Characterizing Nonmodel Microorganisms And Reconstructing Catabolic Pathways For Bio-Productions, Whitney Hollinshead Dec 2016

Characterizing Nonmodel Microorganisms And Reconstructing Catabolic Pathways For Bio-Productions, Whitney Hollinshead

McKelvey School of Engineering Graduate Student Theses & Dissertations

In the 1970s, spikes in oil prices sparked early interests into first-generation biofuels (i.e., the production of ethanol from corn). Four decades later, biofuel manufacturing has under-performed as developed industrial strains suffer from poor performance and high costs of feedstocks. While the advancement in molecular biology has greatly benefited pathway engineering, synthetic biology hosts still cannot overcome the obstacles for profitable microbial bio-production. One approach may be to study non-model organisms and employ their metabolic features towards an improved bio-production pipeline. Here, this thesis aims to develop 13C-metabolism analysis tools to characterize novel organisms and re-program cell metabolism using novel …

Genetic Circuits For Transcriptional Regulation In Synechocystis Sp. Pcc 6803, Cheryl M. Immethun Dec 2016

Genetic Circuits For Transcriptional Regulation In Synechocystis Sp. Pcc 6803, Cheryl M. Immethun

McKelvey School of Engineering Graduate Student Theses & Dissertations

Microbial biosynthesis has produced a variety of complex compounds using processes that are more environmentally-friendly than many conventional methods. The most common hosts are heterotrophs, which require the addition of an organic carbon source; while cyanobacteria possess many traits that make them a more sustainable biotechnology platform. As phototrophs, cyanobacteria can employ sunlight and carbon dioxide to create many value-added compounds. A wealth of tools has been developed to engineer the commonly used heterotrophs for higher yields and titers; yet, few synthetic biology tools have been designed for cyanobacteria. Furthermore, many of the tools created for heterotrophs do not function …

High-Throughput Exploration Of Glass Formation Via Laser Deposition And The Study Of Heterogeneous Microstructure In A Bulk Metallic Glass Alloy, Peter Te-Chuan Tsai Dec 2016

High-Throughput Exploration Of Glass Formation Via Laser Deposition And The Study Of Heterogeneous Microstructure In A Bulk Metallic Glass Alloy, Peter Te-Chuan Tsai

McKelvey School of Engineering Graduate Student Theses & Dissertations

Bulk metallic glasses are a relatively novel class of engineering alloys characterized by a "disordered" atomic structure devoid of long-range translational symmetry. Compared to crystalline alloys, the confluence of metallic bonding and amorphous structure imbues bulk metallic glasses with a unique set of properties that makes them particularly attractive for a wide variety of structural applications. Such properties include exceptional yield strengths, high elastic resilience, resistance to corrosion, and in particular, the unparalleled ability among metals to be thermoplastically formed across a wide range of length scales when heated above the glass transition temperature. Formation of metallic glass from a …

Novel Contrasts In Photoacoustic Tomography, Ruiying Zhang Dec 2016

Novel Contrasts In Photoacoustic Tomography, Ruiying Zhang

McKelvey School of Engineering Graduate Student Theses & Dissertations

Photoacoustic tomography (PAT) combines rich optical contrast and high ultrasonic resolution in optically scattering tissue at depths. Taking advantage of its 100% sensitivity to optical absorption, PAT has been widely applied to structural, functional and molecular imaging, with both endogenous and exogenous contrasts, at superior depths than pure optical methods. This dissertation explores novel absorption contrast mechanisms of PAT based on optical/thermal patterns, endogenous cellular chromophores, nanoparticles, small-molecule dyes and genetically-encoded proteins. With these novel contrasts, the proof-of-concept applications of PAT have been extended to include homogenous flow measurements, targeted angiogenesis imaging and therapy, label-free white blood cell imaging, 3D-whole-organ …

Applications Of Aerosol Technologies In The Silicon Industry, Miguel Vazquez Pufleau

McKelvey School of Engineering Graduate Student Theses & Dissertations

In this dissertation, the focus was on two unit operations that have the potential to significantly lower the cost and energy required for refining silicon. First, the removal of carbon from silicon kerf (sawing dust from wafering) was examined to enable its recycling. Second, silane pyrolysis was studied to better understand the dynamics of the initial stages of silicon aerosol formation and growth.

