Monomethylmercury (MeHg) is a neurotoxic compound that causes adverse health effects in exposed wildlife and humans. It is primarily accumulated through diet and is the only mercury species to build up in aquatic food chains. However, our understanding with respect to MeHg biogeochemical cycling in marine environments and its interactions with marine organisms is still limited. Charcot-Marie-Tooth (CMT) disease is the most commonly inherited neurological disorder, but its molecular mechanisms remain unclear. One variant of CMT, Charcot-Marie-Tooth 2F (CMT2F), is characterized by mutations in heat shock protein 27 (Hsp27). Bioactive sphingolipids have been implicated in many neurodegenerative diseases, but it was unknown if they were dysregulated in CMT. Liquid chromatography/mass spectrometry was used to profile sphingolipids in CMT models. Hsp27 KO mice demonstrated decreases in ceramide in peripheral nerve tissue at an increased age, suggesting that sphingolipid metabolism may be involved in CMT2F. Indeed, the disease associated Hsp27 S135F mutant demonstrated decreases in mitochondrial ceramides. Given …

Optical domain technologies can provide high spatiotemporal resolution for biomedical research and therefore attract tremendous interests in multiple disciplines including psychiatry, neurology, ophthalmology, oncology, urology and many more. Optical coherence tomography (OCT), after more than 20 years of development, has been becoming one of the most widely-used bio-optical modalities not only for laboratory-based research but also for clinical applications. For example, OCT becomes the standard of care for diagnosing glaucoma. Additionally, its derived velocimetry, optical coherence Doppler tomography (ODT), also demonstrates the unique capability of mapping cerebral blood flow velocity (CBFv) of vascular networks with capillary resolution (&phis;<6µm) and thus allows for the widespread utilization in numerous neurology studies. Although ODT has been widely used in biomedical research, there remain several technical limitations including the shallow penetration depth, low sensitivity for imaging slow flow in capillary and the measurement error due to Doppler angle. To address these challenges, viable solutions are proposed in this dissertation. Chapter 1 starts with a comprehensive review over basic principle of OCT and ODT. Chapter 2-4 explicitly address these OCT technical challenges, for instance, using a 1310nm super-luminescent Diode (SLD) laser source to extend optical penetration depth, utilizing optical contrast agent: intralipid to enhance ODT’s sensitivity for capillary flow detection, implementing numerical methods to extend ODT’s dynamic range and decouple Doppler angle. In addition to these technical improvements, Chapter 5-6 address two biomedical challenges in oncology and psychiatry. Specifically, Chapter 5 introduces a longitudinal optical access to rodent cerebral vasculature and a method of tumor boundary detection, which provides a powerful insight into the tumor progression. Chapter 6 reveals the close correlation between cocaine-elicited transient ischemic attack (TIA) and hypo-perfused cortex. Chapter 7 focuses on the development of multi-modality system and novel imaging scheme for the study of neurovascular coupling. Chapter 8 briefly concludes the contents presented in this dissertation and outlines the future development and potential applications of ODT. | 129 pages

