Reef-building coral populations throughout the world are being threatened by numerous stressors and continue to decline. As potent endocrine-disrupting compounds, exogenous sex steroid contamination has been a largely overlooked stressor to corals. Previous research indicates these compounds are prevalent in marine environments, fluctuate annually along with reproductive cycles, can bioaccumulate, and have had variable effects on growth and reproduction in several cnidarian species. This project had three primary objectives: (1) establish environmental estradiol and progesterone concentrations in Broward County and lower Florida Keys reef environments, (2) conduct 17 β-estradiol and progesterone larval assays on P. astreoides larvae to determine the …

Invasive species represent one of the most significant threats to ecosystems, human and animal health, infrastructure, the economy, and cultural resources. Because potentially invasive, non-native species typically enter the United States through ports of entry in urban environments, some of the first observable impacts may be to infrastructure, yet little is known about the economic costs associated with these impacts to the “built” environment. In addition, federal agencies currently lack the authority necessary to effectively prevent, eradicate, and control invasive species that impact the human-built environment. The subject invention concerns gene sequences and the use thereof as markers for large granular lymphocyte (LGL) leukemia. The gene sequences of the invention are differentially expressed in LGL. Another aspect of the invention pertains to therapeutic compositions directed to gene expression and gene products of differentially expressed genes in LGL. The invention also concerns methods for screening and identifying compositions that may be of therapeutic benefit to patients having LGL leukemia and/or autoimmune disorders. Reduced mitochondrial DNA copy number, mitochondrial DNA mutations or disruption of electron transfer chain complexes induce mitochondria-to-nucleus retrograde signaling, which induces global change in nuclear gene expression ultimately contributing to various human pathologies including cancer. Recent studies suggest that these mitochondrial changes cause transcriptional reprogramming of nuclear genes although the mechanism of this cross talk remains unclear. Here, we provide evidence that mitochondria-to-nucleus retrograde signaling regulates chromatin acetylation and alters nuclear gene expression through the heterogeneous ribonucleoprotein A2 (hnRNAP2). These processes are reversed when mitochondrial DNA content is restored to near normal cell levels. We show that the mitochondrial stress-induced …

Transcript levels do not faithfully predict protein levels, due to post-transcriptional regulation of gene expression mediated by RNA binding proteins (RBPs) and non-coding RNAs. We developed a multivariate linear regression model integrating RBP levels and predicted RBP-mRNA regulatory interactions from matched transcript and protein datasets. RBPs significantly improved the accuracy in predicting protein abundance of a portion of the total modeled mRNAs in three panels of tissues and cells and for different methods employed in the detection of mRNA and protein. The presence of upstream translation initiation sites (uTISs) at the mRNA 5' untranslated regions was strongly associated with improvement …

© The Author(s) 2016. Due to the adverse impact of DDTs on ecosystems and humans, a full fate assessment deems a comprehensive study on their occurrence in soils over a large region. Through a sampling campaign across China, we measured the concentrations, enantiomeric fractions (EFs), compound-specific carbon isotope composition of DDT and its metabolites, and the microbial community in related arable soils. The geographically total DDT concentrations are higher in eastern than western China. The EFs and δ13 C of o,p'-DDT in soils from western China show smaller deviations from those of racemic/standard compound, indicating the DDT residues there mainly …

Actin has well established functions in cellular morphogenesis. However, it is not well understood how the various actin assemblies in a cell are kept in a dynamic equilibrium, in particular when cells have to respond to acute signals. Here, we characterize a rapid and transient actin reset in response to increased intracellular calcium levels. Within seconds of calcium influx, the formin INF2 stimulates filament polymerization at the endoplasmic reticulum (ER), while cortical actin is disassembled. This body of work and research is focused upon finding connections between the artist’s personal trajectory, Surrealist ideology, and Feminist texts. Starting with the Surrealist’s ideology of the unconscious, dreams, and use of objects, the artist investigates the eroticism and fetishization of female imagery. Surrealist work is brimming with portrayals of women as man’s mediator, muse, femme-enfant [child-woman], source and object of man’s desire, and the embodiment of l’amour fou [crazy love]. These representations of women depicted through fragmented female bodies transform into objects to be easily used and consumed, like furniture or food. The artist investigates how Surrealist women …