Developing High Purity Embryonic Stem Cell-Derived V2a Interneurons For In Vitro Investigation And Transplantation Following Spinal Cord Injury, Nisha Iyer Dec 2016

Developing High Purity Embryonic Stem Cell-Derived V2a Interneurons For In Vitro Investigation And Transplantation Following Spinal Cord Injury, Nisha Iyer

McKelvey School of Engineering Graduate Student Theses & Dissertations

Functional recovery following spinal cord injury has been attributed to plasticity in local interneuron populations, which are able to create novel circuits that bypass the inhibitory lesion to activate pre-existing motor pathways. Cell replacement strategies and growth factor delivery platforms often attempt to enhance these natural mechanisms of regeneration. The work of this thesis sought to overcome the major obstacle to manipulating specific interneurons for therapy: an inability to obtain large, enriched interneuron subpopulations for drug screening and transplantation. We focused on the generation of V2a interneurons, which normally coordinate a wide range of locomotor tasks in both the spinal …

An Assessment Of Data Related To Inspections Of Risk Factors For Public Swimming Pools, Shanita Shack, Maurice Redmond, R. Christopher Rustin

Environmental Health Sciences: Faculty Publications

Background: The Georgia Department of Public Health (DPH) is mandated to ensure that public swimming pools are safe for those who use them. This mandate is carried out by the DPH and local environmental health professionals through regulations and inspections. In 2015, legislation was introduced proposing to reduce the authority of the DPH to inspect certain pool types (apartments, subdivision, condominiums) and thus reduce regulatory protections in place for swimmers. To ensure that the DPH had current information on the risks associated with pools, the EH team, with assistance from a graduate student, analyzed inspection data to evaluate risk factors …

Explicit Bases Of Motives Over Number Fields With Application To Feynman Integrals, Yu Yang Dec 2016

Explicit Bases Of Motives Over Number Fields With Application To Feynman Integrals, Yu Yang

Arts & Sciences Graduate Student Theses and Dissertations

Chapter 1 contains background material on higher Chow groups, KLM formula and Feynman integrals. In Chapter 2, we construct explicit bases for these extension classes mapping to zeta values. In Chapter 4, we study the Feynman integral of the three spoke wheel graph, reinterpret it as an image of regulator using higher Abel-Jacobi maps and theoretically prove that it is a rational multiple of zeta three. In Chapter 5, a reflexive graph polytope based on the graph polynomial is constructed. In Chapter 6, to generalize the results beyond wheel with three spokes, a criterion is given on the vanishing of …

Development Of A One-Equation Eddy Viscosity Turbulence Model For Application To Complex Turbulent Flows, Timothy Wray Dec 2016

Development Of A One-Equation Eddy Viscosity Turbulence Model For Application To Complex Turbulent Flows, Timothy Wray

McKelvey School of Engineering Graduate Student Theses & Dissertations

Computational fluid dynamics (CFD) is routinely used in performance prediction and design of aircraft, turbomachinery, automobiles, and in many other industrial applications. Despite its wide range of use, deficiencies in its prediction accuracy still exist. One critical weakness is the accurate simulation of complex turbulent flows using the Reynolds-Averaged Navier-Stokes equations in conjunction with a turbulence model. The goal of this research has been to develop an eddy viscosity type turbulence model to increase the accuracy of flow simulations for mildly separated flows, flows with rotation and curvature effects, and flows with surface roughness. It is accomplished by developing a …

A Transactional Model And Platform For Designing And Implementing Reactive Systems, Justin Ross Wilson Dec 2016

A Transactional Model And Platform For Designing And Implementing Reactive Systems, Justin Ross Wilson

McKelvey School of Engineering Graduate Student Theses & Dissertations

A reactive program is one that has "ongoing interactions with its environment." Reactive programs include those for embedded systems, operating systems, network clients and servers, databases, and smart phone apps. Reactive programs are already a core part of our computational and physical infrastructure and will continue to proliferate within our society as new form factors, e.g. wireless sensors, and inexpensive (wireless) networking are applied to new problems.

