Biological systems often generate unique and useful structures, which can have industrial relevance either as direct components or as an inspiration for biomimetic materials. For fabrication of nanoscale silica structures, we explored the use of the silaffin R5 peptide from Cylindrotheca fusiformis expressed on the surface of the fd bacteriophage. By utilizing the biomineralizing peptide component displayed on the bacteriophage surface, we found that low concentrations (0.09 mg/mL of the R5 bacteriophage, below the concentration range used in other studies) could be used to create silica nanofibers. An additional benefit of this approach is the ability of our R5-displaying phage …

Activation Des Éosinophiles Humains Par Des Nanoparticules, Denis Girard, Maxime Marion-Murphy

Marked reductions in population size can trigger corresponding declines in genetic variation. Understanding the precise genetic consequences of such reductions, however, is often challenging due to the absence of robust pre- and post-reduction datasets. Here, we use heterochronous genomic data from samples obtained before and immediately after the 2011 eruption of the Puyehue-Cordón Caulle volcanic complex in Patagonia to explore the genetic impacts of this event on two parapatric species of rodents, the colonial tuco-tuco (Ctenomys sociabilis) and the Patagonian tuco-tuco (C. haigi). Previous analyses using microsatellites revealed no post-eruption changes in genetic variation in C. …

Induction Of Immune Surveillance Of The Dysmorphogenic Lens., Caitlin M. Logan, Caitlin J. Bowen, A. Sue Menko Nov 2017

Induction Of Immune Surveillance Of The Dysmorphogenic Lens., Caitlin M. Logan, Caitlin J. Bowen, A. Sue Menko

Department of Pathology, Anatomy, and Cell Biology Faculty Papers

The lens has been considered to be an immune privileged site not susceptible to the immune processes normally associated with tissue injury and wound repair. However, as greater insight into the immune surveillance process is gained, we have reevaluated the concept of immune privilege. Our studies using an N-cadherin lens-specific conditional knockout mouse, N-cadΔlens, show that loss of this cell-cell junctional protein leads to lens degeneration, necrosis and fibrotic change, postnatally. The degeneration of this tissue induces an immune response resulting in immune cells populating the lens that contribute to the development of fibrosis. Additionally, we demonstrate that the lens …

The Mental Homologies Of Mammals. Towards An Understanding Of Another Mammalsworld View, Marthe Kiley-Worthington Nov 2017

The Mental Homologies Of Mammals. Towards An Understanding Of Another Mammalsworld View, Marthe Kiley-Worthington

Theory of Mind Collection

Mammals’ mental homologies include that they look after their young, suckle and protect them; they acquire information about the world by learning. They have five types of sensory receptors and a brain to analyze the information and they feel: that is they are sentient. Mental homologies have been largely ignored by behavioural scientists since Darwin because of certain historical beliefs. This however has not been the case for people who have had to do with non-human mammals who have long recognized their mental similarities to humans. As a result, behavioural science has sponsored some inappropriate research (examples are given). The …

Our growing awareness of the microbial world's importance and diversity contrasts starkly with our limited understanding of its fundamental structure. Despite recent advances in DNA sequencing, a lack of standardized protocols and common analytical frameworks impedes comparisons among studies, hindering the development of global inferences about microbial life on Earth. Here we present a meta-analysis of microbial community samples collected by hundreds of researchers for the Earth Microbiome Project. Coordinated protocols and new analytical methods, particularly the use of exact sequences instead of clustered operational taxonomic units, enable …

Experimental Analysis Of Reintroduction Strategies To Conserve The Vulnerable Plains Topminnow Fundulus Sciadicus In Nebraska, David A. Schumann, W. Wyatt Hoback, Keith D. Koupal, Casey W. Schoenebeck, Steven C. Schainost, Tammy L. Wilson

