Characterization Of An Al$^{3+}$-Selective Fluorescent Probe Based On A Benzoyl Hydrazine Derivative And Its Application In Cell Imaging, Yuxiang Ji, Chunwei Yu, Shaobai Wen, Jun Zhang

A fluorescent probe based on benzoyl hydrazine was synthesized and characterized as an Al$^{3+}$-selective fluorescent probe. This probe showed good selectivity towards Al$^{3+\, }$compared to other common ions. Under optimized experimental conditions, the probe exhibited a linear dynamic response for Al$^{3+}$ from 5.0 $\times $ 10$^{-7}$ to 4.5 $\times $ 10$^{-6}$ M with a detection limit of 1.3 $\times $ 10$^{-7}$ M in ethanol-water solution (9:1, v:v, pH 6.8, 20 mM HEPES). Furthermore, it was used for imaging of Al$^{3+}$ in living cells with satisfying results.

Hexavalent chromium, Cr(VI), is a highly toxic species, while trivalent chromium, Cr(III), is an essential trace element for humans and other organisms. In the present work, a simple, rapid, and efficient procedure for water treatment of Cr(VI) is described. Active carbon (AC) was chemically treated with HCl and NaOH for the formation of surface modified Acidic-AC and Basic-AC sorbents, respectively. In addition, AC sorbent was treated with HNO$_{3}$ as an oxidizing agent and sodium sulfite as a reducing agent for the formation of Ox-AC and Rd-AC sorbents, respectively. Hypericin (HYP)-mediated photodynamic therapy (PDT) is a new alternative treatment strategy for colon cancer inducing various cell-death pathways. Apoptotic cell death is the desired cellular fate in cancer cells. Therefore, we investigated the apoptotic pathways by determining apoptosis-related proteins (survivin, caspase-3, caspase-9, Bcl-2, and Bax) at the mRNA level using real-time polymerase chain reaction (qPCR), and the percentage of survivin was determined by survivin ELISA in HT-29 and Caco-2 colon cancer cell lines. Human embryonic stem cells (hESCs) and reprogrammed human induced pluripotent stem cells (hiPSCs) proliferate indefinitely in the undifferentiated state (self-renewal) and differentiate into the three germ layers (and ultimately into all cell types) (pluripotency). Branching in the N-glycan core of hESCs/hiPSCs is limited. hESCs/hiPSCs have high mannose-type and biantennary complex-type core structures, indicating immature stages of N-glycoproteins. The two branches on the core consist of type 2 N-acetyllactosamine (LacNAc), completed mainly with α 2,6-linked sialic acid. Complex terminal fucosylation, particularly α 1,2-linked fucosylation, is another characteristic N-glycosylation feature of hESCs/hiPSCs. Laser Speckle Contrast Imaging (LSCI) is a real-time, non-invasive method in used to investigate blood flow and perfusion in biological tissues with high temporal and spatial resolution. A reduction in speckle contrast due to particle motion is the primary contrast mechanism in LSCI. Motion results in speckle fluctuations in time and reduces the contrast over a given camera integration period. There are a variety of parameters that effect contrast besides motion. Magnetoelastic sensors represent a low-cost wireless and battery-less method for monitoring parameters in embedded or implanted applications; however, some limitations still exist preventing their commercial implementation. Presented in this work are a variety of studies that are aimed at improving the feasibility of magnetoelastic materials for sensing and actuating applications. Magnetoelastic resonant sensors of non-standard geometries were investigated to determine if geometry could play a role on the sensitivity of the sensor response to mass loading. Elevated plasma arginine vasopressin (AVP) levels have been found in human hypertension subjects and several salt dependent experimental animal models of hypertension including the Dahl salt sensitive hypertension (SSHTN) model. The orexin system is involved in AVP regulation and its over activation has been implicated in hypertension, however, the role of brain orexin in SSHTN is unknown. The generalized lognormal distribution plays an important role in various aspects of life testing experiments. We examine Bayesian analysis of this distribution using objective priors (in the general sense of priors constructed using some formal rules) for the model parameters in this paper. Specifically, the derivation of explicit expressions for multiple types of the Jeffreys priors, the reference priors with different group ordering of the parameters, and the first-order matching priors. Barataria Bay, one of the largest receiving basins for the Mississippi deltaic complex, is the location of a proposed river-sediment diversion for delta restoration. In order to determine how the sediment in the receiving-basin may respond to diversion flows, twenty-five sediment vibracores were collected from a 115 km2 study area located near Myrtle Grove and Bayou Dupont, southeast of New Orleans, LA. These cores were subject to multiple tests, including gamma bulk density scans, grain size analysis, and loss-on-ignition, in order to identify the lithology and stratigraphy. Biomass or coal gasification are promising processes for renewable energy. However, a major operating problem in gasification is what to do with syngas byproducts such as H2S and tars (heavy hydrocarbons) that cause catalyst deactivation downstream. Studies have shown that rare earth oxides (REOs) doped with transition metals are promising catalysts for tar reforming in the presence of sulfur.(R. Li, Roy, Bridges, & Dooley, 2014) In Chapter 1, propane is studied as a model compound for tar reforming with transition metal-doped (e.g., Mn, Fe) REOs. Fertilizer nutrient losses through leachate and runoff from excessive irrigation in nursery container production and turfgrass management can be high and have negative environmental impacts. The objective of this research was to examine the influence of fertilizer source and irrigation regimen on nutrient losses during nursery container production and turfgrass management. The relationship of arbuscular mycorrhizal (AM) fungi with northern white cedar (NWC) was examined from the perspective of both fundamental questions about habitat specificity in the root fungal community, as well as applied questions regarding AM fungal efficacy in NWC restoration in peat soils. Modeling lake processes and dynamics improves understanding of the system and supports predictions of the response of the lake to perturbations, such as climate change. LAKE2K, a 3-layer surface water quality model, uses a mass balance approach to simulate the physical and biogeochemical cycles in Lake Superior. The model is successfully calibrated with data from offshore Lake Superior in 2011, a year with average meteorological conditions. The thermal regime, phytoplankton populations, and phosphorus cycle of Lake Superior are modeled, resulting in a representation of seasonal trends in this dimictic system. We present results from experiments that emulate atmospheric ice nucleation by aerosols. We have refined experimental techniques to improve measurements of ice forming nuclei in the contact mode. Our results show that atmospherically relevant dusts such as kaolinite, feldspar, rhyolitic ash, and Arizona Test Dust have efficiencies in the contact mode that are higher than the immersion mode. Experiments with bacteria show that biological material has the potential to contribute significantly to ice concentrations, but has large variability. The Northeast Ohio Regional Sewer District (NEORSD, Cleveland, OH) serves over one million citizens through the operation of three waste water treatment plants. NEORSD provides this service through combined and separated sewer systems, which, during rainfall events, are known to deliver large volumes of untreated wastewater containing high concentrations of Escherichia coli (E.coli), ammonia nitrogen, and phosphorus. Proteins are nano-machines that carry out majority of the cellular functions. Thermodynamically they are functional and stable within a very narrow range (1 kcal/mol). External perturbations in the form of pH change, thermal, or oxidative/reducing stress can destabilize the protein resulting in misfolding and aggregation. Prolonged environmental stress can affect the cells adaptive response resulting in loss of ability to refold or recycle proteins. This can lead to accumulation of misfolded or aggregated proteins within the cell. Such accumulation of aggregated proteins have been associated with neurodegenerative disorders such as Amyotrophic Lateral Sclerosis (ALS), Parkinson’s, Huntington’s, and Alzheimer’s disease.