Asynchronous concurrency is a fundamental characteristic of reactive systems that makes them difficult to develop. Threads are commonly used for implementing reactive systems, but they may magnify problems associated with asynchronous concurrency, as …

Knowledge And Fourth Amendment Privacy, Matthew Tokson Dec 2016

Knowledge And Fourth Amendment Privacy, Matthew Tokson

Northwestern University Law Review

This Article examines the central role that knowledge plays in determining the Fourth Amendment’s scope. What people know about surveillance practices or new technologies often shapes the “reasonable expectations of privacy” that define the Fourth Amendment’s boundaries. From early decisions dealing with automobile searches to recent cases involving advanced information technologies, courts have relied on assessments of knowledge in a wide variety of Fourth Amendment contexts. Yet the analysis of knowledge in Fourth Amendment law is rarely if ever studied on its own.

This Article fills that gap. It starts by identifying the characteristics of Fourth Amendment knowledge. It finds, …

Biogenetically Inspired Total Synthesis Of Lingzhiol. Design And Synthesis Of Axially Chiral N-Heterocyclic Carbene Catalysts., Krishna Sharmah Gautam

Arts & Sciences Graduate Student Theses and Dissertations

A stereo- and enantio-selective synthesis of lingzhiol has been achieved, mimicking a proposed biogenetic pathway via Brønsted acid-catalyzed semipinacol rearrangement of a 2,3-epoxy alcohol. A new and improved method was developed for alkylation of anilines using a domestic microwave oven. The resulting racemic chiral anilines were resolved via classical resolution and transformed into axially chiral thiazolylidene-based NHC organocatalysts, which were applied to asymmetric benzoin condensation.

Investigating The Life-Cycle Of Photosystem Ii Using Mass Spectrometry, Daniel Avram Weisz Dec 2016

Investigating The Life-Cycle Of Photosystem Ii Using Mass Spectrometry, Daniel Avram Weisz

Arts & Sciences Graduate Student Theses and Dissertations

Photosystem II (PSII) is a protein complex found embedded in the thylakoid membranes of all organisms that perform oxygenic photosynthesis. PSII converts sunlight into chemical energy, filling our atmosphere with molecular oxygen in the process and supporting nearly all life on Earth. PSII undergoes frequent light-induced damage as an unavoidable result of the electron transfer reactions it catalyzes. When damaged, PSII is disassembled, repaired, and reassembled in an intricate, tightly regulated process. The structure and mechanism of function of active PSII are relatively well-understood, due to the available crystal structures of the active complex and many years of biochemical and …

On Control Systems Of The Brain: A Study Of Their Connections, Activations, And Interactions, Haoxin Sun

McKelvey School of Engineering Graduate Student Theses & Dissertations

Implementation of daily functions in humans crucially relies on both the bottom-up moment-to- moment processing of relevant input and output information as well as the top-down controls that instantiate and regulate goal-directed strategies. The current dissertation focuses on different systems of brain regions related to task control. We are interested in investigating, in detail, some of the basic activity patterns that different control systems carry during simple tasks, and how differences in activity patterns may shed new insight onto the distinctions among the systems' functional roles. In addition, carefully coordinated interactions between brain regions specialized for control-related activity and regions …

Courtly Love And Its Counterparts In The Medieval Mediterranean, Robin William Girard Dec 2016

Courtly Love And Its Counterparts In The Medieval Mediterranean, Robin William Girard

Arts & Sciences Graduate Student Theses and Dissertations

At least since Stendhal's De l'Amour, scholars have argued that the theories of courtly love expressed in the Old French literature of the High Middle Ages were inspired by or drew upon elements of Islamicate culture. In this dissertation, I explore the theories of love represented in works of literature, lover's manuals and medical treatises composed in and around the late twelfth–century court of Marie de Champagne, which I read in conjunction with contemporaneous works from the Islamicate world, ranging from Al‑Andalus to Persia, in order to show how these works reflect the common cultural orientation of the medieval Mediterranean. …

A Proposed Imaging System To Spatially And Temporally Monitor Unburned Hydrocarbons In Jet Engine Exhaust, Emily E. Berkson Dec 2016

