Copyright © 2015 Wolters Kluwer Health, Inc. The Caenorhabditis elegans model is a rapid and inexpensive method to address pharmacologic questions. We describe the use of C. elegans to explore 2 pharmacologic questions concerning candidate antiobesity drugs and illustrate its potential usefulness in pharmacologic research: (1) to determine a ratio of betahistine-olanzapine that blocks the olanzapine-induced intestinal fat deposition (IFD) as detected by Nile red staining and (2) to identify the mechanism of action of a pharmaceutical candidate AB-101 that reduces IFD. Olanzapine (53 mg/mL) increased the IFD (12.1 6 0.1%, P < 0.02), which was blocked by betahistine (763 mg/mL, 39.3 6 0.01%, P < 0.05) in wild-type C. elegans (N2). AB-101 (1.0%) reduced the IFD in N2 (P < 0.05), increased the pharyngeal pumping rate (P < 0.05), and reversed the elevated IFD induced by protease inhibitors atazanavir and ritonavir (P < 0.05). AB-101 did not affect IFD in a ACS null mutant strain acs-4(ok2872) III/hT2[bli-4(e937) let-?(q782) qIs48](I;III) suggesting an involvement of the lipid oxidation pathway and an upregulation of CPT-1. Our studies suggest that C. elegans may be used as a resource in pharmacologic research. This article is intended to stimulate a greater appreciation of its value in the development of new pharmaceutical interventions.

Robustness Of The Potential And Flux Landscapes In Fission Yeast Cell Cycle Networks, Xiaosheng Luo

Using fission yeast cell cycle as an example, we uncovered that the non-equilibrium network dynamics and global properties are determined by two essential features: the potential landscape and the flux landscape. While potential landscape quantifies the probabilities of different states forming hills and valleys, the flux landscape quantifies the probability fluxes of different loops flowing through states. These two landscapes can be quantified through the decomposition of the dynamics into the detailed balance preserving part and detailed balance breaking non-equilibrium part. While the funneled potential landscape is often crucial for the stability of the single attractor networks, we have uncovered …

Linking Transcriptional And Physiological Responses To High Light Stress In A Wild-Type And A Photomorphogenic Tomato Mutant, Jordyn Radke

Tomato (Solanum lycopersicon) is the leading vegetable crop worldwide and an important part of a healthy human diet. In this study we used Next-Generation Sequencing, metabolomics and gene ontologies to study the effects of high light stress on the photomorphogenic hp-2dg tomato mutant. Wild-type (cv. ‘Manapal’) and mutant plants were grown for six weeks under low light and transferred to high light for one week. Illumina-based RNA Sequencing was performed and analysis of differential gene expression identified over 650 transcripts that were strongly regulated by light intensity (FC>4, p2,p<0,01). HPLC analysis of tomato leaves showed differential accumulation of phenylpropanoid metabolites such as chlorogenic acid, rutin, and caffeoylglucaric acid which was strongly dependent on genotype and light condition. Our findings show insightful differences between the wildtype and the photomorphogenic mutant as well as novel genes involved in high light adaptation in plants.

Summary: 1. Warming temperatures cause changes in growing seasons and prey that drive earlier breeding by birds, especially dietary specialists within homogenous habitat. Less is known about how generalists respond to climate-associated shifts in growing seasons or prey, which may be occurring at different rates across land cover types used by predators. 2. The organism is one of the fundamental concepts of biology and has been at the center of many discussions about biological individuality, yet what exactly it is can be confusing. The definition that we find generally useful is that an organism is a unit in which all the subunits have evolved to be highly cooperative, with very little conflict. We focus on how often organisms evolve from two or more formerly independent organisms. Two canonical transitions of this type—replicators clustered in cells and endosymbiotic organelles within host cells—demonstrate the reality of this kind of evolutionary transition and suggest conditions that …

BACKGROUND: Previous research has shown fluid replacement beverages ingested after exercise can affect hydration biomarkers. No specific hydration marker is universally accepted as an ideal rehydration parameter following strenuous exercise. Currently, changes in body mass are used as a parameter during post-exercise hydration. Additional parameters are needed to fully appreciate and better understand rehydration following strenuous exercise. This randomized, double-blind, parallel-arm trial assessed the effect of high-pH water on four biomarkers after exercise-induced dehydration.

