Identification Of Putative Receptors For The Novel Adipokine Ctrp3 Using Ligand-Receptor Capture Technology, Ying Li, Tammy Ozment, Gary L. Wright, Jonathan M. Peterson
ETSU Faculty Works
C1q TNF Related Protein 3 (CTRP3) is a member of a family of secreted proteins that exert a multitude of biological effects. Our initial work identified CTRP3’s promise as an effective treatment for Nonalcoholic fatty liver disease (NAFLD). Specifically, we demonstrated that mice fed a high fat diet failed to develop NAFLD when treated with CTRP3. The purpose of this current project is to identify putative receptors which mediate the hepatic actions of CTRP3.
Methods
We used Ligand-receptor glycocapture technology with TriCEPS™-based ligand-receptor capture (LRC-TriCEPS; Dualsystems Biotech AG). The LRC-TriCEPS experiment with CTRP3-FLAG protein as ligand and insulin as a …

Cell-To-Cell Transmission Of Dipeptide Repeat Proteins Linked To C9orf72-Als/Ftd., Thomas Westergard, Brigid K Jensen, Xinmei Wen, Jingli Cai, Elizabeth Kropf, Lorraine Iacovitti, Piera Pasinelli, Davide Trotti
Farber Institute for Neuroscience Faculty Papers
Aberrant hexanucleotide repeat expansions in C9orf72 are the most common genetic change underlying amyotrophic lateral sclerosis (ALS) and frontotemporal dementia (FTD). RNA transcripts containing these expansions undergo repeat-associated non-ATG translation (RAN-T) to form five dipeptide repeat proteins (DPRs). DPRs are found as aggregates throughout the CNS of C9orf72-ALS/FTD patients, and some cause degeneration when expressed in vitro in neuronal cultures and in vivo in animal models. The spread of characteristic disease-related proteins drives the progression of pathology in many neurodegenerative diseases. While DPR toxic mechanisms continue to be investigated, the potential for DPRs to spread has yet to be determined. …

Background: There are three main dietary groups in mammals: carnivores, omnivores, and herbivores. Currently, there is limited comparative genomics insight into the evolution of dietary specializations in mammals. Due to recent advances in sequencing technologies, we were able to perform in-depth whole genome analyses of representatives of these three dietary groups. Results: We investigated the evolution of carnivory by comparing 18 representative genomes from across Mammalia with carnivorous, omnivorous, and herbivorous dietary specializations, focusing on Felidae (domestic cat, tiger, lion, cheetah, and leopard), Hominidae, and Bovidae genomes. We generated a new high-quality leopard genome assembly, as well as two wild …

Speed And Rhythm Affect Temporal Structure Of Variability In Reaching Poststroke: A Pilot Study, Amit Sethi, Nicholas Stergiou, Tara Patterson, Carolynn Patten, Lorie G. Richards
Journal Articles
Temporal structure reveals the potential adaptive strategies employed during upper extremity movements. The authors compared the temporal structure of upper extremity joints under 3 different reaching conditions: preferred speed, fast speed, and reaching with rhythmic auditory cues in 10 individuals poststroke. They also investigated the temporal structure of these 3 reaching conditions in 8 healthy controls to aid in the interpretation of the observed patterns in the poststroke cohort. Approximate entropy (ApEn) was used to measure the temporal structure of the upper extremity joints. ApEn was similar between conditions in controls. After stroke, ApEn was significantly higher for shoulder, elbow, …

Implementation Of A Mentor Led Physical Activity Program Among Disadvantaged Youth, Karen M. Martin
Dissertations
Physical activity is an integral part of the war on pediatric obesity. Pediatric obesity is a worldwide epidemic affecting children of all races, classes, and gender. According to the Ohio Youth Risk Behavior Survey of 2013 over one third of Ohio children in grades nine through twelve were considered overweight or obese and engage in more than three hours of screen time, television, computers or video games daily. The purposes of this project was to introduce physical activity in the classroom environment in the form of physical activity breaks, led by physical activity leaders (PALs) to achieve improved BMI category, …

