With the increasing number of emerging contaminants that are cationic at environmentally relevant pH, there is a need for robust predictive models for organic cation sorption coefficients (Kd). Column chromatography was validated as a method to obtain organic cation sorption isotherms for environmental solids; thereby enabling a less labor intensive means to generate the extensive datasets of linear Kd values. Current predictive models fail to account for the differences in the abundance and affinity of inorganic exchange ions naturally present on environmental solids. Childhood poverty is a pervasive problem that can alter mental health outcomes. Children from impoverished circumstances are more likely than their middle-income counterparts to develop internalizing problems such as depression and anxiety. To date, however, the emotional-cognitive control processes that link childhood poverty and internalizing symptoms remain largely unexplored. Using the Emotion Go/NoGo paradigm, we examined the association between poverty and emotional response inhibition in middle childhood. We further examined the role of emotional response inhibition in the link between middle childhood poverty and internalizing symptoms. As Arctic environmental conditions fluctuate, ongoing economic-related agreements established for the Barents Region continue to support and attract Norwegian and Russian oil-producing expeditions within the shared maritime zone. Increased industrial activity throughout the Circumpolar North heightens the need to understand the factors that influence policies responsible for protecting the environment – in particular, preventive measures. Agency theory provides the framework for an analysis of various dynamics that influence the Norwegian and Russian governments (principals) as they develop and enforce rules that regulate petroleum industries (agents). Mechanisms underlying the ability of invasive exotic plant species (IES) to establish outside of their native ranges and outcompete native species (NS) are not yet fully understood, especially in low-resource environments. In deciduous forest understories, where light availability is low, but seasonally variable, one potential contributor to success of IES is extended leaf phenology, which may result from earlier leaf flush or later leaf abscission compared to that of NS. If either is the case, IES would tend to exploit a broader temporal niche for assimilating resources, which is particularly beneficial if it results in photosynthetic activity when the canopy …

This paper introduces literature that supports the need for Community Health Centers and management of low income patients with chronic illnesses. Embryonic stem cells can spontaneously differentiate into cell types of all germ layers within embryoid bodies (EBs) in a highly variable manner. Whether there exists an intrinsic differentiation program common to all EBs is unknown. Here, we present a novel combination of high-throughput live two-photon imaging and gene expression profiling to study early differentiation dynamics spontaneously occurring within developing EBs. Onset timing of Brachyury-GFP was highly variable across EBs, while the spatial patterns as well as the dynamics of mesendodermal progression following onset were remarkably similar. A warming climate is expected to cause widespread thawing of discontinuous permafrost, and the co-occurrence of wildfire may function to exacerbate this process. Here, I examined the vulnerability of permafrost to degradation from fire disturbance as it varies across different landscapes of the Interior Alaskan boreal forest using a combination of observational, modeling, and remote sensing approaches. Across all landscapes, the severity of burning strongly influenced both post-fire vegetation and permafrost degradation. The thickness of the remaining surface organic layer was a key control on permafrost degradation because its low thermal conductivity limits ground heat flux. Thus, variation in burn …

This thesis addresses the need to identify more accessible and cost-effective ways for federal food assistance programs to deliver nutrition education to Alaska Native people living in rural and remote communities in Alaska. The ultimate aim is to explore whether technology-based nutrition education is a feasible and acceptable alternative to traditional face-to-face nutrition counseling. I begin this thesis by examining the preliminary efficacy and acceptability of an 11 week text message-based intervention to promote fruit and vegetable (F&V) intake among parents with young children (n=74) using a pre-post study design. Kaposi’s Sarcoma (KS) is a malignancy caused by infection with Kaposi’s Sarcoma-associated Herpesvirus [KSHV; also known as Human Herpesvirus 8 (HHV8)] in which tumor cells show a characteristic ‘spindle-like’ morphology. The transcription factor RTA (Replication and Transcription Activator) is the viral protein responsible for reactivating KSHV from its latent state. Production of RTA in latently infected cells causes a number of viral proteins to be produced and leads to a cascade of gene expression changes in both viral and host genes. I examined post-fire moose habitat dynamics on Kanuti National Wildlife Refuge in interior Alaska with the objective of increasing understanding of local moose habitat characteristics. I estimated browse density, biomass, and summer browse use in a 2005 burn, 1990 burn, 1972 burn, and an unburned area. I revisited each site the following spring to estimate browse availability and removal during winter. In addition to evaluating browse production and use, I estimated proportional habitat use of varying-aged burns by 51 VHF-collared moose. I found that summer browse production and winter browse availability were highest in the 1990 and 2005 burns. I …

