Stochastic kriging (SK) and stochastic kriging with gradient estimators (SKG) are popular approaches to approximate complex simulation models because of their ability to replace the expensive simulation outputs by metamodel values. Obtaining an accurate SK/SKG metamodel is highly desirable in practice. This dissertation studies the monotonicity properties of the mean squared error (MSE) of optimal SK and SKG predictors. In particular, we show that in both SK and SKG, the MSEs of the corresponding optimal predictors are non-increasing functions of the numbers of design points. Based on these findings, we design an adaptive sequential sampling approach to obtain SK/SKG predictors …

Recent developments in next generation DNA sequencing and optical restriction mapping involve manipulation of DNA molecules on surfaces. Here we propose a novel method that can control the adsorbed DNA density on polymer surfaces by applying an electric field. The efficiency of deposition was optimized with respect to DNA concentration in solution, electric field type and electric field strength. Enhancement of adsorption density of greater than twenty-fold was found. In addition, DNA molecules are fragmented on a polymer surface by soft lithography. Several experimental conditions have been tested to optimize the polydimethylsiloxane (PDMS) stamp fabrication and DNA cutting method. Fragmented …

Nano-particles are great additives to the thermal properties of the polymers, however, they sometimes have some disadvantages on the mechanical properties. The mixing of polymers and nano-particles such as cloisite clays, graphene, melamine polyphosphate and molybdenum disulfide, are mostly physical reactions between them. Therefore, the dispersion of the nano-particles inside the polymers is very important. As for the frame retardant of the polymers, the additives are used as three most important components during the mixing, i.e. the acid source, the carbonization agent (or char forming agent), and a blowing agent. As Rebecca Whisnant has noted, notions of “national…and…bodily (especially sexual) sovereignty are routinely merged in rhetoric and metaphor.†(2008:155) Departing from this observation, this project explores the ‘existential infrastructure’ of the ‘logic of sovereign integrity’ in which personhood is thought as analogous to jurisdiction over a bounded territorial enclosure. It traces the philosophical history of this logic, and unfolds the post-Heideggerian French critique of sovereignty as an analysis of how sovereign imperatives are animated by a drive to deny the vulnerabilities of constitutive ontological relation, and are thus implicated in violent cycles of disavowal and appropriation. This dissertation focuses on the Oper Frankfurt, a musical institution that has played an influential role in the development of operatic staging since the 1980s. Through an examination of productions staged at the house in two different periods (1979 – 1987 and 2010 – 2015), I pay particular attention to the ways in which these contemporary stagings contribute to a discourse about recent opera— particularly in Germany— and what has frequently been called Regietheater. This polemical term, often translated as “director’s theater,†connotes a provocative, theatrically experimental, even massively reenvisioned reading of an operatic work, propelled by a visionary stage …

The evidence for dark matter is overwhelming. We know where the dark matter is, how much there is, but we do not know what type of particle it consists of. Experimental efforts, as well as theoretical, have tried to shed light on its identity. On the experimental side, “indirect detection†aims to detect stable leptons, baryons, or photons resulting from the annihilation or decay of dark matter particles in outer space. The mammalian target of rapamycin (mTOR) signaling pathway, which integrates extracellular and intracellular signals, plays a key role in cell metabolism, growth and survival; therefore, the proteins in the signaling pathway must be tightly regulated in order to maintain cellular homeostasis. Conversely, dysregulation of key components of mTOR signaling pathway leads to increased cellular growth and proliferation, which is implicated in many human cancers. Protein kinase C (PKC) is a family of serine/ threonine kinases that regulate many cellular processes including proliferation and cell survival. This research deals with a general kinematic synthesis problem of how to generate the motion of a spatial platform subject to one or more spherical con- straints. Each spherical constraint can be modeled by a SS dyad, which is a spatial two-link kinematic chain connected by two spherical (S) joints. It defines a five-degree-of-freedom motion of the end link such that a point stays on the surface of a sphere. This thesis deals with verification of complex systems, which involve probabilistic choices. In total, we explore three interrelated problems. First, we explore probabilistic extensions of mu-calculus. GPL extends mu-calculus by having all the probabilistic choices made first; this keeps the model checking procedure decidable for any property. We extend GPL to a logic, XPL, which is undecidable, in general. We define a syntactic property of an XPL formula, separability, as a sufficient condition for model checking. As computers become faster and more parallel, the number of computations a process executes is becoming too simplistic a predictor of performance. In many circumstances, modern algorithmic research can no longer rely on the assumption that each computation roughly corresponds to a set amount of time. For example, large-scale computers may spend far more time moving data around than performing computation, in which case minimizing data transfers is the best way to improve wall-clock performance. This thesis concerns simulations of Inertial Confinement Fusion. Inertial confinement is carried out in a large scale facility at National Ignition Facility. The experiments have failed to reproduce design calculations, and so uncertainty quantification of calculations is an important asset. Uncertainties can be classified as aleatoric or epistemic. This thesis is concerned with aleatoric uncertainty quantification. Among the many uncertain aspects that affect the simulations, we have narrowed our study of possible uncertainties. Turbulent mixing from hydrodynamical instabilities, such as Richtmyer-Meshkov (RMI) and Rayleigh-Taylor (RTI) instabilities, plays a critical role in numerous applications ranging from performance degradation in inertial confinement capsules to supernova explosions. At high Reynolds numbers (Re), for which experimental data is not available, numerical simulations are paramount in studying these instabilities. However, the algorithmic differences due to differences in numerical modeling often give solutions that are converged but not unique, that is, different codes converge to different solutions. Since the beginning of the 21st century, the jun’ai (pure-love) genre has flourished in Japan, both in works of popular literature and in film. This phenomenon coincides with a time when the country is seen by the media as being characterized by soshitsukan (sense of loss). In jun’ai films, the heroine is often the object of loss. This theme of loss in jun’ai therefore resonates with the Japanese social context, but this connection has not yet been fully elucidated. The simulation campaign to model Inertial Confinement Fusion (ICF) capsules has come a long way since experiments with the potential to achieve net energy gain began in 2010. A remaining area of uncertainty is the modeling of instability growth and mixing of ablator material into the core, degrading the capsules. In this dissertation, I study the intersection of community gardens with gentrification. In much of the gentrification scholarship, gentrification was studied from the macro level and was thought to result from changes in the housing market, hence being viewed as a top-down process in which the elite revitalized decaying urban neighborhoods by bringing capital into these areas and building higher end housing and services or was driven by consumer choices of privileged individuals (Smith 1979; Ley 1981).