Poor growth with underweight for age, decreased length/height for age, and underweight-for-height are all relatively common in children with CHD. The underlying causes of this failure to thrive may be multifactorial, including innate growth potential, severity of cardiac disease, increased energy requirements, decreased nutritional intake, malabsorption, and poor utilisation of absorbed nutrition. These factors are particularly common and severe in low- and middle-income countries. Although nutrition should be carefully assessed in all patients, failure of growth is not a contraindication to surgical repair, and patients should receive surgical repair where indicated as soon as possible. Tropical storm intensity prediction remains a challenge in tropical meteorology. Some tropical storms undergo dramatic rapid intensification and rapid decline. Hurricane researchers have considered particular ambient environmental conditions including the ocean thermal and salinity structure and internal vortex dynamics (e.g., eyewall replacement cycle, hot towers) as factors creating favorable conditions for rapid intensification. At this point, however, it is not exactly known to what extent the state of the sea surface controls tropical cyclone dynamics. Theoretical considerations, laboratory experiments, and numerical simulations suggest that the air-sea interface under tropical cyclones is subject to the Kelvin-Helmholtz type instability. Predominantly localized in plant secondary cell walls, lignin is a highly crosslinked, aromatic polymer that imparts structural support to plant vasculature, and renders biomass recalcitrant to pretreatment techniques impeding the economical production of biofuels. Lignin is synthesized via the phenylpropanoid pathway where the primary precursor phenylalanine (Phe) undergoes a series of functional modifications catalyzed by 11 enzyme families to produce p-coumaryl, coniferyl, and sinapyl alcohol, which undergo random polymerization into lignin. Several metabolic engineering efforts have aimed to alter lignin content and composition, and make biofuel feedstock more amenable to pretreatment techniques. Skeletal muscles are composed of muscle cells or myofibers which are classified as four types (I, IIa, IIx/d, and IIb), with different physiological, biochemical, structural and metabolic characteristics. The molecular regulation of myofiber identify is not completely understood. I identified methyltransferase like 21 (Mettl21) e and Mettl21c as the type IIb and type I myofiber- specific protein, respectively. Sperm chromatin not only has a unique structure to condense and protect the paternal DNA in transit, but also provides epigenetic information that supports embryonic development. Most of the unique sperm nuclear architecture is formed during the sweeping postmeiotic chromatin remodeling events in spermiogenesis, where the majority of nucleosomes are removed and replaced by protamines. The remaining histones and other chromatin proteins are located in structurally and transcriptionally relevant positions in the genome and carry diverse post-translational modifications relevant to the control of embryonic gene expression. The purpose of this study is to measure effects of exercise environments on stress in premedical and predental students, and how to provide a method that would strive to reduce stress levels in college students. To further understand the different ways stress levels can be reduced, a method was performed to see if exercise environments have an effect on stress in premedical and predental students. Tissue engineered grafts would ideally provide more availability, while also replicating the mechanical properties of the native grafts. Ruthenium polypyridyl complexes (RPCs) represent a promising therapeiutic class of drugs for transition-metal based anticancer drug development. The complex [(phen)2Ru(tatpp)Ru(phen)2] (44+) is a RPC which has shown success in both cell free and cellular assay. Unlike many other RPCs, 44+ can selectively kill malignant cells without light irradiation from external sources. This binuclear complex 44+ is a DNA intercalator containing redox-active ligand tatpp (9,11,20,22-Tetraaza tetrapyrido[3,2-a:2'3'-c:3'',2''- 1:2''',3''']-pentacene) which cleaves DNA in presence of a mild reducing agent, i.e. glutathione (GSH).1,2,3 The cellular assay (MTT assay) with different types of malignant cells has also established this as an equally potent cytotoxic agent …

This dissertation seeks to understand the geological and climatological processes that have promoted diversification on continental and island systems. Using molecular genetic data generated using Sanger sequencing and Next Generation Sequencing platforms, I conduct phylogenetic and biogeographic analyses, estimate gene flow, and conduct species delimitation. Using these analyses, I explore diversification processes on continents and islands using reptile and amphibian systems. In Chapter 2 I evaluate the role of geographical features to whipsnake diversification. Long non-coding RNA (lncRNA) are emerging as contributors to malignancies. Little is understood about the contribution of lncRNA to epithelial-to-mesenchymal transition (EMT), which correlates with metastasis. Ovarian cancer is usually diagnosed after metastasis. Here we report an integrated analysis of >700 ovarian cancer molecular profiles, including genomic data sets, from four patient cohorts identifying lncRNA DNM3OS, MEG3, and MIAT overexpression and their reproducible gene regulation in ovarian cancer EMT. Genome-wide mapping shows 73% of MEG3-regulated EMT-linked pathway genes contain MEG3 binding sites. DNM3OS overexpression, but not MEG3 or MIAT, significantly correlates to worse overall patient survival. DNM3OS knockdown results in …