Animal Sentience
Several types of objection have been raised against the arguments I presented in my target article, “Are animals persons?” Among the objections are the following: (1) the claim that animals are persons is of little significance, (2) my use of the Lockean conception of the person is questionable, (3) whether a creature qualifies as a person is a matter of social construction rather than objective fact, (4) reflective consciousness is more important than I realize, (5) my reliance on implicit self-awareness in the account of personhood is ill-advised, (6) my account entails that too many creatures qualify as persons, and …

St. John's Law Review
(Excerpt)
In Part I, this Article introduces the new and emerging technologies, including the Internet, cloud computing, three-dimensional (“3D”) printing, and synthetic biology, which will bring this radical change. Part II provides an overview of the innovation cycle, including the stages of basic research, inventing and prototyping, product development, marketing, and distribution. It also describes, in detail, how these new technologies are dramatically lowering the costs and risks of all stages in the innovation cycle.
Part III considers how lawmakers might adapt patent law to account for the new age of innovation and its lower costs of innovation. This Article …

Testing Modeling Assumptions In The West Africa Ebola Outbreak, Matthew C. Ingram, Keith Burghardt, Christopher Verzijl, Junming Huang, Binyang Song, Marie-Pierre Hasne
Political Science Faculty Scholarship
The Ebola virus in West Africa has infected almost 30,000 and killed over 11,000 people. Recent models of Ebola Virus Disease (EVD) have often made assumptions about how the disease spreads, such as uniform transmissibility and homogeneous mixing within a population. In this paper, we test whether these assumptions are necessarily correct, and offer simple solutions that may improve disease model accuracy. First, we use data and models of West African migration to show that EVD does not homogeneously mix, but spreads in a predictable manner. Next, we estimate the initial growth rate of EVD within country administrative divisions and …

Multiple Discrete Soluble Aggregates Influence Polyglutamine Toxicity In A Huntington's Disease Model System, Wen Xi, Xin Wang, Clyde L. Denis
New Hampshire Agricultural Experiment Station Publications
Huntington’s disease (HD) results from expansions of polyglutamine stretches (polyQ) in the huntingtin protein (Htt) that promote protein aggregation, neurodegeneration, and death. Since the diversity and sizes of the soluble Htt-polyQ aggregates that have been linked to cytotoxicity are unknown, we investigated soluble Htt-polyQ aggregates using analytical ultracentrifugation. Soon after induction in a yeast HD model system, non-toxic Htt-25Q and cytotoxic Htt-103Q both formed soluble aggregates 29S to 200S in size. Because current models indicate that Htt-25Q does not form soluble aggregates, reevaluation of previous studies may be necessary. Only Htt-103Q aggregation behavior changed, however, with time. At 6 hr …

Atomic Tiles: Manipulative Resources For Exploring Bonding And Molecular Structure, Alan L. Kiste, Rebecca G. Hooper, Gregory E. Scott, Seth Bush
Chemistry and Biochemistry
A simple manipulative resource, Atomic Tiles, is described for scaffolding the learning of Lewis structures without using algorithmic, rule-based methods of drawing. Students use Atomic Tiles to (1) create models of bonding that lead to drawing Lewis structures, (2) use the structures they create to infer patterns required for rational structures and common organic functional groups, (3) translate between Lewis structures and molecular models, and (4) use molecular models to identify isomers.

Exploring How Secondary Pre-Service Teachers’ Use Online Social Bookmarking To Envision Literacy In The Disciplines, Jamie Colwell, Kristen Gregory
Reading Horizons: A Journal of Literacy and Language Arts
This study considers how pre-service teachers envision disciplinary literacy through an online social bookmarking project. Thirty secondary pre-service teachers participated in the project through an undergraduate literacy course. Online bookmarks and post-project reflections were collected and analyzed using a constant comparative approach to determine emergent themes. Results suggest varying levels of disciplinary knowledge among pre-service teachers, influences of pre-service teachers' envisionments on posted bookmarks, and considerations about standardized testing in disciplinary literacy instruction. Implications for teacher education are discussed in light of these results.
