Matrix metalloproteinases (MMPs) play critical roles in cancer invasion and metastasis by digesting basement membrane and extracellular matrix (ECM). Much attention has focused on the enzymatic activities of MMPs; however, the regulatory mechanism of MMP expression remains elusive. By employing bioinformatics analysis, we identified a potential p53 response element within the MMP-14 promoter. Experimentally, we found that p53 can repress MMP-14 promoter activity, whereas deletion of this p53 response element abrogated this effect. Furthermore, we found that p53 expression decreases MMP-14 mRNA and protein levels and attenuates MMP-14-mediated cellular functions. In geological outcrops and drill cores from reef frameworks, the skeletons of scleractinian corals are usually leached and more or less completely transformed into sparry calcite because the highly porous skeletons formed of metastable aragonite (CaCO3) undergo rapid diagenetic alteration. Upon alteration, ghost structures of the distinct annual growth bands often allow for reconstructions of annual extension ( = growth) rates, but information on skeletal density needed for reconstructions of calcification rates is invariably lost. This report presents the bulk density, extension rates and calcification rates of fossil reef corals which underwent minor diagenetic alteration only. The corals derive from …

Energy harvesting using ferrofluid in OHP. Characterization of as-synthesized (bare) and surface-modified ferrofluid samples was performed using Fourier transform infrared spectroscopy, dynamic light scattering, X-ray powder diffraction, transmission electron microscopy, and atomic force microscopy. These ferrofluids were tested in a novel oscillating heat pipe set-up was utilized to harvest electricity, demonstrating the concept of ferrofluidic induction. Cobalterrite nanoparticles surface-modified with citric acid demonstrated good magnetic strengths and generated voltages close to those of the as-synthesized ferrofluids while maintaining dispersion. Amphiphiles are molecules that contain both water-soluble and water-insoluble components. The dual nature of these molecules engenders remarkable properties including the ability to self-assemble into ordered structures. Cell membranes are composed of amphiphilic phospholipids that organize into a bilayer motif. Synthetic amphiphiles can interact with natural membranes and influence the transport of molecules across the cell membrane. The work elaborated in this report employs amphiphiles to co-assemble with DNA and transport the genetic material across cell membranes. A series of cyclic peptides containing a number of tryptophan (W) and glutamic acid (E) residues were synthesized and evaluated as pH-sensitive agents for targeting of acidic tissue and pH-dependent cytoplasmic delivery of molecules. Biophysical studies revealed the molecular mechanism of peptides action and localization within the lipid bilayer of the membrane at high and low pHs. The symmetric, c[(WE)4WC], and asymmetric, c[E4W5C], cyclic peptides translocated amanitin, a polar cargo molecule of similar size, across the lipid bilayer and induced cell death in a pH- and concentration-dependent manner. Science is also defined as, “knowledge about or study of the natural world based on facts learned through experiments and observation” by Merriam-Webster dictionary. Whether the science is Physics or Geology, Biology or Chemistry, scientists seem to get excited when science happens. But science is always happening, so scientists should always be excited and exuberant, which if math serves me correctly would definitely equal happy.

© 2016 by The American Society for Biochemistry and Molecular Biology, Inc. Oncostatin M (OSM) is a multifunctional gp130 cytokine. Although OSM is produced in adipose tissue, it is not produced by adipocytes. OSM expression is significantly induced in adipose tissue from obese mice and humans. The OSM-specific receptor, OSM receptor ô (OSMR), is expressed in adipocytes, but its function remains largely unknown. To better understand the effects of OSM in adipose tissue, we knocked down Osmr expression in adipocytes in vitro using siRNA. Saturn’s moon, Titan, has been a scientific marvel since Cassini’s flyby discovered methane-ethane lakes in the northern hemisphere. Several science missions to explore these lakes have been proposed, but none have been launched. Using these previous mission designs, as well as the success of the Huygens probe, this paper will discuss the development of a deployable multi-buoy system with the intent of studying the methane-ethane lakes. The buoys will study the chemical makeup of the lakes, determine meteorology of Titan atmosphere, and map the depth and floor of the targeted lakes. This thesis is a concept study on the multi-buoy …

