Functional Features Of Hyperactive Deg/Enac Channels Involved In Cell Death, Cristina Matthewman
Functional Features Of Hyperactive Deg/Enac Channels Involved In Cell Death, Cristina Matthewman
Open Access Dissertations
Ion channels of the DEG/ENaC family can induce neurodegeneration under conditions in which they become hyperactivated. The C. elegans DEG/ENaC channel MEC-4(d) encodes a mutant channel with a substitution in the pore domain that causes swelling and death of the six touch neurons in which it is expressed. Dominant mutations in homolog channel UNC-8 result in uncoordinated movement. Here we show that this unc-8(d) movement defect is correlated with the selective death of cholinergic motor neurons in the ventral nerve cord, a milder toxicity compared with MEC-4(d). UNC-8(G387E), denoted UNC-8(d), and UNC-8(A586T) mutants encode hyperactivated channels that are strongly inhibited …

Diet Composition Of Age-0 Fishes In Created Habitats Of The Lower Missouri River, Trevor A. Starks, James M. Long
Diet Composition Of Age-0 Fishes In Created Habitats Of The Lower Missouri River, Trevor A. Starks, James M. Long
United States Geological Survey: Staff Publications
Channelization of the Missouri River has greatly reduced the availability of shallow water habitats used by many larval and juvenile fishes and contributed to imperilment of floodplain-dependent biota. Creation of small side channels, or chutes, is being used to restore shallow water habitat and reverse negative environmental effects associated with channelization. In the summer of 2012, the U.S. Army Corps of Engineers collected early life stages of fishes from constructed chutes and nearby unrestored shallow habitats at six sites on the Missouri River between Rulo, Nebraska and St. Louis, Missouri. We compared the diets of two abundant species of fishes …

The Performance Analysis Of Two Relatively Small Capacity Urban Retrofit Stormwater Controls, James J. Houle, Thomas P. Ballestero, Timothy A. Puls
The Performance Analysis Of Two Relatively Small Capacity Urban Retrofit Stormwater Controls, James J. Houle, Thomas P. Ballestero, Timothy A. Puls
UNH Stormwater Center
This paper details field investigations that were conducted on the performance of small capacity urban retrofit stormwater control measures. The objective of the two year study (2013–2015) was to provide performance data on stormwater retrofits that could not be fully sized according to conventional standards due to space constraints. In many states performance credits are not granted to stormwater management controls that are not designed to manage regionally derived water quality volumes. In retrofit applications there may exist numerous limitations to conventionally sized systems such as limited rights of way, setback distances or existing utilities. The larger scale objective of …

Two New Genera Of Songbirds Represent Endemic Radiations From The Shola Sky Islands Of The Western Ghats, India, Sushma Reddy, V. V. Robin, C. K. Vishnudas, Pooja Gupta, Frank E. Rheindt, Daniel M. Hooper, Uma Ramakrishnan
Biology: Faculty Publications and Other Works
A long-standing view of Indian biodiversity is that while rich in species, there are few endemics or in-situ radiations within the subcontinent. One exception is the Western Ghats biodiversity hotspot, an isolated mountain range with many endemic species. Understanding the origins of the montane-restricted species is crucial to illuminate both taxonomic and environmental history.

Effect Of Phytosanitary Irradiation On The Quality Of Two Varieties Of Pummelos (Citrus Maxima (Burm.) Merr.), A. Jain, J. Ornelas-Paz, D. Obenland, K. Rodriguez (Friscia), Anuradha Prakash
Food Science Faculty Articles and Research
Phytosanitary treatments prevent the introduction of pests such as fruit flies into pest free zones, and are often required for international trade. Irradiation is increasingly being considered as an alternative to cold and chemical phytosanitary treatments, such as methyl bromide. In this study, the effect of low dose gamma irradiation on the post-harvest quality of two varieties of pummelos (Citrus maxima (Burm.) Merr.), an emerging crop of interest in the US was evaluated. Two varieties of pummelos grown in California were irradiated at the phytosanitary target dose of 150 Gy and a higher dose of 1000 Gy to exaggerate …

Approaches to quantifying and predicting soil biogeochemical cycles mostly consider microbial biomass and community composition as products of the abiotic environment. Current numerical approaches then primarily emphasise the importance of microbe–environment interactions and physiology as controls on biogeochemical cycles. Decidedly less attention has been paid to understanding control exerted by community dynamics and biotic interactions. Yet a rich literature of theoretical and empirical contributions highlights the importance of considering how variation in microbial population ecology, especially biotic interactions, is related to variation in key biogeochemical processes like soil carbon formation. We demonstrate how a population and community ecology perspective can …

