Myelodysplastic syndromes (MDS) are clonal disorders of haematopoiesis characterised by dysplastic changes of major myeloid cell lines. However, the mechanisms underlying these dysplastic changes are poorly understood. Here, we used a genetically modified mouse model and human patient data to examine the physiological roles of H2AX in haematopoiesis and how the loss of H2AX contributes to dyserythropoiesis in MDS. H2AX knockout mice showed cell-autonomous anaemia and erythroid dysplasia, mimicking dyserythropoiesis in MDS. Also, dyserythropoiesis was increased in MDS patients with the deletion of chromosome 11q23, where H2AX is located. Although loss of H2AX did not affect the early stage of …

Striped bass (Morone saxatilis), white bass (Morone chrysops), and their hybrid are an important group of fish prized for recreational angling in the United States, and there and abroad as a high-value farmed fish. Regardless of habitat, it is not uncommon for fish of the genus Morone to encounter and cope with conditions of scarce oxygen availability. Previously, we determined that hybrid striped bass reared under conditions of chronic hypoxia exhibited reduced feed intake, lower lipid and nutrient retention, and poor growth. To better understand the molecular mechanisms governing these phenotypes, in the present study, we examined the …

Cross-Platform Normalization Of Microarray And Rna-Seq Data For Machine Learning Applications, Jeffrey A. Thompson, Jie Tan, Casey S. Greene
Dartmouth Scholarship
Large, publicly available gene expression datasets are often analyzed with the aid of machine learning algorithms. Although RNA-seq is increasingly the technology of choice, a wealth of expression data already exist in the form of microarray data. If machine learning models built from legacy data can be applied to RNA-seq data, larger, more diverse training datasets can be created and validation can be performed on newly generated data. We developed Training Distribution Matching (TDM), which transforms RNA-seq data for use with models constructed from legacy platforms. We evaluated TDM, as well as quantile normalization, nonparanormal transformation, and a simple log …

The 40-Residue Insertion In Vibrio Cholerae Fadr Facilitates Binding Of An Additional Fatty Acyl-Coa Ligand, Wei Shi, Gabriela Kovacikova, Wei Lin, Ronald. K. Taylor, Karen Skorupski, F. Jon Kull
Dartmouth Scholarship
FadR is a master regulator of fatty acid metabolism and influences virulence in certain members of Vibrionaceae. Among FadR homologues of the GntR family, the Vibrionaceae protein is unusual in that it contains a C-terminal 40-residue insertion. Here we report the structure of Vibrio cholerae FadR (VcFadR) alone, bound to DNA, and in the presence of a ligand, oleoyl-CoA. Whereas Escherichia coli FadR (EcFadR) contains only one acyl-CoA-binding site in each monomer, crystallographic and calorimetric data indicate that VcFadR has two. One of the binding sites resembles that of EcFadR, whereas the other, comprised residues from the insertion, has not …

Processes Controlling Carbon Cycling In Antarctic Glacier Surface Ecosystems, Elizabeth A. Bagshaw, Martyn Tranter, Jemma L. Wadham, Andrew G. Fountain, A. Dubnick, S. Fitzsimons
Geology Faculty Publications and Presentations
Glacier surface ecosystems, including cryoconite holes and cryolakes, are significant contributors to regional carbon cycles. Incubation experiments to determine the net production (NEP) of organic matter in cryoconite typically have durations of 6-24 hours, and produce a wide range of results, many of which indicate that the system is net heterotrophic. We employ longer term incubations to examine the temporal variation of NEP in cryoconite from the McMurdo Dry Valleys, Antarctica to examine the effect of sediment disturbance on system production, and to understand processes controlling production over the lifetimes of glacier surface ecosystems. The shorter-term incubations have durations of …

Development Of Cad-Related Items For A Concept Inventory For Engineering Graphics, Steven Nozaki, Nancy Study, Heidi Steinhauer, Sheryl Sorby, Mary Sadowski, Ron Miller
ASEE EDGD Midyear Conference
This research paper describes the development of a concept inventory for Engineering Graphics. As instructional methods continue to evolve in the area of graphics, there is a need for a concept inventory instrument that is able to measure the students’ misconceptions of important graphical concepts. In a Delphi study, CAD was identified as an important component of the graphics curriculum to be included in the concept inventory. There have been unanticipated challenges in the creation of multiple-choice items designed to assess the understanding of the concepts in the CAD construct. This paper is a work-in-progress describing the evolution of possible …

Models For Hsv Shedding Must Account For Two Levels Of Overdispersion, Amalia Magaret
UW Biostatistics Working Paper Series
We have frequently implemented crossover studies to evaluate new therapeutic interventions for genital herpes simplex virus infection. The outcome measured to assess the efficacy of interventions on herpes disease severity is the viral shedding rate, defined as the frequency of detection of HSV on the genital skin and mucosa. We performed a simulation study to ascertain whether our standard model, which we have used previously, was appropriately considering all the necessary features of the shedding data to provide correct inference. We simulated shedding data under our standard, validated assumptions and assessed the ability of 5 different models to reproduce the …