Nebraska Game and Parks Commission: Staff Research Publications

Plains topminnow Fundulus sciadicus are endemic to the Great Plains (USA), and because of declines in their geographic range and local abundance, are granted protection throughout their native range. Experimental reintroductions were conducted to improve conservation techniques and enhance the long-term outlook for plains topminnow persistence in Nebraska. Reintroductions were attempted at 17 extirpated stream reaches using low and high founder densities (no. of fish per suitable area) and during 2 seasons (spring and fall) to identify successful techniques for future conservation efforts. Reintroduced populations were sampled monthly (excluding winter) for 2 yr to monitor population persistence. Repeated presence− absence …

Systolic Blood Pressure Response In Sprint (Systolic Blood Pressure Intervention Trial) And Accord (Action To Control Cardiovascular Risk In Diabetes): A Possible Explanation For Discordant Trial Results, Khurram Nasir

All Publications

BACKGROUND:

SPRINT (Systolic Blood Pressure Intervention Trial) and the ACCORD (Action to Control Cardiovascular Risk in Diabetes) blood pressure trial used similar interventions but produced discordant results. We investigated whether differences in systolic blood pressure (SBP) response contributed to the discordant trial results.

METHODS AND RESULTS:

We evaluated the distributions of SBP response during the first year for the intensive and standard treatment groups of SPRINT and ACCORD using growth mixture models. We assessed whether significant differences existed between trials in the distributions of SBP achieved at 1 year and the treatment-independent relationships of achieved SBP with risks of primary …

Hydrogen Cyanide Produced By Pseudomonas Chlororaphis O6 Exhibits Nematicidal Activity Against Meloidogyne Hapla, Beom Ryong Kang, Anne J. Anderson, Young Cheol Kim

Biology Faculty Publications

Root-knot nematodes (Meloidogyne spp.) are parasites that attack many field crops and orchard trees, and affect both the quantity and quality of the products. A root-colonizing bacterium, Pseudomonas chlororaphis O6, possesses beneficial traits including strong nematicidal activity. To determine the molecular mechanisms involved in the nematicidal activity of P. chlororaphis O6, we constructed two mutants; one lacking hydrogen cyanide production, and a second lacking an insecticidal toxin, FitD. Root drenching with wild-type P. chlororaphis O6 cells caused juvenile mortality in vitro and in planta. Efficacy was not altered in the fitD mutant compared to the wild-type but was reduced in …

Access to a geographically diverse set of modern human samples from the present time and from ancient remains, combined with archaic hominin samples, provides an unprecedented level of resolution to study both human history and adaptation. The amount and quality of ancient human data continues to improve, and enables tracking the trajectory of genetic variation over time. These data have the potential to help us redefine or generate new hypotheses of how human evolution occurred, and revise previous conjectures. In this review, we argue that leveraging all these data will help us better detail adaptive histories in humans. As a …

The field of paleogenomics (the study of ancient genomes) is rapidly advancing with more robust methods of isolating ancient DNA and increasing access to next-generation DNA sequencing technology. As these studies progress, many important ethical issues have emerged that should be considered when ancient Native American remains, whom we refer to as ancestors, are used in research. We highlight a recent article by Kennett et al. (2017), “Archaeogenomic evidence reveals prehistoric matrilineal dynasty,” that brings several ethical issues to light that should be addressed in paleogenomics research (Kennett et al. 2017). The study helps elucidate the matrilineal relationships in ancient …

Off-Target Effects Of Drugs That Disrupt Human Mitochondrial Dna Maintenance., Matthew J Young Nov 2017

Off-Target Effects Of Drugs That Disrupt Human Mitochondrial Dna Maintenance., Matthew J Young

Articles

Nucleoside reverse transcriptase inhibitors (NRTIs) were the first drugs used to treat human immunodeficiency virus (HIV) the cause of acquired immunodeficiency syndrome. Development of severe mitochondrial toxicity has been well documented in patients infected with HIV and administered NRTIs. In vitro biochemical experiments have demonstrated that the replicative mitochondrial DNA (mtDNA) polymerase gamma, Polg, is a sensitive target for inhibition by metabolically active forms of NRTIs, nucleotide reverse transcriptase inhibitors (NtRTIs). Once incorporated into newly synthesized daughter strands NtRTIs block further DNA polymerization reactions. Human cell culture and animal studies have demonstrated that cell lines and mice exposed to NRTIs …

