There are many segmentation algorithms in the computer vision field whose general purpose is to separate between object and background in an image. In this work, we take advantage of extra knowledge prior to run-time to speed up the segmentation. Specifically, in the rigid case, we have a three dimensional model of the entire object, and we use the Bhattacharyya coefficient from information theory to segment an image containing a two dimensional projection of that object as well as determine its location relative to the camera. The outer layer of the cell wall of Candida albicans contains glycoproteins made up of mannose-rich glycans that are critical for host-fungal interactions, including adhesion, virulence, and drug resistance. Asparagine (N)-linked glycans are added to a core oligosaccharide by mannosyltransferases in the Golgi to create the long outer chain α1,6-linked mannose “backbone” that branches with α1,2-, α1,3-, and β1,2- linked mannoses. In Saccharomyces cerevisiae, the mannosyltransferases responsible for these modifications have been well studied and characterized. Orthologs of these mannosyltransferases have been identified in C.albicans and in this study we examine VAN1, a mannosyltransferase that acts very early in elongation …

Carbon-carbon and carbon-heteroatom bond-forming reactions lie at the heart of synthetic organic chemistry. They allow construction of complex molecular frameworks and introduction of new substituents. Despite the tremendous progress in the area over the past decades, there has been a growing demand for development of novel methods to construct C–C and C–X bonds under mild and eco-compatible reaction conditions. In order to form a new C–C bond without using stoichiometric amounts of toxic and unstable reagents, we developed a photocatalytic protocol for the coupling between alkenylpyridines and carbonyl derivatives with a Lewis acid co-catalyst. Sperm acrosomal exocytosis plays an important role in mammalian fertilization. This exocytosis process can be triggered by a subset of terminal carbohydrate residues on the zona pellucida around the mouse egg. Although acrosomal exocytosis (AE) has been successfully induced in vitro using purified mouse zona pellucida protein, the mechanism of AE is unclear. Previous studies in the Sampson laboratory used a series of glycopolymers that mimic the sugars displayed by the zona pellucida and applied these polymers in an in vitro acrosome assay. This dissertation mainly investigates the estimation of dynamic factor model in the Bayesian framework, using the techniques of particle filter with online parameter learning such as marginalized particle filter and particle learning. Compatibility challenges occur when sharing an application across system interfaces such as Linux and Windows APIs, or reusing an application on a disruptive hardware such as Intel SGX. Existing approaches require either exhaustively porting applications to new APIs or preserving all previous APIs for backward compatibility. Since both approaches are time-consuming, developers urgently need a solution to the compatibility issues on innovative OSes or hardware, to promptly benefit average users. Population genetics is a model based science with three complementary approaches: theoretical, empirical, and experimental. The last of these employs complex experimental designs to directly estimate model parameters or, when possible, tests causal hypotheses through direct manipulation. Building on the long tradition of experimental population genetics in Drosophila species I propose new mechanisms in evolution of chromosomal rearrangements (Chapters 1-3) and then describe a series of experiments directly testing the role of metabolic genes in temperate adaptation of Drosophila melanogaster (Chapters 4-6). In phylogenetic series of gene arrangements, younger chromosomal inversions tend to be distal to older inversions. After Chapter …

According to Joseph North’s Literary Criticism: A Concise Political History, the titular discipline is today the site of a hegemonic historicist/contextualist practice. It ascended to this hegemony by edging out a different literary practice, what I call textualism, first thought through by the New Critics but also yoked by them to religious themes. North argues that since historicism/contextualism transforms literary criticism into a site of knowledge production and the commodification of literature as part of the “neoliberalization” of the university, the time has come to return to textualist practice so as to combat this trend, if only at a discursive …

My dissertation examines diverse perspectives that literary authors, popular science writers, and academics bring to bear on the question of who can contribute to public science discourse. That discourse has a vexed history; it inherits assumptions of incompatibility from debates about a “Two Cultures” divide, and has been the battleground of the “Science Wars” between scientists and cultural theorists. I move beyond discord and valuation, in favor of a “participatory model,” where both scientists and nonscientists promote the scientific literacy of the lay public. This thesis includes some newly-proposed adaptive search algorithms for simulation optimization in discrete and continuous domains. We first consider the ranking and selection problem of allocating a given simulation budget among a set of design alternatives in order to maximize the probability of correct selection. Previous research used to take static approaches to allocate certain number of simulation budget on different designs, i.e. pre-calculating the allocation plan before simulation, and among them, sequential optimal computing budget allocation (OCBA) shows the best allocation efficiency so far. This thesis examines the diet of loggerhead sea turtles (Caretta caretta) in New York between 1995 – 2014 using stomach contents analysis. Loggerheads are one of four sea turtle species that occur in the temperate waters of New York during summer months. Estuaries of Long Island provide foraging habitat for juvenile loggerheads from June to November. In order to quantify loggerhead diet, I examined individual and inter-annual variation in the stomach contents of 123 individual turtles that stranded along Long Island. The eastern oyster, Crassostrea virginica, is an economically- and ecologically-valuable species. Both commercial fisheries and ecological services are impacted by disease-associated mortality. Perkinsosis, caused by the protozoan parasite Perkinsus marinus, is one of the major diseases that cause mass mortalities. This dissertation aimed to investigate the role of oyster blood cells (hemocytes) in P. marinus infection. Hemocytes associated with mucosal surfaces lining the pallial (shell) cavity were characterized through functional assays and cell surface epitope labeling in conjunction with flow cytometry. Bi-directional transit of hemocytes across mucosal pallial epithelia was also evaluated using flow cytometry and fluorescence microscopy. In-vitro studies …

