Road Dust Correlated With Decreased Reproduction Of The Endangered Utah Shrub Hesperidanthus Suffrutescens, Matthew B. Lewis, Eugene W. Schupp, Thomas A. Monaco Dec 2017

Road Dust Correlated With Decreased Reproduction Of The Endangered Utah Shrub Hesperidanthus Suffrutescens, Matthew B. Lewis, Eugene W. Schupp, Thomas A. Monaco

Western North American Naturalist

Roads associated with energy development have fragmented much of the Uinta Basin, the Colorado Plateau in general, and other areas of western North America. Beyond reducing available habitat, spreading exotic species, and creating barriers to dispersal, unpaved roads also increase dust loads on plants and pollinators, which may reduce plant growth and reproduction. We studied the effects of an unpaved road on reproduction of an endangered Utah endemic shrub. We measured the size and reproductive output of 156 plants and the dust deposition in plots at increasing distances from the road. We also hand outcrossed 240 flowers from 80 plants …

Traditional Ecological Knowledge In Collaborative Forest Restoration In The Sierra Nevada, Michael Boero Dec 2017

Traditional Ecological Knowledge In Collaborative Forest Restoration In The Sierra Nevada, Michael Boero

Master's Theses

Forest restoration in the Sierra Nevada is receiving increased attention due to climate change, increasing populations in the wildland-urban interface, and the historical mismanagement of some forests, evidenced in part by the increasing frequency and severity of wildfires in recent years. In response to an urgency to implement restoration in the absence of consensus on the best ways to proceed, public land managers practice collaborative management approaches to ameliorate antagonism between stakeholders and allow restoration projects to proceed when agreement is difficult to achieve. Stakeholders often include representatives from local community groups, environmental organizations, private industry, and—notably—tribal groups, who hold …

The purpose of this work is to develop an experimental setup with associated mathematical analysis to analyze the changes in diffusivity through calcium alginate gel beads made under different polymerization conditions. The diffusivity of the solutes glucose and tryptophan was calculated by measuring the concentration change over time as solute diffused out of the alginate spheres and into the bulk. The alginate beads were generated using different polymerization conditions following a two-level, three-factor, full-factorial matrix with alginate concentration, cross-linker concentration, and cross-linking time as the factors. The diffusivities showed variations between different conditions, which were analyzed to determine the main …

Developmental Effects Of Nicotine Exposure In Drosophila Melanogaster, Lisa Janet Wong Dec 2017

Developmental Effects Of Nicotine Exposure In Drosophila Melanogaster, Lisa Janet Wong

Master's Theses

Approximately 12%-20% of pregnant women smoke at some point during pregnancy, and 10% of pregnant women are reported to have smoked during the last 3 months of pregnancy. Smoking during pregnancy leads to developmental health risks for the fetus and child, including increased mortality, low birth weight, and developmental delays. The direct molecular targets of nicotine are nicotinic acetylcholine receptors (nAChRs) due to the similarities in structure between nicotine and acetylcholine. However, in many cases, it remains unclear what molecular events downstream of nAChRs lead to the deleterious effects of nicotine on development. We have established Drosophila melanogaster as a …

Metabolic Reprogramming Of Pancreatic Ductal Adenocarcinoma Cells In Response To Chronic Low Ph Stress, Jaime Abrego Dec 2017

Metabolic Reprogramming Of Pancreatic Ductal Adenocarcinoma Cells In Response To Chronic Low Ph Stress, Jaime Abrego

Theses & Dissertations

Pancreatic ductal adenocarcinoma (PDAC) is among the most lethal of all cancers with a 5-year survival rate of only 8.2%. This is because PDAC is diagnosed in its advanced stages and is characterized by radio and chemotherapy resistance. Aggressiveness of PDAC tumors is attributed to its high metabolic phenotype, which is characterized by increased glycolysis rate and lactate secretion, while oxidative metabolism is reduced. These metabolic features are required to fulfill the biosynthetic demands of proliferating PDAC cells. However, this increase in metabolic activity results in acidification of the extracellular space because the dense fibrotic stroma of PDAC tumors limits …

Focus: A Graph Approach For Data-Mining And Domain-Specific Assembly Of Next Generation Sequencing Data, Julia Sommer Dec 2017

Focus: A Graph Approach For Data-Mining And Domain-Specific Assembly Of Next Generation Sequencing Data, Julia Sommer

