Background: CD100, also known as Sema4D, is an immune semaphorin constitutively expressed on natural killer (NK) cells and T cells. As an immune activation molecule, CD100 has important immunoregulatory effects on NK functions by enhancing the interactions between NK cells and target cells. The aim of this study was to investigate whether hepatitis C virus (HCV) infection affects CD100 expression, and whether interferon-α treatment enhances NK killing activity to facilitate HCV clearance via CD100. Methods: Expression of CD100 on NK cells was evaluated by flow cytometry in patients with chronic HCV infection, with or without pegylated interferon-α-based therapy. NK cell …

Urosepsis occurs in 20-30 % of septic case. Early diagnosis were undoubtfully important to improve the results of sepsis management. Bacteriological confirmation may be difficult to obtain and negative cultures do not exclude the presence of infection. Increased understanding of inflammatory cascade mechanisms provided several indicator of infection and prognosis. Lactate measurement in patients with infection and possibly severe sepsis to help identify patients at high risk of death in order to apply aggressively therapy. Elevation in serum C-reactive protein (CRP) is also associated with an increasing risk of death in patients with infection. This study was to determine the …

Water, Sanitation, And Citizenship: Perceptions Of Water Scarcity, Reuse, And Sustainability In Valparaiso De Goias, Brazil, Paola Andrea Gonzalez
USF Tampa Graduate Theses and Dissertations
Access to reliable water and sanitation are two important goals to improve livelihoods around the world. Providing access to improved and safe water resources that are equitable and appropriate to local needs is important to improve sustainability long-term. In addition, framing access to water and sanitation as basic human rights is often used as a rationale in developing new water, sanitation, and hygiene interventions in developing countries around the world. But not all countries consider access to safe water and sanitation as a human right. In the thesis, the politics of improving and investment in water access and sanitation provision …

Mechanism Elucidation And Inhibitor Discovery Against Serine And Metallo-Beta-Lactamases Involved In Bacterial Antibiotic Resistance, Orville A. Pemberton
USF Tampa Graduate Theses and Dissertations
The emergence and proliferation of Gram-negative bacteria expressing β-lactamases is a significant threat to human health. β-Lactamases are enzymes that degrade the β-lactam antibiotics (e.g., penicillins, cephalosporins, monobactams, and carbapenems) that we use to treat a diverse range of bacterial infections. Specifically, β-lactamases catalyze a hydrolysis reaction where the β-lactam ring common to all β-lactam antibiotics and responsible for their antibacterial activity, is opened, leaving an inactive drug. There are two groups of β-lactamases: serine enzymes that use an active site serine residue for β-lactam hydrolysis and metalloenzymes that use either one or two zinc ions for catalysis. Serine enzymes …

Road Map To Revolution? Patent-Based Open Science, Lee Petherbridge
Maine Law Review
The contemporary approach to innovation in the life sciences relies on a patent-based proprietary model. Limitations on patent rights and business concerns often focus innovation to markets where the near-term monetary rewards are highest. This is “efficient” under an austere understanding of the term, but the proprietary model can be problematic from a practical perspective because it may not focus innovation to certain deserving markets. This Article contends that the property rights conferred by patent law may still serve as a positive base for innovation directed to underserved markets. The comparatively strong rights conferred by patent law provide upstream or …

NLRP10 is a nucleotide-binding oligomerization domain-like receptor that functions as an intracellular pattern recognition receptor for microbial products. Here, we generated a Nlrp10−/− mouse to delineate the role of NLRP10 in the host immune response and found that Nlrp10−/− dendritic cells (DCs) elicited sub-optimal IFNγ production by antigenspecific CD4+ T cells compared to wild-type (WT) DCs. In response to T-cell encounter, CD40 ligation or Toll-like receptor 9 stimulation, Nlrp10−/− DCs produced low levels of IL-12, due to a substantial decrease in NF-κB activation. Defective IL-12 production was also evident in vivo and affected IFNγ production by CD4+ T cells. Upon …

