Specific Binding Affinity Of The Non-Catalytic Domain Of Eukaryotic Like Type Ib Topoisomerase Of Vaccinia Virus, Benjamin R. Reed

Topoisomerases are ubiquitous proteins that alter supercoiling in double stranded DNA (dsDNA) during transcription and replication and. vaccinia and the closely related poxvirus variola virus, at 314 amino acids in length, encode the smallest of the type I topoisomerases(TopIB). TopIB is a two domain protein that recognizes the sequence 5’-T/CCCTT, cleaves at the 3’-end and relaxes supercoiling through rotation. The C-terminal domain (CTD) alone contains the catalytic activity and specificity. Deletion of the N-terminal domain results in a greatly reduced rate of relaxation and rapid dissociation. Biochemical data suggests that the N-terminal domain (NTD) is important for pre-cleavage binding and …

This study examined the prevalence and impact (or intensity) of four different bullying-victimization forms (physical, verbal, relational, cyber) as experienced by the LGBTQ college-age population. In addition, this study also investigated LGBTQ college students’ bully victimization experiences and their links to depressive symptomatology. The relationship between self-rated victimization and its impact and depression was also explored. There is still a large knowledge gap regarding the etiology of cybercrime. Intoxication as a poetic principle is often identified with the romantic imagination. The literature of the intoxicated reverie is commonly thought of as synonymous with works such as Coleridge’s “Kubla Khan,” DeQuincey’s accounts of numerous nightmares and reveries, a number of Keats’ odes, Novalis’ hymns, E.T.A. Hoffmann’s stories, and Poe’s oneiric Gothic tales. Humor is widely believed to be an adaptive method of regulating emotions; however, the empirical literature remains inconclusive. One potential explanation for inconsistent results is that humor may be a multidimensional construct. Correlational research suggests that “adaptive” humor styles (Self-Enhancing and Affiliative) are more beneficial than “maladaptive” humor styles (Self-Defeating and Aggressive). The current study examined the effects of humor styles on positive and negative emotion in a sample of 146 young adults. It begins with a seeming paradox: on the one hand, since Judith Butler (1999; 2011) we can no longer think gender as ontological in any simple sense; on the other, clinical experience and the voices of transgender and gender-queer individuals shows gender to function on the order of reality, and one exceeding the social. In high-profile violent incidents, there appears to be a disproportionate focus on the perpetrator’s mental health status in relation to the incident (Angermeyer & Matschinger, 1996). Several studies have highlighted the biased nature of the media in reporting news on mental illness and its negative impact on general consensus (Corrigan et al., 2013; Wahl, 1992, 2003). Researchers have also suggested that the media is a significant source of knowledge for the public (Jorm, 2000; Wahl, 2003). Don DeLillo’s 1997 novel Underworld stands as the framing text for this study of fiction, cultural affect, and resistance in the later part of the 1980’s – the exhausted, waning years of the Cold War – and the 1990’s, the period immediately following its collapse. DeLillo’s book is situated in the 1990’s, a period of what I term “intra-anxiety” following the Cold War and prior to the attacks of September 11th and the ensuing “War on Terror.” The Cold War had provided an organizing myth for America and American culture, absorbing and structuring anxieties and governing affect. “The Cold …