Polycystic ovarian syndrome (PCOS) is a common endocrine disorder in women of reproductive age. The pathophysiology of PCOS has conventionally thought to originate from androgen excess. However, recent evidence suggests that androgen excess is a downstream consequence to inflammatory dysregulation and subsequent metabolic abnormalities. Inflammatory mRNA gene expression of TNFa and IL-1ß in mononuclear cells isolated from women diagnosed with PCOS was explored using qPCR. Additionally, the correlations between body mass index (BMI) and fasting glucose on mRNA expression of TNFa and IL-1ß were explored. mRNA expression of both TNFa and IL-1ß were found to be significantly higher in PCOS …

Postpartum depression is recognized as the most common complication of childbearing; however, its etiology remains fairly undetermined. Many different influences have been hypothesized as to what may cause postpartum depression, including changes in levels of various hormones (such as estrogen and progesterone), a decrease in serotonin, low levels of vitamin D, social factors, and iron deficiency. The lack of strong evidence for one specific cause makes it fairly clear that there are many factors that play a role in the development of postpartum depression. Quality of interpersonal relationships is a strong predictor of mental and physical health outcomes (Cacioppo, & Hawkley, 2003) and individuals with social anxiety disorder (SAD) report increased relationship impairment (Schneier et al., 1994). Evidence from the interpersonal literature suggests that individuals with SAD exhibit interpersonal constraint, in that they rate themselves as colder and more restricted in the amount of warmth they display with close others (Rodebaugh, Bielak, Vidovic, & Moscovitch, 2016). Accurate classification of astronomical objects currently relies on spectroscopic data. Acquiring this data is time-consuming and expensive compared to photometric data. Hence, improving the accuracy of photometric classification could lead to far better coverage and faster classification pipelines. This paper investigates the benefit of using unsupervised feature-extraction from multi-wavelength image data for photometric classification of stars, galaxies and QSOs. An unsupervised Deep Belief Network is used, giving the model a higher level of interpretability thanks to its generative nature and layer-wise training. Given the increasing diversity of religious beliefs and outlooks in the United States, John Dewey’s proposals regarding “a common faith” can help educators provide the tools for their students to think critically about these and other issues related to the changing religious landscape. Particular attention is given to three groups of students: those who are adherents of dogmatic or exclusivist religious communities; those who share the belief that no legitimate value judgments about religious faith are possible; and those religiously unaffiliated students who feel excluded from the possibility of religious faith.

Diet mediates many life history trade-offs and therefore is an important environmental component to consider. Because some organisms share food sources with humans, they encounter anthropogenic changes to their diet, such as genetically modified crops and chemical applications, which can contribute to additional complexity in their environment. To understand the effect of diet on the life history of the soybean looper, Chrysodeixis includens, we exposed larvae to several different food sources, including genetically modified soybean leaves and the pesticide spinosad. Anthelmintic-resistant gastrointestinal nematode parasites are a threat to small ruminant industry sustainability. Meat goat does were administered one of four anthelmintics orally (ivermectin (n = 18), moxidectin (n = 18), levamisole hydrochloride (n = 17), or albendazole (n = 19)) or water (n = 18). Fecal samples were collected pretreatment and 12 days post-treatment. Fecal egg counts (FEC) were determined by the modified McMaster technique. The FEC reduction percentages (FECR%) were calculated using three equations. Log transformed FEC means were analyzed by treatment, sire breed of doe, and doe age. Sire breed affected (P < 0.05) pretreatment FEC, but not post-treatment FEC (P = 0.12). Pretreatment FEC did not differ (P = 0.21) by treatment group. Posttreatment FEC varied (P < 0.05) by treatment. Anthelmintic resistance determinations were based on FECR% falling below 90% or 80%, dependent on equation applied. Resistance was detected to all four anthelmintics using each equation. These results suggest the need for alternative methods of internal parasite control in goats.

ADAM10 and ADAM17 have been shown to contribute to the acquired drug resistance of HER2-positive breast cancer in response to trastuzumab. The majority of ADAM10 and ADAM17 inhibitor development has been focused on the discovery of compounds that bind the active site zinc, however, in recent years, there has been a shift from active site to secondary substrate binding site (exosite) inhibitor discovery in order to identify non-zinc-binding molecules. In the present work a glycosylated, exosite-binding substrate of ADAM10 and ADAM17 was utilized to screen 370,276 compounds from the MLPCN collection. As a result of this uHTS effort, a selective, …
