In 2010, researchers observed an abrupt decline in nesting numbers of the Kemp’s ridley following 35 years of positive growth. In wake of this halt, and subsequent lowered nesting averages, evaluation of the genetic diversity of remaining individuals is relevant to investigate the plausibility of a genetic bottleneck, and determine the reproductive stability of the Kemp’s ridley.

Here, I evaluate the genetic diversity within a sampling of the Kemp’s ridleys on South Padre Island using two methods: targeting the mitochondrial control region and targeting the complete mitochondrial genome. Opportunistic samples were donated from juvenile ridleys on the East coast, and …

The PI3K pathway controls many cellular processes such as cell growth, survival, metabolism, apoptosis, and differentiation. Canonically, PI3K activates AKT leading to subsequent nuclear exportation of FOXO transcription factors. Dysregulation of the PI3K pathway is a hallmark in many cancers and often is attributable to a null mutation in PTEN. PTEN is a phosphatase that reverses the reactions catalyzed by PI3K. Loss of PTEN leads to constitutive inactivation of tumor suppressor FOXO proteins. Despite continued widespread use, the genomic effects of cisplatin-based chemotherapy and implications for subsequent treatment are incompletely characterized. Here, we analyze whole exome sequencing of matched pre- and post-neoadjuvant cisplatin-based chemotherapy primary bladder tumor samples from 30 muscle-invasive bladder cancer patients. We observe no overall increase in tumor mutational burden post-chemotherapy, though a significant proportion of subclonal mutations are unique to the matched pre- or post-treatment tumor, suggesting chemotherapy-induced and/or spatial heterogeneity. We subsequently identify and validate a novel mutational signature in post-treatment tumors consistent with known characteristics of cisplatin damage and repair. Mosquito-borne illnesses present a great threat to human health, and result in millions of cases and hundreds of thousands of deaths annually. This study aimed to determine the effectiveness of extracted essential oils from three neotropical plants traditionally used in Costa Rican folk medicine as mosquito repellents. I extracted the essential oils of Siparuna decipiens (Monimiaceae), Gliricidia sepium (Fabaceae), and Neuroloena lobata (Asteraceae) via simple steam distillation. To change public perceptions, particularly from urban populations, of agriculture and natural resources (ANR) fields, the University of Tennessee developed a model of strategic educational recruitment to extend beyond traditional college fair experiences to educate underrepresented candidates. The High School Agricultural Education Initiative (HSAI) provided 10th-grade students with opportunities to: (a) gain knowledge of and appreciation for disciplines in agriculture and natural resources, (b) learn about college admissions standards, (c) network with college students and staff to learn about campus resources, and (d) understand how a college education improves professional opportunities. The plant-soil nitrogen cycle plays a significant role in allocation of available N to plants, and improved understanding of N cycling helps sustainably increase fertilizer use efficiency. There are various processes (nitrogen mineralization and nitrification) involved in the availability and mobility of nitrogen in the soil. The primary objective of this study was to determine the NUE under contrasting nitrogen treatments over a period of five years. Atherosclerosis is a chronic inflammatory disorder of medium and large vessels. Immune signaling and dyslipidemia are two of several processes which influence lesion development in atherosclerosis. Cannabinoids, such as those found in marijuana, exert their effects through two cannabinoid receptors, CB1 and CB2. Recent studies using CB2 knockout mice and CB2-selective ligands have shed light on a protective role of CB2 in early stages of atherosclerosis. The purpose of this quantitative study was to determine if a relationship existed between the quality of school-community partnerships and the average score of each ACT subtest for rural Tennessee high school students. Specifically, the researcher examined the following school-community partnerships: business partnerships, university partnerships, service learning partnerships, school-linked service integration, and faith-based partnerships (Sanders, 2006). Administrators from 62 rural Tennessee high schools rated the partnerships present in their schools using the Improving Community Partnership Quality rating scale developed by Sanders (2006). Thus, new technologies are needed to combat these troublesome weeds. A new non-GMO, herbicide-resistant rice type is under development that is resistant to quizalofop, an acetyl coenzyme A carboxylase (ACCase)-inhibiting herbicide that will allow for selective grass weed control in rice. The purpose of this study was to examine whether different habitual activity levels may affect balance ability in older adults. Balance must be studied as poor balance increases likelihood of falls. This study examined activity levels in older adults and effect on balance ability. Fifty-nine older adults aged 66.5 ± 9.5 participated. Three groups were separated by physical activity level. First group had 13 masters athletes, older adults performing least 150 minutes of aerobic activity a week and competing in nationally sanctioned event within the last six months. This thesis offers the first global history of the Cold War in the eastern Mediterranean. It examines the international linkages that bound Greece, Turkey, and Cyprus with superpowers, non-aligned states, and transnational movements during the second half of the twentieth century, and it considers the effects of such linkages upon the eastern Mediterranean’s domestic arenas. Throughout, it demonstrates that two forces – synthesis of outside influence alongside consolidation of internal identities – dictated the region’s experiences during the Cold War. Biotherapeutic drugs, derived from biological molecules such as proteins and DNA, are becoming an integral and exceptionally critical aspect of modern medicine. Compared to common pharmaceutical drugs, biotherapeutics are much larger in size and have greater target specificity, allowing them to treat many chronic diseases ranging from cancer to rheumatoid arthritis. The major issue with protein based therapeutics is that they readily undergo proteolysis, or enzymatic degradation, when administered through subcutaneous injections. Histone-like nucleoid structuring proteins (HNS) play significant roles in shaping the chromosomal DNA, regulation of transcriptional networks in microbes, as well as bacterial responses to environmental changes such as temperature fluctuations. In this work, the intracellular organization of HNS proteins in E. coli bacteria was investigated utilizing super-resolution fluorescence microscopy, which surpasses conventional microscopy by 10–20 fold in spatial resolution. Because of the evolution of herbicide-resistant weeds, such as barnyardgrass and red rice in rice, there is a need for alternative herbicide sites of action. Very-long-chain fatty acid (VLCFA)-inhibiting herbicides are not labeled for use in U.S. rice production; however, this site of action (SOA) has been used with success in Asian rice. The VLCFA-inhibiting herbicides pethoxamid, pyroxasulfone, acetochlor, and S-metolachlor were evaluated for rice tolerance and control of commonly problematic weeds in Arkansas rice at various rates and application timings. The ability of beta-amyloid peptide (Aβ) to disrupt the plasma membrane through formation of pores and membrane breakage has been previously described. However, the molecular determinants for these effects are largely unknown. In this study, we examined if the association and subsequent membrane perforation induced by Aβ was dependent on GM1levels. Pretreatment of hippocampal neurons with D-PDMP decreased GM1 and Aβ clustering at the membrane (Aβ fluorescent-punctas/20 μm, control = 16.2 ± 1.1 vs. D-PDMP = 6.4 ± 0.4, p < 0.001). Interestingly, membrane perforation with Aβ occurred with a slower time course when the GM1 content was diminished (time to establish perforated configuration (TEPC) (min): control = 7.8 ± 2 vs. low GM1 = 12.1 ± 0.5, p < 0.01), suggesting that the presence of GM1 in the membrane can modulate the distribution and the membrane perforation by Aβ. On the other hand, increasing GM1 facilitated the membrane perforation (TEPC: control = 7.8 ± 2 vs. GM1 = 6.2 ± 1 min, p < 0.05). Additionally, using Cholera Toxin Subunit-B (CTB) to block the interaction of Aβ with GM1 attenuated membrane perforation significantly. Furthermore, …

