ASEAN Journal on Science and Technology for Development
A strain of Bacillus subtilis FS2 isolated from a traditional fish source in Vietnam has been shown to produce an enzyme possessing both a gelatinase and a lipase A (LipA) activity when tested on agar plates containing 0.1% tributyrin and 0.3% gelatine. A gene encoding LipA was isolated and expressed in E. coli BL21 (DE3). The enzyme has got an apparent Mr of 24 kDa, a pI of 9.2 and was purified to electrophoretical homogeneity by affinity chromatography. Both the lipase and the gelatinase showed optimum activity at pH 10 and at 30 0 C, with highest activity in the …

Msaabcr Operon Is Involved In Persister Cell Formation In Staphylococcus Aureus, Gyan S. Sahukhal, Shanti Pandey, Mohamed O. Elasri

Background
Persister cells comprise a phenotypic variant that shows extreme antibiotic tolerance resulting in treatment failures of bacterial infections. While this phenomenon has posed a great threat in public health, mechanisms underlying their formation in Staphylococcus aureus remain largely unknown. Increasing evidences of the presence of persister cells in recalcitrant infections underscores the great urgency to unravel the mechanism by which these cells develop. Previously, we characterized msaABCR operon that plays roles in regulation of virulence, biofilm development and antibiotic resistance. We also characterized the function of MsaB protein and showed that MsaB is a putative transcription factor that binds …

Mounting evidence suggests that weight management and physical activity (PA) improve overall health and well being, and reduce the risk of morbidity and mortality among cancer survivors. Although many opportunities exist to include weight management and PA in routine cancer care, several barriers remain. This review summarizes key topics addressed in a recent National Academies of Science, Engineering, and Medicine workshop entitled, "Incorporating Weight Management and Physical Activity Throughout the Cancer Care Continuum." Discussions related to body weight and PA among cancer survivors included: 1) current knowledge and gaps related to health outcomes; 2) effective intervention approaches; 3) addressing the …

Evaluating Student Growth In High School Science Education Through The Lens Of Science Identity, Vanessa Tran
University Honors Theses
The Physics First sequence has been known to increase competence and test scores of students, but how is science identity developed through Physics First? Particularly, this study compares the differences between males and females to better understand if competence makes a difference in how science identity develops through Physics First. The Colorado Learning Attitudes about Science Survey (CLASS) is utilized to understand how science attitudes may reflect science identity. In focusing on the self-perceived aspect of science identity, real-world connections and self-efficacy categories in which science identity are evaluated. The study found significant negative shifts in real world connection attitudes …

For The Greater Good: The Subordination Of Reproductive Freedom To State Interests In The United States And China, Marisa S. Cianciarulo
Akron Law Review
This Article provides a comparative analysis of two very different restrictions on reproductive freedom that have startling parallels and similarities. Both China and the United States impose limits on reproductive freedom: China restricts the number of children that families can have, often in ways that violate international law, while some U.S. states have attempted to restrict access to abortion in ways that violate the precepts of Roe v. Wade as well as international law. Both China and U.S. states impose restrictions on reproductive freedom in order to achieve compelling state goals: protecting development and sustainability in China, and protecting prenatal …

A Comparison Of Water Potential, Photosynthetic Rate, Electron Transport Rate, And Stomatal Conductance Between Native Malosma Laurina And Exotic Schinus Molle, Karagan L. Smith, Agatha C. Heng, Viridiana Hernandez-Lopez, Stephen D. Davis
Biology
Schinus molle is a relatively new invasive chaparral shrub in the Santa Monica Mountains and has only recently been observed to displace native shrubs like Malosma laurina. To investigate the probable cause of S. molle’s competitive displacement of M. laurina, we compared their water status, photosynthetic rates, electron transport rates, and stomatal conductance to water vapor diffusion during the fall months of 2016. We hypothesized that S. molle would physiologically outperform M. laurina indicated by higher photosynthetic rates, electron transport rates, stomatal conductance rates, and less negative water potential. We found that there was no significant difference between dry …

Evaluating Mobile-Based Citizen Science In Increasing Citizen Participants In Environmental Management, Wenqi Ou
Community and Regional Planning Program: Theses
Mobile-based citizen science as thriving citizen education tool increases the non-expert citizen involvement in the scientific world by encouraging the public upload nature observation to assist scientific research while learning scientific knowledge. Mobile-base citizen science as social media has potential to facilitate citizen engagement in the environmental management. Based on the conceptual framework of citizen science incorporation with environmental management, large users in the program foster the citizen involvement in the environmental management. Research here applies SWOT (Strength, Weakness, Opportunity, Threats) analysis to evaluate mobile based citizen science regarding data management, citizen participation, and partnership. It revealed the current status …

