Localization Of Sip470, A Plant Lipid Transfer Protein In Nicotiana Tabacum, Shantaya Andrews

SABP2-interacting protein 470 (SIP470), a non-specific lipid transfer protein (nsLTP), was discovered in a yeast two-hybrid screening using SABP2 as bait and tobacco leaf proteins as prey. SABP2 is an important enzyme in systemic acquired resistance that converts salicylic acid to methyl salicylate. Localization studies are an important aspect to understanding the biological function of proteins. nsLTPs are generally considered apoplastic proteins and has been localized intracellularly and extracellularly. Transient expression shows highest expression of SIP470-eGFP at 2 days post infiltration into Nicotiana benthamiana. Confocal microscopy showed localization near the periphery of the cell. Insulin resistance and obesity are major public health concerns. The impact of diet and genetics on insulin resistance and obesity is well accepted. Additionally, the gut microbiota has been shown to influence obesity and metabolic disorders. However, much remains to be understood about the role of gut microbiota in the development of insulin resistance and obesity. We utilized a mouse model lacking PAS kinase, a protein involved in cellular metabolism, in order to better understand the relationship between diet, genetics and the gut microbiota. In natural ecosystems, like deciduous and coniferous forests, soil CO2 flux or soil respiration is highly variable and influenced by multiple factors including temperature, precipitation, dissolved soil organic carbon (DOC), dissolved organic matter (DOM), and bacterial and fungal biomass and diversity. However, as the human population continues to grow rapidly, so too do urbanized landscapes with unknown consequences to soil respiration. To determine the extent urbanization influences seasonal shifts in microorganisms and environmental drivers alter soil respiration, we evaluated bacterial and fungal communities, soil physiochemical characteristics, and respiration in forested and urbanizing ecosystems in three watersheds across northern Utah, USA. …

Animating by hand can be a long and challenging process in part because of the necessity of drawing every frame by hand. 3D animation media minimize this problem with the use of automatically interpolated frames, but despite significant research no universally acceptable techniques have been demonstrated for 2 dimensional interpolation. In this paper we explore computer-assisted optimizations to the animation pipeline. Specifically, we utilize 3D motion fields to create more realistic in-between frames for sets of 2D ``key frames.'' We demonstrate our method by using it to create 2D special effects for a 30-second clip of an animated short film.

Heart failure is the main cause of death for both men and women in the United States. The only proven treatment for patients with heart failure is heart transplantation. The goal of this research is to create patches of tissue that could mimic the function of the native heart to repair the damaged portions of the heart. In this study, whole porcine hearts were decellularized to create a 3D construct that was recellularized with cardiomyocytes (CM) differentiated from human induced pluripotent stem (IPS) cells. Context: Thuja occidentalis is prescribed in Homoeopathy in treating rheumatoid arthritis. We speculated the anti-arthritic mechanism of Homoeopathic dilutions of Thuja occidentalis against the complete Freund's adjuvant (CFA)-induced arthritis in rats. Materials and Methods: Arthritis was induced (n = 28) by subplantar injection of 0.1 ml CFA in the right hind paw of rats. The oral dose of crude Thuja occidentalis was 30 mg/kg/b. i. d and that of Homoeopathic dilutions was 0.1 ml/b. i. d. Orally administered diclofenac at 5 mg/kg/day served as a standard. Pulmonary complications are a significant cause for morbidity and mortality in osteogenesis imperfecta (OI). However, to date, there have been few studies that have systematically evaluated pulmonary function in individuals with OI. We analyzed spirometry measurements, including forced vital capacity (FVC) and forced expiratory volume in the first second (FEV1 ), in a large cohort of individuals with OI (n = 217) enrolled in a multicenter, observational study. Peace of Mind Dog Rescue (POMDR) relies heavily on its volunteers to carry out important operational needs that allow them to fulfill their mission of being a resource and advocate for local senior dogs and senior citizens. From overseeing adoption events to foster care to administrative duties, volunteers serve as the arms-and-legs of the organization. Despite having a volunteer base of over 700 individuals, POMDR faces a challenge in preparing new volunteers for their role within the organization. A 51-year-old white female presented with a painful ulcer of the left ventrolateral tongue. An incisional biopsy confirmed a diagnosis of adenosquamous carcinoma. The adenosquamous carcinoma is an uncommon malignant tumor with histopathological features of a squamous carcinoma and an adenocarcinoma. A definitive diagnosis requires histopathological examination of a deep biopsy involving the submucosal tissue. This malignant tumor shows aggressive behavior with early invasive growth and a poor prognosis. The histopathological findings and differential diagnosis of a case of adenosquamous carcinoma of the tongue are discussed.

Vegetative environmental buffers, or VEBs, are rows of trees and shrubs purposefully planted to mitigate livestock odor. In this article, we present VEB-econ, a free-to-use geographic information system–based decision support tool Extension professionals can implement when working with livestock producers in designing site-specific VEBs. A soil database links tree and shrub species recommendations to soil-based suitability guidelines. VEB-econ estimates annualized cost for tree establishment and management and opportunity costs and factors in potential Natural Resources Conservation Service cost-share payments. We delivered to growers and Extension educators a workshop addressing good agricultural practices (GAP) for produce safety. To assess the workshop's effects as applicable to behavioral intervention theory, we studied past behaviors, behavioral intentions, and changes in knowledge. Workshop participants had been aware of but did not fully understand certain GAP, such as implementing written food safety plans; after the workshop, both groups reported improved understanding of various concepts. Producers were mixed regarding intent to make GAP-related changes after workshop participation, whereas the educators overall were likely to make changes related to teaching GAP. Small-scale science activities provide an opportunity for engagement of diverse, large audiences at settings such as 4-H fairs. We present practical information for implementation of small-scale food science experiences in the Extension education context. Our focus is description of a tool for adaptation of activities for inquiry-based learning and tips for miniaturization of activities to save on costs and resources.

