Debris, Diatoms, And Dolphins: Tracking Child Engagement At A Public Science Festival, Kaya Van Beynen, Theresa G. Burress
USF St. Petersburg Campus Faculty Publications
Visitors to public science festivals have a tremendous amount of free choice to decide how to navigate through the festival, as well as when, where, and how long to stop at an exhibit. This study examines how elementary-aged children individually or collaboratively engaged with festival exhibits at a public science festival in St. Petersburg, Florida. Although many exhibit activities are designed to appeal to children, no research has been done with regard to child engagement with one-day, outdoor science festivals, such as this one. Engagement can be measured by unobtrusive observation of the behavior and interactions of children. Factors that …

Justice John Paul Stevens retired from the Supreme Court almost a decade ago and turned ninety-eight years old in April 2018. How should we remember his legacy on the Supreme Court? This Article places his legacy within his election law jurisprudence. Specifically, Justice Stevens provided a consistent theory, which we term “impartial governance,” that has had a lasting impact on the field. This theory undergirds Justice Stevens’s creation of the important Anderson-Burdick-Crawford balancing test that federal courts use to construe the constitutionality of laws that impact the right to vote, such as voter ID laws. It is part of his …

Going Gaga: Pop Fandom As Online Community Of Practice, John D. N. Carter
Theses and Dissertations--Linguistics
Among various fan sites dedicated to pop stars, GagaDaily is one prominent online collective that centers around Lady Gaga. This study is a piece of ethnographic research focused on two claims – GagaDaily constitutes a Community of Practice (Eckert, 2006) in an online setting, and the regular use of humor by users fulfills social and pragmatic roles in the discourse. Communicative phenomena (both textual and graphic) that characterize the linguistic repertoire of GagaDaily members were catalogued from the first 100 pages of one thread within the forums. These data were grouped into categories corresponding to different dimensions of language use …

Phages (viruses that infect bacteria) play important roles in the gut ecosystem through infection of bacterial hosts, yet the gut virome remains poorly characterized. Mammalian gut viromes are dominated by double-stranded DNA (dsDNA) phages belonging to the order Caudovirales and single-stranded DNA (ssDNA) phages belonging to the family Microviridae. Since the relative proportion of each of these phage groups appears to correlate with age and health status in humans, it is critical to understand both ssDNA and dsDNA phages in the gut. Building upon prior research describing dsDNA viruses in the gut of Ciona robusta, a marine invertebrate model system …

Zooplankton dominate the abundance and biomass of multicellular animals in pelagic marine environments; however, traditional methods to characterize zooplankton communities are invasive and laborious. This study compares zooplankton taxonomic composition revealed through metabarcoding of the cytochrome oxidase I (COI) and 18S rRNA genes to traditional morphological identification by microscopy. Triplicates of three different sample types were collected from three coral reef sites in the Florida Keys National Marine Sanctuary: (1) 1 L surface seawater samples prefiltered through 3 μm filters and subsequently collected on 0.22 μm filters for eDNA (PF‐eDNA); (2) 1 L surface seawater samples filtered on …

Measurements of the status and trends of key indicators for the ocean and marine life are required to inform policy and management in the context of growing human uses of marine resources, coastal development, and climate change. Two synergistic efforts identify specific priority variables for monitoring: Essential Ocean Variables (EOVs) through the Global Ocean Observing System (GOOS), and Essential Biodiversity Variables (EBVs) from the Group on Earth Observations Biodiversity Observation Network (GEO BON) (see Data Sheet 1 in Supplementary Materials for a glossary of acronyms). Both systems support reporting against internationally agreed conventions and treaties. GOOS, established under the auspices …

The expression of the Clostridium difficile binary toxin CDT is generally observed in the RT027 (ST1) and RT078 (ST11) C. difficile isolates, which are associated with severe C. difficile infection (CDI). However, we recently reported that the non-RT027 and non-RT078 C. difficile strain LC693 (TcdA+TcdB+ CDT+, ST201) caused severe diarrhea in a patient in Xiangya Hospital in China. C.difficile LC693 is a member of Clade 3, and in this study, we identified LC693 as RT871 and compared its virulence and pathogenicity to those of C.difficile R20291 (TcdA+TcdB+CDT+, ST1/RT027), UK6 (TcdA+TcdB+CDT+, ST35/RT027), CD630 (TcdA+TcdB+CDT−, ST54, RT012), and 1379 (TcdA+TcdB+CDT−, ST54/RT012), with …

Domain III (DIII) of the tick-borne encephalitis virus (TBEV) protein E contains epitopes, which induce antibodies capable of neutralizing the virus. To enhance the immunogenicity of this protein, which has a low molecular weight, the aim of the present work was to express, isolate, and characterize a chimeric protein based on the fusion of the bacterial chaperone HSP70 of Yersinia pseudotuberculosis and EIII (DIII + stem) as a prospective antigen for an adjuvanted delivery system, the tubular immunostimulating complex (TI-complex). The chimeric construction was obtained using pET-40b(+) vector by ligating the respective genes. The resulting plasmid was transformed into DE3 …