Over the past twenty years, technological advances have driven the development of media consumption in both home and mobile contexts. While not ubiquitous, multi-channel audio home cinema systems have become more prevalent, as has the consumption of broadcast and gaming media on smartphone and tablet technology via mobile telecommunications networks. This has created new possibilities and poses new challenges for audio content delivery such as how the same content can be presented to greatest effect given that it may be consumed via either a surround-sound home entertainment system or in a mobile context using stereo headphones. This paper outlines research …

Niche Cadherins Control The Quiescence-To-Activation Transition In Muscle Stem Cells., Aviva J. Goel, Marysia-Kolbe Rieder, Hans-Henning Arnold, Glenn L. Radice, Robert S. Krauss
Department of Medicine Faculty Papers
Many adult stem cells display prolonged quiescence, promoted by cues from their niche. Upon tissue damage, a coordinated transition to the activated state is required because non-physiological breaks in quiescence often lead to stem cell depletion and impaired regeneration. Here, we identify cadherin-mediated adhesion and signaling between muscle stem cells (satellite cells [SCs]) and their myofiber niche as a mechanism that orchestrates the quiescence-to-activation transition. Conditional removal of N-cadherin and M-cadherin in mice leads to a break in SC quiescence, with long-term expansion of a regeneration-proficient SC pool. These SCs have an incomplete disruption of the myofiber-SC adhesive junction and …

Cancer immunotherapy has transformed the treatment of cancer. However, increasing use of immune-based therapies, including the widely used class of agents known as immune checkpoint inhibitors, has exposed a discrete group of immune-related adverse events (irAEs). Many of these are driven by the same immunologic mechanisms responsible for the drugs' therapeutic effects, namely blockade of inhibitory mechanisms that suppress the immune system and protect body tissues from an unconstrained acute or chronic immune response. Skin, gut, endocrine, lung and musculoskeletal irAEs are relatively common, whereas cardiovascular, hematologic, renal, neurologic and ophthalmologic irAEs occur much less frequently. The majority of irAEs …

The Toxoplasma genome encodes the capacity for distinct architectures underlying cell cycle progression in a life cycle stage-dependent manner. Replication in intermediate hosts occurs by endodyogeny, whereas a hybrid of schizogony and endopolygeny occurs in the gut of the definitive feline host. Here, we characterize the consequence of the loss of a cell cycle-regulated ovarian tumor (OTU family) deubiquitinase, OTUD3A of Toxoplasma gondii (TgOTUD3A; TGGT1_258780), in T. gondii tachyzoites. Rather than the mutation being detrimental, mutant parasites exhibited a fitness advantage, outcompeting the wild type. This phenotype was due to roughly one-third of TgOTUD3A-knockout (TgOTUD3A-KO) tachyzoites exhibiting deviations from endodyogeny …

Delivering isotopic tracers for metabolic studies in rodents without overt stress is challenging. Current methods achieve low label enrichment in proteins and lipids. Here, we report noninvasive introduction of 13C6-glucose via a stress-free, ad libitum liquid diet. Using NMR and ion chromatography-mass spectrometry, we quantify extensive 13C enrichment in products of glycolysis, the Krebs cycle, the pentose phosphate pathway, nucleobases, UDP-sugars, glycogen, lipids, and proteins in mouse tissues during 12 to 48 h of 13C6-glucose feeding. Applying this approach to patient-derived lung tumor xenografts (PDTX), we show that the liver supplies glucose-derived Gln …

Social Anxiety Disorder (SAD) and Alcohol Use Disorder (AUD) are highly prevalent and frequently co-occur. The results of population studies suggest that SAD tends to precede AUD, and the results of laboratory studies suggest that alcohol use facilitates social behaviors in socially anxious individuals. Therefore, we posited that, in a modern context, a tendency to consume alcohol may be positively selected for among socially anxious individuals by its effect on the likelihood of finding a partner and reproducing. We tested the hypothesis that a higher proportion of individuals with a lifetime diagnosis of SAD and AUD reproduce (i.e., have at …

