Physicochemical Modulation Of Huntingtin Aggregation On Lipid Membranes: Implications For Huntington's Disease, Maxmore Chaibva Jan 2016

Physicochemical Modulation Of Huntingtin Aggregation On Lipid Membranes: Implications For Huntington's Disease, Maxmore Chaibva

Graduate Theses, Dissertations, and Problem Reports (ETD)

Huntington disease is a genetic, neurodegenerative disease caused by an expanded polyglutamine (polyQ) in the first exon of the huntingtin (htt) protein, facilitating its aggregation, leading to the formation of a diverse population of potentially toxic aggregate species, such as oligomers, fibrils, and annular aggregates. Htt interacts with a variety of membranous structures within the cell, and the first seventeen amino acids (Nt17) of htt directly flanking the polyQ domain is an amphipathic alpha-helix (AH) lipid-binding domain. AHs are also known to detect membrane curvature. Nt17 also promotes diverse aggregate species of htt and undergoes a number of posttranslational modifications …

Comparative Analysis Of Uninhibited And Constrained Avian Wing Aerodynamics, Jordan A. Cox Jan 2016

Comparative Analysis Of Uninhibited And Constrained Avian Wing Aerodynamics, Jordan A. Cox

Graduate Theses, Dissertations, and Problem Reports (ETD)

The flight of birds has intrigued and motivated man for many years. Bird flight served as the primary inspiration of flying machines developed by Leonardo Da Vinci, Otto Lilienthal, and even the Wright brothers. Avian flight has once again drawn the attention of the scientific community as unmanned aerial vehicles (UAV) are not only becoming more popular, but smaller. Birds are once again influencing the designs of aircraft. Small UAVs operating within flight conditions and low Reynolds numbers common to birds are not yet capable of the high levels of control and agility that birds display with ease. Many researchers …

Endocannabinoid Modulation Of Secretory And Inflammatory Causes Of Gastric Pathology, Molly S. Crowe Jan 2016

Endocannabinoid Modulation Of Secretory And Inflammatory Causes Of Gastric Pathology, Molly S. Crowe

Graduate Theses, Dissertations, and Problem Reports (ETD)

Non-steroidal anti-inflammatory drugs (NSAIDs) are common analgesic drugs that also cause well-known, negative gastrointestinal (GI) side effects. The specific physiological mechanism(s) of NSAID-induced GI damage are unknown, and are likely due to multiple causes. The most studied components include: increased gastric acid secretions, inflammation of the gastric epithelium, and increased gastric motility. Currently, proton pump inhibitor (PPI) antacids are used by chronic NSAID users, with the expectation that decreasing gastric acid output will reduce gastric epithelial irritation. However, PPIs are associated with side effects like infection, gastric polyps, and dementia. Therefore, new gastroprotective treatments that prevent NSAID-induced gastropathy are needed. …

Four Days That Transformed Spain 11-M Impact On Memory Recovery Examined Through The Lens Of Duality, Thomas J. Divins

Graduate Theses, Dissertations, and Problem Reports (ETD)

This thesis studies the controversial aftermath of the Madrid terrorist attack of March 11, 2004 and how it impacted the history of memory and silence in Spain since the Spanish Civil War. The terrorist attack had surprising and significant short-term political impacts, which have been well documented by historians, political scientists, and journalists. However, surveying the attack aftermath through the lens of the history of duality in Spain has been less studied. The "lens of duality" is the viewpoint that Spain's history in the twentieth and twenty-first centuries can be explained as a clash between deeply-divided sectors within society which …

Elucidating Mechanisms Of Canonical Wnt - Ephrin-B Crosstalk, William Tyler Koch Jan 2016

Elucidating Mechanisms Of Canonical Wnt - Ephrin-B Crosstalk, William Tyler Koch

Graduate Theses, Dissertations, and Problem Reports (ETD)