A Proposed Imaging System To Spatially And Temporally Monitor Unburned Hydrocarbons In Jet Engine Exhaust, Emily E. Berkson

Theses

The local air quality surrounding airports is significantly impacted by ground-based airplane operations, such as landing, taxiing, idling, and takeoff. In recent years, studies performed across the country revealed that people living near major airports suffer from increased cancer risks and higher likelihoods of developing respiratory issues. Jet engine emissions have been monitored extensively by direct probing of the exhaust, but there are currently no inexpensive, portable, and unobtrusive methods with which to study exhaust plume dynamics. This study assesses the feasibility of designing a multispectral imaging system to track the spatial-temporal nature of a jet engine plume. Ideally, the …

Harvesting A Sustainable Energy Future: Examining The Effect Of Chemical Composition On The Electromechanical Properties Of Polymer Gel Beads, Kaushik Kudtarkar Dec 2016

Harvesting A Sustainable Energy Future: Examining The Effect Of Chemical Composition On The Electromechanical Properties Of Polymer Gel Beads, Kaushik Kudtarkar

Theses

Clean energy is required to ensure global prosperity and economic growth. Increased industrialization is expected to increase energy demand by 50% by 2035. This will result in increased air pollution. Carbon dioxide emissions have been linked to global warming and other pollution related problems. These emissions can be reduced by recapturing the waste energy in the form of heat, vibration, and friction.

Common applications like transportation can lose as much as 78% of the energy they generate. Energy harvesters can be used to recapture energy lost through vibration, heat, etc. This recaptured energy will be reused and hence, we don’t …

Image Politics: U.S. Aid To The Spanish Republic And Its Refugees, Sebastiaan Faber Dec 2016

Image Politics: U.S. Aid To The Spanish Republic And Its Refugees, Sebastiaan Faber

Faculty & Staff Scholarship

The Spanish Civil War marks an important chapter in the development of non-governmental refugee aid as we know it. But studying the case of U.S. aid for Spanish Republican refugees also brings into focus some of the tensions and contradictions associated with the concept of humanitarianism, including the role played by documentary images. What were the ideals that moved those who fought against fascism or aided antifascist refugees in the 1930s and ‘40s? Were they properly political, or did they transcend politics, becoming humanitarian? The distinction is crucial to understanding the efforts undertaken to aid Spanish Civil War refugees and …

The Second Quantized Approach To The Study Of Model Hamiltonians In Quantum Hall Regime, Li Chen Dec 2016

The Second Quantized Approach To The Study Of Model Hamiltonians In Quantum Hall Regime, Li Chen

Arts & Sciences Graduate Student Theses and Dissertations

The study of model Hamiltonians, whose exact ground states are well known quantum Hall states, have been investigated solely in the literature from the point of view of first quantized approach in the last few decades. However, a second quantized approach to such Hamiltonians has not been taken in the same measure. In this dissertation, we study Haldane pseudopotential, which is the parent Hamiltonian of 1/M Laughlin state, and Trugman-Kivelson pseudopotential, which is the parent Hamiltonian of 2/5 unprojected Jain state. We find that the study of these Hamiltonians in their second quantized forms not only reproduces all properties of …

Modern Assessment Of The High-Energy Background Environment At Small Atmospheric Depths Using The X-Calibur X-Ray Polarimeter And Its Implications, Rashied Baradaran Amini Dec 2016

Modern Assessment Of The High-Energy Background Environment At Small Atmospheric Depths Using The X-Calibur X-Ray Polarimeter And Its Implications, Rashied Baradaran Amini

Arts & Sciences Graduate Student Theses and Dissertations

Since the discovery of cosmic rays, Earth's upper atmosphere at depths of 1-10 g cm2 has been used for balloon-borne observations probing the high-energy universe. However, the interaction of cosmic rays with the earth atmosphere generates numerous particles, each with their own flux and interaction physics that contribute to instrument background. In Fall 2014, the X-Calibur X-ray polarimeter designed and built at Washington University in St. Louis was launched from Ft. Sumner, New Mexico. While no astrophysical observation was performed as a result of a failure in telescope mechanisms, X-Calibur was able to record hours of instrument background with an …