Workers on prison farms in the United States commonly grow produce and row crops and raise livestock. Teaching good agricultural practices to prisoners and prison staff increases the safety of the food produced and reduces the likelihood of an illness outbreak. We developed a food safety curriculum for use on prison farms in Iowa. In developing the curriculum, we encountered many challenges that resulted in modifications in the content and delivery method. We present the content of the curriculum and provide educators with a better understanding of challenges and potential solutions related to working with prison farms.

Our goal with this article is to present a visual aid and tool for assessing local food systems. We propose that local food systems comprise four essential components represented by the acronym FOOD: in(F)rastructure, pr(O1)duction, (O2)rganizations, and (D)emand. The FOOD assessment tool provides a visual overview of the statuses of these four essential components relative to a particular food system. It is also useful for comparing one food system to another or for tracking changes over time. Within the last decade, the positive effects of Pueraria mirifica on osteoporosis have been established in rats and monkeys. However, the effect of P. mirifica on bone healing has not been elucidated. In this study, post-menopausal osteoporotic monkeys were subjected to an iliac crest biopsy, divided into two groups (5 animals/group), and fed daily with standard monkey diet alone (PMP0 group) or mixed with 1,000 mg/kg body weight of P. mirifica powder (PMP1000 group) for 16 months. The use of sentinel animals in animal research facilities is the cornerstone of animal health monitoring for exposure to natural pathogens of laboratory animals. This is because infection with these pathogens produces no overt signs of disease yet the infection may affect the outcome of research utilising these animals. One of the important pathogens includes Klebsiella pneumoniae, whose prevalence can be high in laboratory animals. Human knee kinematics, especially during gait, are an important analysis tool. The current "gold standard" for kinematics measurement is a multi-camera, marker-based motion capture system with 3D position tracking. These systems are accurate but expensive and their use is limited to a confined laboratory environment. High deflection strain gauges (HDSG) are a novel class of sensors that have the potential to measure kinematics and can be inexpensive, low profile, and are not limited to measurements within a calibrated volume. This research applies techniques from the field of optimization to spatial positioning and energy management in Unmanned Aerial Vehicles (UAVs). Two specific areas are treated: optimization of UAV view plans for 3D modeling of infrastructure, and trajectory optimization of solar powered high-altitude long-endurance (HALE) UAVs. Structure-from-Motion (SfM) is a computer vision technique for creating 3D models from 2D images. View planning is the process of planning image sets that will effectively model a given scene. At the beginning of World War I, the only military working dogs the United States owned were sled dogs. In comparison, European nations in World War I used canines as sentries, messengers, ambulance, and draft dogs. In 1942, members of the American public, created Dogs for Defense Inc. to help recruit dogs for military use. By the end of the Vietnam War, dogs no longer were donated by the American public for use, rather the American military owned the dogs they deployed.

Presented is the first field survey and laboratory-based study focused on Harpellales gut fungi found in Culicidae (mosquito) larvae collected from various parts of Idaho. Overall, 34 sites were sampled ranging from urban storm drains and irrigation puddles to pristine stream-side puddles. These sites yielded 17 different species of mosquitoes and three previously described species of gut fungi. Three species of mosquitoes were the first recorded observations as hosts of the following gut fungi: Culiseta alaskaensis was infested with Zancudomyces culisetae (from Renwyck Creek), Culex tarsalis with Smittium culicis (from Cottonwood Creek), and Ochleratus sp. with Smittium minutisporum (from Bear …

Archaeologists describe the Uinta Fremont (A.D. 0 – 1300) as a mixed foraging-farming society that underwent a dramatic social change from A.D. 700 – 1000. Researchers observe through different architectural styles and subsistence activity a change from large, aggregated settlements to more dispersed and defensively oriented villages and hamlets. The Ideal Free Distribution (IFD) model provides an explanatory framework through which to interpret these changes. Fluids in computer generated imagery can add an impressive amount of realism to a scene, but are particularly time-consuming to simulate. In an attempt to run fluid simulations in real-time, recent efforts have attempted to simulate fluids by using machine learning techniques to approximate the movement of fluids. We explore utilizing machine learning to simulate fluids while also integrating the Fluid-Implicit-Particle (FLIP) simulation method into machine learning fluid simulation approaches.

The innate immune system is the first line of host defense against external exposures. During these initial encounters, antigen presenting cells - specifically monocytes and macrophages - modulate further inflammatory responses. Macrophages exist along a spectrum of phenotypic programs; on the inflammatory M1 end they enhance immune activity while on the anti-inflammatory M2 end they suppress further immune activation. Furthermore, within M2 macrophages there exist many subpopulations, namely M2a and M2d, each with specific roles during infection or exposure. We sought to compare the epigenetic profiles of these subpopulations of macrophages to determine key regulatory gene networks and factors that …
