Recent developments in next generation DNA sequencing and optical restriction mapping involve manipulation of DNA molecules on surfaces. Here we propose a novel method that can control the adsorbed DNA density on polymer surfaces by applying an electric field. The efficiency of deposition was optimized with respect to DNA concentration in solution, electric field type and electric field strength. Enhancement of adsorption density of greater than twenty-fold was found. In addition, DNA molecules are fragmented on a polymer surface by soft lithography. Several experimental conditions have been tested to optimize the polydimethylsiloxane (PDMS) stamp fabrication and DNA cutting method. Fragmented …

Interpenetrations (or entanglements) between chemically identical polymers are important parameters in fundamental physics and industrial applications. However, little is known about the interaction between free polymer chains and chemically identical, but otherwise highly adsorbed chains on solid surfaces. Here we report an unusual interaction between free chains and chemically identical flattened chains which have many surface-segmental contacts with solids. Based on the mechanical adhesion measurements, we found the 2.5 nm-thick equilibrium polyethylene-oxide (PEO) flattened layer show no adhesion with the PEO melts even at T >Tm. Liquid contact angle measurements further revealed that the flattened layer has the same macroscopic …

However, the relative high density of structural defects in those crystals has hampered the performance of devices built on them. In this work curing process for glass fiber reinforced polymer composite laminates is studied. Curing temperature, time and pressure were selected as the cure parameters for optimization. Central composite design (CCD) of design of experiment (DOE) approach was adopted for conducting the experiments. Flexural strength and flexural modulus of the composite laminate were the response measures to select the final cure parameters. Analysis of variance (ANOVA), surface plots and contour plots clearly demonstrate that temperature, time and pressure contribute greatly to the response functions. This establishes two models that can be used to predict the mechanical properties of composite laminated …

Nanocrystalline metals have useful mechanical properties such as high strength, improved wear resistance, and longer fatigue life; however, they are relatively unstable – grain boundary doping is a viable method towards stabilization [1-3]. After comparing the work of Murdoch and Schuh, that used a thermodynamic model to estimate grain boundary segregation enthalpy to the experimental work of Umbrajkar et al. | it was realized that mechanical alloying is a processing route for grain boundary stabilization [10,11]. This thesis presents a Synchrotron White Beam X-ray Topography study of dislocations in 4H- SiC single crystals. Basic introduction is given including the crystal structure and atomic arrangement, crystal growth techniques, and the dislocation types. The commonly observed dislocations, including Micropipes (MPs), Basal Plane Dislocations (BPDs), Threading Screw Dislocations (TSDs), Threading Edge Dislocations (TEDs), Threading Mixed Dislocations (TMDs), in 4H-SiC single crystals have been reviewed. There is an increasing need for precise molecular detection as a diagnostic tool for early identification of diseases, pathogens, and abnormal protein levels in the body. Typical chemical analytical methods are generally costly, unstable, and time-consuming. Molecular imprinting (MI) technique,based on the “lock and key model†, could be a simple method to overcome those shortcomings. In this study, a self-assembled monolayer (SAM) was employed as a platform to fabricate MI biosensor for detection of macromolecules. Superconducting materials are promising for future applications on energy transport and storage. The key properties for superconductors are critical temperature Tc, critical current density Jc and upper critical field Hc2. In this dissertation, detailed transport studies were performed on two superconducting materials: FeSe0.5Te0.5 thin films and the second generation YBa2Cu3O7-δ coated conductors, in order to determine what limits the broad applications of these superconductors. High quality of FeSe0.5Te0.5 thin films were grown by pulsed laser deposition. Low temperature oxygen annealing and proton irradiation were conducted on the films, as the post-treatment techniques in order to improve the transport performance. Jc …

Infinitely renormalizable quadratic polynomials have been heavily studied. In the context of quadratic-like renormalization, one may try to prove the existence of a priori bounds, a definite thickness for the annuli corresponding to the renormalizations. In 1997, M. Lyubich showed that a priori bounds imply local connectivity of the Julia set and combinatorial rigidity for the corresponding quadratic polynomial. In a paper from 2006, J. Kahn showed that infinitely renormalizable quadratic polynomials of bounded primitive type admit a priori bounds. In 2002, H. Inou generalized some of the polynomial-like renormalization theory to polynomials of higher degree with several critical points. …

The moduli spaces of pseudo-holomorphic maps into an almost Kahler manifold are fundamental to Gromov-Witten theory. It has been speculated since the early days of Gromov-Witten theory that these moduli spaces contain natural closed subspaces, whenever the genus is positive, that also give rise to curve-counting invariants. This speculation was confirmed in genus 1 over a decade ago. In this dissertation, we describe a natural subspace of every moduli space of genus 2 pseudo-holomorphic maps that has strata of the correct virtual dimension to give rise to curve-counting invariants. In this dissertation, we describe a new continuity path introduced by X. Chen aiming to attack the existence problem of constant scalar curvature problem via a direct PDE approach. The path connects the solution of J-equation to the constant scalar curvature Kaehler metric. We will present various openness results about the continuity path. Let $S\to C$ be a smooth projective surface with numerically trivial canonical bundle fibered onto a curve. We prove the multiplicativity of the perverse filtration with respect to the cup product on $H^*(S^{[n]},\mathbb{Q})$ for the natural morphism $S^{[n]}\to C^{(n)}$. We also prove the multiplicativity for five families of Hitchin systems obtained in a similar way and compute the perverse numbers of the Hitchin moduli spaces. We show that for small values of $n$ the perverse numbers match the predictions of the numerical version of the de Cataldo-Hausel-Migliorini $P=W$ conjecture and of the conjecture by Hausel-Letellier-Villegas. | 48 pages

As healthcare grows more advanced, the next step is an individualized approach to patient care. Skeletal tumor resection provides complex challenges for surgeons. The goal of providing an accurate wide resection in sarcomas from preoperative plans while trying to preserve as much functionality remains a difficult task. Tinnitus means objective somatosounds, and subjective auditory or sensorineural perception of noise without external physical sounds. There are several tinnitus treatments, but it is difficult to ascertain how otorhinolaryngologists rank the alternatives of treatments due to the insufficiency of available evidence and standards. A hierarchical decision model (HDM), a popular multi-criteria decision analysis (MCDA) method, can help to implement a comprehensive assessment of tinnitus treatments and rational decision-making to select the appropriate tinnitus treatments. Richard Foreman’s Lava (1989) is chosen to be studied in terms of stream of consciousness as one of the representative plays of Ontological-Hysteric Theatre. Given the nature of stream of consciousness, continuation of experiences, this play is analyzed through its manifestation process itself. The stream of consciousness is found to consist of three main features; Fragmentation, Mismatch, and Rituality. These features are not only belong to Richard Foreman’s Lava but also have been descended and regarded as the main characteristics of avant-garde theaters.
