Clinical and Translational Science Institute

Mitochondrial dysfunction is thought to play a significant role in neurodegeneration observed in Parkinson’s disease (PD), yet the mechanisms underlying this pathology remain unclear. Here, we demonstrate that loss of mitoNEET (CISD1), an iron–sulfur containing protein that regulates mitochondrial bioenergetics, results in mitochondrial dysfunction and loss of striatal dopamine and tyrosine hydroxylase. Mitochondria isolated from mice lacking mitoNEET were dysfunctional as revealed by elevated reactive oxygen species (ROS) and reduced capacity to produce ATP. Gait analysis revealed a shortened stride length and decreased rotarod performance in knockout mice, consistent with the loss of striatal dopamine. Together, these data suggest that …

Objective: Regular physical activity (PA) reduces the risk of disease and premature death. Knowing factors associated with PA might help reducing the disease and economic burden caused by low activity. Studies suggest that socio-cultural factors may affect PA, but systematic overviews of findings across the life course are scarce. This umbrella systematic literature review (SLR) summarizes and evaluates available evidence on socio-cultural determinants of PA in children, adolescents, and adults. Gender identity shapes the ways transgender adults experience themselves and relate to the world around them. Although research and theory suggest that gender identity is a multidimensional construct, most measures of gender identity have viewed gender as primarily a unitary construct tied to the gender binary. The aim of this study was to develop and validate the Multilayered Gender Identity Questionnaires (MGIQ), a set of measures of gender identity, in a sample of transgender adults. Purpose

Treatment of oropharyngeal squamous cell carcinoma (OPSCC) is evolving toward risk-based modification of therapeutic intensity, which requires patient-specific estimates of overall survival (OS) and progression-free survival (PFS).

Methods

To develop and validate nomograms for OS and PFS, we used a derivation cohort of 493 patients with OPSCC with known p16 tumor status (surrogate of human papillomavirus) and cigarette smoking history (pack-years) randomly assigned to clinical trials using platinum-based chemoradiotherapy (NRG Oncology Radiation Therapy Oncology Group [RTOG] 0129 and 0522). Two cAMP signaling compartments centering around adenylyl cyclase (AC) exist in human airway smooth muscle (HASM) cells, one containing ß2AR-AC6 and another containing E prostanoid receptors (EPR)-AC2. We hypothesized that different phosphodiesterase (PDE) isozymes selectively regulate cAMP signaling in each compartment. According to RNA-seq data, 18 of 24 PDE genes were expressed in primary HASM cells derived from age- and gender-matched donors with and without asthma. PDE8A was the third most abundant of the cAMP-degrading PDE genes, after PDE4A and PDE1A. Knockdown of PDE8A using shRNA evoked 2-fold greater cAMP responses to 1 DM forskolin in the presence of IBMX. …

A Duplex Endpoint Pcr Assay For Rapid Detection And Differentiation Of Leptospira Strains, D. Benacer, S.N. Mohd Zain, J.W. Lewis, M.K.N.M. Khalid, Kwai Lin Thong

INTRODUCTION: This study aimed to develop a duplex endpoint PCR assay for rapid detection and differentiation of Leptospira strains. METHODS: Primers were designed to target the rrs (LG1/LG2) and ligB (LP1/LP2) genes to confirm the presence of the Leptospira genus and the pathogenic species, respectively. RESULTS: The assay showed 100% specificity against 17 Leptospira strains with a limit of detection of 23.1pg/µl of leptospiral DNA and sensitivity of 103 leptospires/ml in both spiked urine and water. CONCLUSIONS: Our duplex endpoint PCR assay is suitable for rapid early detection of Leptospira with high sensitivity and specificity.

Recently, geologists in Southern Nevada discovered new deposits of naturally occurring asbestos and microscopic fibers in rocks and soil. The danger is that inhaling them can lead to mesothelioma. One problem is that this rare cancer often takes decades to manifest. This discovery abruptly stalled a highway project near Las Vegas. Due to this condition, management developed numerous protocols to keep workers safe. Between October 2013 and July 2016, over 156,000 children travelling without their guardians were apprehended at the U.S.–Mexico border and transferred to the care of the Office of Refugee Resettlement (ORR). During that same period, ORR placed over 123,000 unaccompanied migrant youth – predominantly from Central America – with a parent or other adult sponsor residing in the U.S. Following placement, local communities are tasked with integrating migrant youth, many of whom experience pre- and in-transit migration traumas, family separation, limited/interrupted schooling, and unauthorised legal status, placing them at heightened risk for psychological distress, academic disengagement, maltreatment, and human trafficking. …

Previous investigations have revealed a range of cognitive, physiological, and psychological benefits following mindfulness training in young, middle-aged, and older adults. The aim of this thesis was to provide new insights into three issues that have not been adequately addressed in the extant literature. First, the application and integration of mindfulness training within conventional educational contexts is limited. In study 1, I provide evidence of the utility and effectiveness of mindfulness training incorporated as a part of traditional college curriculum. The Impact Of Copyright Exceptions For Researchers On Scholarly Output, Michael Palmedo

High prices restrict access to academic journals and books that scholars rely upon to author new research. One possible solution is the expansion of copyright exceptions allowing unauthorized access to copyrighted works for researchers. I test the link between copyright exceptions for health and science researchers and their publishing output at the country-subject level. I find that scientists residing in countries that implement more robust research exceptions publish more papers and books in subsequent years. This relationship between copyright exceptions and publishing is stronger in lower-income countries, and stronger where there is stricter copyright protection of existing works.

This qualitative, narrative study examined the lived experiences of transgender students at four-year institutions in the South. The college transition process and academic and social integration for transgender students was explored through the frameworks of Transition Theory (Schlossberg, 1995), Minority Stress Theory (Meyer, 1995; Breslow, Brewster, Velez, Wong, Geiger, & Soderstrom, 2015), and Academic and Social Integration theories (Tinto, 1975; Braxton, Hirschy, & McClendon, 2004). Substance abuse counseling has many counselors and supervisors who are in recovery from a personal history of substance abuse. Approximately 37% of supervisors in the substance abuse field reported being in personal recovery (Eby, Burke, & Birkelbach, 2009). Little is known about how a clinical supervisor’s personal recovery influences his or her clinical supervision.