Theses & Dissertations

Next Generation Sequencing (NGS) has emerged as a key technology leading to revolutionary breakthroughs in numerous biomedical research areas. These technologies produce millions to billions of short DNA reads that represent a small fraction of the original target DNA sequence. These short reads contain little information individually but are produced at a high coverage of the original sequence such that many reads overlap. Overlap relationships allow for the reads to be linearly ordered and merged by computational programs called assemblers into long stretches of contiguous sequence called contigs that can be used for research applications. Although the assembly of the …

Self-Efficacy For Symptom Management In Adult Hematopoietic Stem Cell Transplant Patients, Lynn White

Theses & Dissertations

Hematopoietic stem cell transplant (HSCT) is a treatment for hematologic cancers and other hematologic conditions that causes severe treatment-related symptoms. The first 30 days after HSCT, or the acute phase, is when symptoms are most intense. During this time, the ability of patients to manage their symptoms, in collaboration with their health care providers, is crucial to reduce the distress caused by the symptoms. Self-efficacy is the person’s confidence in their ability to perform a behavior, such as symptom management. This body of work describes the concept of self-efficacy for symptom management (SESM), presents an integrated literature review on self-efficacy …

Novel Therapeutic Strategies For Treatment Of Castration-Resistant Prostate Cancer, Matthew A. Ingersoll Dec 2017

Novel Therapeutic Strategies For Treatment Of Castration-Resistant Prostate Cancer, Matthew A. Ingersoll

Theses & Dissertations

Prostate cancer (PCa) remains the most commonly diagnosed solid tumor and is the third leading cause of cancer-related death in United States men. While androgen deprivation therapy is the current standard-of-care treatment for metastatic PCa, most patients eventually relapse and develop castration-resistant (CR) tumors, for which there is currently no effective treatment. Therefore, synthesis of novel therapeutic agents and identification of alternative target proteins are necessary to improve treatment. Herein, I investigate the efficacy of novel imidazopyridine and statin derivatives as alternative therapeutic compounds. These molecules not only inhibit androgen receptor signaling, but also block activation of the AKT axis, …

Comparison Of The Bi-Directional Performance Of Micro-Channel Sieve And Thin-Film Time Peripheral Nerve Interfaces, Robert Coker Dec 2017

Comparison Of The Bi-Directional Performance Of Micro-Channel Sieve And Thin-Film Time Peripheral Nerve Interfaces, Robert Coker

McKelvey School of Engineering Graduate Student Theses & Dissertations

Sophisticated motorized prosthetic limbs contain multiple degrees of freedom of motion as well as embedded pressure and angle transducers to provide sensory feedback in amputees. Although several central neural recording and stimulation modalities exist for both controlling these motions and providing sensory feedback from a prosthetic limb, directly interfacing the peripheral nerves which originally innervated the limb has many advantages. A difficulty with this bi-directional approach is that electrically stimulating axons to provide haptic feedback creates stimulation artifacts at neighboring recording sites within the nerve that are several orders of magnitude larger than the electroneurogram used for control. In this …

Coupling Of Oxidation-Reduction Reactions Of Chromium, Iron And Manganese: Implications For The Fate And Mobility Of Chromium In Aquatic Environments, Chao Pan Dec 2017

Coupling Of Oxidation-Reduction Reactions Of Chromium, Iron And Manganese: Implications For The Fate And Mobility Of Chromium In Aquatic Environments, Chao Pan

McKelvey School of Engineering Graduate Student Theses & Dissertations

Both within the United States and internationally, hexavalent chromium (Cr(VI)) is a contaminant of concern in drinking water supplies. The U.S. Environmental Protection Agency is considering a Cr(VI)-specific standard. Thus improved technologies for Cr(VI) removal in drinking water are needed. Iron electrocoagulation for Cr(VI) removal was examined at conditions directly relevant to drinking water treatment, and humic acid (HA) affects the performance of electrocoagulation in multiple ways. The success of the chromium treatment or remediation also relies on the stability of the Cr(III)-containing solids with respect to reoxidation under groundwater conditions. Manganese is ubiquitous in aquatic and terrestrial environments, and …

Analysis Of Argonaute-Small Rna-Transcription Factor Circuits Controlling Leaf Development, John Steen Hoyer Dec 2017

Analysis Of Argonaute-Small Rna-Transcription Factor Circuits Controlling Leaf Development, John Steen Hoyer

Arts & Sciences Graduate Student Theses and Dissertations

Experimental studies of plant development have yielded many insights into gene regulation, revealing interactions between core transcriptional and post-transcriptional regulatory pathways present in all land plants. This work describes a direct connection between the three main small RNA-transcription factor circuits controlling leaf shape dynamics in the reference plant Arabidopsis thaliana. We used a high-throughput yeast 1-hybrid platform to identify factors directly binding the promoter of the highly specialized ARGONAUTE7 silencing factor. Two groups of developmentally significant microRNA-targeted transcription factors were the clearest hits from these screens, but transgenic complementation analysis indicated that their binding sites make only a small contribution …