Ice particles are ubiquitous in the atmosphere, either coexisting with supercooled water droplets in mixed-phase clouds or existing as the sole inhabitant of cirrus clouds. Aerosol particles act as heterogeneous ice nucleating particles (INPs) and influence ice crystal formation, affecting both cloud radiative properties and climate. Heterogeneous ice nucleation is not a well understood process and is cause for great uncertainty in climate models. Organic aerosol is ubiquitous in the atmosphere. Catalytic performance of supported metal NPs, including activity, selectivity, and stability, is influenced by many different factors. These factors include properties of NPs and support materials. In the first part of this project, supported Rh nanocatalysts were produced by a novel doping-segregation method. Different ex-situ and in-situ characterization techniques were utilized to systematically investigate the doping-segregation process in order to gain fundamental knowledge on this phenomenon. Driven by a strong desire for clean and renewable fuel, effective methods of hydrogen production have been sought after for a long time. Solar water splitting within a photoelectrochemical (PEC) cell has been one of the most promising methods. The key component of a PEC cell, the semiconductor photoelectrode, is responsible for carrier generation through light absorption, which initiates redox reactions in water to produce hydrogen and oxygen. The development of sequencing since the Human Genome Project has greatly advanced. Current Next Generation Sequencing (NGS) platforms can produce more cost-effective sequencing in a shorter amount of time by using massively parallel sequencers. However, current NGS platforms have limitations in that only DNA fragments of size 100-300 bases may be analyzed. When producing these fragments of DNA, the positional order is often lost due to random fragmentation. Additionally, with repeats, insertions, and deletions of the genome, it further complicates the construction of contigs. The genesis of Food from Stein was a commission from an ensemble in Vancouver, British Columbia. While that concert ultimately did not take place, it nonetheless dictated the instrumentation of the piece: flute, viola, harp and voice. This genesis also informed the structure of the piece. The commission required that the work reflect the concept of biodiversity (the performance was to occur in the Beatty Biodiversity Museum at the University of British Columbia). This thesis takes as its central object of analysis The Birth of a New World – A Cooperative Tribute, an illustration spread featured in the November 1926 issue of New Masses, a notable leftist magazine in the United States during the interwar period of the 1920’s and 1930’s. As a collaborative piece by the artists Hugo Gellert, William Gropper, I. Klein, Louis Lozowick and William Siegel, this illustration demonstrates how a model for artistic collaboration and a collective vision for an alternative society emerged during this period by drawing inspirations from political and artistic revolutions elsewhere. This dissertation explores the trajectory of the Chilean State’s statistical projects from the mid-eighteenth century to the beginning of the twentieth century. It begins in a moment when the scientific concerns of the Enlightenment dovetailed with the State’s demand for knowledge after the dissolution of the Spanish Empire. It analyzes the construction of a specialized bureaucracy entrusted with the mission of producing the statistical information the Chilean State required for effective government. Big Picture is a contemporary Broadway-style musical comedy with a story adapted from Claudio Monteverdi’s 1643 opera L’incoronazione di Poppea (“The Coronation of Poppea”). Monteverdi’s opera is a story of lust, power, and corruption, set in the imperial Roman Empire. The opera follows the love affair of Emperor Nero and his mistress Poppea as they conspire to banish Nero’s wife, the empress Ottavia, and crown Poppea queen. “Please See” is a work of literary fiction and is partially based on a cold case from 2007, when the bodies of four prostitutes were found in a drainage easement in the salt marsh behind an Atlantic City motel. This thesis examines recent theories of sound art and ecoacoustics, with an emphasis on object-oriented ontologies and new materialism. Following Timothy Morton’s suggestion that, toward the end of the Anthropocene, subjects are implicated in object systems that are non-local, temporally protracted, and viscous, it explores the deployment of sonic resonances to structure an encounter with the nonhuman cosmos. In keeping with the critique of Western ocularcentrism, which prioritizes vision over other sense modalities in epistemic claims, it argues that a materialist rhythmicity, or a manipulation of exhibitionary or auditory limits, introduces radical materialist and structural alterity to the spectatorial subject. …

The ideals of universal design have profoundly impacted instruction, policy, and infrastructure in course architecture and design within elementary education and at some universities. Within international and global studies, however, these principles have not deeply affected either pedagogy or scholarship despite the fact that classes in international studies may include more international students and third culture kids than classes in other programs.
