Wild-bird origin influenza A viruses (IAVs or avian influenza) have led to sporadic outbreaks among domestic poultry in the United States and Canada, resulting in economic losses through the implementation of costly containment practices and destruction of birds. We used evolutionary analyses of virus sequence data to determine that 78 H5 low-pathogenic avian influenza viruses (LPAIVs) isolated from domestic poultry in the United States and Canada during 2001 to 2017 resulted from 18 independent virus introductions from wild birds. Within the wild-bird reservoir, the hemagglutinin gene segments of H5 LPAIVs exist primarily as two cocirculating genetic sublineages, and our findings …

European Starling (Sturnus Vulgaris) Reproduction Undeterred By Predator Scent Inside Nest Boxes, Bradley Blackwell, Thomas W. Seamans, Morgan B. Pfeiffer, Bruce N. Buckingham Jan 2018

European Starling (Sturnus Vulgaris) Reproduction Undeterred By Predator Scent Inside Nest Boxes, Bradley Blackwell, Thomas W. Seamans, Morgan B. Pfeiffer, Bruce N. Buckingham

United States Department of Agriculture Wildlife Services: Staff Publications

Indirect predator cues near nests have been shown to enhance perceived predation risk and associated antipredator behaviours in breeding animals across taxa and particularly with birds. We hypothesized that scent from the raccoon (Procyon lotor (Linnaeus, 1758)) inside nest boxes would, despite being an evolutionarily unique predator, enhance perceived risk to the European Starling (Sturnus vulgaris (Linnaeus, 1758)), thus reducing use of treated sites and reproduction. During early spring, starlings selected from nest boxes treated with equal volumes of predator scent, a novel odour, or water (n = 40 boxes per treatment). We evaluated effects of treatment …

Experimental Tests Of Nonsurgical Reproductive Inhibitors To Prevent Coyote Reproduction, Julie K. Young, Marjorie J. Macgregor, Eric M. Gese, Douglas C. Eckery

United States Department of Agriculture Wildlife Services: Staff Publications

Sterilization is an effective nonlethal tool to reduce livestock depredation by coyotes (Canis latrans) because adults without pups to provision are less likely to kill livestock. Surgical sterilization is costly and invasive, so identifying nonsurgical methods for canids that allow wide-scale application is important. We conducted a preliminary assessment of 2 types of reproductive inhibitors (gonadotropin releasing hormone [GnRH] vaccine and deslorelin, a GnRH agonist) on coyote reproductive capabilities. We treated captive coyotes with a GnRH vaccine (n = 6 males, n = 6 females) or deslorelin (n = 6 males), measured number of litters and …

United States Department of Agriculture Wildlife Services: Staff Publications

Concentrations of 26 trace elements including essential (Mg, Ca, Cr, V, Mn, Fe, Co, Ni, Cu, Zn, Se, Sr and Mo) and toxic (As, Cd and Pb), were determined in the liver, kidney, brain, hair, muscle, and stomach contents of the small Indian mongooses inhabiting eight areas on three Hawaiian Islands, Oahu, Maui and Hawaii. There were significant differences in concentrations of some metals among the habitats. Cadmium concentrations in mongooses from the macadamia nut orchards on Island of Hawaii were relatively higher than those in populations from other seven areas. Lead concentrations in mongooses from the Ukumehame firing range …

Estimating Densities For Sympatric Kit Foxes (Vulpes Macrotis) And Coyotes (Canis Latrans) Using Noninvasive Genetic Sampling, R. C. Lonsinger, P. M. Lukacs, E. M. Gese, R. N. Knight, L. P. Waits

United States Department of Agriculture Wildlife Services: Staff Publications

Kit fox (Vulpes macrotis Merriam, 1888) populations in the Great Basin Desert have declined and are of increasing concern for managers. Increasing coyote (Canis latrans Say, 1823) abundance and subsequent intraguild interactions may be one cause for this decline. Concurrent monitoring of carnivores is challenging and therefore rarely conducted. One possible solution for monitoring elusive carnivores is using noninvasive genetic sampling. We used noninvasive genetic sampling to collect fecal DNA from kit foxes and coyotes and estimate their densities from 2013–2014 in Utah, USA. We identified individuals based on microsatellite genotypes and estimated density with multisession spatially explicit …

Contagious disease is a major threat to survival, and the cost of relying on the immune system to defeat pathogens is high; therefore, behavioral avoidance of contagious individuals is arguably an adaptive strategy. Animal findings demonstrate the ability to detect and avoid sick individuals by the aid of olfactory cues, and a recent study indicated that human axillary odor also becomes more aversive as a function of immune activation. By injecting healthy human participants with lipopolysaccharide (0.6 ng/kg body weight) to experimentally induce inflammation, this study demonstrates that natural daily rhythms of urine odor—its perceived dimensions and volatile profile—are altered …

Persistent Spillback Of Bovine Tuberculosis From White-Tailed Deer To Cattle In Michigan, Usa: Status, Strategies, And Needs, Kurt C. Vercauteren, Michael J. Lavelle, Henry Campa Iii Jan 2018

