Polo-like Kinse 1 (PLK1) is a serine/threonine protein kinase that is expressed in dividing cells. It is a widely studied protein that plays an important role in regulating mitotic progression. PLK1 is over-expressed in several tumor types, and studies have shown that down-regulating PLK1 expression inhibits cancer cell proliferation, validating PLK1 as an oncotarget. Efforts to target this protein therapeutically has led to the development of several ATP-based inhibitors, although these are not exclusively specific. Because other PLK family members are tumor suppressors, it is important that PLK1 inhibitors selectively bind to PLK1 and not the other PLKs, particularly PLK3. …

The management of saltmarshes requires detailed knowledge of the underlying processes driving their distribution in both time and space to make appropriate management decisions. With most of the world’s population living in the coastal zone and rising sea levels, one of our most important natural resources in the coastal zone faces increasing threat of collapse. This study uses the current state of Light Detection and Ranging (LiDAR) technology to model and predict saltmarsh distribution at a landscape-scale and provide evidence that a terrestrial laser scanner (TLS) can be used to estimate saltmarsh biomass for inclusion into existing models.

Disordered eating has become a significant issue among children and adolescents; nearly 14% of all youth displaying disordered eating patterns. Vibrio gazogenes, a Gram-negative species of marine bacteria, was capable of transporting 20 nm (diameter) surface-carboxylated, polystyrene, fluorescent FluoSpheres® microspheres (excitation/emission = 505/515) through the outer membrane, which was indicated by a detectable decrease in the fluorescence intensity of the nanoparticles in the culture medium. A mechanism of transport was investigated involving the ATP-binding cassette (ABC) protein transporters that traverse the outer membrane. Inhibition of the ABC transporters did not prevent the entry of the nanoparticles into the cell, suggesting there was an alternate mechanism of transport. The addition of nanoparticles to the culture medium also did not provide any …

Age-Related Liquefaction Resistance Of Pleistocene Coastal Plain Sands In South Carolina, Michael J. Hasek

Soils of Pleistocene age in the South Carolina Coastal Plain have experienced liquefaction due to historic and pre-historic earthquakes. Numerous field and laboratory studies have shown that aged soil deposits maintain a greater resistance to liquefaction than younger soil deposits. The currently available methods for assessing liquefaction potential are based on cases in which soils are of Holocene age or younger (< 10,000 yrs). The Pleistocene age soils that were tested and characterized varied in age from about 200,000 years old to 1,400,000 years old. Several sites were investigated using field methods that included the seismic cone penetration test, cone penetration test, standard penetration test, and flat plate dilatometer. Piezometers were installed at the sites. Undisturbed soil samples were retrieved from the subsurface and frozen ex situ to minimize sample disturbance during transportation and laboratory handling. The undisturbed samples were used for cyclic triaxial testing in the laboratory and were tested for shear wave velocity and compression wave velocity using in-cell transducers. Laboratory tests were performed to determine the specific gravity, grain size distribution, moisture content, unit weight, Atterberg limits, Unified Soil Classification, and visual-manual description. Optical petrography and scanning electron microscopy were used to determine the mineral content of the soils, to view grain characteristics, and to view microscopic features that were part of the soil aging process.

Laboratory index tests showed that Pleistocene soils consisted predominately of poorly-graded fine sands, silty sands, and clayey sands. Shear wave velocities from the cyclic triaxial test specimens were comparable to the in situ shear wave velocities measured using …

With worldwide petroleum resources dwindling and greenhouse gas emissions rising, it is urgent to find renewable replacements for petroleum-derived products. A biomass-derived chemical with high potential as a platform intermediate, γ-valerolactone (GVL) can be readily synthesized by hydrogenation of levulinic acid (LA), itself a common biomass intermediate, using supported Ru catalysts. Overall, vapor phase hydrogenation is more energy sensitive as the higher boiling point of LA (~245°C) and requires a high energy input, comparable to liquid phase hydrogenation, which is more economical. This dissertation describes fundamental and applied studies that have advanced the understanding and application laser induced breakdown spectroscopy (LIBS) to the deep-ocean environment and to operation on a deep-ocean submersible. Chapter one is an overview of LIBS including a brief history, LIBS theory, the instrumentation typically used, and a specific application. Chapter two analyzes the effect that monodisperse suspended particles have on LIBS emission intensity and on the formation of a LIBS plasma. "Numerous sandstone filled depressions hosted in upper Cambrian to Middle Ordovociain dolostones in south-central Missouri have historically been identified as Pennsylvanian paleokarst structures. U-Pb detrital zircon geochronology is at odds the time of formation and sedimentological evidence challenges their classification. "Filled sink" deposits yield primary zircon age populations of 2.8-2.6 Ga (~50% of all analyzed grains), 1.2-1.1 Ga (~25%), 1.8-1.6 Ga (~15%) and 1.5-1.3 Ga (~10%). These zircon populations most likely originated from the Superior Craton and Midcontinent Rift and/or Grenville orogen, respectively. In this work, the synthesis of radioactive nanostructures by water radiolysis was studied. The irradiation processes were done in the Missouri University of Science and Technology research nuclear reactor (MSTR).

Radioactive gold nanoparticles (AuNPs) were synthesized from aqueous solutions containing the metal salt precursors by radiolysis of water. Seven different samples were irradiated at 200kW of thermal power for 0.5, 1, 3, 5, 10, 30, and 60 minutes. The average sizes of the obtained nanoparticles ranged from 3 nm to 400 nm, it was found that the particle size decreased with the irradiation time. Some agglomerations of particles were found …

Monte Carlo experiments aimed at assessing the statistical power of structural equation modelling (SEM) techniques typically focus on true population path coefficients, ignoring true sample path coefficients. We demonstrate the limitations stemming from such practice in statistical power assessments. This is done in the context of SEM techniques employing the partial least squares (PLS) method, where power claims have led to much recent debate. "In order to maximize bioethanol production an efficient pretreatment method for the hydrolysis of polysaccharides to fermentable sugars is necessary. Commonly used pretreatment methods are the slow enzymatic hydrolysis and the corrosive mineral acid hydrolysis process that requires a post-treatment neutralization step and generates waste stream. We investigated the high temperature water with carbonic acid catalyst as an alternative method. "Acetaminophen (N-acetyl-p-aminophenol, APAP) is one of the most widely used over-the-counter antipyretic analgesic medications. It is safe at therapeutic doses, but an overdose can result in severe hepato-nephrotoxicity, a leading cause of drug-induced acute liver failure in the U.S. Although a few different mechanisms have been proposed for APAP-induced toxicity, a significant amount of evidence has pointed to the potential involvement of oxidative stress in acetaminophen toxicity. Adaptive resonance theory (ART), an unsupervised learning clustering algorithm, is a clustering method that can learn arbitrary input patterns in a stable, fast and self-organizing way.
