a significant role at the dynamic interface of free-surface flows especially at the microscale in capillary-dominated flows. A model for accurately predicting the formation of two-dimensional viscous droplets in vacuum or gas of negligible density and viscosity resulting from axisymmetric oscillation due to surface tension is solved using smoothed particle hydrodynamics composed of the Navier-Stokes system and appropriate interfacial conditions for the free-surface boundaries. The evolution of the droplet and its free-surface interface is tracked over time to investigate the effects of surface tension forces implemented using a modified continuous surface force method and is compared with …

Environmental issues are global challenges that our students will have to address in their future. Addressing these issues through curriculum helps bring purpose to students’ learning. We have developed a capstone project in our introductory biology class that allows students to engage in these issues in a creative and personalized manner, and which asks students to not only address the biology at the core of these issues but also suggest possible solutions to the problem. In this session, we will share how we scaffold our curriculum to prepare students for this experience, as well as show examples of student work.

Extreme weather conditions challenge pig thermoregulation during transport and are addressed by the National Pork Board (NPB) Transport Quality Assurance® (TQA) program that provides guidelines for trailer boarding, bedding, and misting. These guidelines are widely applied, yet very little is known about the microenvironment within the trailer. In this study, TQA guidelines (V4) were evaluated via extensive thermal environment measurements during transport in order to evaluate spatial variability and implications on ventilation pattern. Faced with issues, such as drought and climate change, educators around the world acknowledge the need for developing students’ ability to solve problems within and across contexts. A systems thinking pedagogy, which recognizes interdependence and interconnected relationships among concrete elements and abstract concepts (Meadows, 2008; Senge et al., 2012), has potential to transform the classroom into a space of observing, theorizing, discovering, and analyzing, thus linking academic learning to the real world. Building Automation System (BAS) is a complex distributed control system that is widely deployed in commercial, residential, industrial buildings for monitoring and controlling mechanical/electrical equipment. Through increasing industrial and technological advances, the control components of BAS are becoming increasingly interconnected. Along with potential benefits, integration also introduces new attack vectors, which tremendous increases safety and security risks in the control system. Historically, BAS lacks security design and relies on physical isolation and "security through obscurity". These methods are unacceptable with the "smart building" technologies. I was composing a new blog post on my computer when I heard a clunk, chunk coming from my outdated air conditioner. Because the wall AC unit faced the outside, occasional bugs would appear in my living space. Sometimes a random bird would fly into the unit, but they were too big to break through. What came through the vent now was not a bug or a bird. A bat with a wingspan of about 12 inches had somehow squeezed through a small opening and was now speeding in circles around my living room.

The Transportation Wallet is one of Portland’s newest parking and transportation demand management (TDM) strategies, designed to reduce parking demand while simultaneously offering new mobility options by bundling transit and bike share passes into one consumer product. Two of Portland’s managed parking districts have elected to add a surcharge to the cost of on-street parking permits, a portion of which is used to subsidize the cost of the Transportation Wallet. Residents and employees in the parking districts are eligible to purchase the Transportation Wallet at a fraction of the retail cost. As an added incentive to increase parking supply, people …

Machine learning (ML) models trained for various purposes are generally kept confidential, e.g., due to their commercial value, proprietary nature of training data, etc. Therefore, commercial cloud-based machine-learning service providers protect their ML models even as they provide one or more services to customers that employ ML models. For example, a service enables a customer to upload an observation, e.g., an image, and receive a label for the observation, generated by a ML model that’s trained to determine labels for images. An electron paramagnetic resonance (EPR)- based method for noninvasive three-dimensional extracellular pH mapping was developed using a pH-sensitive nitroxyl radical as an exogenous paramagnetic probe. Fast projection scanning with a constant magnetic field sweep enabled the acquisition of four-dimensional (3D spatial +1D spectral) EPR images within 7.5 min. Three-dimensional maps of pH were reconstructed by processing the pH-dependent spectral information on the images. To demonstrate the proposed method of pH mapping, the progress of extracellular acidosis in tumor-bearing mouse legs was studied. Characteristics common to animals living in subterranean environments include the reduction or absence of eyes, lessened pigmentation and enhanced sensory systems. How these characteristics have evolved is poorly understood for the majority of cave dwelling species. In order to understand the evolution of these changes, this study uses an invertebrate model system, the freshwater isopod crustacean, Asellus aquaticus, to examine whether adult differences between cave and surface dwelling individuals first appear during embryonic development. We hypothesized that antennal elaboration, as well as eye reduction and pigment loss, would be apparent during embryonic development. We found that differences in pigmentation, eye …

Habitat loss, wind energy development, and the disease white-nose syndrome are major threats contributing to declines in bat populations in North America. In the southeastern US in particular, the recent arrival of white-nose syndrome and changes in landscape composition and configuration have driven shifts in bat species populations and distributions. Effective management strategies which address these large-scale, community-level threats require landscape-scale analyses. Our objective was to model the relationship between ecoregional and landscape factors and occupancy by all bat species in South Carolina, USA, during summer. Building on the successful 2017 presentation, A Primer in Science and Engineering Collection Development, this presentation will explore a content type that stumps the engineering librarian and acquisitions librarian alike - standards. Technical standards are documents that establish a uniform practice or process for materials, procedures, and products. Each of us uses standards daily - from USB ports to elevators to lights, automobiles, bridges, standardized train tracks, and much more. Standards are prepared and issued by a professional groups, committees, societies, or governmental agencies, and can influence safety and performance and the role of business and manufacturing.

Background: Lignin is a promising source of building blocks for upgrading to valuable aromatic chemicals and materials. Endocarp biomass represents a non-edible crop residue in an existing agricultural setting which cannot be used as animal feed nor soil amendment. With significantly higher lignin content and bulk energy density, endo-carps have significant advantages to be converted into both biofuel and bioproducts as compared to other biomass resources. Deep eutectic solvent (DES) is highly effective in fractionating lignin from a variety of biomass feedstocks with high yield and purity while at lower cost comparing to certain ionic liquids.

Eight bat species were known and documented from the California Channel Islands when J.C. Von Bloeker (1967) presented at the First California Islands Symposium in 1965. Additional bat surveys by P.E. Brown (1980) in the 1970s resulted in “Distribution of bats of the California Channel Islands,” which was presented at the second symposium in 1978. Methods of detecting and identifying bats have changed over the past century. Museum collection methods using shotguns have been replaced by mist-netting and recording of echolocation calls. Currently, capture or acoustic records have identified 14 bat species (56% of the 25 species known to occur …

Medical simulation competitions have become an increasingly popular method to provide a hands-on "gamified" approach to education and training in the health professions. The most well-known competition, SimWars, consists of well-coordinated teams that are tasked with completing a series of mind-bending clinical scenarios in front of a live audience through 'bracket-style' elimination rounds. Mutations in ubiquilin2 (UBQLN2) have been linked to abnormal protein aggregation in amyotrophic lateral sclerosis (ALS). The mechanisms underlying UBQLN2-related neurodegenerative diseases remain unclear. Using a tetracycline-regulated gene expression system, the ALS-linked UBQLN2P497H mutant was selectively expressed in either the spinal motor neurons or astrocytes in rats. We found that selectively expressing mutant UBQLN2P497H in the spinal motor neurons caused several core features of ALS, including the progressive degeneration of motor neurons, the denervation atrophy of skeletal muscles, and the abnormal protein accumulation.