Persistent Spillback Of Bovine Tuberculosis From White-Tailed Deer To Cattle In Michigan, Usa: Status, Strategies, And Needs, Kurt C. Vercauteren, Michael J. Lavelle, Henry Campa Iii

United States Department of Agriculture Wildlife Services: Staff Publications

Free-ranging white-tailed deer (Odocoileus virginianus) are believed to be a self-sustaining reservoir for bovine tuberculosis (bTB) in northeastern Lower Michigan, USA. Although a comprehensive control program is in place and on-farm mitigation strategies to curtail bTB transmission between cattle and deer have been implemented for over a decade, cattle and deer continue to become infected with the disease. Thus, renewed motivation to eradicate bTB is needed if that is truly the goal. Recurrent detection of bTB in cattle in the region is of mounting concern for state and federal agricultural agencies, producers, and wildlife managers. Current on-farm mitigation …

Enclosure Utilization And Enrichment Structure Preferences Of Captive Coyotes, Jeffrey T. Schultz, Julie K. Young Jan 2018

Enclosure Utilization And Enrichment Structure Preferences Of Captive Coyotes, Jeffrey T. Schultz, Julie K. Young

United States Department of Agriculture Wildlife Services: Staff Publications

Environmental enrichment improves well-being of captive animals using a variety of tools, including adding complexity to the physical environment. Designing enrichment structures requires an understanding of behavioral and biological responses to enrichment efforts. Captive coyotes (Canis latrans) utilize shelter structures to hide, rest, and display vigilant behavior. Because these simple structures are regularly used, new and more complex enrichment structures could enhance enclosure enrichment. This study examined the time captive coyotes spent at discrete, complex enclosure features to determine: (1) how coyotes utilize enclosure space and shelter structures; and (2) if coyotes have a preferred enrichment structure design. …

Anticoagulant Rodenticide Residues In Game Animals In California, Stella C. Mcmillin, Robert H. Poppenga, Shannon C. Chandler, Deana L. Clifford Jan 2018

Anticoagulant Rodenticide Residues In Game Animals In California, Stella C. Mcmillin, Robert H. Poppenga, Shannon C. Chandler, Deana L. Clifford

United States Department of Agriculture Wildlife Services: Staff Publications

Anticoagulant rodenticides (ARs) are used to control rodents around homes, buildings, and in agriculture. They have been found widely in predatory and scavenging wildlife as a result of secondary exposure and less commonly in herbivores and omnivores from primary exposure. While predators and scavengers have been monitored for AR exposure, very little information is available about AR residues in edible muscle tissue of game animals. Game animals may be exposed to ARs through direct consumption of bait, ingestion of contaminated food or vegetation, or consumption of contaminated prey items. Carcasses of three species of game animals (black bear, wild pigs, …

A Simple And Scalable Hydrogel-Based System For Culturing Protein-Producing Cells, Qiang Li, Qiaofeng Wang, Ou Wang, Kaifeng Shao, Haishuang Lin, Yuguo Lei

Department of Chemical and Biomolecular Engineering: Faculty Publications

Recombinant protein therapeutics have become important components of the modern medicine. Majority of them are produced with mammalian cells that are cultured either through adherent culturing, in which cells are cultured on substrates, or suspension culturing, in which cells are suspended and cultured in agitated cell culture medium in a culture vessel. The adherent cell culturing method is limited by its low yield. In suspension culturing, cells need extensive genetic manipulation to grow as single cells at high density, which is time and labor-consuming. Here, we report a new method, which utilizes a thermoreversible hydrogel as the scaffold for culturing …

Colorectal cancer (CRC) remains one of the leading causes of cancer related deaths in the United States. Currently, there are limited therapeutic options for patients suffering from CRC, none of which focus on the cell signaling mechanisms controlled by the popular kinase family, cyclin dependent kinases (CDKs). Here we evaluate a Pfizer developed compound, CP668863, that inhibits cyclin-dependent kinase 5 (CDK5) in neurodegenerative disorders. CDK5 has been implicated in a number of cancers, most recently as an oncogene in colorectal cancers. Our lab synthesized and characterized CP668863 – now called 20-223. In our established colorectal cancer xenograft model, 20-223 reduced …

Pancreatic Cancer Associated With Obesity And Diabetes: An Alternative Approach For Its Targeting, Ramesh Pothuraju, Satyanarayana Rachagani, Wade M. Junker, Sanjib Chaudhary, Saraswathi Viswanathan, Sukhwinder Kaur, Surinder K. Batra

Journal Articles: Biochemistry & Molecular Biology

BACKGROUND: Pancreatic cancer (PC) is among foremost causes of cancer related deaths worldwide due to generic symptoms, lack of effective screening strategies and resistance to chemo- and radiotherapies. The risk factors associated with PC include several metabolic disorders such as obesity, insulin resistance and type 2 diabetes mellitus (T2DM). Studies have shown that obesity and T2DM are associated with PC pathogenesis; however, their role in PC initiation and development remains obscure.