Throughout development, canonical Wnt signaling contributes to the formation and maintenance of a wide array of cells, tissues, and organs. Dys-regulated Wnt signaling during embryonic development is implicated in developmental defects known as neurochristopathies, including craniofacial and heart defects, as well as defects in neural development. Due to its roles in stem cell maintenance and self-renewal, tissue homeostasis, and regeneration, aberrant Wnt signaling in adult tissues can result in various forms of cancer, including colorectal cancer, breast cancer, lung cancer, and gastro-intestinal cancer, among others. Dys-regulated Wnt signaling is also implicated in other pathologies including bone disease, and metabolic diseases, …

Assessment Of Molecular Action Of Direct Gating And Allosteric Modulatory Effects Of Meprobamate (Miltownrtm) On Gaba A Receptors, Manish Kumar

Graduate Theses, Dissertations, and Problem Reports (ETD)

Meprobamate (tradenames Miltown, Equanil) is a schedule IV anxiolytic and the primary metabolite of the muscle relaxant carisoprodol. Meprobamate modulates GABAA (gamma-aminobutyric acid type A) receptors, and has been described as having barbiturate-like activity. A thorough understanding of its mechanism remains ill-defined, however. To gain insight into its actions, we have conducted a series of studies using recombinant GABAA receptors. In alphaxbetazgamma2 GABAA receptors (where x = 1-6 and z = 1-3), the ability to enhance GABA-mediated current was evident for all alpha subunit isoforms, with the largest effect observed in alpha5-expressing receptors. Direct gating was comparable with all alpha …

Distribution And Efficacy Of Chemotherapeutics In The Treatment Of Preclinical Brain Metastases Of Breast Cancer, Tori Terrell Hall

Graduate Theses, Dissertations, and Problem Reports (ETD)

Brain metastases are a critical, life-threatening problem for women with advanced metastatic breast cancer. Approximately 80% of women with disseminated central lesions are unable to survive the first year after diagnosis. Despite the breakdown of the blood-brain barrier, chemotherapeutics have limited penetration and distribution into brain metastases and are unable to induce cytotoxicity in the tumor. Limiting the development of new treatments for brain metastases of breast cancer, there are no commercially available in vitro models available that accurately model, and mimic the functionality of, the in vivo blood-tumor barrier (BTB). In an attempt to address the aforementioned problem, the …

Graduate Theses, Dissertations, and Problem Reports (ETD)

Ischemic heart disease (IHD) continues to be a leading cause of morbidity and mortality in increasingly more countries. Function and structural damage to the heart are likely consequences of ischemic insults and may even lead to death. The body's physiological response to ischemia is by increasing nourishment or perfusion to the deprived organ in an attempt to decrease the potential ischemia-induced damage and achieve faster recovery. This reaction to ischemia is termed reactive hyperemia (RH), and in the heart, it is coronary reactive hyperemia (CRH). Epoxyeicosatrienoic acids (EETs) are oxylipins derived from arachidonic acid (AA) and possess distinct cardiovascular benefits, …

The Relationship Between Plasma Osmolarity And Feed Efficiency In Beef Cattle And Effect Of Feeding Cows Medicated Feed Through Mineral Containing Altosidrtm Igr On The Growth Of Beef Calves, Taylor Diane Harrison

Graduate Theses, Dissertations, and Problem Reports (ETD)

Dietary cation-anion modification in cattle has effects on productivity through various means. Alterations of dietary cation-anion balances has been shown to affect plasma osmolarity. Plasma osmolarity is directly related to renal function and metabolism in all animals. In a feeding operation with a single diet fed to a similar population of animals, plasma osmolarity should fall within a somewhat normal range (approximately 270-310 mOsm). However, subtle, yet very distinguishable differences will exist between animals. In theory, variances in osmolarity may reflect differences in metabolism and as such, efficiency of feed utilization. Many researchers believe that utilizing residual feed intake (RFI) …

Graduate Theses, Dissertations, and Problem Reports (ETD)

