Microbiology, Immunology, and Tropical Medicine Faculty Publications

A new generation of vaccines for the neglected tropical diseases (NTDs) have now advanced into clinical development, with the Na-GST-1/Alhydrogel Hookworm Vaccine already being tested in Phase 1 studies in healthy adults. The current manuscript focuses on the often overlooked critical aspects of NTD vaccine product development, more specifically, vaccine stability testing programs. A key measure of vaccine stability testing is "relative potency" or the immunogenicity of the vaccine during storage. As with most NTD vaccines, the Na-GST-1/Alhydrogel Hookworm Vaccine was not developed by attenuation or inactivation of the pathogen (Necator americanus), so conventional methods for measuring relative potency are …

Malondialdehyde-Acetaldehyde (Maa) Adducted Surfactant Protein Induced Lung Inflammation Is Mediated Through Scavenger Receptor A (Sr-A1)., Muna Sapkota, Jane M. Devasure, Kusum K. Kharbanda, Todd A. Wyatt

Journal Articles: Environmental, Agricultural & Occupational Health

BACKGROUND: Co-exposure to cigarette smoke and alcohol leads to the generation of high concentrations of acetaldehyde and malondialdehyde in the lung. These aldehydes being highly electrophilic in nature react with biologically relevant proteins such as surfactant protein D (SPD) through a Schiff base reaction to generate SPD adducted malondialdehyde-acetaldehyde adduct (SPD-MAA) in mouse lung. SPD-MAA results in an increase in lung pro-inflammatory chemokine, keratinocyte chemoattractant (KC), and the recruitment of lung lavage neutrophils. Previous in vitro studies in bronchial epithelial cells and macrophages show that scavenger receptor A (SR-A1/CD204) is a major receptor for SPD-MAA. No studies have yet examined …

Dual-Functional-Tag-Facilitated Protein Labeling And Immobilization, Xinyi Zhang, Wei Lu, Kevin Kwan, Dibakar Bhattacharyya, Yinan Wei

Chemistry Faculty Publications

An important strategy in the construction of biomimetic membranes and devices is to use natural proteins as the functional components for incorporation in a polymeric or nanocomposite matrix. Toward this goal, an important step is to immobilize proteins with high efficiency and precision without disrupting the protein function. Here, we developed a dual-functional tag containing histidine and the non-natural amino acid azidohomoalanine (AHA). AHA is metabolically incorporated into the protein, taking advantage of the Met-tRNA and Met-tRNA synthetase. Histidine in the tag can facilitate metal-affinity purification, whereas AHA can react with an alkyne-functionalized probe or surface via well-established click chemistry. …

Background: The prevailing paradigm of host-parasite evolution is that arms races lead to increasing specialisation via genetic adaptation. Insect herbivores are no exception and the majority have evolved to colonise a small number of closely related host species. Remarkably, the green peach aphid, Myzus persicae, colonises plant species across 40 families and single M. persicae clonal lineages can colonise distantly related plants. This remarkable ability makes M. persicae a highly destructive pest of many important crop species. Results: To investigate the exceptional phenotypic plasticity of M. persicae, we sequenced the M. persicae genome and assessed how one clonal lineage responds …

Body Size And Lower Limb Posture During Walking In Humans, Martin Hora, Libor Soumar, Herman Pontzer, Vladimír Sládek

Publications and Research

We test whether locomotor posture is associated with body mass and lower limb length in humans and explore how body size and posture affect net joint moments during walking. We acquired gait data for 24 females and 25 males using a three-dimensional motion capture system and pressure-measuring insoles. We employed the general linear model and commonality analysis to assess the independent effect of body mass and lower limb length on flexion angles at the hip, knee, and ankle while controlling for sex and velocity. In addition, we used inverse dynamics to model the effect of size and posture on net …

Potent Single-Domain Antibodies That Arrest Respiratory Syncytial Virus Fusion Protein In Its Prefusion State, Iebe Rossey, Morgan Gilman, Stephanie Kabeche, Koen Sedeyn, Daniel Wrapp

Dartmouth Scholarship

Human respiratory syncytial virus (RSV) is the main cause of lower respiratory tract infections in young children. The RSV fusion protein (F) is highly conserved and is the only viral membrane protein that is essential for infection. The prefusion conformation of RSV F is considered the most relevant target for antiviral strategies because it is the fusion-competent form of the protein and the primary target of neutralizing activity present in human serum. Here, we describe two llama-derived single-domain antibodies (VHHs) that have potent RSV-neutralizing activity and bind selectively to prefusion RSV F with picomolar affinity. Crystal structures of these VHHs …

Impact Of Colonialism On Contemporary African Art, Jayna Clemens

Africana Studies Student Research Conference (2013-2019)

“If you don’t know history, then you don’t know anything. You are a leaf that doesn’t know it is part of a tree.” – Michael Crichton

Studying history provides insight and explanations for what is present in today’s society. Colonization is not the only piece of Africa’s history, but it is a major contributor. Studying the history of Africa, pre and post colonialism, provides explanations for how Africa arrived at its present and where it is headed in the future. Colonization had a major impact on Africa’s culture and history, including contemporary African art. Contemporary African art, including description, materials, …

Representative Bureaucracy, Street-Level Bureaucrats And Bureaucratic Discretion In Federal Disaster Assistance, Jason David Rivera Feb 2017

Representative Bureaucracy, Street-Level Bureaucrats And Bureaucratic Discretion In Federal Disaster Assistance, Jason David Rivera

Journal of Public Management & Social Policy

This paper explores the role of representative bureaucracy and bureaucratic discretion in the allocation of federal disaster assistance to Hurricane Sandy survivors. Through the analysis of focus groups and key informant interviews, this study finds that FEMA home inspectors are not diverse in reference to race and gender; however, inspectors are found to be diverse in reference to place of origin. Although the role that race and gender play in the allocation of resources is found to be unclear, the region from which inspectors come from is found to be influential in the allocation of aid to survivors. As such, …

Tissue Engineering Of The Temporomandibular Joint : Where Do We Stand Now?, Camille Haddad, Dalia Fleihan Feb 2017

Tissue Engineering Of The Temporomandibular Joint : Where Do We Stand Now?, Camille Haddad, Dalia Fleihan

International Arab Journal of Dentistry

Tissue engineering is an alternative to traditional strategies to repair and regenerate temporomandibular joints (TMJ). Nowadays, patients suffering from severe dysfunctions of the TMJ may undergo discectomy, a procedure that consists of removing the damaged disc in hopes of reducing the symptoms. However, tissue engineering presents a potential solution for patients suffering from these disorders, due to the lack of safety and effectiveness of TMJ disc implants. Since 1991, several studies have investigated the possibility of regenerating the articular disc. This literature review aims to expose the new challenges and techniques in TMJ disc tissue engineering whether it concerns cell sourcing, …

The tomato (Solanum lycopersicum) ripening process from mature green (MG) to turning and then to red stages is accompanied by the occurrences of physiological and biochemical reactions, which ultimately result in the formation of the flavor, color and texture of ripe fruits. The two trivalent metal ions Al3+ and La3+ are known to induce different levels of phytotoxicity in suppressing root growth. This paper aims to understand the impacts of these two metal ions on tomato fruit proteomes. Tomato ‘Micro-Tom’ plants were grown in a hydroponic culture system supplemented with 50 μM aluminum sulfate (Al2 (SO4)3 …
