Not Anywhere, Elsewhere: An Ethnographic Look At A Communal Arts Space, Ava Zelkowitz Howard

Theses & ETDs

This thesis is an ethnography of Elsewhere Museum, a nonprofit artist residency and public meeting space in Greensboro, North Carolina. I learned how this living museum, a former thrift store, negotiated its relations to its residents and visitors through participant observation and informal interviews. Elsewhere engages artists and visitors with art and their surroundings’ past, present and future(s) through forms of play, learning, and self and collective reflection. These methods are used purposefully to resist dominant frameworks of power distribution in intentional communities and in the art world.

“They Do Evil Because Of Fear”: Anti-Heroism And American Anxieties In Fear The Walking Dead, Rachael A. Murphy Jan 2018

“They Do Evil Because Of Fear”: Anti-Heroism And American Anxieties In Fear The Walking Dead, Rachael A. Murphy

Theses & ETDs

This thesis explores the current state of the anti-hero in American television. Several popular anti-hero series have recently ended, begging the question of what comes next for the genre. This thesis uses a popular anti-hero TV show, Fear the Walking Dead, as a case study to address the issue. It relies on Joseph Campbell’s conception of the traditional hero in order to trace the development of the anti-hero in twenty-first century American television. Psychoanalyst Carl Jung’s concept of the Shadow is also used to explore the ways in which three anti-heroes in Fear the Walking Dead interact with their own …

Visualizing Structural Transformation And Guest Binding In A Flexible Metal–Organic Framework Under High Pressure And Room Temperature, Hui Yang, Feng Guo, Prem Lama, Wen-Yang Gao, Hui Wu, Leonard J. Barbour, Wei Zhou, Jian Zhang, Briana Aguila, Shengqian Ma

Chemistry Faculty Publications

Understanding the effect of gas molecules on the framework structures upon gas sorption in porous materials is highly desirable for the development of gas storage and separation technologies. However, this remains challenging for flexible metal–organic frameworks (MOFs) which feature “gate-opening/gate-closing” or “breathing” sorption behaviors under external stimuli. Herein, we report such a flexible Cd-MOF that exhibits “gating effect” upon CO2 sorption. The ability of the desolvated flexible Cd-MOF to retain crystal singularity under high pressure enables the direct visualization of the reversible closed-/open-pore states before and after the structural transformation as induced by CO2 adsorption/desorption through in situ single-crystal X-ray …

Microbial Function And Hydrochemistry Within A Stratified Anchialine Sinkhole: A Window Into Coastal Aquifer Interactions, Madison Davis, James R. Garey Jan 2018

Microbial Function And Hydrochemistry Within A Stratified Anchialine Sinkhole: A Window Into Coastal Aquifer Interactions, Madison Davis, James R. Garey

Molecular Biosciences Faculty Publications

Anchialine sinkholes provide insight into coastal aquifer systems and coastal mixing processes. Aquifer microbial community function is usually inferred from hydrochemical information, but there are few direct studies of microbial communities in the Floridan Aquifer. Hospital Hole is a 43 m-deep stratified sinkhole under the Weeki Wachee River, FL, with three distinct brackish layers: a hypoxic layer, a chemocline and a sulfidic anoxic layer. Illumina sequencing and bioinformatic tools were used to reconstruct metabolic functions and interactions of microbial communities in each layer. Each layer appears to originate from different parts of the coastal mixing zone and has a distinct …

Mbd2-Cp2c Loop Drives Adult-Type Globin Gene Expression And Definitive Erythropoiesis, Min Young Kim, Ji Sook Kim, Seung Han Son, Chang Su Lim, Hea Young Eum, Dae Hyun Ha, Mi Ae Park, Eun Jung Baek, Buom-Yong Ryu, Ho Chul Kang, Vladimir N. Uversky, Chul Geun Kim Jan 2018