These pigments range in color from orange-red to blue-violet and could serve as natural colorants to replace artificial additives. Whether reduced supraspinal activation contributes to age-related reductions in maximal torque during dynamic contractions is not known. The purpose was to determine whether there are age differences in voluntary activation and its variability when assessed with stimulation at the motor cortex and the muscle during maximal isometric, concentric, and eccentric contractions. Thirty young (23.6 ± 4.1 years) and 31 old (69.0 ± 5.2 years) adults performed maximal isometric, shortening (concentric) and lengthening (eccentric) contractions with the elbow flexor muscles. Despite the fact that the recent rice policy has been moving to a strategy of self-sufficiency while the status quo of the national rice economy remains ambiguous, Malaysia has made an extreme policy decision to pursue an autarky economy in its rice sector, thus closing borders from the international markets in the future. The goal of this dissertation research is to comprehensively evaluate a deep-rooted rice policy in Malaysia and analyze the holistic impacts of the self-sufficiency and international trade policies at the national and farm-household levels, utilizing economic frameworks. Protein-protein interaction (PPI) networks play an important role in studying the functional roles of proteins, including their association with diseases. However, protein interaction networks are not sufficient without the support of additional biological knowledge for proteins such as their molecular functions and biological processes. To complement and enrich PPI networks, we propose to exploit biological properties of individual proteins. More specifically, we integrate keywords describing protein properties into the PPI network, and construct a novel PPI-Keywords (PPIK) network consisting of both proteins and keywords as two different types of nodes.
