Department of Cancer Biology Faculty Papers

Despite ubiquitous activation in human cancer, essential downstream effector pathways of the MYC transcription factor have been difficult to define and target. Using a structure/function-based approach, we identified the mitochondrial RNA polymerase (POLRMT) locus as a critical downstream target of MYC. The multifunctional POLRMT enzyme controls mitochondrial gene expression, a process required both for mitochondrial function and mitochondrial biogenesis. We further demonstrate that inhibition of this newly defined MYC effector pathway causes robust and selective tumor cell apoptosis, via an acute, checkpoint-like mechanism linked to aberrant electron transport chain complex assembly and mitochondrial reactive oxygen species (ROS) production. Fortuitously, MYC-dependent …

Adaptive Water Governance: Integrating The Human Dimensions Into Water Resource Governance, Kofi Akamani Aug 2016

Adaptive Water Governance: Integrating The Human Dimensions Into Water Resource Governance, Kofi Akamani

Journal of Contemporary Water Research and Education

The command-and-control paradigm of water resource management that dominated 20th century water policy has resulted in vulnerabilities due to its inadequate consideration of the human dimensions as well as its failure to prioritize the need for learning and adapting to change and uncertainties. In response to these shortfalls, recent years have seen the emergence of alternative water policy frameworks, notably integrated water resource management (IWRM) and adaptive management. However, while IWRM broadens the planning goals and scope of water resource governance through integration and coordination across scales and sectors, it does not adequately emphasize learning to deal with uncertainty. On …

Water And Society: Interdisciplinary Education In Natural Resources, Laszlo J. Kulcsar, Joseph A. Aistrup, Thomas Bulatewicz, Jeffrey M. Peterson, Stephen M. Welch, David R. Steward

Journal of Contemporary Water Research and Education

Natural resource management and education must account for both the natural and human components of a complex system, yet examples of such interdisciplinary approaches are still relatively rare, especially in education. This study discusses a graduate seminar on water management, developed from an interdisciplinary National Science Foundation (NSF) grant. The seminar applied constructivist pedagogy which is not only best suited for interdisciplinary work, but also allows flexibility to redesign the course components and deliverables after each evaluation cycle. While the seminar was a successful experiment, several challenges remain when implementing such courses. These challenges include not only the organization and …

Dynamic Assessment Of Current Management In An Intensively Managed Agroecosystem, Christopher G. Wilson, Kenneth M. Wacha, A.N. (Thanos) Papanicolaou, Heather A. Sander, Violet B. Freudenberg, Benjamin K.B. Abban, Chang Zhao Aug 2016

Dynamic Assessment Of Current Management In An Intensively Managed Agroecosystem, Christopher G. Wilson, Kenneth M. Wacha, A.N. (Thanos) Papanicolaou, Heather A. Sander, Violet B. Freudenberg, Benjamin K.B. Abban, Chang Zhao

Journal of Contemporary Water Research and Education

To assess management impacts on the functionality of intensively managed agroecosystems, a modeling framework was developed with a bottom-up approach and spatially distributed, process-based models. The framework is equipped with dynamic, data-informed indicators and indices to illuminate the factors influencing sustainability. The proposed dynamic indices consider natural and human aspects of an agroecosystem such as erosion, biogeochemistry, and economics. Most current indicators are static, or slow-changing, soil characterization parameters that reflect better long-term interactions between landscape features, climate, and biology. However, the ever-changing land management and climate necessitates the use of dynamic parameters that reflect agroecosystem responses to different land …

Chase Home For Children: Childhood In Progressive New England, Katherine M. Evans

Graduate Masters Theses

This thesis aims to further the study of childhood in archaeology through the examination of a children’s aid institution in Progressive New England. Specifically, this research explores how the Progressive and Victorian aims of Chase Home for Children, as expressed in primary sources, are manifested in the material culture. Chase Home participated in the larger Progressive movement in its mission to train children “in the practical duties, to encourage habits of honesty, truthfulness, purity and industry, to prepare them to take their position in life as useful members of society” (Children’s Home Pamphlet 1878). An analysis of small finds from …