Molecular Biosciences Faculty Publications

During hematopoiesis, red blood cells originate from the hematopoietic stem cell reservoir. Although the regulation of erythropoiesis and globin expression has been intensively investigated, the underlining mechanisms are not fully understood, including the interplay between transcription factors and epigenetic factors. Here, we uncover that the Mbd2-free NuRD chromatin remodeling complex potentiates erythroid differentiation of proerythroblasts via managing functions of the CP2c complexes. We found that both Mbd2 and Mbd3 expression is downregulated during differentiation of MEL cells in vitro and in normal erythropoiesis in mouse bone marrow, and Mbd2 downregulation is crucial for erythropoiesis. In uninduced MEL cells, the Mbd2-NuRD …

Septal Secretion Of Protein A In Staphylococcus Aureus Requires Seca And Lipoteichoic Acid Synthesis, Wenqi Yu, Dominique Missiakas, Olaf Schneewind Jan 2018

Septal Secretion Of Protein A In Staphylococcus Aureus Requires Seca And Lipoteichoic Acid Synthesis, Wenqi Yu, Dominique Missiakas, Olaf Schneewind

Molecular Biosciences Faculty Publications

Surface proteins of Staphylococcus aureus are secreted across septal membranes for assembly into the bacterial cross-wall. This localized secretion requires the YSIRK/GXXS motif signal peptide, however the mechanisms supporting precursor trafficking are not known. We show here that the signal peptide of staphylococcal protein A (SpA) is cleaved at the YSIRK/GXXS motif. A SpA signal peptide mutant defective for YSIRK/GXXS cleavage is also impaired for septal secretion and co-purifies with SecA, SecDF and LtaS. SecA depletion blocks precursor targeting to septal membranes, whereas deletion of secDF diminishes SpA secretion into the cross-wall. Depletion of LtaS blocks lipoteichoic acid synthesis and …

Primary Liver Cancers, Part 2: Progression Pathways And Carcinogenesis, Kun Jiang, Barbara A. Centeno

Pathology and Cell Biology Faculty Publications

Hepatocellular carcinoma (HCC) and primary intrahepatic cholangiocarcinoma (ICC) have been increasing in incidence worldwide and are leading causes of cancer death. Studies of the molecular alterations leading to these carcinomas provide insights into the key mechanisms involved. A literature review was conducted to identify articles with information relevant to current understanding of the etiologies and molecular pathogenesis of HCC and ICC. Chronic inflammatory diseases are the key etiological risk factors for both HCC and ICC, although other diseases play a role, and for many ICCs, an underlying risk factor is not identified. Mutations in catenin beta 1 (CTNBB1) …

Plasma Derived From Human Umbilical Cord Blood: Potential Cell-Additive Or Cell-Substitute Therapeutic For Neurodegenerative Diseases, Jared Ehrhart, Paul R. Sanberg, Svitlana Garbuzova-Davis

Neurosurgery and Brain Repair Faculty Publications

Limited efficacy of current therapeutic approaches for neurodegenerative disease has led to increased interest in alternative therapies. Cord blood plasma (CBP) derived from human umbilical cord blood (hUCB) may be a potential therapeutic. Benefits of CBP injection into rodent models of aging or ischaemic stroke have been demonstrated, though how benefits are elicited is still unclear. The present study evaluated various factors within the same samples of CBP and human adult blood plasma/sera (ABP/S). Also, autologous CBP effects vs. ABP/S or foetal bovine serum supplements on mononuclear cells from hUCB (MNC hUCB) in vitro were determined. Results showed significantly low …

Neurosurgery and Brain Repair Faculty Publications

Blood-spinal cord barrier (BSCB) alterations, including capillary rupture, have been demonstrated in animal models of amyotrophic lateral sclerosis (ALS) and ALS patients. To date, treatment to restore BSCB in ALS is underexplored. Here, we evaluated whether intravenous transplantation of human bone marrow CD34+ (hBM34+) cells into symptomatic ALS mice leads to restoration of capillary integrity in the spinal cord as determined by detection of microhemorrhages. Three different doses of hBM34+ cells (5 × 104, 5 × 105 or 1 × 106) or media were intravenously injected into symptomatic G93A SOD1 mice …

Human Umbilical Cord Blood Serum–Derived Α-Secretase: Functional Testing In Alzheimer’S Disease Mouse Models, Ahsan Habib, Huayan Hou, Takashi Mori, Jun Tian, Jin Zeng, Shengnuo Fan, Brian Giunta, Paul R. Sanberg, Darrell Sawmiller, Jun Tan