Climate change is predicted to affect the larval stages of many marine organisms. Ocean warming can reduce larval survival and hasten larval development, whereas ocean acidification can delay larval development. Ocean acidification is especially concerning for marine organisms that develop and grow calcified shells or skeletons in an environment undersaturated with calcium carbonate minerals. This study assessed the effects of ocean warming and acidification on the fertilization and larval development of the green sea urchin, Lytechinus variegatus, a tropical species common in Florida and the Caribbean. Coral recruitment in Southeast Florida is being outpaced by mortality, resulting in population declines in many species. Identifying the coral species most likely to recruit and survive on Southeast Florida reefs and evaluating spatial variation in recruitment and survivorship is crucial for managing decreasing coral populations. This study focuses on 12 sites in Broward and Miami-Dade counties that have served as long-term stations for monitoring adult coral cover and demographics. The use and misuse of opioid pain medication has reached epidemic proportions in the United States. More than 16,000 people die each year due to the misuse or abuse of prescription opioid medications, which is more than from heroin, cocaine, and benzodiazepines combined. The purpose of this evidence-based practice project is to establish processes enabling a primary care clinic to implement and adhere to the 2016 Centers for Disease Control’s guidelines for opioid prescription practices in the primary care setting. Currently, there is a lack of surveys that report the occurrence of gastrointestinal parasites in the whiteheaded capuchin monkey (Cebus albifrons). We therefore assessed the presence and richness (= number of different parasite genera) of parasites in C. albifrons in wildlife refuges (n = 11) and in a free-ranging group near a human village (n = 15) in the Ecuadorian Amazon. In the 78 samples collected (median of 3 samples per animal), we identified a total of 6 genera of gastrointestinal parasites, representing protozoa, nematodes, acanthocephalans and cestodes. The Proteomics Standards Initiative (PSI) of the Human Proteome Organization (HUPO) has now been developing and promoting open community standards and software tools in the field of proteomics for 15 years. Under the guidance of the chair, cochairs, and other leadership positions, the PSI working groups are tasked with the development and maintenance of community standards via special workshops and ongoing work. Among the existing ratified standards, the PSI working groups continue to update PSI-MI XML, MITAB, mzML, mzIdentML, mzQuantML, mzTab, and the MIAPE (Minimum Information About a Proteomics Experiment) guidelines with the advance of new technologies and techniques. NADPH oxidase 5 (NOX5) generated reactive oxygen species (ROS) have been implicated in signaling cascades that regulate cancer cell proliferation. To evaluate and validate NOX5 expression in human tumors, we screened a broad range of tissue microarrays (TMAs), and report substantial overexpression of NOX5 in malignant melanoma and cancers of the prostate, breast, and ovary. In human UACC-257 melanoma cells that possesses high levels of functional endogenous NOX5, overexpression of NOX5 resulted in enhanced cell growth, increased numbers of BrdU positive cells, and increased γ-H2AX levels. The objective of this work is to quantify the one-dimensional spatial distribution of radionuclides in field lysimeters from the Radionuclide Field Lysimeter Experiment (RadFLEX) facility at the Savannah River Nationals Laboratory (SRNL). The lysimeters, containing 137Cs, 60Co, 133Ba and 152Eu incorporated either into solid wasteforms (Portland cement and reducing grout) or introduced into soil via a filter paper wasteform, were weathered for three to four years. The initial contaminant activities range from 4.0 to 9.0 MBq for the cementitious wasteforms and 0.25 to 0.47 MBq for the filter paper wasteform. An analytical method was developed to …

Background: As more women joined the armed forces the population of women veterans inevitably increased and with it so did the rates of women veteran homelessness. Women veterans are a unique group of women who also possess protective factors from homelessness, yet are 3 to 4 times likely to become homeless than non-veteran women. The purpose of this systematic review is to glean from other articles and studies what the risk factors for women veteran homelessness are. This Article examines recent claims that the fetus be afforded the status of a person under the Fourteenth Amendment. It shows that such claims do not carry the necessary objectivity to operate reasonably in a pluralistic society. It then goes on to afford what a better view of personhood that could so operate might actually look like. Along the way, this Article takes seriously the real deep concerns many have for the sanctity of human life. By the end, it attempts to find a balance for those concerns with the view of personhood offered that should engage current debates about …
