We recently identified a novel vancomycin-resistant Enterococcus faecium (VREfm) clone ST736 with reduced daptomycin susceptibility. The objectives of this study were to assess the population dynamics of local VREfm strains and genetic alterations predisposing to daptomycin resistance in VREfm ST736 strains. Multilocus sequence typing and single nucleotide variant data were derived from whole-genome sequencing of 250 E. faecium isolates from 1994-1995 (n = 43), 2009-2012 (n = 115) and 2013 (n = 92). This study is about how art constitutes an invitation for marginalized people. Refugees are the uninvited, and often remain on the periphery of host communities. This research examines how artmaking acts as an invitation for refugee preschool students and their families to connect with the broader community. I approached data collection through the method of educational connoisseurship and educational criticism and the theory of relational aesthetics. I analyzed data from observations conducted in a refugee preschool classroom and from interviews with six families. There are countless aspects of water to be explored, from the sheer physical qualities of water to the spiritual, political, and practical associations with water. The position of an author on the byline of a paper affects the inferences readers make about their contributions to the research. We examine gender differences in authorship in the ecology literature using two datasets: submissions to six journals between 2010 and 2015 (regardless of whether they were accepted), and manuscripts published by 151 journals between 2009 and 2015. Women were less likely to be last (i.e., “senior”) authors (averaging ~23% across journals, years, and datasets) and sole authors (~24%), but more likely to be first author (~38%), relative to their overall frequency of authorship (~31%). In many applications data are collected sequentially in time with very short time intervals between observations. If one is interested in using new observations as they arrive in time then non-sequential Bayesian inference methods, such as Markov Chain Monte Carlo (MCMC) sampling, can be too slow. Increasingly, state space models are being used to model nonlinear and non-Gaussian systems. The structure of state space models allows for sequential Bayesian inference so that an approximation to the posterior distribution of interest can be updated as new observations arrive. This dissertation presents a collection of studies that investigate the genetic and epigenetic associations to ascites phenotype in broiler chickens. Ascites is a significant metabolic disease associated with fast-growing meat-type chickens (broilers) and is a terminal result of pulmonary hypertension syndrome PHS. It is a multi-factorial syndrome caused by interactions between genetic, physiological, environmental, and managemental factors. It was estimated that ascites accounts for losses of about US$1 billion annually worldwide and for over 25% of broilers mortality. The Food and Drug Administration (FDA) have set new standards that apply to agriculturalists producing crops eaten fresh and/or raw by consumers. This new produce safety rule, known as the Food Safety Modernization Act (FSMA), has established science-based standards for all areas of production in agriculture with regards to microbial contamination. Escherichia coli (E. coli) O157:H7 is a particular bacterium of concern under FSMA guidelines. Grower compliance is mandatory and therefore vital to the continuation of any farm. Historically speaking, the South has maintained rigid, male dominated constructions of gender, which have manifested themselves in almost every aspect of the southern individual’s actions, sense of self, and appearance. While much of southern culture has been centered on this rigid identity, the southern gothic literary tradition strays from this stark binary, especially in its depiction of female characters. Through the use of grotesque representations, these characters are able to form a unique identity and deviate from the phallocentric social norms, though it often comes at the cost of social alienation. Background. Vinyl chloride (VC) is an abundant environmental contaminant that causes steatohepatitis at high exposure levels. We have shown previously that low concentrations of VC exacerbate high fat diet (HFD)-induced liver injury in mice. The mechanisms involved in the progression of liver injury caused by VC and HFD include oxidative stress, inflammation, metabolic and mitochondrial dysfunction. Mitochondrial aldehyde dehydrogenase 2 (ALDH2) serves as a key line of defense against most reactive aldehydes, including lipid aldehydes (e.g., 4-HNE) and chloroacetaldehyde (VC metabolite). There are thirteen known endogenous EGF-like ligands. We previously reported that Betacellulin (BTC) increases ligand-mediated corneal wound healing more than Epidermal Growth Factor (EGF) [Peterson et al. (2014) IOVS 55(5):2870-80], although the molecular reason for this is unknown. Despite being better at promoting wound healing via enhanced cell migration, BTC has reduced receptor affinity and weaker induction of EGFR phosphorylation. These data indicate that BTC’s response is not due to enhanced affinity or EGFR-kinase activity. Receptor phosphorylation and proximity ligation assays indicate that BTC treatment significantly increases ErbB3 phosphorylation and EGFR:ErbB3 heterodimers. BTC traffics EGFR at a faster rate than …

