Rapidly Restoring Biological Soil Crusts And Ecosystem Functions In A Severely Disturbed Desert Ecosystem, Lindsay P. Chiquoine, Scott R. Abella, Matthew A. Bowker

Life Sciences Faculty Research

Restoring biological soil crusts (biocrusts) in degraded drylands can contribute to recovery of ecosystem functions that have global implications, including erosion resistance and nutrient cycling. To examine techniques for restoring biocrusts, we conducted a replicated, factorial experiment on recently abandoned road surfaces by applying biocrust inoculation (salvaged and stored dry for two years), salvaged topsoil, an abiotic soil amendment (wood shavings), and planting of a dominant perennial shrub (Ambrosia dumosa). Eighteen months after treatments, we measured biocrust abundance and species composition, soil chlorophyll a content and fertility, and soil resistance to erosion. Biocrust addition significantly accelerated biocrust recovery on disturbed …

Electrical and Computer Engineering Senior Theses

Sound Localization is very useful for many real world applications, ranging from the military, to the fall detection of the elderly, to field biology. However, the purpose of our design was mainly a proof of concept. In this paper, we show the methods and algorithm we used to find a direction of arrival of a basic 1khz continuous sine wave. Our results show how a direction of sound can be calculated to a difficult yet condensed algorithm that was originally designed to triangulate cellular devices using cell towers. We have concluded that our margins of error have come from the …

3d Dynamic Motion Planning For Robot-Assisted Cannula Flexible Needle Insertion Into Soft Tissue, Yanjiang Zhao, Wenqiang Wu, Yongde Zhang, Ruixue Wang, Jingchun Peng, Yan Yu

Department of Radiation Oncology Faculty Papers

In robot-assisted needle-based medical procedures, insertion motion planning is a crucial aspect. 3D dynamic motion planning for a cannula flexible needle is challenging with regard to the nonholonomic motion of the needle tip, the presence of anatomic obstacles or sensitive organs in the needle path, as well as uncertainties due to the dynamic environment caused by the movements and deformations of the organs. The kinematics of the cannula flexible needle is calculated in this paper. Based on a rapid and robust static motion planning algorithm, referred to as greedy heuristic and reachability-guided rapidly-exploring random trees, a 3D dynamic motion planner …

Increased Delta As A Compensatory Strategy During Working Memory In High-Performing Patients With Schizophrenia, Faith Steffen-Allen

Master's Theses

Working memory (WM) impairment is a core cognitive deficit in schizophrenia and has been linked to dysfunction in the gamma, alpha, and theta bands. However, successful WM performance in high-performing patients is less well characterized. This study aims to investigate modulation of the EEG signal during successful WM performance in schizophrenia and to examine how that modulation differs from controls.

Patients with schizophrenia (n=7) and controls (n=9) had scalp EEG recorded during a modified Sternberg WM task with two difficulty levels. The task was subdivided into four stages (pre-trial baseline, encoding, retention, and probe). Task-induced amplitudes were computed for the …

Human Locomotion: Examining Energy Costs And Human Behavior Associated With Load-Carrying, Gailynn Benjamin Jun 2016

Human Locomotion: Examining Energy Costs And Human Behavior Associated With Load-Carrying, Gailynn Benjamin

Honors Projects

The energy expenditure during load-carrying depends on the mass of the load, the position of the load on the body, and if tools are used to help transfer the load. When the load being carried is a human child, strategies employed to cope with the increased energetic burden should include males carrying older and heavier children, arm use decreasing as children become older, and tool use increasing as children become older. To uncover whether people make energetically-mediated decisions to carry children, 236 adult males and 314 adult females were observed walking around parks in Seattle, WA, while carrying a child. …

Mimicking The Bioactivation Of The Antitubercular Agent Ethionamide Using Peracetic Acid, Isabelle E. Logan Jun 2016

Mimicking The Bioactivation Of The Antitubercular Agent Ethionamide Using Peracetic Acid, Isabelle E. Logan

University Honors Theses

Tuberculosis still affects a large number of the world’s population and drug resistance is becoming an increasing problem. Second line of defense antitubercular agents such as ethionamide (ETD) are thus becoming increasingly important. Since ethionamide is a prodrug, its metabolization through oxidation by a monooxygenase was successfully mimicked using peracetic acid. The reaction was found to be biphasic, with a fast initial oxidation to yield ethionamide sulfoxide (ETD-SO), which was relatively stable and was successfully isolated. We derived a bimolecular rate constant of 3.08 ± 0.72 × 102 M-1 s-1 for this initial phase of the reaction. …

