The transition of sessile epithelial cells to a migratory, mesenchymal phenotype is essential for metazoan development and tissue repair, but this program is exploited by tumor cells in order to escape the confines of the primary organ site, evade immunosurveillance, and resist chemo-radiation. In addition, epithelial-to-mesenchymal transition (EMT) confers stem-like properties that increase efficiency of colonization of distant organs. This review evaluates the role of cell-cell junctions in suppressing EMT and maintaining a quiescent epithelium. We discuss the conflicting data on junctional signaling in cancer and recent developments that resolve some of these conflicts. We focus on evidence from breast …

Rewriting The How-To Of Parenting: What Is Really Modern About Abc’S Modern Family, Sylvia Henneberg
Rewriting The How-To Of Parenting: What Is Really Modern About Abc’S Modern Family, Sylvia Henneberg
Journal of Interdisciplinary Feminist Thought
Despite its unconventional family structures and playful gestures at redefining traditional gender scripts, ABC’s domestic comedy, Modern Family, perpetuates perceived notions of femininity and masculinity within the context of three families whose differences from the nuclear family are far from radical. Reassuring even its most conservative viewers on these counts, the show does, however, portray a potentially unsettling approach to parenting. The dynamics of parenting are unstable, bending gender rules and disregarding boundaries normally set by age, race, and social class. As normative notions of parenting are questioned and expanded, parenthood is no longer an immutable institution built on hard …

Ocean acidification (OA) effects on photophysiology and calcification were examined in Halimeda discoidea, a calcifying macroalga that produces tropical reef sediments. Photosynthetic parameters, including maximum photosynthetic rate (Pmax), photosynthetic efficiency (α) and compensating irradiance (Ic) were determined in short-term assays on live thalli after a 10 d exposure to 4 levels of CO2 partial pressures (pCO2; 491, 653, 982 and 1201 µatm) under saturating (300 µmol photons m-2 s-1) and sub-saturating (90 µmol photons m-2 s-1) irradiance in an aquaria study. Injury-Induced Neuronal Turnover With Zinc Sulfate Affects Cilitated Olfactory Sensory Neurons More Than Microvillous Olfactory Sensory Neurons In The Adult Zebrafish, James Hentig
Honors Theses
Zinc sulfate is a known olfactory toxicant, although its effects on the olfactory epithelium of zebrafish have not been examined. Previous observations utilizing Triton X-100 found selective retention of some olfactory sensory neurons and loss of others. Based on those findings, we hypothesized that the retention of the ability to sense amino acids, detected by microvillous olfactory sensory neurons, and loss of the ability to perceive bile salts, detected by ciliated olfactory sensory neurons, is a universal response to chemical ablation. Fish were treated with zinc sulfate and survived for 2, 5, 10 and 14 days post treatment and processed …

Dissolved lignin is a well-established biomarker of terrigenous dissolved organic matter (DOM) in the ocean, and a chromophoric component of DOM. Although evidence suggests there is a strong linkage between lignin concentrations and chromophoric dissolved organic matter (CDOM) absorption coefficients in coastal waters, the characteristics of this linkage and the existence of a relationship that is applicable across coastal oceans remain unclear. Here, 421 paired measurements of dissolved lignin concentrations (sum of nine lignin phenols) and CDOM absorption coefficients [ag(λ)] were used to examine their relationship along the river-ocean continuum (0–37 salinity) and across contrasting coastal oceans (sub-tropical, temperate, high-latitude). …

The hyperphosphorylated microtubule-associated protein tau is present in several neurodegenerative diseases, although the causal relationship remains elusive. Few mouse models used to study Alzheimerlike dementia target tau phosphorylation. We created an inducible pseudophosphorylated tau (Pathological Human Tau, PH-Tau) mouse model to study the effect of conformationally modified tau in vivo. Leaky expression resulted in two levels of PH-Tau: low basal level and higher upon induction (4% and 14% of the endogenous tau, respectively). Unexpectedly, low PH-Tau resulted in significant cognitive deficits, decrease in the number of synapses (seen by EM in the CA1 region), reduction of synaptic proteins, and localization …

Drug Repurposing To Target Ebola Virus Replication And Virulence Using Structural Systems Pharmacology, Zheng Zhao, Che Martin, Raymond Fan, Philip E. Bourne, Lei Xie
Publications and Research
Background: The recent outbreak of Ebola has been cited as the largest in history. Despite this global health crisis, few drugs are available to efficiently treat Ebola infections. Drug repurposing provides a potentially efficient solution to accelerating the development of therapeutic approaches in response to Ebola outbreak. To identify such candidates, we use an integrated structural systems pharmacology pipeline which combines proteome-scale ligand binding site comparison, protein-ligand docking, and Molecular Dynamics (MD) simulation.
Results: One thousand seven hundred and sixty-six FDA-approved drugs and 259 experimental drugs were screened to identify those with the potential to inhibit the replication and virulence …

Proteome Based Construction Of The Lymphocyte Function-Associated Antigen 1 (Lfa-1) Interactome In Human Dendritic Cells, C Eich, E Lasonder, Lj Cruz, I Reinieren-Beeren, Et Al
School of Biomedical Sciences
The β2-integrin lymphocyte function-associated antigen 1 (LFA-1) plays an important role in the migration, adhesion and intercellular communication of dendritic cells (DCs). During the differentiation of human DCs from monocyte precursors, LFA-1 ligand binding capacity is completely lost, even though its expression levels were remained constant. Yet LFA-1-mediated adhesive capacity on DCs can be regained by exposing DCs to the chemokine CCL21, suggesting a high degree of regulation of LFA-1 activity during the course of DC differentiation. The molecular mechanisms underlying this regulation of LFA-1 function in DCs, however, remain elusive. To get more insight we attempted to identify specific …