As students increasingly use online chemistry animations and simulations, it is becoming more important to understand how students independently engage with such materials and to develop a set of best practices for students’ use of these resources outside of the classroom. Most of the literature examining students’ use of animations and simulations has focused on classroom use with some studies suggesting that better outcomes are obtained when students use simulations with minimal guidance while others indicate the need for appropriate scaffolding. This study examined differences with respect to (1) student understanding of the concept of dissolution of ionic and covalent …

Although hydroxyapatite (HAp) has been doped with dozens of different ions, the quest for an ion imparting a combination of properties conducive to bone healing is still ongoing. Because of its protean potency and the similarity in size and shape to the phosphate tetrahedron, the selenite ion presents a natural ionic substitute in HAp. The incorporation of selenite into synthetic HAp using two different methods – co-precipitation and ion-exchange sorption – was studied for its effect on crystal properties and on a triad of biological responses: antibacterial, anticancer and osteoinductive. Co-precipitation yielded HAp with a higher selenite content than sorption …

Biofilms are communities of microbes embedded in a matrix of extracellular polymeric substances, largely polysaccharides. Multiple types of extracellular polymeric substances can be produced by a single bacterial strain. The distinct polymer components of biofilms are known to provide chemical protection, but little is known about how distinct extracellular polysaccharides may also protect biofilms against mechanical stresses such as shear or phagocytic engulfment. Decades-long infections of Pseudomonas. aeruginosa biofilms in the lungs of cystic fibrosis patients are natural models for studies of biofilm fitness under pressure from antibiotics and the immune system. In cystic fibrosis infections, production of the extracellular …

An Analysis Of Stable Isotopes From N-Alkanes Derived From Leaf Waxes In North America, Katherine T. Truong Ms.
Master's Theses
Leaf wax biomarkers are increasingly becoming a fundamental aspect of paleoclimate research. The strength of the relationship between their molecular and isotopic properties and environmental conditions is critical to their effectiveness in reconstructing past environments. To address the current lack of data pertaining to arid and semi-arid environments, soil and modern plant samples (grasses, trees, forbs, and shrubs) from sites in the western United States were analyzed to assess relationships between normal alkane molecular distribution, δD and δ13C and seasonal/annual climatic conditions in semi-arid ecosystems. The data show a stronger relationship between δD of leaf waxes (δDwax …

FadR is a master regulator of fatty acid (FA) metabolism that coordinates the pathways of FA degradation and biosynthesis in enteric bacteria. We show here that a ΔfadR mutation in the El Tor biotype of Vibrio cholerae prevents the expression of the virulence cascade by influencing both the transcription and the posttranslational regulation of the master virulence regulator ToxT. FadR is a transcriptional regulator that represses the expression of genes involved in FA degradation, activates the expression of genes involved in unsaturated FA (UFA) biosynthesis, and also activates the expression of two operons involved in saturated FA (SFA) biosynthesis. …

FSD-C10, a Fasudil derivative, was shown to reduce severity of experimental autoimmune encephalomyelitis (EAE), an animal model of multiple sclerosis (MS), through the modulation of the immune response and induction of neuroprotective molecules in the central nervous system (CNS). However, whether FSD-C10 can promote neuroregeneration remains unknown. In this study, we further analyzed the effect of FSD-C10 on neuroprotection and remyelination. FSD-C10-treated mice showed a longer, thicker and more intense MAP2 and synaptophysin positive signal in the CNS, with significantly fewer CD4(+) T cells, macrophages and microglia. Importantly, the CNS of FSD-C10-treated mice showed a shift of activated macrophages/microglia from …

Engineering A Fluorescent Protease Sensor For In Vivo Protein Detection, Thomas C. Kinard
Honors Scholar Theses
This report details the results of an ongoing project to engineer a mutant form of Red Fluorescent Protein (RFP) variant mCherry that acts as a real-time in vivo protease sensor. The sought-after mutant only becomes fluorescent when exposed to Tobacco Etch Virus (TEV) Protease, this system’s model protease. This will be accomplished via the insertion of the TEV Protease Recognition Site (TEV-PRS) in such a position that, before cleavage, will prevent the protein from folding to fluorescent conformation, but upon cleavage, will allow for fluorescent conformation to occur. The cylindrical structure of the protein, composed of beta-pleated sheets, contains “loops” …