MAIN BODY: Several biochemical and physiological factors associated with obesity and/or T2DM including adipokines, inflammatory mediators, and altered microbiome are involved in PC progression and metastasis albeit by different …

The Bi-Directional Relationship Between Gut Microbiota And Autoimmunity, Trevor O. Kirby Jan 2018

The Bi-Directional Relationship Between Gut Microbiota And Autoimmunity, Trevor O. Kirby

EWU Masters Thesis Collection

Humans serve as a major reservoir for a vast number of microbiota. These microbes have evolved symbiotic relationships with humans due to their close proximity with their host. As a result, the immune system adapts to the microbiota thus modulating immunological function. Autoimmunity is a state in which there are aberrant immune responses produced against host tissue. Intestinal bacteria are directly impacted by instances of inflammation brought on by autoimmunity. The complicated nature between autoimmunity and bacterial modulation demonstrates a bi-directional relationship. Here, we utilize experimental autoimmune encephalomyelitis, a model for multiple sclerosis, to explore the bi-directional relationship that disease …

The Source Of Excess Nutrients To Pine Draw, Turnbull National Wildlife Refuge, Henry Price Jan 2018

The Source Of Excess Nutrients To Pine Draw, Turnbull National Wildlife Refuge, Henry Price

EWU Masters Thesis Collection

Nitrogen and phosphorus are the most common limiting nutrients for biological activity in freshwater ecosystems. Applying inorganic nitrogen fertilizer has increased crop productivity but caused excess nitrogen inputs to the hydrosphere. Nitrate contamination is a worldwide environmental problem. The fate of nitrogen in ecosystems is variable based on land type and hydrogeological interactions. Excess nitrogen can be retained in soils, sequestered in stream organisms, denitrified or transported downstream. The goals of this study were to monitor nitrogen concentrations in Pine Draw, Turnbull National Wildlife Refuge (TNWR), Washington, U.S.A., and to determine the source of nitrogen loading. Pine Draw is unique …

Informal Science In The Classroom: How Informal And Formal Educational Institutions Support Science Instruction, Kellie Michelle Crawford Jan 2018

Informal Science In The Classroom: How Informal And Formal Educational Institutions Support Science Instruction, Kellie Michelle Crawford

EWU Masters Thesis Collection

Science education has seen a major overhaul with the implementation of ideas from the NRC’s (2012) report, A Framework for K-12 Science Education: Practices, Crosscutting Concepts, and Core Ideas. At the same time, we have seen a rise in the number of formal educational facilities partnering with informal facilities to bring these practices to students. This study explored the effect these blended approaches had on both student attitudes and their achievement in science content areas. To do this, surveys were designed to assess these areas in upper elementary students participating in a one-year blended science program. First, the literature was …

Optimisation Of Ion Exchange Chromatography Purification Protocols For A Staphylococcal Peptidoglycan Degrading Hydrolase Enzyme, Fiona Maher Jan 2018

Optimisation Of Ion Exchange Chromatography Purification Protocols For A Staphylococcal Peptidoglycan Degrading Hydrolase Enzyme, Fiona Maher

Theses

Bacteriophage (phage) are the most abundant biological entities on earth and were first discovered by d’Herelle in 1917. They are found wherever their hosts live and, like all viruses they do not have the ability to make their own protein. Therefore, in order to reproduce, phage must invade and infect bacterial cells. This project focused on the optimisation of Ion Exchange Chromatography purification protocols for a staphylococcal peptidoglycan degrading hydrolase enzyme (CHAPk). The project objective was to obtain the greatest yield of enzyme from the growth of the E.coU XL 1-Blue expression system into which the vector pQE60 was previously …

Predicting Energy And Water Consumption On Dairy Farms Through Statistical Analysis And Machine-Learning Methods, Philip Shine Jan 2018

Predicting Energy And Water Consumption On Dairy Farms Through Statistical Analysis And Machine-Learning Methods, Philip Shine

Theses

With the abolishment of milking quotas across all European Union member states in April 2015, dairy farmers must adjust their farming practises to minimise milk production costs to adequately prepare for potential periods of reduced revenue. Milk production is an intense energy and water consuming process. Coupled with challenging European greenhouse gas reduction targets and legislation regarding the prevention of groundwater pollution and deterioration, increasing the production of milk in Ireland must be met with the sustainable consumption of on-farm energy and direct water resources, to ensure the future monetary and environmental sustainability of Ireland’s dairy industry. Thus, this body …

Development Of Multi-Class Analytical Methods For Agrochemical Contaminants, Sarah Tuck Jan 2018

Development Of Multi-Class Analytical Methods For Agrochemical Contaminants, Sarah Tuck

Theses

In this project, cost-effective multi-class analytical methods were developed to detect a range of antiparasitic, feed additive and pesticide substances in foodstuffs of animal origin. These substances have the potential to enter the food chain through their occurrence as residues either through the animals themselves or other routes e.g. contamination. To ensure the continued safety of food destined for human consumption regulatory control limits have been established for permitted levels of these residues in foods of animal origin. Despite these controls the continued monitoring of foodstuffs is necessary to ensure these limits are not breached and that substances are not …