Neurosurgery and Brain Repair Faculty Publications

Alzheimer’s disease (AD) is an age-related disorder that affects cognition. Our previous studies showed that the neuroprotective fragment of amyloid procurer protein (APP) metabolite, soluble APPα (sAPPα), interferes with β-site APP-cleaving enzyme 1 (BACE1, β-secretase) cleavage and reduces amyloid-β (Aβ) generation. In an attempt to identify approaches to restore sAPPα levels, we found that human cord blood serum (CBS) significantly promotes sAPPα production compared with adult blood serum (ABS) and aged blood serum (AgBS) in Chinese hamster ovary cells stably expressing wild-type human APP. Interestingly, CBS selectively mediated the α-secretase cleavage of human neuron-specific recombinant APP695 in a cell-free …

Department of Animal Science: Faculty Publications

Background: Phenotypic networks describing putative causal relationship among multiple phenotypes can be used to infer single-nucleotide polymorphism (SNP) effects in genome-wide association studies (GWAS). In GWAS with multiple phenotypes, reconstructing underlying causal structures among traits and SNPs using a single statistical framework is essential for understanding the entirety of genotype-phenotype maps. A structural equation model (SEM) can be used for such purpose.

Methods: We applied SEM to GWAS (SEM-GWAS) in chickens, taking into account putative causal relationships among body weight (BW), breast meat (BW), hen-house production (HHP), and SNPs. We assessed the performance of SEM-GWAS by comparing the model results …

The Influence Of Fat And Hemicellulose On Methane Production And Energy Utilization In Lactating Jersey Cattle, O. R. Drehmel, T. M. Brown-Brandl, J. V. Judy, S. C. Fernando, P. S. Miller, K. E. Hales, Paul J. Kononoff Jan 2018

The Influence Of Fat And Hemicellulose On Methane Production And Energy Utilization In Lactating Jersey Cattle, O. R. Drehmel, T. M. Brown-Brandl, J. V. Judy, S. C. Fernando, P. S. Miller, K. E. Hales, Paul J. Kononoff

Department of Animal Science: Faculty Publications

Feeding fat to lactating dairy cows may reduce methane production. Relative to cellulose, fermentation of hemicellulose is believed to result in less methane; however, these factors have not been studied simultaneously. Eight multiparous, lactating Jersey cows averaging (±SD) 98 ± 30.8 d in milk and body weight of 439.3 ± 56.7 kg were used in a twice-replicated 4 × 4 Latin square to determine the effects of fat and hemicellulose on energy utilization and methane production using a headbox-type indirect calorimetry method. To manipulate the concentration of fat, porcine tallow was included at either 0 or 2% of the diet …

Predictive Ability Of Genome-Assisted Statistical Models Under Various Forms Of Gene Action, Mehdi Momen, Ahmad Ayatollahi Mehrgardi, Ayyub Sheikhi, Andreas Kranis, Llibertat Tussell, Gota Morota, Guilherme J. M. Rosa, Daniel Gianola Jan 2018

Predictive Ability Of Genome-Assisted Statistical Models Under Various Forms Of Gene Action, Mehdi Momen, Ahmad Ayatollahi Mehrgardi, Ayyub Sheikhi, Andreas Kranis, Llibertat Tussell, Gota Morota, Guilherme J. M. Rosa, Daniel Gianola

Department of Animal Science: Faculty Publications

Recent work has suggested that the performance of prediction models for complex traits may depend on the architecture of the target traits. Here we compared several prediction models with respect to their ability of predicting phenotypes under various statistical architectures of gene action: (1) purely additive, (2) additive and dominance, (3) additive, dominance, and two-locus epistasis, and (4) purely epistatic settings. Simulation and a real chicken dataset were used. Fourteen prediction models were compared: BayesA, BayesB, BayesC, Bayesian LASSO, Bayesian ridge regression, elastic net, genomic best linear unbiased prediction, a Gaussian process, LASSO, random forests, reproducing kernel Hilbert spaces regression, …

Effect Of Differing Ingredients And Packaging Technologies On The Color Of High-Pressure Processed Ground Beef, Jhinuk Gupta, Chad G. Bower, Gary Sullivan, George A. Cavender