Insulin-Like Growth Factor Pathway Described In Austrofundulus Limnaeus Diapause And Escape Embryos, Steven Cody Woll Aug 2016

Insulin-Like Growth Factor Pathway Described In Austrofundulus Limnaeus Diapause And Escape Embryos, Steven Cody Woll

Dissertations and Theses

Development in the annual killifish Austrofundulus limnaeus can follow two distinct developmental trajectories. Typical development includes the entrance of embryos into a state of metabolic and developmental arrest termed diapause. Alternately, embryos can escape diapause and develop directly without pause. These two trajectories are characterized by differences in the rate and timing of developmental, morphological, and physiological traits. Insulin and Insulin-like growth factor (IGF) signaling (IIS) is known to regulate entrance into diapause in a variety of invertebrates. In this thesis I explore the possible role of IGFs in the regulation of development and diapause in embryos of A. limnaeus …

Mitochondrial Dna Sequence And Lack Of Response To Anoxia In The Annual Killifish Austrofundulus Limnaeus, Josiah T. Wagner, Florisela H. Chavez, Jason E. Podrabsky

Biology Faculty Publications and Presentations

The annual killifish Austrofundulus limnaeus inhabits ephemeral ponds in regions of Venezuela, South America. Permanent populations of A. limnaeus are maintained by production of stress-tolerant embryos that are able to persist in the desiccated sediment. Previous work has demonstrated that A. limnaeus have a remarkable ability to tolerate extended periods of anoxia and desiccating conditions. After considering temperature, A. limnaeus embryos have the highest known tolerance to anoxia when compared to any other vertebrate yet studied. Oxygen is completely essential for the process of oxidative phosphorylation by mitochondria, the intracellular organelle responsible for the majority of adenosine triphosphate production. Thus, …

The Importance Of Mammalogy, Infectious Disease Research, And Biosafety In The Field, Matthew R. Mauldin, Jeffrey B. Doty, Yoshinori Nakazawa, Ginny L. Emerson, Darin S. Carroll

MANTER: Journal of Parasite Biodiversity

Large amounts of data and multitudes of publications have been independently generated by researchers in mammalogy and infectious diseases. The frequent confluence of these fields in epidemiological research as well as the facility of the data generated to be used in applied methods (e.g., conservation, public outreach, public health interventions) suggests that the intersection of these fields is important not only to their committed scientists but also to other areas of investigation, including public health. Given the increased frequency with which researchers in these fields interact with potentially infected humans, animals, and tissues, their occupations present a higher risk of …

Institutional Constraints To Collaborative Ecosystem Management Within A Wetlands Conservation Partnership, Jennifer A. Behnken, John W. Groninger, Kofi Akamani Aug 2016

Institutional Constraints To Collaborative Ecosystem Management Within A Wetlands Conservation Partnership, Jennifer A. Behnken, John W. Groninger, Kofi Akamani

Journal of Contemporary Water Research and Education

Recent decades have seen an evolution in resource management paradigms from the multiple use and sustained yield toward ecosystem management. This ongoing paradigm shift has generated interest in collaborative decision-making as a more promising alternative to conventional top down institutions. While a collaborative approach to ecosystem management promises several benefits, including equity, efficiency, and sustainability, institutional constraints often impede the success of these initiatives. Using the Cache River Joint Venture Partnership in southern Illinois, USA as a case study, this study employed a qualitative research approach to understand how various institutional attributes influence prospects for conflict and cooperation in collaborative …

Examining Valve Shape Variation In The Freshwater Diatom Genus Eunotia Over Time And Space, Jordan M. Bishop Aug 2016

Examining Valve Shape Variation In The Freshwater Diatom Genus Eunotia Over Time And Space, Jordan M. Bishop

Master's Theses

A geometric morphometrics landmark analysis was used to investigate evolutionary shape change of Eunotia, a cosmopolitan freshwater diatom genus. Modern records from temperate and tropical habitats were compared with fossils originating from a locality in the Northwest Territories of Canada. The Giraffe Pipe sediment core from a middle Eocene tropical lake contains abundant siliceous remains of diatoms and other taxa, including Eunotia, making these the oldest known representatives of the genus. The purpose of this study was to investigate the shape of Eunotia when comparing modern tropical specimens to analogous fossil specimens. Over 600 specimens were examined in total, …