LIM homeobox 8 (Lhx8) is an important transcription factor that is preferentially expressed in germ cells. Lhx8 null mice are infertile due to lack of oocytes and impairment of the transition from primordial follicles to primary follicles. Lhx8 deficiency also affects the expression of many important oocyte-specific genes. To date, no attempts have been made to investigate the existence of any cellular factors that might interact with Lhx8 protein in oocytes and early embryos. In this study, we report the characterization of rainbow trout and bovine Lhx8 genes and identification of important germ cell-specific nuclear factors that interact with Lhx8 …

Spatial And Temporal Distribution Of The Brown Marmorated Stink Bug (Hemiptera: Pentatomidae), Jakob Goldner

Graduate Theses, Dissertations, and Problem Reports (ETD)

An obstacle to efficient management of the brown marmorated stink bug, Halyomorpha halys (Hemiptera: Pentatomidae), is the lack of knowledge of its seasonal movements at the landscape level. H. halys is highly mobile and utilizes various commodities throughout the growing season and winter, making it difficult to manage. We hypothesized that H. halys displayed an annual spatial pattern at the farm scale and as they moved into overwintering habitats. To test for a spatial pattern at the farm scale, we conducted a two-year spatial study on three organic farms in West Virginia and New Jersey. A total of 92-127 sample …

Development Of A Cobinamide-Based Optical Sensor For Hydrogen Cyanide And Hydrogen Sulfide, Lee Ann Greenawald

Graduate Theses, Dissertations, and Problem Reports (ETD)

In an occupational or military environment, a personal air-purifying respirator must be provided when breathing-air is contaminated by harmful dust, fumes, gases, aerosols, or vapors. Too often, a respirator user does not have enough information to know when to change his/her cartridge/canister and thus is potentially exposed to toxic gases. Currently, there is no definitive way to determine when respirators' carbon beds have begun to fail. When an end-of-service-life indicator (ESLI) is incorporated into the carbon bed, it informs one, in real-time, when imminent breakthrough is occurring and to replace the cartridge/canister. ESLIs are a more reliable and safer way …

Paradoxical Roles Of Nanoparticles In Cancer Therapeutics And Carcinogenesis, Emily Despeaux Jan 2016

Paradoxical Roles Of Nanoparticles In Cancer Therapeutics And Carcinogenesis, Emily Despeaux

Graduate Theses, Dissertations, and Problem Reports (ETD)

Nanoparticles (NPs) are becoming increasingly common in consumer goods and are under investigation for a variety of industrial and biomedical applications. However, challenges in determining NP toxicity may prevent them from reaching their full potential. NPs cannot be treated as single class for toxicity evaluations. Even among particles made from the same material, particle-specific physical properties, including size, shape, surface charge, agglomeration state, and surface modifications have a strong effect on the toxicity. Even so, the obstacles to conclusively and reproducibly evaluating toxicity span all NP classes. NP literature is riddled with confusing and often contradictory reports regarding the biocompatibility …

Using Tree-Ring Data To Analyze The Effects Of Volcanic Eruptions On Climate In Inner Asia From 500 Bce To Present, Jennie Zhu

Graduate Theses, Dissertations, and Problem Reports (ETD)

Volcanic eruptions have influenced regional and global climate on annual, interannual, decadal, and multi-decadal time scales. However, past studies have largely focused on the response of global temperature to modern eruptions with the aid of satellite records. This approach limits the number of eruptions that can be studied and restricts our knowledge to relatively small events when compared to the volcanic history of the late Holocene. I used modern climate records to evaluate the effect of recent eruptions on Mongolian climate. I then extended my analysis to the last 2500 years by examining existing and new tree-ring data from Mongolia …

Multiwalled carbon nanotubes (MWCNT) are one of the most exciting industrial materials of our time. They are used in life-saving medical therapeutics and many commercial products that could make day-to-day life seem effortless. However, previous toxicological research has demonstrated this material to be significantly genotoxic in both in vitro and in vivo models and potentially carcinogenic in the lung. The unique physiochemical properties of MWCNT make respiratory exposures likely in workers. Combining the genotoxic effects with the potential for lung deposition in the workplace, MWCNT should be considered as a potential health hazard. Altering the physiochemical properties of MWCNT has …
