Selective Binding Of Airapl Tandem Uims To Lys48-Linked Tri-Ubiquitin Chains, Simin Rahighi, Ilauna Braunstein, Nicola Ternette, Benedikt Kessler, Masato Kawasaki, Ryuichi Kato, Tsutomu Matsui, Ariel Stanhill, Soichi Wakatsuki

Pharmacy Faculty Articles and Research

Lys48-linked ubiquitin chains act as the main targeting signals for protein degradation by the proteasome. Here we report selective binding of AIRAPL, a protein that associates with the proteasome upon exposure to arsenite, to Lys48-linked tri-ubiquitin chains. AIRAPL comprises two ubiquitin-interacting motifs in tandem (tUIMs) that are linked through a flexible inter-UIM region. In the complex crystal structure UIM1 binds the proximal ubiquitin, whereas UIM2 (the double-sided UIM) binds non-symmetrically to the middle and distal ubiquitin moieties on either side of the helix. Specificity of AIRAPL for Lys48-linked ubiquitin chains is determined by UIM2, and the flexible inter-UIM linker increases …

Systems Perturbation Analysis Of A Large-Scale Signal Transduction Model Reveals Potentially Influential Candidates For Cancer Therapeutics, Bhanwar L. Puniya, Laura Allen, Colleen Hochfelder, Mahbubul Majumder, Tomáš Helikar Feb 2016

Systems Perturbation Analysis Of A Large-Scale Signal Transduction Model Reveals Potentially Influential Candidates For Cancer Therapeutics, Bhanwar L. Puniya, Laura Allen, Colleen Hochfelder, Mahbubul Majumder, Tomáš Helikar

Mathematics Faculty Publications

Dysregulation in signal transduction pathways can lead to a variety of complex disorders, including cancer. Computational approaches such as network analysis are important tools to understand system dynamics as well as to identify critical components that could be further explored as therapeutic targets. Here, we performed perturbation analysis of a large-scale signal transduction model in extracellular environments that stimulate cell death, growth, motility, and quiescence. Each of the model’s components was perturbed under both loss-of-function and gain-of-function mutations. Using 1,300 simulations under both types of perturbations across various extracellular conditions, we identified the most and least influential components based on …

An Evaluation Of Extraction Techniques For Arsenic In Staple Diets (Fish And Rice) Utilising Both Classical And Enzymatic Extraction Methods, Bashdar A. Sadee, Mike E. Foulkes, Steve J. Hill Feb 2016

An Evaluation Of Extraction Techniques For Arsenic In Staple Diets (Fish And Rice) Utilising Both Classical And Enzymatic Extraction Methods, Bashdar A. Sadee, Mike E. Foulkes, Steve J. Hill

School of Geography, Earth and Environmental Sciences

Enzymatic extraction methods were evaluated with classical extraction approaches for the determination of arsenic in food. The extraction efficiency for total arsenic was determined by analysing CRM materials DORM-3 fish protein, NIES 106 rice flour and GBW10015 spinach. These were compared with total arsenic concentration determined using microwave-assisted acid digestion and ICP-MS. The total arsenic concentrations in the CRM materials were in good agreement with the certified values. Enzymatic hydrolysis using trypsin has been successfully employed to extract arsenic species in DORM-3 and fish samples. Whilst this method of hydrolysing the proteins worked well for the fish samples, an alternative …

Use Of Galvanic Skin Responses, Salivary Biomarkers, And Self-Reports To Assess Undergraduate Student Performance During A Laboratory Exam Activity, Idalis Villanueva, Maria Valladares, Wade H. Goodridge Feb 2016

Use Of Galvanic Skin Responses, Salivary Biomarkers, And Self-Reports To Assess Undergraduate Student Performance During A Laboratory Exam Activity, Idalis Villanueva, Maria Valladares, Wade H. Goodridge

Engineering Education Faculty Publications

Typically, self-reports are used in educational research to assess student response and performance to a classroom activity. Yet, addition of biological and physiological measures such as salivary biomarkers and galvanic skin responses are rarely included, limiting the wealth of information that can be obtained to better understand student performance. A laboratory protocol to study undergraduate students' responses to classroom events (e.g., exams) is presented. Participants were asked to complete a representative exam for their degree. Before and after the laboratory exam session, students completed an academic achievement emotions self-report and an interview that paralleled these questions when participants wore a …

Our Flight From Death Is Killing Us: Applying A Psychological Framework To A Christian Understanding Of The Human Condition, G. Mark Griffith Feb 2016

Our Flight From Death Is Killing Us: Applying A Psychological Framework To A Christian Understanding Of The Human Condition, G. Mark Griffith

Doctor of Ministry

Our awareness of mortality motivates us to subconsciously act in ways that are damaging. This dissertation engages the problems to individual and communities generated by our unexamined awareness of death. This awareness motivates us to attempt to flee death's grasp—even though we know there is no escape. The flight from death robs us not only of our ability to live full lives but also robs us of our ability to faithfully follow Jesus into the world. At times, we even distort religion and use it to shield us from death. Faith practices can be manipulated to draw us away from …

Signed, Sealed, Delivered: Baptism As Beginning In Christ, And Becoming Like Christ, Eric E. Peterson Feb 2016

Signed, Sealed, Delivered: Baptism As Beginning In Christ, And Becoming Like Christ, Eric E. Peterson

Doctor of Ministry

This dissertation involves applied research, relying on the foundation of sacramental
theology, to answer the question, "What is the sign that has the power to help people
embrace not only a sacred self-identity, but to live holy, meaningful, and abundant
lives in a largely broken and sacrilegious world?" Beginning with a review of the major
and various assumptions people have historically adhered to in the pursuit of a good
life, the dissertation proceeds to argue for the superiority of a sacramental
perspective. Employing a semiotics of baptism, chapter two provides a critical analysis
of the opposing forces of "cultural baptism" …

Biblical Hospitality: An Environment For Leader Development, Joanne L. Holz Feb 2016

Biblical Hospitality: An Environment For Leader Development, Joanne L. Holz

Doctor of Ministry

The Salvation Army in the southeastern United States is currently facing challenges with retention of its ordained leaders called Officers. Causes for post- commissioning defection center around the need for changes in the context in which training and learning occurs. This study examines the need for Biblical Hospitality as an environment that hosts transformational opportunities and creates the context needed for leadership development across the system and within individuals in the U. S. A. Southern Territory.

There is a return to Biblical Hospitality, which is finding its place in multiple areas of life with life-transforming opportunities and results. For the …

Matthews views Myers as a "well-educated liberal arts specialist...who can talk with anybody intelligently about anything that person is interested in." Myers recommended book purchases to Matthews and he summarizes the effect that and conversations about French history had on him. He recounts why and how Myers recruited him to be Dean of Students and describes changes to Student Affairs that followed, what he saw in Cabinet and Board of Trustee meetings about budget decisions, an incidence of racism involving Sodexho (ca 2000), and remarks Myers made on the first anniversary of September 11, 2001 [n.b., there are no recordings …