Hexavalent chromium [Cr(VI)] is a widespread environmental and occupational carcinogen. The mechanism of carcinogenesis remains poorly understood, but chromosome instability (CIN) is the dominant theory. How numerical CIN arises is unclear, but it correlates with centrosome amplification. Both phenotypes are hallmarks of cancers, early events in carcinogenesis, and have been shown to occur after Cr(VI) exposure to human skin and lung fibroblasts. In this study, we investigate numerical CIN and centrosome amplification in whale cells. Whales are our closest marine relatives, have long lifespans, and are exposed to environmental Cr(VI). Achieving state wildlife agency biological goals for deer (Odocoileus spp.) management may often conflict with hunter desires. Concomitantly, better information is needed to optimize agency deer herd management goals with hunters’ social goals. In 2016, we surveyed 3,000 North Dakota, USA, resident deer hunters using a self-administered mail survey to gain a better understanding of motivations, satisfaction, and hunter demographics that may be used to inform hunter recruitment and retention (HRR) efforts during a period of reduced statewide deer populations. An increase in the property tax rate of a school district creates an increase in local revenues for the district (Missouri Department of Elementary and Secondary Education. [MODESE], 2017). African Wild Dogs (Lycaon pictus) are an endangered species of canid from Sub-Saharan Africa. They are very social communal hunters, and are capable of chasing down prey for long stretches of time. Wild dogs benefit a savannah ecosystem by regulating the populations of their prey so that it does not become unhealthy and overgrown. Like many organisms, wild dogs are also in competition with other predators for resources, namely lions and hyenas, who are capable of stealing their kills and occasionally injuring and killing them. The use of genetics in recent years has brought to light the need to reevaluate the classification of many gorgonian octocorals. In 2017, Poliseno et al. conducted a comprehensive, phylogenetic study of the genus Leptogorgia and found that it, too, was in need of taxonomic revision. To expand on their study, 25 samples representing two Leptogorgia species—L virgulata and L. hebes —from the northwestern Gulf of Mexico (GOM) were examined. Ten novel, complete mitochondrial genomes were sequenced using Next Generation sequencing and 24 novel mtMutS sequences were obtained from both species. In amyotrophic lateral sclerosis (ALS), mitochondrial dysfunction and oxidative stress form a vicious cycle that promotes neurodegeneration and muscle wasting. To quantify the disease-stage-dependent changes of mitochondrial function and their relationship to the generation of reactive oxygen species (ROS), we generated double transgenic mice (G93A/cpYFP) that carry human ALS mutation SOD1G93A and mt-cpYFP transgenes, in which mt-cpYFP detects dynamic changes of ROS-related mitoflash events at individual mitochondria level. Irisin, a muscle-origin protein derived from the extracellular domain of the fibronectin domain-containing 5 protein (FNDC5), has been shown to modulate mitochondria welfare through paracrine action. Here, we test the hypothesis that irisin contributes to cardioprotection after myocardial infarction by preserving mitochondrial function in cardiomyocytes. Animal model studies show that intravenous administration of exogenous irisin produces dose-dependent protection against ischemia/reperfusion (I/R)-induced injury to the heart as reflected by the improvement of left ventricular ejection fraction and the reduction in serum level of cTnI (n = 15, P < 0.05). I/R-induced apoptosis of cardiomyocytes is reduced after irisin treatment. The irisin-mediated protection has, at least in part, an effect on mitochondrial function because administration of irisin increases irisin staining in the mitochondria of the infarct area. Irisin also reduces I/R-induced oxidative stress as determined by mitochondrial membrane potential evaluation and superoxide FLASH event recording (n = 4, P < 0.05). The interaction between irisin and superoxide dismutase2 (SOD2) plays a key role in the protective process because irisin treatment increases SOD activity (n = 10, P < 0.05) and restores the mitochondria localization of SOD2 in cardiomyocytes (n = 5, P < 0.05). These results demonstrate that irisin plays a protective role against I/R injury to the heart. Targeting the action of irisin in mitochondria presents a novel therapeutic intervention for myocardial infarction.

Invasive species represent a global threat to ecosystems, human health, and the economy. A basic knowledge of invasive species biology is crucial to understand current and future impacts and implications. The purpose of this book is to provide a broad background on invasive species, and also details on specific examples through case studies.
The students in the course Aquatic Invasive Species (MAR 442) at the University of New England in Biddeford, Maine, have researched and reviewed scientific literature to educate readers about these issues. Diversity And Inclusion As Essential Elements Of 4-H Youth Development Programs, Rachel Sumner, Andy Turner, Anthony Burrow

Diversity and inclusion are essential elements of 4-H’s goals related to positive youth development, workforce development, and organizational sustainability. Previous research has examined Cooperative Extension professionals’ attitudes on this topic, demonstrating that most of them recognize the importance of diversity and inclusion in 4-H. Here we present results from a survey of adolescents participating in New York state 4-H programs assessing their endorsement of racial/ethnic diversity and inclusion in 4-H. Results revealed that, overall, youth felt positively toward this kind of diversity and inclusion in 4-H, and endorsement was most strongly predicted by a learning orientation that emphasizes acquiring new …

The emergence and spread of antimicrobial resistance is a natural evolutionary reaction to antimicrobial exposure. However, the misuse and overuse of antimicrobial drugs in human medicine and in agriculture are speeding up the process. Antimicrobials have been used in food-producing animals for therapeutic purposes as well as to promote growth by applying low concentrations in animal feed. Antimicrobial resistant pathogens can enter the food chain through food containing residues of antibiotic-resistant bacteria and cause infections in humans. Organic products that have historically been viewed as waste products may improve soil health by adding carbon (C) and nutrients to soil. Two such products are woodchips, generated from forest or rangeland management activities, and livestock manure. In Nebraska, eastern redcedar (Juniperus virginiana L.) is a native but invasive tree species inhibiting rangeland productivity. Livestock manure that is underutilized while inorganic fertilizers are imported for crop production presents a water quality risk by contributing to local- and regional-scale nutrient imbalances. In this paper, we explore the reasons why Hilbert’s axiomatic program to unify gravitation theory and electromagnetism failed and outline a plausible resolution of this problem. The latter is based on Gödel’s incompleteness theorem and Newton’s aether stream model.

Invasive plants can alter pollination dynamics in invaded communities by disrupting patterns of pollinator visitation, pollen transfer dynamics (conspecific [CP] and heterospecific [HP]), and reproductive success. The direction of invasive effects (competitive, neutral, and facilitative) may be partially determined by spatial scale and species’ floral traits. Here, we investigated pollinator visitation, CP and HP receipt, and pollen tube growth for species in a C. arvense present community and non-present community at two scales. At the community-level, the effect of C. arvense on pollinator visitation varied among species. Floral symmetry seemed to explain this variation. At the floral neighborhood-level, we found …
