Naturally-occurring antibiotic resistance genes in soil bacteria represent a potentially important reservoir of genes that could contribute to antibiotic resistance of human pathogens. It has been reported that over 40 genes in bacterial genomes are controlled by concentrations of ferric iron. We examined the effect of soil metal content on the level of resistance to two antibiotics, ampicillin (Amp) and tetracycline (Tet), and the presence of multiple genes that code for efflux pump-mediated resistance. These pumps act to export toxins (e.g. heavy metals and antibiotics, perhaps). Because of this, growth in heavy metal-contaminated soils might select for antibiotic resistance. Ninety-six …

Families experiencing out-of-home placement (OHP) are in crisis. The fact that a child has been removed from the home means the family has become involved with the child welfare or juvenile justice system. Families that experience OHP are disproportionately poor, and the reasons for OHP often stem from poverty. Because OHP is expensive, and society values parental responsibility, federal and state laws require that parents be referred to the child support system to help offset the cost of OHP through child support collections. Despite HLA allele matching, significant acute GvHD remains a major barrier to successful unrelated donor BMT. We conducted a genome-wide association study (GWAS) to identify recipient and donor genes associated with the risk of acute GvHD. A case-control design (grade III-IV versus no acute GvHD) and pooled GWA approach was used to study European-American recipients with hematological malignancies who received myeloablative conditioning non-T-cell-depleted first transplantation from HLA-A, -B, -C, -DRB1, -DQB1 allele level (10/10) matched unrelated donors. Biological molecules and biologically active small molecules are complex three dimensional structures. Current flat screen monitors are limited in their ability to convey the full three dimensional characteristics of these molecules. Augmented reality devices, including the Microsoft HoloLens, offer an immersive platform to change how we interact with molecular visualizations. We describe a process to incorporate the three dimensional structures of small molecules and complex proteins into the Microsoft HoloLens using aspirin and the human leukocyte antigen (HLA) as examples. Watson and Crick’s discovery of the structure of DNA (deoxyribonucleic acid) in 1953 launched a truth-finding mission not only in science but also in the law. Just thirty years later–after the science had evolved–DNA evidence was being introduced in criminal courts. Today, DNA evidence is heavily relied on in criminal and related cases. It is routinely introduced in murder and rape cases as evidence of guilt; DNA databases have grown as even arrestees have been required to surrender DNA samples; and this evidence has been used to exonerate hundreds of convicted individuals. Background: Appendectomy is considered the gold standard treatment for acute appendicitis. Recently the need for surgery has been challenged in both adults and children. In children there is growing clinician, patient and parental interest in non-operative treatment of acute appendicitis with antibiotics as opposed to surgery. To date no multicentre randomised controlled trials that are appropriately powered to determine efficacy of non-operative treatment (antibiotics) for acute appendicitis in children compared with surgery (appendectomy) have been performed.

The Hippo pathway is an important signaling pathway that controls cell proliferation and apoptosis. It is evolutionarily conserved in mammals and is stimulated by cell-cell contact, inhibiting cell proliferation in response to increased cell density. During early embryonic development, the Hippo signaling pathway regulates organ development and size, and its functions result in the coordinated balance between proliferation, apoptosis, and differentiation. Its principal effectors, YAP and TAZ, regulate signaling by the embryonic stem cells and determine cell fate and histogenesis. A plasma component is responsible for altered glomerular permeability in patients with focal segmental glomerulosclerosis. Evidence includes recurrence after renal transplantation, remission after plasmapheresis, proteinuria in infants of affected mothers, transfer of proteinuria to experimental animals, and impaired glomerular permeability after exposure to patient plasma. Therapy may include decreasing synthesis of the injurious agent, removing or blocking its interaction with cells, or blocking signaling or enhancing cell defenses to restore the permeability barrier and prevent progression. Agents that may prevent the synthesis of the permeability factor include cytotoxic agents or aggressive chemotherapy. Poly (ADP-Ribose) Polymerase I (PARP-1) is a first responder to DNA damage and participates in the regulation of gene expression. The interaction of PARP-1 with chromatin and DNA is complex and involves at least two different modes of interaction. In its enzymatically inactive state, PARP-1 binds native chromatin with similar affinity as it binds free DNA ends. Automodification of PARP-1 affects interaction with chromatin and DNA to different extents. Here we describe a series of biochemical and biophysical techniques to quantify and dissect the different binding modes of PARP-1 with its various substrates. BACKGROUND/AIMS: Youth with type 2 diabetes (T2D) have poor compliance with medical care. This study aimed to determine which demographic and clinical factors differ between youth with T2D who receive care in a pediatric diabetes center versus youth lost to follow-up for >18 months.

METHODS: Data were analyzed from 496 subjects in the Pe-diatric Diabetes Consortium registry. Enrollment variables were selected a priori and analyzed with univariable and multivariable logistic regression models.