Cmg Helicase Assembly And Activation: Regulation By C-Myc Through Chromatin Decondensation And Novel Therapeutic Avenues For Cancer Treatment, Victoria Bryant Jun 2016

Cmg Helicase Assembly And Activation: Regulation By C-Myc Through Chromatin Decondensation And Novel Therapeutic Avenues For Cancer Treatment, Victoria Bryant

USF Tampa Graduate Theses and Dissertations

The CMG (Cdc45, MCM, GINS) helicase is required for cellular proliferation and functions to unwind double-stranded DNA to allow the replication machinery to duplicate the genome. Cancer cells mismanage helicase activation through a variety of mechanisms, leading to the potential for the development of novel anti-cancer treatments. Mammalian cells load an excess of MCM complexes that act as reserves for new replication origins to be created when replication forks stall due to stress conditions, such as drug treatment. Targeting the helicase through inhibition of the MCM complex has sensitized cancer cells to drugs that inhibit DNA replication, such as aphidicolin …

Investigating The Origin And Functions Of A Novel Small Rna In Escherichia Coli, Fenil Rashmin Kacharia Jun 2016

Investigating The Origin And Functions Of A Novel Small Rna In Escherichia Coli, Fenil Rashmin Kacharia

Dissertations and Theses

Non-coding small RNAs (sRNAs) regulate various cellular processes in bacteria. They bind to a chaperone protein Hfq for stability and regulate gene expression by base-pairing with target mRNAs. Although the importance of sRNAs in bacteria has been well established, the mode of origination of novel sRNA genes is still elusive, mainly because the rapid rate of evolution of sRNAs obscures their original sources. To overcome this impediment, we identified a recently formed sRNA (EcsR2) in E. coli, and show that it evolved from a degraded bacteriophage gene. Our analyses also revealed that young sRNAs such as EcsR2 are expressed …

Image Spam Analysis, Annapurna Sowmya Annadatha Jun 2016

Image Spam Analysis, Annapurna Sowmya Annadatha

Master's Projects

Image spam is unsolicited bulk email, where the message is embedded in an image. This technique is used to evade text-based spam lters. In this research, we analyze and compare two novel approaches for detecting spam images. Our rst approach focuses on the extraction of a broad set of image features and selection of an optimal subset using a Support Vector Machine (SVM). Our second approach is based on Principal Component Analysis (PCA), where we determine eigenvectors for a set of spam images and compute scores by projecting images onto the resulting eigenspace. Both approaches provide high accuracy with low …

Determining The Effectiveness Of Soil Treatment On Plant Stress Using Smart-Phone Cameras, Anurag Panwar

USF Tampa Graduate Theses and Dissertations

Plants are vital to the health of our biosphere, and effectively sustaining their growth is fundamental to the existence of life on this planet. A critical aspect, which decides the sustainability of plant growth is the quality of soil. All other things being fixed, the quality of soil greatly impacts the plant stress, which in turn impacts overall health. Although plant stress manifests in many ways, one of the clearest indicators are colors of the leaves. In this thesis, we conducted an experimental study in a greenhouse for detecting plant stress caused by nutrient deficienceies in soil using smartphone cameras, …

Bolstering The Cell Membrane Of S. Cerevisiae For Efficient Bioethanol Production From Lignocellulosic Biomass, Conary Meyer, Matt Kubit

Bioengineering Senior Theses

With the need for alternatives to fossil fuels becoming more prevalent, biofuels has become an increasingly attractive alternative. Traditional biofuel production was quickly halted as a result of its ethical complications, leading to the development of secondgeneration biofuels. This system utilizes plant waste instead of food as its starting material, allowing for rapid recycling of this widely available and cheap carbon source. This switch was, however, coupled with complications. Of those, the most prominent is the inevitable release of acetic acid resulting from the breakdown of the lignocellulosic waste. This acetic acid is challenging to neutralize or extract in a …

Preclinical Study For Targeted Breast Cancer Therapy, Katie Bond, Lauren Mccormick, Jordan Karroll

Bioengineering Senior Theses

Cancer is a devastating disease which affects millions. For this reason, many scientists are working to develop drugs that can specifically target cancer cells within the body. Preclinical trials are an important phase in the drug development process, as they allow scientists to determine whether or not a drug candidate may be viable for human use. Targeted therapies can be achieved by using monoclonal antibodies as well as small molecule drugs. In this study we compared the effects of two control monoclonal antibody drugs, “mAb1” and “mAb2,” with one small molecule drug, “Drug S,” on the MDAMB231 human breast cancer …