Incorporation Of Acoustic Sensors On A Coastal Ocean Monitoring Platform For Measurements Of Biological Activity, James Locascio, David Mann, Kyle Wilcox, Mark Luther
Marine Science Faculty Publications
Ocean observing stations have mainly focused on data collection of physical parameters measured in the ocean and atmosphere and also, to some extent, of biogeochemical parameters. Robust sensors capable of measuring biological data reflective of higher tropic level function at the same time scales as other parameters are not yet commonly incorporated into the sensor array used on observing platforms. In this project, we reengineered a coastal ocean observatory to include two hydrophones for this purpose. One hydrophone (HTI 96-MIN) was used to record ambient acoustic signals of fish reproductive sounds, and a second hydrophone (Vemco VR2C) was used to …
Comparing Two Flywheel-Piston Linkages, Anthony Sanchez
Comparing Two Flywheel-Piston Linkages, Anthony Sanchez
Undergraduate Journal of Mathematical Modeling: One + Two
The main idea of this paper is to compare two different ways of linking a flywheel to a piston: the Scotch Yoke linkage and the Eccentric linkage. The Scotch Yoke linkage is a way to convert the linear motion of a piston into rotational motion by using a flywheel, or vice versa. The Eccentric mechanism, on the other hand, consists of a circular wheel that is fixed to a rotating axle that makes it rotate. We compare two methods by expressing both motions as functions of three variables (R, L and θ) and use these functions to …

Analyzing Children's Literature Awards, Literary Criteria, And District Demographics To Create A Diverse And Inclusive Elementary School Classroom Library, Casey L. Lavallee
Honors Undergraduate Theses
Future elementary school teachers are tasked with the responsibility to ensure that the children in their classrooms have access to high quality books every day. This can be accomplished by building classroom libraries with titles that are not only of high literary quality but are also inclusive of the students in the classroom as well as the people these children will encounter outside of the classroom. Children should be able to see themselves represented in the literature they read, and they should also be able to develop empathy for people who are different than they are. Diverse literature can have …

The Six-Legged Subject: A Survey Of Secondary Science Teachers’ Incorporation Of Insects Into U.S. Life Science Instruction, Erin Ingram, Douglas Golick
Department of Entomology: Faculty Publications
To improve students’ understanding and appreciation of insects, entomology education efforts have supported insect incorporation in formal education settings. While several studies have explored student ideas about insects and the incorporation of insects in elementary and middle school classrooms, the topic of how and why insects are incorporated in secondary science classrooms remains relatively unexplored. Using survey research methods, this study addresses the gap in the literature by (1) describing in-service secondary science teachers’ incorporation of insects in science classrooms; (2) identifying factors that support or deter insect incorporation and (3) identifying teachers’ preferred resources to support future entomology education …

Loss Of Α-Calcitonin Gene-Related Peptide (Αcgrp) Reduces Otolith Activation Timing Dynamics And Impairs Balance, Sherri M. Jones, Sarath Vijayakumar, Samantha A. Dow, Joseph C. Holt, Paivi M. Jordan, Anne E. Luebke
Department of Special Education and Communication Disorders: Faculty Publications
Calcitonin gene-related peptide (CGRP) is a neuroactive peptide that is thought to play a role at efferent synapses in hair cell organs including the cochlea, lateral line, and semicircular canal. The deletion of CGRP in transgenic mice is associated with a significant reduction in suprathreshold cochlear nerve activity and vestibulo– ocular reflex (VOR) gain efficacy when compared to littermate controls. Here we asked whether the loss of CGRP also influences otolithic end organ function and contributes to balance impairments. Immunostaining for CGRP was absent in the otolithic end organs of aCGRP null (-/-) mice while choline acetyltransferase (ChAT) immunolabeling appeared …

There is widespread agreement that agricultural antibiotic resistance should be reduced, however, it is unclear from the available literature what an appropriate target for reduction would be. Organic farms provide a unique opportunity to disentangle questions of agricultural antibiotic drug use from questions of antibiotic resistance in the soil. In this study, soil was collected from 12 certified organic farms in Nebraska, evaluated for the presence of tetracycline and sulfonamide resistance genes (n = 15 targets), and correlated to soil physical, chemical, and biological parameters. Tetracycline and sulfonamide antibiotic resistance genes (ARGs) were found in soils from all 12 …
Measuring Visual Acuity In Awake Mice Using Visually Evoked Potentials (Veps), Emanuel Drutu
Measuring Visual Acuity In Awake Mice Using Visually Evoked Potentials (Veps), Emanuel Drutu
CUP Undergraduate Research
We investigated a means to measure visual acuity in awake mice using visually evoked potentials (VEPs). Using counter-phasing sinusoidal gratings as stimuli, we compared the effectiveness of vertical and horizontal orientations in generating VEPs. Using stereotaxic implanted electrodes in the animal’s primary visual cortex, the evoked VEPs were recorded and analyzed. At the lowest spatial frequency, vertical stimuli evoked the largest VEP amplitude. However, at higher spatial frequencies, a reversal occurs where horizontal gratings evoked larger VEPs. These data suggest vertical and horizontal stimuli have different effects on generating VEPs although further investigation is required to determine which stimulus is …

