Space flight exposes astronauts to stressors that alter the immune response, rendering them vulnerable to infections and diseases. In this study, we aimed to determine the levels of inflammasome activation in the brains of mice that were housed in the International Space Station (ISS) for 37 days. C57BL/6 mice were launched to the ISS as part of NASA’s Rodent Research 1 Mission on SpaceX-4 CRS-4 Dragon cargo spacecraft from 21 September 2014 to 25 October 2014. Dissected mouse brains from that mission were analyzed by immunoblotting of inflammasome signaling proteins and Electrochemiluminescence Immunoassay (ECLIA) for inflammatory cytokine levels. Our data …

Infrared Laser Ablation Of Biomolecules From Formalin-Fixed Tissue, Blessing Chisom Egbejiogu
LSU Doctoral Dissertations
In the research described in this dissertation, a high-precision infrared laser ablation microsampling method was utilized for spatially resolved proteomic and lipidomic analyses of formalin fixed paraffin embedded (FFPE) tissue, a widely used archival sample in biomedical research. Despite its extensive use, FFPE tissue presents challenges for proteomic analysis due to protein cross-linking and modifications introduced during fixation, which can hinder extraction and identification. Developing a streamlined high-resolution method to recover biomolecules from FFPE samples is critical for advancing biomarker discovery, retrospective studies, and multi-omics research. To address these challenges, this study employs an infrared (IR) laser ablation microdissection system …

What makes a wine exceptional enough to score a perfect 10 from experts? This study explores a data-driven approach to identify the ideal physicochemical composition for wines that could achieve this highest possible rating. Using a dataset of 11 measurable attributes, including alcohol, sulfates, residual sugar, density, and citric acid, for wines rated up to a maximum quality score of 8 by expert tasters, we sought to predict compositions that might enhance wine quality beyond current observations. Our methodology applies a second-degree polynomial ridge regression model, optimized through an exhaustive evaluation of feature combinations. Furthermore, we propose a specific chemical …

Antimicrobial resistance (AMR) is a major global health problem. The growing number of antibiotic-resistant microbes needs the development of novel antibiotics. Nanotechnology shows potential for overcoming bacterial resistance. Nanoparticles, particularly those around 100 nm, display higher antibacterial activity by using methods that differ from traditional antibiotics. When particle size reduces, materials such as zinc and silver improve their antibacterial action. There are several techniques to evaluate nanoparticle antibacterial activity, each having merits and downsides. This study critically evaluates selected literature, identifies bactericidal mechanisms, and discusses assessment procedures, emphasizing the importance of innovative approaches to bacterial infections.

Systems and methods for automated stereology using deep learning are disclosed. The systems include an update in the form of a semi-automatic approach for ground truth preparation in 3D stacks of microscopy images (disector stacks) for generating more training data. The systems also present an exemplary disector-based MIMO framework where all the planes of a 3D disector stack are analyzed as opposed to a single focus-stacked image (EDF image) per stack. The MIMO approach avoids the costly computations of 3D deep learning-based methods by using the 3D context of cells in disector stacks; and prevents stereological bias in the previous …

The removal of color arising from water-soluble dyes in wastewaters is necessary to counter the threat to health. Because of thedifficulty in elimination of these dyes by conventional methods, their photodegradation using photosensitizers such as phthalocyanines(Pcs) has been employed recently. When compared to peripherally substituted derivatives, silicon Pcs are reported to be morebiocompatible with low toxicity. Thus a new silicon Pc substituted with two quaternized dimethylamino phenoxy units (compound 1)and its quaternized derivative (compound 2) were synthesized to test its photocatalytic ability employing the dyes methylene blue (MB),eosin B (EB), erythrosine (ERB), sulforhodamine B (SRB), and brilliant blue FCF (BRB), …

Couples who may have been unsuccessful in their efforts to conceive naturally have found ways to parenthood through assisted reproductive technologies. In vitro fertilization is one of the most complex processes these couples pursue, and it involves creating an embryo outside of the human body and then inserting it into the woman with the hopes of successful implantation. Under the circumstances, many embryos may be created and then frozen—or cryopreserved—to prolong the availability of the process to the couple for months or even years. However, a conflict can arise if the couple gets divorced, which begs the question: what happens …

Task-free brain activity affords unique insight into the functional structure of brain network dynamics and has been used to identify neural markers of individual differences. In this work, we present an algorithmic optimization framework that directly inverts and parameterizes brain-wide dynamical-systems models involving hundreds of interacting neural populations, from single-subject M/EEG time-series recordings. This technique provides a powerful neurocomputational tool for interrogating mechanisms underlying individual brain dynamics ("precision brain models") and making quantitative predictions. We extensively validate the models' performance in forecasting future brain activity and predicting individual variability in key M/EEG metrics. Last, we demonstrate the power of our …

Methods for assessing compound identification confidence in metabolomics and related studies have been debated and actively researched for the past two decades. The earliest effort in 2007 focused primarily on mass spectrometry and nuclear magnetic resonance spectroscopy and resulted in four recommended levels of metabolite identification confidence─the Metabolite Standards Initiative (MSI) Levels. In 2014, the original MSI Levels were expanded to five levels (including two sublevels) to facilitate communication of compound identification confidence in high resolution mass spectrometry studies. Further refinement in identification levels have occurred, for example to accommodate use of ion mobility spectrometry in metabolomics workflows, and alternate …

The azomethine ligand and its metal complexes have been identified by using mass spectrometry, 1H and 13C NMR spectroscopy, C, H, N, and S elemental analysis, X-ray diffraction (XRD), (FESEM) microscopy, atomic absorbance spectroscopy. UV-visible spectroscopy, Fourier-transform infrared spectroscopy (FT-IR), magnetic susceptibility, and ionic conductance analysis. The chemical composition and additional measurement data of the metal complexes validate the stoichiometry of the metal: ligand ratio types that include; [M (L)], …

Drosophila melanogaster Meigen (Diptera: Drosophilidae) was used as a model organism and fed on an artificial diet with different clinically important drugs under laboratory conditions. The effects of fluconazole, acyclovir, and piperazine on the survivorship, development, adult longevity, fecundity, and hatching rate of the insect were investigated. The results showed that the highest concentration of fluconazole (1250 mg/L) significantly decreased both the rate of larvae reaching the 3rd instar and the rate of pupation from 93.00 ± 2.95% to 11.00 ± 2.59% (χ2 = 134.696; df = 1; p = 0.0001). The highest concentration of acyclovir (200 mg/L) significantly reduced …

We shall investigate Gompertz dynamic equations within the context of time scales calculus, by exploring the mathematical foundations and applications of the Gompertz model, which is commonly used to describe growth phenomena in various fields such as biology and economics. This research seeks to analyze the Gompertz cumulative distribution functions (CDF) and probability density functions (PDF) across different time scales, including the real numbers R and integer multiples hN. Probability techniques will be used to derive the CDF and PDF associated with the Gompertz dynamic equations, and we will examine how varying the time scale impacts the characteristics …

Applications need to be modeled for analyses. With the availability of many alternate data Models, choosing one needs to be done carefully by considering the complexity of data to be modeled and its analyses requirements. Social networks and other newer applications are primarily relationship-oriented and also have multiple types of entities and relationships. Although simple and attributed graphs have been used historically for modeling these applications, recently, multilayer networks (or MLNs) have been shown to be more effective in preserving the semantics of the application better and further provide flexibility of analyses. However, choice of MLN as a data model …