ASEAN Journal on Science and Technology for Development
This research aimed at studying the property of Euglena Sanguinea Ehrenberg (E. sanguinea) before and after being stimulated by multi-level electromagnetic fields (MLEM). After the stimulation, it was found that physical properties were changed, i.e., cell membrane and components deformed. Main biochemical compositions were changed in accordance with new electrochemical equation which was estimated from FTIR bands. Moreover, the estimated electrical results before and after being stimulated, i.e., impedance and complex relative permittivity of main biochemical compositions from extraction, were then measured, simulated, and compared by means of Cole-Cole plot technique which yielded the statistic correlation 0.89 at 95% confidence …

Metabolic homeostasis is coordinately controlled by diverse inputs. Understanding these regulatory networks is vital to combating metabolic disorders. The nematode Caenorhabditis elegans has emerged as a powerful, genetically tractable model system for the discovery of lipid regulatory mechanisms. Here we introduce DBL-1, the C. elegans homolog of bone morphogenetic protein 2/4 (BMP2/4), as a significant regulator of lipid homeostasis. We used neutral lipid staining and a lipid droplet marker to demonstrate that both increases and decreases in DBL-1/BMP signaling result in reduced lipid stores and lipid droplet count. We find that lipid droplet size, however, correlates positively with the level …

No Evidence For Trace Metal Limitation On Anaerobic Carbon Mineralization In Three Peatland Soils, Jason K. Keller, Jillian Wade
No Evidence For Trace Metal Limitation On Anaerobic Carbon Mineralization In Three Peatland Soils, Jason K. Keller, Jillian Wade
Biology, Chemistry, and Environmental Sciences Faculty Articles and Research
Peatlands store roughly one-third of the terrestrial soil carbon and release the potent greenhouse gas methane (CH4) to the atmosphere, making these wetlands among the most important ecosystems in the global carbon cycle. Despite their importance, the controls of anaerobic decomposition of organic matter to carbon dioxide (CO2) and CH4 within peatlands are not well understood. It is known, however, that the enzymes responsible for CH4 production require cobalt, iron and nickel, and there is a growing appreciation for the potential role of trace metal limitation in anaerobic decomposition. To explore the possibility of …

Bringing Santa Rosa Island Into Channel Islands National Park: The Written Documents 1979–1987, Kate Roney Faulkner
Western North American Naturalist
The National Park Service (NPS) purchased Santa Rosa Island from private landowners Vail & Vickers in 1986. Immediately following the purchase, NPS personnel stated that they and the former landowners were negotiating a 5- to 10-year phaseout of cattle ranching and hunting of introduced deer and elk. Later, NPS staff stated that the former landowners had a 25-year right to presale land uses. This unsubstantiated “right” became the justification for the NPS issuing a series of permits that continued the operations of the former landowner. Over time, questions arose regarding the environmental impacts of the cattle, deer, and elk on …

How Socioeconomic Disadvantages Get Under The Skin And Into The Brain To Influence Health Development Across The Lifespan, Pilyoung Kim, Gary W. Evans, Edith Chen, Gregory Miller, Teresa Seeman
Psychology: Faculty Scholarship
Socioeconomic disadvantage (SED) has adverse impacts on physical (Adler and Rehkopf 2008; Blair and Raver 2012; Braverman and Egerter 2008; Cohen et al. 2010; Poulton et al. 2002) and psychological (Adler and Rehkopf 2008; Bradley and Corwyn 2002; Grant et al. 2003) health development. SED is similar to low socioeconomic status (SES) which is based on occupation, income, and education or a composite of more than one of these indicators (McLoyd 1998). However, we conceptualize SED more broadly than socioeconomic status to also include subjective perception of social position and contextual indicators of disadvantage, such as neighborhood deprivation. One of …

Toward Improving Metalloenzyme Inhibitor Design: A Thermodynamic Study Of Small Molecule Interactions With Copper(Ii), Cobalt(Ii) And Manganese(Ii), Fatimah Aldakheel
College of Science and Health Theses and Dissertations
This work aimed at understanding the interactions of 1) three physiologically relevant transition metal ions copper(II), cobalt(II) and manganese(II) with five tri- or tetra-dentate metal chelators (ligand or L) and 2) the resulting ML complexes with two known metalloenzyme inhibitors, 8-hydroxyquinoline (8-HQ) and acetohydroxamic acid (AHA). The ultimate goal of the work was to identify ligands that can form enzyme active site M2+ structural mimetics capable of interacting with metalloenzyme active site inhibitors. The ligands studied were N-(2-(1-methylimidazolyl)methyl)iminodiacetic acid (DA2Im), nitrilotriacetic acid (NTA), tris(2-aminoethyl)amine (TREN), tris(2-pyridylmethyl)amine (TPA), and bis(2-picolyl)amine (BPA) and they were chosen to mimic the coordination environment …