Glaucoma is one of the leading cause of blindness in the modern age, with more than 65,000,000 afflicted individuals world-wide. High intraocular pressure (IOP) is the main risk factor of the disease. Current methods of monitoring IOP require a visit to an opthamologist and expensive medical equipment which greatly reduces the frequency of measurement. We utilize 3D printing to develop an apparatus that will allow us to take accurate pictures of an implantable IOP sensor with a smartphone camera. This apparatus has a lens to magnify the image and an illumination system to provide appropriate lighting. Our results show that …

Learning To Live With The Trickster: Narrating Climate Change And The Value Of Resilience Thinking, Robin Kundis Craig Jun 2016

Learning To Live With The Trickster: Narrating Climate Change And The Value Of Resilience Thinking, Robin Kundis Craig

Pace Environmental Law Review

This article is based on the 2015 Pace Garrison Lecture that occurred on April 1, 2015. Fittingly for a talk given on April Fool’s Day, this article focuses on tricksters. It posits that framing climate change as one incarnation of a mythological trickster can give us a better cultural narrative framework for thinking about environmental, natural resources, and energy law and policy in a climate change era. The trickster narrative can helpfully displace the dominant engineering framework that informs most of American10 environmental, natural resources, and energy law and policy and open the way to a more productive policy context …

Ramadan & Pregnancy, Omar Rizvi

Celebration of Learning

Fasting during Ramadan, the ninth month of the Muslim calendar, is one of the five pillars of Islam and is fard (obligatory) for all Muslims physically and mentally capable of doing so. In the Qur’an, it is made abundantly clear that religion and its acts of worship are not meant to pose undue difficulty or hardship (Qur’an 22:78, 5:6, 2:185). My paper explores the Islamic definitions for “unnecessary difficulty” and “physically capable” specifically in regards to pregnant women. At what point does fasting incur significant, if any, damage to the fetus that it should prevent the mother from participating in …

Effects Of Mutating The Mrub_1345 Gene Found In Meiothermus Ruber, Brandon M. Wills, Lori R. Scott

Celebration of Learning

Meiothermus ruber is a unique, red-pigmented, thermophilic bacterium that preferentially grows in high-temperature environments ranging from 35-70°C. Due to the lack of studies performed on this organism, there is quite a bit of information missing in regard to the genes found within this organism’s genome and their function. This study focuses in on the Mrub_1345 gene in M. ruber, which has been suggested to be orthologous to the proC gene found of E. coli proline biosynthesis pathway. To test if these genes are orthologs, we performed the complementation assay on wild-type proC. Next, we performed site-directed mutagenesis on amino acids …

Mof Acetylates The Histone Demethylase Lsd1 To Suppress Epithelial-To-Mesenchymal Transition., Huacheng Luo, Anitha K Shenoy, Xuehui Li, Yue Jin, Lihua Jin, Edward Seto, +10 Additional Authors

Biochemistry and Molecular Medicine Faculty Publications

The histone demethylase LSD1 facilitates epithelial-to-mesenchymal transition (EMT) and tumor progression by repressing epithelial marker expression. However, little is known about how its function may be modulated. Here, we report that LSD1 is acetylated in epithelial but not mesenchymal cells. Acetylation of LSD1 reduces its association with nucleosomes, thus increasing histone H3K4 methylation at its target genes and activating transcription. The MOF acetyltransferase interacts with LSD1 and is responsible for its acetylation. MOF is preferentially expressed in epithelial cells and is downregulated by EMT-inducing signals. Expression of exogenous MOF impedes LSD1 binding to epithelial gene promoters and histone demethylation, thereby …

A surveillance study in 1996 identified the USA100 clone (ST5/SCCmecII)-also known as the "New York/Japan" clone-as the most prevalent MRSA causing infections in 12 New York City hospitals. Here we update the epidemiology of MRSA in seven of the same hospitals eighteen years later in 2013/14. Most of the current MRSA isolates (78 of 121) belonged to the MRSA clone USA300 (CC8/SCCmecIV) but the USA100 clone-dominant in the 1996 survey-still remained the second most frequent MRSA (25 of the 121 isolates) causing 32% of blood stream infections. The USA300 clone was most common in skin and soft tissue infections (SSTIs) …