It has been well documented that cognitive conflict is sensitive to the relative proportion of congruent and incongruent trials. However, few studies have examined whether affective conflict processing is modulated as a function of proportion congruency (PC). To address this question we recorded eventrelated potentials (ERP) while subjects performed both cognitive and affective face-word Stroop tasks. By varying the proportion of congruent and incongruent trials in each block, we examined the extent to which PC impacts both cognitive and affective conflict control at different temporal stages. The objective of this study was to evaluate learning outcomes and teaching strategies and the effect on certification exam pass rate in dietetic education programs. A non-experimental associational cross-sectional mixed methods design was chosen for the study in order to gather objective data through the use of the previously validated Constructivist Learning Environment Survey. River otters (Lontra Canadensis) were extirpated in Nebraska by the early 1900’s, but in 1986, the Nebraska Game and Parks Commission (NGPC) began reintroduction efforts. Following reintroductions, the otter was listed as a tier-1 at-risk species in Nebraska. With increasing otter populations, NGPC is evaluating a de-listing plan. In order to inform de-listing efforts, I surveyed Nebraska’s rivers documenting otter sign and used modeling techniques to estimate otter distribution and habitat suitability.
Otter sign surveys were conducted on the navigable rivers of Nebraska. Occupancy modeling techniques were used to examine patterns in otter detections. The best model incorporated distance …

The preparation of α-glucosidase inhibitory peptides was gotten, which was derived from ginkgo biloba by enzymatic hydrolysis with papain. A factorial design experiment was conducted, while, the effects of concentration of substrate, pH, E/S ratio, extraction temperature and time were studied. And then, it was fractioned by using membrane ultrafiltration with molecular weight cutoffs of 100, 50, 30, 10 and 3 kDa. The results showed: with the concentration of substrate 1.5%, pH 7.36, E/S ratio of 5 g/100 g, temperature of 57.9 ℃ and extraction time of 4.4 h, the inhibitory activities ofα-glucosidase was the highest, and reached 17.18%. The CarajaÂs National Forest contains some of the largest iron ore deposits in the world. The majority of the minerals are found below a plant community known as Savana Metalo fila, or ªCangaº, which represents only 3% of the landscape within the CarajaÂs National Forest (CNF). olid stress and tissue stiffness affect tumour growth, invasion, metastasis and treatment. Unlike stiffness, which can be precisely mapped in tumours, the measurement of solid stresses is challenging. Here, we show that 2D spatial maps of the solid stress and the resulting elastic energy in excised or in situ tumours with arbitrary shapes and a wide range of sizes can be obtained via three distinct and quantitative techniques that rely on the measurement of tissue displacement after disruption of the confining structures. The mining of lead and zinc ores in the Tri-State Mining District of Kansas, Missouri, and Oklahoma resulted in some of the most severe environmental degradation in the country. Large amounts of waste rock, known as ‘chat’, were produced during mining and left in surface piles in the surrounding area. These chat piles, located mostly near the town of Picher, Oklahoma contain potentially harmful elements that can be leached from the rock into local streams and sediment. Objective—Breastfeeding is linked to lower rates of childhood obesity. Human milk contains cortisol, known to regulate glucose storage and metabolism. We aimed to test the hypothesis that early exposure to cortisol in human breast milk helps to modulate infant BMI trajectories over the first two years of life.
Methods—Growth curve modeling was used to examine whether infant exposure to cortisol in human milk at 3 months predicted changes in child body mass index percentile (BMIP) at 6, 12, and 24 months of age in 51 breastfeeding mother-child pairs.

Based on the raw materials of mussel meat after taking out the pearl, ultra high pressure is used to extract mussel polysaccharide. The response surface experimental design was used to get the training samples for the neutral network, while the sophisticated neutral network was used for training and stimulating, analyzing extraction factors (pressure strength, solid- liquid ratio and pressure holding time), the interaction between the factors that affected the extraction rate of mussel polysaccharide, and optimizing the ultra high pressure extraction process of mussel polysaccharide. With a low-power microwave treatment on Huangguan pear fruits, different levels of low-power microwave treatment on fruit storage quality and the influence of the core browning were studied. Through the determinations of total phenols, polyphenol oxidase (PPO), malondialdehyde (MDA), DPPH in Huangguan pear core and ethylene, respiration intensity, hardness in storage period, the optimal treating conditions were gotten. Essential oil was extracted from Lysimachia foenum-graecum Hance using steam distillation method. The extraction conditions of essential oil were optimized by orthogonal experimental design, and its preservation on longan was studied. The results showed that the optimum extraction technology were: the concentration of sodium chloride 4%, the distilling time was 6 h, and the solid-liquid ratio of 1∶10 (g/mL). Under the condition, the extraction rate of essential oil reached 0.167 2%. The preservation results indicated: the antibacterial effects of 4 treatments, the content of Vitamin C and soluble solid decreased gradually, and weight loss and corruption rate of longan increased …

Puffy fruit and soluble solid content (SSC) are important indexes for evaluating the quality of citrus. The feasibility was discussed for detecting Puffing disease and SSC of intact citrus simultaneously by online visible-near infrared (visible-NIR) transmittance spectroscopy. The spectra were recorded with the integration time of 100 ms in the wavelength range of 350~1 150 nm when the samples were conveyed at the speed of five samples per second.