Advances in allogeneic hematopoietic cell transplantation for sickle cell disease have improved outcomes, but there is limited analysis of healthcare utilization in this setting. We hypothesized that, compared to late transplantation, early transplantation (at age <10 >years) improves outcomes and decreases healthcare utilization. We performed a retrospective study of children transplanted for sickle cell disease in the USA during 2000-2013 using two large databases. Univariate and Cox models were used to estimate associations of demographics, sickle cell disease severity, and transplant-related variables with mortality and chronic graft-versus-host disease, while Wilcoxon, Kruskal-Wallis, or linear trend tests were applied for the estimates of …10>

ARUP is a reference laboratory that is part of the University of Utah. When a physician wants to run a test on a patients blood or other fluid for diagnostic purposes, there are a few options for where the test is run. Some hospitals are equipped to run many common tests. When a test exceeds what the hospital can do however, a patient’s sample is then sent to a better equipped lab, such as ARUP. ARUP is one of the largest reference laboratories in the nation, and has hundreds of thousands of samples that are tested there every week. These …

Lyme disease is a tick-borne, multi-systemic disease, caused by the bacterium Borrelia burgdorferi. Though antibiotics are used as a primary treatment, relapse often occurs after the discontinuation of antimicrobial agents. The reason for relapse remains unknown, however previous studies suggest the possible presence of antibiotic resistant Borrelia round bodies, persisters and attached biofilm forms. Thus, there is an urgent need to find antimicrobial agents suitable to eliminate all known forms of B. burgdorferi. In this study, natural antimicrobial agents such as Apis mellifera venom and a known component, melittin, were tested using SYBR Green I/PI, direct cell counting, biofilm …

Suppurative Complications Of Acute Hematogenous Osteomyelitis In Children, Jennifer J. Johnston, Cristina Murray-Krezan, Walter N. Dehority
Pediatrics Research and Scholarship
We carried out a case-control study in children with acute hematogenous osteomyelitis (AHO) with and without suppurative complications discharged from our institution over an 11-year period to test the hypothesis that abscess formation was associated with a delayed presentation to care. Of 102 children with AHO, 54 abscesses were documented in 46 patients (25 bone, 29 muscle). A delay in presentation was not associated with abscess formation (6.5 vs. 5.0 days, P=0.26). Overall, 78% of all bone abscesses were visible on initial MRI. Consistent use of MRI at presentation may identify children with suppurative complications of AHO.

High-throughput sequencing (HTS) of immunoglobulin (B-cell receptor, antibody) and T-cell receptor repertoires has increased dramatically since the technique was introduced in 2009 (1-3). This experimental approach explores the maturation of the adaptive immune system and its response to antigens, pathogens, and disease conditions in exquisite detail. It holds significant promise for diagnostic and therapy-guiding applications. New technology often spreads rapidly, sometimes more rapidly than the understanding of how to make the products of that technology reliable, reproducible, or usable by others. As complex technologies have developed, scientific communities have come together to adopt common standards, protocols, and policies for generating …

Six herds of 45 to 90 cow/calf pairs grazing on upland range were used to examine the efficacy of an injectable extended release eprinomectin parasite control on production traits, activity behavior, and efficacy against internal parasitism. In 2016, treatment cows were given a subcutaneous injection of LongRangetm. In 2017, all cows in the study were treated with a short acting Synanthictm treatment and only treatment cows were given an additional LongRangetm treatment. In both 2016 and 2017, fecal egg counts were significantly lower (P < 0.05) in eprinomectin treated cows compared to control cows. Calf gains were 4.8 kg and 8.7 kg greater (P < 0.1) for the calves of dams treated with eprinomectin compared to calves of control cows in 2016 and 2017, respectively. Activity characteristics of cattle were inconclusive, with treated cows having lower (P < 0.01) grazing and traveling times compared to control cows in 2016, and more (P < 0.01) grazing and traveling in 2017.