A resource guide was created, designed for general education teachers in upper elementary classrooms. In human stool, a large population of bacterial genes and transcripts from hundreds of genera coexist with host genes and transcripts. Assessments of the metagenome and transcriptome are particularly challenging, since there is a great deal of sequence overlap among related species and related genes. We sequenced the total RNA content from stool samples in a neonate using previously-described methods. We then performed stepwise alignment of different populations of RNA sequence reads to different indices, including ribosomal databases, the human genome, and all sequenced bacterial genomes. Cocor bebek adalah tumbuhan sukulen yang mampu memproduksi tunas adventif (reproduksi vegetatif) pada tepian daunnya. Kemampuan reproduksi vegetatif ini menghasilkan tanaman yang sama dalam waktu yang singkat, sehingga memungkinkan untuk dijadikan sebagai bioreaktor protein rekombinan. Transformasi dilakukan menggunakan Agrobacterium tumefaciens pada tunas daun cocor bebek dari spesies Kalanchoe mortagei dan Kalanchoe daigremontiana. Optimasi dilakukan mencakup: galur A. tumefaciens, kerapatan optis dari kultur A. Tumefaciens, konsentrasi acetosyringone, teknik ko-kultivasi, pH medium dan komposisi medium ko-kultivasi. This Master’s Thesis establishes what motivates a group of Mississippi farmers to participate in sustainable agriculture instead of industrial agriculture. A database of sustainable farmers was constructed in collaboration with the Gaining Ground Sustainability Institute of Mississippi. This research project used social network analysis with 28 farmers and participant observation and semi-structured interviewing with a purposively selected sample of 14 farmers. This project also explores the sustainable agricultural practices of participants. A map of the social network of sustainable agriculturalists in Mississippi is presented and shows that some farmers are well connected, some moderately connected, and others are isolated. As …

A Petrographic Analysis Of The Microbial Thrombolite Buildup In The Oxfordian Smackover Formation, Little Cedar Creek Field, Alabama, Natalie Samai-Odegaarden

The Jurassic (Oxfordian) Smackover Formation in Little Cedar Creek Field, Alabama is composed of microbial thrombolitic buildups. Core description, petrography, SEM, and isotopic analysis were used to identify the succession of organisms, microbial carbonate deposition, and diagenesis that contributed to formation of these thrombolitic buildups. The microbial thrombolite reef facies in this study accounts for 38.5% of the total Smackover Formation. This facies was deposited 0.5 to 6.75 miles from the paleo-coastline. Today it is located 10,225 to 11,750 feet in the subsurface and contains buildups 26 to 50 feet thick. Four microfacies were defined: A- Black Renalcis-like layers, B- …

This research has been completed to determine whether plasmid DNA will bind to a simple borosilicate filter in a reusable filter holder, and whether it can be released in a controlled procedure after a wash has been done to eliminate impurities.
The motivation for this is to help small research groups, both in educational settings as well as in other small research settings be able to study isolated DNA without much of the expense generally associated with purification kits and to remove the limitations on trial numbers imposed by the expense of each individual isolation kit.

Adipose tissue, also known as fat tissue are the lipid filled cells with various functions such as, shock absorber, energy storage, and thermal insulation. In this study, the impact of compressive stress at the microstructural level on the porcine abdominal adipose tissue is analyzed under different strain rates of 0.1%, 1%, and 10%. The compressed tissues were fixed in formalin followed by hematoxylin and eosin staining, which were further analyzed at the cellular level using an optical microscope. Large amounts of dredged sediment are removed from ports and river channels annually to maintain necessary depths in the maritime industry. The most common management approach for dredged soils throughout the southeast US is disposal in confined facilities. This may be the most feasible approach for ports with modest amounts of dredged soil and ample capacity for disposal. However, there is likely a more feasible option for some ports desiring to increase dredged soil containment capacity. The spectral signatures of vehicles in hyperspectral imagery exhibit temporal variations due to the preponderance of surfaces with material properties that display non-Lambertian bi-directional reflectance distribution functions (BRDFs). These temporal variations are caused by changing illumination conditions, changing sun-target-sensor geometry, changing road surface properties, and changing vehicle orientations. To quantify these variations and determine their relative importance in a sub-pixel vehicle reacquisition and tracking scenario, a hyperspectral vehicle BRDF sampling experiment was conducted in which four vehicles were rotated at different orientations and imaged over a six-hour period. Chromatin immunoprecipitation followed by massively parallel DNA sequencing (ChIP-seq) or microarray hybridization (ChIP-chip) has been widely used to determine the genomic occupation of transcription factors (TFs). We have previously developed a probabilistic method, called TIP (Target Identification from Profiles), to identify TF target genes using ChIP-seq/ChIP-chip data. To achieve high specificity, TIP applies a conservative method to estimate significance of target genes, with the trade-off being a relatively low sensitivity of target gene identification compared to other methods.