This paper draws upon the principles of critical discourse analysis in order to examine the production of capitalist and consumerist discourses within contemporary nonhuman animal rights activism. The analysis presents evidence to suggest that the discourses being produced via the websites of People for the Ethical Treatment of Animals (PETA) and The Vegan Society are consistently being constructed through market-centric ideologies that treat activists mainly as middle-class consumers. This paper argues that the consistent presence of neoliberal discourse signals an instructive entanglement with broader sociopolitical issues. Psychodynamic assessment of object relations on projective tests has consistently been shown to contribute to a better understanding of children’s psychological functioning and to guide therapeutic interventions (Tuber, 1992). This research examines the enhanced utility of applying a psychodynamically-derived scale of children’s object relations to a commonly used projective assessment tool, the Thematic Apperception Test (TAT) (Morgan & Murray, 1935; Murray, 1943). This study examined the extent to which maternal rejection, paternal rejection, maternal care, and maternal overprotection predict extrinsic life aspirations as well as maladaptive money attitudes in young adults. Additionally, this study sought to determine if the Adult Parental Acceptance-Rejection Questionnaire correlates with the Parental Bonding Instrument. The variable, extrinsic life aspirations, was examined to determine if it mediates between parental rejection and maladaptive money attitudes. Recent debates on immigration policies have included a discursive contest over the representation of unauthorized immigrants, in both the news media and the Library of Congress Subject Headings (LCSH), a subject indexing system administered by the Library of Congress. Using a mixed methods approach from a critical discourse analysis perspective, I examine the responses of the news media and the Library of Congress to societal pressures for change, showing how the Library’s complex institutional position can constrain its responses. Understanding the process of speciation is of central interest to evolutionary biologists. Speciation can be studied using a phylogeographic approach, by identifying regions that promote lineage divergence, addressing whether speciation has occurred with gene flow, and when extended to multiple taxa, addressing if the same patterns of speciation are shared across codistributed groups with different ecologies. Here I examine the comparative phylogeographic histories and population genomics of thirteen snake taxa that are widely distributed and co-occur across the arid southwest of North America. Two types of aggression in children and adolescents have been identified: reactive aggression (RA) and proactive aggression (PA). Despite the accumulating evidence suggesting differential temperamental, behavioral, cognitive, social-environmental, and neurobiological correlates in relation to the two types of aggression, no study has examined emotion regulation in children with RA vs. PA using psychophysiological approaches. In this study a sample of eight to 10 years old children participated in an emotion regulation task in which they were required to either induce or inhibit their emotions. This project evaluates four mainstream taggers on a representative collection of child-adult’s dialogues from Child Language Data Exchange System. The nine children’s files from Valian corpora and part of Eve corpora have been manually labeled, and rewrote with LARC tagset. They served as gold standard corpora in the training and testing process. Four taggers: CLAN MOR tagger, ACOPOST trigram tagger, Stanford parser, and Ver. 1.14 of Brill tagger have been tested by 10-fold cross validation. This dissertation provides an ethnographic account of the food and parenting practices of a diverse group of middle-class families in the Mount Airy neighborhood of Philadelphia. It starts from the basic premise that the economic pressures on the American middle classes find expression in family life around the socially reproductive work of choosing food and parenting.

The Saccharomyces cerevisiae gene, PAH1, encodes a phosphatidate (PA) phosphatase that plays a fundamental role in lipid metabolism. PA phosphatases are key enzymes that catalyze the PA dephosphorylation reaction to form diacylglycerides, the first step in the synthesis of triacylglycerols. Pah1p, one of the main PA phosphatases in yeast, has not only emerged as a key player in lipid biosynthetic pathways, but also acts as an important regulator of nuclear membrane biogenesis, the transcriptional regulation of many inositol-sensitive upstream activating sequence (UASINO)containing genes needed for phospholipid synthesis, vacuole homeostasis, and lipid droplet formation. Feralisation occurs when a domestic population recolonizes the wild, escaping its previous restricted environment, and has been considered as the reverse of domestication. We have previously shown that Kauai Island's feral chickens are a highly variable and admixed population. Here we map selective sweeps in feral Kauai chickens using whole-genome sequencing. The detected sweeps were mostly unique to feralisation and distinct to those selected for during domestication. To ascribe potential phenotypic functions to these genes we utilize a laboratory-controlled equivalent to the Kauai population—an advanced intercross between Red Junglefowl and domestic layer birds that has been used previously for both …

Potato tubers are protected from dehydration and pathogens by a covering peel (periderm) impregnated with suberin, a complex cross-linked biopolymer that contains both polyaliphatic and lignin-like aromatic domains. Current models describing the macromolecular structure of suberin assume that ferulic acid cross-links both domains as it may form carboxyl ester bonds with aliphatic monomers and non-ester radical-coupled bonds with phenolics. Ferulic acid also links by ester bonds to glycans and acts in cross-linking polysaccharides and lignin. Fatty alcohol/ω-hydroxyacid hydroxycinnamoyl transferase (FHT) is a BADH acyltransferase responsible for the synthesis of akyl-ferulates that is necessary for suberin biosynthesis. Suberin is a biopolyester constituent of specialized plant periderm tissues formed within the phellem cell walls. Suberin and waxes of the periderm layer act to prevent water diffusion, mechanical breakdown and pathogenic invasion in plants. Ferulic esters, ω-hydroxyacids and α,ω-fatty diacids are considered to be the most important linkage between aliphatic and aromatic suberin domains and also linked with cell-wall polysaccharides. The potato gene FHT (fatty ω- hydroxyacid/fatty alcohol hydroxycinnamoyl transferase) esterifies ferulic acid to suberin. In nature, peptide toxins are an abundant resource, produced both by marine and terrestrial organisms. A major target of these peptide toxins is the group of the highly important voltage-gated ion channels. Due to their high specificity and affinity, peptide toxins have been used for over a decade in discovery and characterization of voltage-gated ion channels. Although peptide toxins have been extensively characterized structurally, the structural characterization of eukaryotic voltage-gated sodium channels has seen much less progress, due to their large size and high hydrophobicity.