Department of Medical Oncology Faculty Papers

Preclinical models suggest that histone deacetylase (HDAC) and mammalian target of rapamycin (mTOR) inhibitors have synergistic anticancer activity. We designed a phase I study to determine the safety, maximum tolerated dose (MTD), recommended phase II dose (RP2D), and dose-limiting toxicities (DLTs) of combined mTOR inhibitor sirolimus (1 mg-5 mg PO daily) and HDAC inhibitor vorinostat (100 mg-400 mg PO daily) in patients with advanced cancer. Seventy patients were enrolled and 46 (66%) were evaluable for DLT assessment since they completed cycle 1 without dose modification unless they had DLT. DLTs comprised grade 4 thrombocytopenia (n = 6) and grade 3 …

Presence Of The Cyanotoxin Microcystin In Arctic Lakes Of Southwestern Greenland, Jessica Trout-Haney, Zachary Wood, Kathryn Cottingham

Dartmouth Scholarship

Cyanobacteria and their toxins have received significant attention in eutrophic temperate and tropical systems where conspicuous blooms of certain planktonic taxa release toxins into fresh water, threatening its potability and safe use for recreation. Although toxigenic cyanobacteria are not confined to high nutrient environments, bloom-forming species, or planktonic taxa, these other situations are studied les often studied. For example, toxin production in picoplankton and benthic cyanobacteria—the predominant photoautotrophs found in polar lakes—is poorly understood. We quantified the occurrence of microcystin (MC, a hepatotoxic cyanotoxin) across 18 Arctic lakes in southwestern Greenland. All of the focal lakes contained detectable levels of …

Thirty-four microsatellite loci were isolated from three reef fish species; golden snapper Lutjanus johnii, blackspotted croaker Protonibea diacanthus and grass emperor Lethrinus laticaudis using a next generation sequencing approach. Both IonTorrent single reads and Illumina MiSeq paired-end reads were used, with the latter demonstrating a higher quality of reads than the IonTorrent. From the 1–1.5 million raw reads per species, we successfully obtained 10–13 polymorphic loci for each species, which satisfied stringent design criteria. We developed multiplex panels for the amplification of the golden snapper and the blackspotted croaker loci, as well as post-amplification pooling panels for the grass …

Towards Safer Seafood: What Indonesian Law Should “Say” About Mercury-Contaminated Fish, Margaretha Quina Aug 2016

Towards Safer Seafood: What Indonesian Law Should “Say” About Mercury-Contaminated Fish, Margaretha Quina

Indonesia Law Review

Fish is a popular culinary dish in Indonesian culture and a major economic resource on which many people depend their livelihood. However, with severe pollution in Indonesian water, including uncontrolled mercury pollution which persists in the food chain and eventually gets into humans’ body as the top predator, fish safety is particularly worrying – especially taking into account the frequency of average Indonesians’ consumption of fish. In various jurisdictions, the management tool used by lawmakers and regulators with regard to this issue is information disclosure, or known as “fish advisory warning,” to cover the failure of command and control. This …

Supervisory Dyads In School Psychology Internships: Does Personality Difference Affect Ratings Of Supervisory Working Alliance, Supervision Satisfaction, And Work Readiness?, Sheila P. Desai

Graduate Doctoral Dissertations

The internship is a critical part of graduate training and often the only opportunity to receive on-site clinical supervision during school psychology practice. Nonetheless, the process of pairing interns with field supervisors is not standardized and sometimes relies on factors such as logistics and supervisor credentials rather than a consideration of interpersonal variables that could optimize the internship experience. Related fields have found mixed evidence for a relationship between personality similarity within a supervisory dyad and outcomes such as a strong supervisory relationship, satisfaction with supervision, and supervisee effectiveness. This study examined the influence of personality similarity on ratings of …