Hyaluronidase And Hyaluronan Oligosaccharides Promote Neurological Recovery After Intraventricular Hemorrhage, Govindaiah Vinukonda, Preeti Dohare, Arslan Arshad, Muhammad T. Zia, Sanjeet Panda, Ritesh Korumilli, Praveen Ballabh
NYMC Faculty Publications
Intraventricular hemorrhage (IVH) in premature infants results in inflammation, arrested oligodendrocyte progenitor cell (OPC) maturation, and reduced myelination of the white matter. Hyaluronan (HA) inhibits OPC maturation and complexes with the heavy chain (HC) of glycoprotein inter-α-inhibitor to form pathological HA (HC-HA complex), which exacerbates inflammation. Therefore, we hypothesized that IVH would result in accumulation of HA, and that either degradation of HA by hyaluronidase treatment or elimination of HCs from pathological HA by HA oligosaccharide administration would restore OPC maturation, myelination, and neurological function in survivors with IVH. To test these hypotheses, we used the preterm rabbit model of …

Reversible Recruitment Of A Homeostatic Reserve Pool Of Synaptic Vesicles Underlies Rapid Homeostatic Plasticity Of Quantal Content, Xueyong Wang, Martin J. Pinter, Mark M. Rich
Neuroscience, Cell Biology & Physiology Faculty Publications
Homeostatic regulation is essential for the maintenance of synaptic strength within the physiological range. The current study is the first to demonstrate that both induction and reversal of homeostatic upregulation of synaptic vesicle release can occur within seconds of blocking or unblocking acetylcholine receptors at the mouse neuromuscular junction. Our data suggest that the homeostatic upregulation of release is due to Ca2+-dependent increase in the size of the readily releasable pool (RRP). Blocking vesicle refilling prevented upregulation of quantal content (QC), while leaving baseline release relatively unaffected. This suggested that the upregulation of QC was due to mobilization …

Hyperfine Rather Than Spin Splittings Dominate The Fine Structure Of The B 4Σ-–X 4Σ- Bands Of Aic, Dennis J. Clouthier, Aimable Kalume
Chemistry Faculty Publications
Laser-induced fluorescence and wavelength resolved emission spectra of the B 4Σ−–X 4Σ− band system of the gas phase cold aluminum carbide free radical have been obtained using the pulsed discharge jet technique. The radical was produced by electron bombardment of a precursor mixture of trimethylaluminum in high pressure argon. High resolution spectra show that each rotational line of the 0-0 and 1-1 bands of AlC is split into at least three components, with very similar splittings and intensities in both the P- and R-branches. The observed structure was reproduced by assuming b …

Modeling Postpartum Depression In Rats: Theoretic And Methodological Issues, Ming Li, Shinn-Yi Chou
Modeling Postpartum Depression In Rats: Theoretic And Methodological Issues, Ming Li, Shinn-Yi Chou
Department of Psychology: Faculty Publications
The postpartum period is when a host of changes occur at molecular, cellular, physiological and behavioral levels to prepare female humans for the challenge of maternity. Alteration or prevention of these normal adaptions is thought to contribute to disruptions of emotion regulation, motivation and cognitive abilities that underlie postpartum mental disorders, such as postpartum depression. Despite the high incidence of this disorder, and the detrimental consequences for both mother and child, its etiology and related neurobiological mechanisms remain poorly understood, partially due to the lack of appropriate animal models. In recent decades, there have been a number of attempts to …

Over the last half-century, coral diseases have contributed to the rapid decline of coral populations throughout the Caribbean region. Some coral diseases appear to be potentially infectious, yet little is known about their modes of transmission. This study experimentally tested whether dark-spot syndrome on Siderastrea siderea was directly or indirectly transmissible to neighboring coral colonies. We also tested whether open wounds were necessary to facilitate disease transmission. At the completion of the experiments, we sampled bacterial communities on diseased, exposed, and healthy coral colonies to determine whether bacterial pathogens had transmitted to the susceptible colonies. We saw no evidence of either direct …
Understanding The Interaction Between Ulk1 And Atg9 And Autophagy, Ryan Hyatt, Joshua L. Andersen
Understanding The Interaction Between Ulk1 And Atg9 And Autophagy, Ryan Hyatt, Joshua L. Andersen
Journal of Undergraduate Research
Autophagy is an adaptive catabolic process of self-digestion, a process by which the cell recycles aged organelles and other structures. Interestingly, changes in the regulation of autophagy have been linked to infections, cancers, neurodegeneration, aging, and heart disease (Arroyo, Daniela S. et al. 2014). Understanding the mechanisms which control autophagy will aid in better treatment of those diseases, specifically in the development of effective pharmaceuticals.