RESULTS: After a median of 1.3 years from enrollment, 55% of patients were lost to follow-up. The final model included age, race/ethnicity, parent education, and estimated distance to …

Many theories are used to conceptualize adolescent substance use, yet none adequately assist mental health professionals in assessing adolescents’ strengths and risk factors while incorporating cultural factors. The authors reviewed common adolescent substance abuse theories and their strengths and limitations, and offer a new model to conceptualize adolescent substance use: The Adolescent Substance Use Risk Continuum. We posit that this strengths-based continuum enables clinicians to decrease stigma and offer hope to adolescents and their caregivers, as it integrates relevant factors to strengthen families and minimize risk. Activated PI3K-δ syndrome refers to a recently described primary immunodeficiency syndrome consisting of recurrent sinopulmonary infections, lymphadenopathy, mucosal lymphoid aggregates, increased susceptibility to Epstein-Barr virus and cytomegalovirus, and increased incidence of B-cell lymphomas. Variants in PIK3CD, which encodes the phosphatidylinositol-4,5-bisphosphate 3-kinase catalytic subunit delta isoform, enhance membrane association and kinase activity, resulting in increased signal transduction through the PI3K-Akt pathway. Whole-exome sequencing revealed a pathogenic PIK3CD variant in a patient with history of immunologic impairment, recurrent sinopulmonary infections, and lymphoid hyperplasia presenting as intussusception. From 1924 to 1932, Louisiana lawmakers considered five bills that would have granted superintendents of state institutions and some private hospitals the authority to forcibly sterilize their patients. Based on similar legislation passed in thirty-six other states, the bills cited eugenics as evidence that stripping these patients of their ability to reproduce would prevent the conditions such as feeblemindedness from passing on to the next generation. Although none of the bills passed both houses of the Louisiana legislature, a couple of them came dangerously close to becoming law. Ecology and the role of natural selection in lineage diversification has been a central topic in evolutionary biology since Darwin. At the macroevolutionary scale, this idea is embodied in the ecological theory of adaptive radiation, which posits that rapid diversification is driven by ecological adaptive radiation in which speciation is coupled with niche divergence. Within species, the theory of ecological speciation proposes that local adaptation drives speciation by reducing gene flow among populations occupying different environments either by directly reducing migration or by reducing the fitness of migrants. Metabolic inflexibility is an emerging physiological marker indicative of metabolic dysfunction and associated with type 2 diabetes (T2D) and obesity. Exercise is a potent stimulus to improve metabolic health, however, not much is known about the acute effects of exercise on metabolic flexibility (METFLEX). Ectopic lipid accumulation in skeletal muscle is linked with insulin resistance and type 2 diabetes (T2D). Neutral lipid is stored as lipid droplets, which are coated by lipid droplet coat proteins. One important class of these proteins are the perilipins. Little research has focused on perilipin 3 (PLIN3) and coatomer gtp-ases, theorized to be involved in PLIN3 directed lipid oxidation. Under lipolytic stimulation with epinephrine or palmitate, forskolin, and ionomysin (PFI), primary human myotubes taken from health non-T2D donors showed increased protein content levels of PLIN3. Antifreeze proteins (AFPs) are a class of ice binding proteins (IBPs) that are expressed by different cold-adapted organisms to increase their freezing tolerance. AFPs have two major properties: thermal hysteresis and ice recrystallization inhibition. Here we report the functional and structural analyses of a bacterial AFP, IBPv. IBPv was originally secreted by a bacterium recovered from a deep glacial ice core drilled at Vostok Station, Antarctica. Our study showed that the recombinant protein rIBPv exhibited a thermal hysteresis of 2°C at concentrations higher than 50 µM, effectively inhibited ice recrystallization, and enhanced bacterial viability during freeze-thaw cycling. Lignocellulosic syrup from dilute ammonia pretreated energy cane bagasse is a promising fermentable sugar feedstock that can improve the logistics associated with long-distance transportation, long-time storage, and year-round supply of lignocellulosic biomass to processing industries. However, non-sugar compounds such as organic acids, furans and phenolic compounds formed during pretreatment can have negative effects on downstream processes such as fermentation. The research described in this dissertation was aimed at expanding understanding of the diversity and functional roles of microorganisms able to anaerobically transform chlorinated compounds. During the course of the research, a novel strictly anaerobic bacterial isolate originating from chlorinated solvent contaminated groundwater at a Superfund Site in south Louisiana (USA) was characterized. Phylogenetic analysis based on 16S rRNA gene sequencing placed the newly isolated strain, designated as NSZ-14 T, within the genus Dehalogenimonas but within a separate lineage from previously described species. Protein separation by polyacrylamide gel electrophoresis (PAGE) and identification by matrix assisted laser desorption ionization-mass spectrometry (MALDI-MS) are primary tools of protein analysis. In these techniques, surfactants are used in protein sample preparation in order to enhance the protein solubility. Conventional surfactants have shown limitations in protein analysis due to the structural complexity of proteomes, resulting in low resolution. The research goal of this dissertation is to address some of these limitations by applying novel cationic ionic liquid surfactants (ILS), N-alkyl-4-methyl pyridinium bromide (CnPBr where n=4, 8, 11).