New Techniques And Optimizations Of Short Echo-Time 1h Mri With Applications In Murine Lung, Jinbang Guo Dec 2016

New Techniques And Optimizations Of Short Echo-Time 1h Mri With Applications In Murine Lung, Jinbang Guo

Arts & Sciences Graduate Student Theses and Dissertations

Although x-ray computed tomography (CT) is a gold standard for pulmonary imaging, it has high ionizing radiation, which puts patients at greater risk of cancer, particularly in a longitudinal study with cumulative doses. Magnetic resonance imaging (MRI) doesn't involve exposure to ionizing radiation and is especially useful for visualizing soft tissues and organs such as ligaments, cartilage, brain, and heart. Many efforts have been made to apply MRI to study lung function and structure of both humans and animals. However, lung is a unique organ and is very different from other solid organs like the heart and brain due to …

Structure, Dynamics, And Regulation Of Collective Cell Migration, Andrew J. Loza Dec 2016

Structure, Dynamics, And Regulation Of Collective Cell Migration, Andrew J. Loza

Arts & Sciences Graduate Student Theses and Dissertations

Collective migration is the process by which cells organize individual motions to productively migrate as a group and plays a fundamental role in organism development, tissue regeneration, and cancer invasion. In development, coordinated migration facilitates the formation of complex organ structures and is required for proper dissemination of neural crest cells throughout an organism. After injury, this process allows breaches in epithelial layers to be repaired while maintaining tissue integrity, and in cancer, collective behavior enhances invasion of tumor cells into the surrounding tissue. Chapter 1 provides an introduction for the role of collective migration across an organism’s lifespan, the …

Neural Dynamics Tracking Subjective Cognitive Effort, John Andrew Westbrook Dec 2016

Neural Dynamics Tracking Subjective Cognitive Effort, John Andrew Westbrook

Arts & Sciences Graduate Student Theses and Dissertations

What patterns of brain activity reflect engagement with highly demanding cognitive tasks? How do these patterns relate to subjective, phenomenal effort? Answering these questions is critical to understanding what causes some people to experience cognitive tasks as more effortful than others. Subjective experience, in turn, is vital, with trait tendencies to exert effort having been linked to career and academic success. High subjective effort, as in schizophrenia and depression, can thus be extremely problematic. And yet, poor operational definitions have constrained research into basic questions about what neural dynamics track subjective effort. Here, a powerful, new behavioral economic operationalization is …

How Do The Largest And Smallest Baboon Species Compete For Reproductive Success In A Natural Hybrid Zone?, Monica Mcdonald

Arts & Sciences Graduate Student Theses and Dissertations

This dissertation examines hybridization between two of the most divergent baboons, the kinda baboon (Papio kindae) and the grayfooted chacma baboon (P. ursinus griseipes), which differ markedly in body size and in some social behavior. Preliminary research revealed hybridization between males of the smaller species (kinda) and females of the larger species (grayfoots), but not the reverse. Using behavioral, phenotypic, and genetic data collected from a single hybrid group in Kafue National Park from May 2012 to July 2013, I evaluated whether a similar asymmetry was borne out in this group and whether phenotypic markers of species assignation matched genotypic …

Epigenomics Of Cell Fate In Development And Disease, Rebecca Faith Lowdon Dec 2016

Epigenomics Of Cell Fate In Development And Disease, Rebecca Faith Lowdon

Arts & Sciences Graduate Student Theses and Dissertations

Epigenetic features at regulatory elements provide instructive cues for transcriptional regulation during development. However, the particular epigenetic alterations necessary for proper cell fate acquisition and differentiation are not well understood. This dissertation explores the epigenetic dynamics of regulatory elements during development and uses epigenome annotations to document inappropriate transcriptional regulation in disease. First, I summarize my contributions to developing a new algorithm for detecting differential DNA methylation, M&M. I report the application of the M&M algorithm to identify distinct classes of DNA methylation dynamics in surface ectoderm (SE) progenitor cells and SE-derived lineages: epigenome alterations, and differential DNA methylation in …