A Generalized Biophysical Model Of Transcription Factor Binding Specificity And Its Application On High-Throughput Selex Data, Shuxiang Ruan

Arts & Sciences Graduate Student Theses and Dissertations

The interaction between transcription factors (TFs) and DNA plays an important role in gene expression regulation. In the past, experiments on protein–DNA interactions could only identify a handful of sequences that a TF binds with high affinities. In recent years, several high-throughput experimental techniques, such as high-throughput SELEX (HT-SELEX), protein-binding microarrays (PBMs) and ChIP-seq, have been developed to estimate the relative binding affinities of large numbers of DNA sequences both in vitro and in vivo. The large volume of data generated by these techniques proved to be a challenge and prompted the development of novel motif discovery algorithms. These algorithms …

Regulation Of Gene Expression By Rna Binding Proteins And Micrornas, Kyle Cottrell Dec 2017

Regulation Of Gene Expression By Rna Binding Proteins And Micrornas, Kyle Cottrell

Arts & Sciences Graduate Student Theses and Dissertations

Regulation of gene expression is essential to life. Post-transcriptional regulation of gene expression is a complex process with many inputs that lead to changes in localization, translation and stability of mRNAs. The translation and stability of many mRNAs is regulated by cis-elements, such as mRNA-structure or codon optimality; and by trans-acting factors such as RBPs and miRNAs. Here I report on the complex interactions between RBPs, miRNAs and characteristics of their target mRNAs in respect to effects on translation and RNA stability.Using a reporter based approach we studied modulation of microRNA-mediated repression by various mRNA characteristics. We observed the influence …

The Regulation Of Extracellular Amyloid-Β Levels By Ionotropic Glutamatergic Transmission In An Alzheimer’S Disease Mouse Model, Jane Cecelia Hettinger Dec 2017

The Regulation Of Extracellular Amyloid-Β Levels By Ionotropic Glutamatergic Transmission In An Alzheimer’S Disease Mouse Model, Jane Cecelia Hettinger

Arts & Sciences Graduate Student Theses and Dissertations

Brain extracellular concentration of the peptide amyloid-β (Aβ) is a major contributor to Alzheimer’s disease (AD) pathogenesis. High Aβ levels in the extracellular space precipitate aggregation of the peptide into soluble and insoluble toxic species. This process begins decades before cognitive impairment and triggers the cascade of pathology that eventually leads to AD. Synaptic activity is key to the regulation of extracellular Aβ levels. Presynaptic activity drives the production of Aβ, while postsynaptic receptor activation exhibits more nuanced regulation. For example, high levels of NMDA receptor (NMDA-R) activation have been shown to decrease Aβ production through the extracellular signal-regulated kinase …

Longitudinal Cerebrospinal Fluid Biomarkers Of Alzheimer Disease: Movement Toward The Diagnosis, Prognosis And Staging Of Disease, Courtney Sutphen

Arts & Sciences Graduate Student Theses and Dissertations

Alzheimer’s disease (AD) is a devastating neurodegenerative disease that slowly claims the memories and experiences that comprise the life experiences of individuals that suffer from the disease. Despite a continually accelerating pace of research and discovery, a viable therapeutic intervention for AD has yet to be realized. There are a multitude of factors that may contribute to this difficulty including the challenge of separating the overall disease of Alzheimer’s from the clinically recognizable memory loss that occurs in what is now known to be the end-stage of the disease. Efforts to treat AD have increasingly turned toward very early disease …

Roles Of Peroxisomes And Peroxisome-Derived Products In Controlling Plant Growth And Stress Responses, Elizabeth May Frick Dec 2017

Roles Of Peroxisomes And Peroxisome-Derived Products In Controlling Plant Growth And Stress Responses, Elizabeth May Frick

Arts & Sciences Graduate Student Theses and Dissertations

The peroxisome is a vital organelle conserved through the entire eukaryotic lineage. In all examined species, peroxisomes are responsible for such essential processes as fatty acid beta-oxidation and metabolism of reactive oxygen species (ROS). In plants, peroxisomes have taken on additional specialized roles, such as production of some plant hormones and vitamins. In this work, I have uncovered novel factors regulating peroxisome number in model species Arabidopsis thaliana, and novel mechanisms governing how peroxisomes respond to salt stress. I discovered a role for Arabidopsis MAP KINASE17 (MPK17) as a negative regulator of peroxisome division that acts in the salt-stress response …