The Impact Of Higher Education, Ethnicity, & The Intersection Of Both On Perceptions Of People With Mental Illness, Natalie Crocker
MA Community Psychology Theses
The purpose of this study was to explore how higher education and ethnicity may individually impact perceptions of people with mental illness, as well as to explore the intersection of higher education and ethnicity on such perceptions. Descriptive phenomenological in design, this study explores the experiences of ten students at a university in Portland, Oregon. In semi-structured, open-ended interviews participants recalled experiences in which higher education, ethnicity, and the intersection of both impacted how they thought about mental illness. Interview transcripts were coded and analyzed using Colaizzi’s (1978) seven-step process. The analysis resulted in several themes supporting the current literature …

Sivcpz Closely Related To The Ancestral Hiv-1 Is Less Or Non-Pathogenic To Humans In A Hu-Blt Mouse Model, Zhe Yuan, Guobin Kang, Lance Daharsh, Wenjin Fan, Qingsheng Li
Nebraska Center for Virology: Faculty Publications
The HIV-1 pandemic is a consequence of the cross-species transmission of simian immunodeficiency virus in wild chimpanzees (SIVcpz) to humans. Our previous study demonstrated SIVcpz strains that are closely related to the ancestral viruses of HIV-1 groups M (SIVcpzMB897) and N (SIVcpzEK505) and two SIVcpz lineages that are not associated with any known HIV-1 infections in humans (SIVcpzMT145 and SIVcpzBF1167), all can readily infect and robustly replicate in the humanized-BLT mouse model of humans. However, the comparative pathogenicity of different SIVcpz strains remains unknown. Herein, we compared the pathogenicity of the above four SIVcpz strains with HIV-1 using humanized-BLT mice. …

Characterizing the transmitted/founder (T/F) viruses of multi-variant SIV infection may shed new light on the understanding of mucosal transmission. We intrarectally inoculated six Chinese rhesus macaques with a single high dose of SIVmac251 (3.1 × 104 TCID50) and obtained 985 full-length env sequences from multiple tissues at 6 and 10 days post-infection by single genome amplification (SGA). All 6 monkeys were infected with a range of 2 to 8 T/F viruses and the dominant variants from the inoculum were still dominant in different tissues from each monkey. Interestingly, our data showed that a cluster of rare T/F viruses …

Zika virus (ZIKV), an emerging arbovirus, has become a major human health concern globally due to its association with congenital abnormalities and neurological diseases. Licensed vaccines or antivirals against ZIKV are currently unavailable. Here, by employing a structure-based approach targeting the ZIKV RNA-dependent RNA polymerase (RdRp), we conducted in silico screening of a library of 100,000 small molecules and tested the top ten lead compounds for their ability to inhibit the virus replication in cell-based in vitro assays. One compound, 3-chloro-N-[({4-[4-(2-thienylcarbonyl)-1-piperazinyl]phenyl}amino)carbonothioyl]-1-benzothiophene-2-carboxamide (TPB), potently inhibited ZIKV replication at submicromolar concentrations. Molecular docking analysis suggests that …

The Doctorate of Philosophy in Visual Arts has been a subject of debate in the United States, where the currently accepted terminal degree in visual and fine arts is the Master of Fine Arts. In the United Kingdom, as in much of Western Europe, Australia, New Zealand, and Asia the PhD in visual arts has been well developed and accepted. In a global economy, where students in the higher Education marketplace will have to compete on an international level, it may be inevitable that the higher Education system in the United States ultimately create and adopt a PhD in visual …

A natural impulse for all humans when perceiving data from our surroundings is to sort through the information in order to make sense of our world. Using this information, we evaluate where we stand in relation to our environment. Assumptions based on previous experiences and culturally learned preconceptions affect how we relate with and treat others in a social environment. Those who have dissimilar beliefs and characteristics are met with fear and enmity, while those that are similar to ourselves help to form and justify parts of our individual identities. Using our natural proclivity to sort using prior information, society …

Not Anywhere, Elsewhere: An Ethnographic Look At A Communal Arts Space, Ava Zelkowitz Howard
Theses & ETDs
This thesis is an ethnography of Elsewhere Museum, a nonprofit artist residency and public meeting space in Greensboro, North Carolina. I learned how this living museum, a former thrift store, negotiated its relations to its residents and visitors through participant observation and informal interviews. Elsewhere engages artists and visitors with art and their surroundings’ past, present and future(s) through forms of play, learning, and self and collective reflection. These methods are used purposefully to resist dominant frameworks of power distribution in intentional communities and in the art world.