Department of Animal Science: Faculty Publications

High-pressure processing (HPP) is a nonthermal pasteurization technique to control pathogens, like Escherichia coli. However, color changes in raw beef induced by HPP restrict its use within the beef industry. e objectives of this study were to investigate the effects of adding curing agents (nitrite) and packaging with or without reducing compounds (ascorbic acid/erythorbate) on color retention in high-pressure processed ground beef. Color was measured (CIE L*a*b*) before HPP and on days 3, 7, 12, 14, 19, and 21 after HPP. Statistical analysis (SAS GLIMMIX) was run to identify the main effects of adding curing agents, packaging, and …

Do Stronger Measures Of Genomic Connectedness Enhance Prediction Accuracies Across Management Units?, Haipeng Yu, Matthew L. Spangler, Ronald M. Lewis, Gota Morota

Department of Animal Science: Faculty Publications

Genetic connectedness assesses the extent to which estimated breeding values can be fairly compared across management units. Ranking of individuals across units based on best linear unbiased prediction (BLUP) is reliable when there is a sufficient level of connectedness due to a better disentangling of genetic signal from noise. Connectedness arises from genetic relationships among individuals. Although a recent study showed that genomic relatedness strengthens the estimates of connectedness across management units compared with that of pedigree, the relationship between connectedness measures and prediction accuracies only has been explored to a limited extent. In this study, we examined whether increased …

Porcine circovirus associated disease (PCVAD) is a term used to describe the multifactorial disease syndromes caused by porcine circovirus type 2 (PCV-2), which can be reproduced in an experimental setting through the co-infection of pigs with PCV-2 and porcine reproductive and respiratory syndrome virus (PRRSV). The resulting PCVAD-affected pigs represent a subpopulation within the co-infected group. In co-infection studies, the presence of increased microbiome diversity is linked to a reduction in clinical signs. In this study, fecal microbiota transplantation (FMT) was investigated as a means to prevent PCVAD in pigs co-infected with PRRSV and PCV-2d. The sources of the FMT …

Function And Dysfunction Of Fatty Acid Mobilization: A Review, Joslyn K. Beard, Travis Mulliniks, Dustin T. Yates

Department of Animal Science: Faculty Publications

Western populations have a growing obesity epidemic due in part to excessive nutrient intake from high-fat diets, which are increasingly common. Overindulgence of nutrients is associated with a greater incidence of metabolic dysfunction and a greater risk for obesity, diabetes, hypertension, and other metabolic disorders that lower quality of life. Research in humans and animal models has improved our understanding of how excess circulating free fatty acids negatively impact the ability of muscle and other tissues to regulate nutrient uptake and utilization. It is generally accepted by the scientific community that excess circulating fatty acids lead to insulin resistance, but …

Dietary β-adrenergic agonists (β-AA) are used in livestock to increase muscle protein accretion and decrease adipose deposition during the last 20 to 40 d of the finishing period (Johnson et al., 2014). These β-AA act through specific seven transmembrane receptors and are classified by the receptor isoform to which they primarily bind (Mersmann, 1998). Two β-AA are approved for use in beef cattle in the United States: ractopamine HCl (RAC, β1 agonist) and zilpaterol HCl (ZH, β2 agonist) (Johnson et al., 2014). Supplementation of β-AA increases efficiency of the animal and results in a leaner carcass (Elam et al., 2009). …

Impact Of Pre-And Postpartum Nutrition On March-Calving Cow And Progeny Productivity, Jacki A. Musgrave, Devin L. Broadhead, L. A. Stalker Stalker, Richard N. Funston

Nebraska Beef Cattle Reports

March-calving cows were fed hay or grazed corn residue prepartum, and then either fed hay or grazed subirrigated meadow postpartum. Combinations of these different feeding strategies impacted body weight (BW) and body condition score (BCS) during the pre-and postpartum period; however, resulted in similar pregnancy rates. Although differences were not observed in cow pregnancy rates, a benefit in pre-weaning growth was observed for calves from the dams in postpartum meadow treatment. A tendency for an improvement in marbling score was observed for steers born to cows fed hay prepartum, perhaps indicating a higher plane of nutrition prepartum may improve quality …