Rates of production and carbon cycling in northern ecosystems depend heavily on nitrogen (N) availability across the landscape. Since much of the available N enters these systems through biological N-fixation, Alnus, with its capacity to fix large amounts of N, plays a critical role in ecosystem response to environmental change. However, because of its high phosphorus (P) demands, the abundance, distribution, and N-fixing capacity of Alnus is tightly controlled by the availability of P and its ability to assimilate P by associating with ectomycorrhizal fungal (EMF) symbionts. Since the majority of jobs are done at a desk, those in the workforce have become particularly vulnerable to becoming sedentary. Workplace wellness programs are beginning to become a more common practice, and Kaiser Permanente is leading the way. However, they are discovering that simply providing resources is not enough. Estuaries in Southeast Alaska (SEAK) are linked to terrestrial ecosystems by the flow of freshwater from plentiful precipitation and glacial runoff. This thesis examined the trophic ecology of nearshore fishes in SEAK estuaries to advance our understanding of how deglaciation and resulting shifts in the timing and magnitude of freshwater runoff will affect estuarine food webs. Many crab fisheries around the world are managed by size, sex and season regulations, where male crabs are given at least one opportunity to reproduce before being harvested. Therefore, to set minimum legal size and fishing season for harvest, information on size at maturity and reproductive timing is needed. Lithodes aequispinus has supported a commercial fishery in Southeast Alaska since 1972, with an average annual harvest of 207 t. The current legal size and season for harvest are based L. aequispinus growth and maturity information from other parts of the range and limited information on reproduction. Large-scale reference data sets of human genetic variation are critical for the medical and functional interpretation of DNA sequence changes. Here we describe the aggregation and analysis of high-quality exome (protein-coding region) DNA sequence data for 60,706 individuals of diverse ancestries generated as part of the Exome Aggregation Consortium (ExAC). This catalogue of human genetic diversity contains an average of one variant every eight bases of the exome, and provides direct evidence for the presence of widespread mutational recurrence. We have used this catalogue to calculate objective metrics of pathogenicity for sequence variants, and to identify genes subject to strong …

High-latitude ecosystems store large amounts of carbon in soil organic matter and are among the most vulnerable to climate change. In particular, fire severity and frequency are increasing in boreal ecosystems, and these events are likely to have direct and indirect effects on climate feedbacks via increased emission of carbon (C) from soil and changes in vegetation composition, respectively. Sustainably managing wildlife with diverse utilization values is one of the greatest challenges facing contemporary wildlife management. These challenges can be amplified under changing environmental and socio-economic conditions. In Alaska, boreal forest systems are experiencing rapid change as a result of climate warming. Alaska’s boreal region has warmed twice as rapidly as the global average, affecting a host of processes including an increase in wildfire frequency, extent, and severity. Wildfire is the most common ecological disturbance in the Alaskan boreal forest and an important driver of landscape heterogeneity, burning on average 1 to 2 million acres per year. Fire severity …

The development of an embryo from a fertilised egg to a multicellular organism proceeds through numerous steps, with the formation of the three germ layers (endoderm, mesoderm, ectoderm) being one of the first. In this paper we study the mesendoderm (the tissue that collectively gives rise to both mesoderm and endoderm) gene regulatory network for two species, \textit{Xenopus laevis} and the axolotl (\textit{Ambystoma mexicanum}) using Boolean networks. We find that previously-established bistability found in these networks can be reproduced using this Boolean framework, provided that some assumptions used in previously-published differential equations models are relaxed. This paper analyses and deconstructs the transhumanist commitment to animal rights and the well-being of all sentient beings. Some transhumanists have argued that such a commitment entails a moral imperative to help non-human animals overcome their biological limitations by enhancing their cognitive abilities and generally “uplifting” them to a more human-like existence. Young adult females who participate in endurance sports are at risk of developing Female Athlete Triad (FAT). The high prevalence of FAT among collegiate athletes and its potentially lifelong consequences substantiate the importance of relevant research. Ecological production functions (EPFs) link ecosystems, stressors, and management actions to ecosystem services (ES) production. Although EPFs are acknowledged as being essential to improve environmental management, their use in ecological risk assessment has received relatively little attention. Ecological production functions may be defined as usable expressions (i.e., models) of the processes by which ecosystems produce ES, often including external influences on those processes. We identify key attributes of EPFs and discuss both actual and idealized examples of their use to inform decision making. Whenever possible, EPFs should estimate final, rather than intermediate, ES. Butyrylcholinesterase (BChE) polypeptide variants of the presently-disclosed subject matter have enhanced catalytic efficiency for (-)-cocaine, as compared to wild-type BChE. Pharmaceutical compositions of the presently-disclosed subject matter include a BChE polypeptide variant having an enhanced catalytic efficiency for (-)-cocaine. A method of the presently-disclosed subject matter for treating a cocaine-induced condition includes administering to an individual an effective amount of a BChE polypeptide variant, as disclosed herein, to lower blood cocaine concentration.