Barriers To And Facilitators Of Help-Seeking Behavior Among Men Who Experience Sexual Violence, Martina Delle Donne, Joseph Deluca, Pavel Pleskach, Christopher Bromson, Marcus P. Mosley, Edward T. Perez, Shibin G. Matthews, Rob Stephenson, Victoria Frye

Publications and Research

Research on sexual violence and related support services access has mainly focused on female victims; there is still a remarkable lack of research on men who experience sexual violence. Research demonstrates that people who both self-identify as men and are members of sexual-orientation minority populations are at higher risk of sexual violence. They are also less likely to either report or seek support services related to such experiences. The present study is an exploratory one aimed at filling the gap in the literature and better understanding how men, both straight and gay as well as cisgender and transgender, conceptualize, understand, …

The major histocompatibility complex is directly involved in the immune response and thus the genes coding for its proteins are useful markers for the study of genetic diversity, susceptibility to disease (autoimmunity and infections), transplant medicine, and pharmacogenetics, among others. The polymorphism of the system also allows researchers to use it as a proxy for population genetics analysis, such as genetic admixture and genetic structure. In order to determine the immunogenetic characteristics of a sample from the northern part of Mexico City and to use them to analyze the genetic differentiation from other admixed populations, including those from previous studies …

Microbial Diversity In Trace Water Of Jet Fuel In Vietnam, Hien Lai Thuy, Hang Pham Thi, Nga Vuong Thi, Hai Hoang, Yamaguchi Nobuyasu, Tani Katsuji

ASEAN Journal on Science and Technology for Development

Microorganisms in petroleum product, including jet fuel, have been found by some English andAmerican authors since the 20’s of the last century. In 1958, after Bakanaukas isolated many microorganisms in jet fuel JP4, this topic was especially concentrated. Most of microorganisms living in jet fuel are able to degrade hydrocarbon of the fuel to obtain energy and carbon source for their growth. The presence of microorganisms in jet fuel not only brings about the lost of fuel in store process, but also briefly changes the component and structure of jet fuel. Moreover, the products of their metabolism, such as organic …

Bioinformatic And Biophysical Analyses Of Proteins, Jonathan Catazaro Nov 2017

Bioinformatic And Biophysical Analyses Of Proteins, Jonathan Catazaro

Department of Chemistry: Dissertations, Theses, and Student Research

The prevailing dogma in structural genomics is the existence of a strong correlation between protein sequence, structure, and biological function. Proteins with high sequence similarity typically have a similar, if not the same, structure and function. In many cases this logic can fail due to distantly related proteins having very low sequence similarity, a lack of a representative structure, structural novelty, or the absence of a characterized function. Further, the paradigm fails to account for dynamics, which have a significant effect on structural stability and enzymatic efficacy.

Nuclear magnetic resonance (NMR) spectroscopy is uniquely capable of solving the structure, assisting …

Development Of A Broadly Protective Modified-Live Virus Vaccine Candidate Against Porcine Reproductive And Respiratory Syndrome Virus, Haiyan Sun, Aspen Workman, Fernando A. Osorio, David J. Steffen, Hiep L.X. Vu

Nebraska Center for Virology: Faculty Publications

Modified-live virus (MLV) vaccines are widely used to protect pigs against porcine reproductive and respiratory syndrome virus (PRRSV). However, current MLV vaccines do not confer adequate levels of heterologous protection, presumably due to the substantial genetic diversity of PRRSV isolates circulating in the field. To overcome this genetic variation challenge, we recently generated a synthetic PRRSV strain containing a consensus genomic sequence of PRRSV-2. We demonstrated that our synthetic PRRSV strain confers unprecedented levels of heterologous protection. However, the synthetic PRRSV strain at passage 1 (hereafter designated CON-P1) is highly virulent and therefore, is not suitable to be used as …
