Impacts From The Use Of Antibiotics In Livestock: Methods Of Transmission Of Antibiotic Resistance From Livestock To Humans, Kristin M. Walden
Impacts From The Use Of Antibiotics In Livestock: Methods Of Transmission Of Antibiotic Resistance From Livestock To Humans, Kristin M. Walden
Biology: Student Scholarship & Creative Works
Antibiotic use in livestock production has been around since the 1950s. Antibiotic feed is used in livestock and other meat producing animals for three reasons: illness prevention, illness treatment, and growth promotion. Unfortunately, since the time that antibiotics were first invented, antibiotic resistant bacteria have become a threat to public health. There are many studies showing methods of transmission of antibiotic resistance from livestock to humans. Antibiotic resistance can spread from livestock to soil, water, insects, and food, which ultimately comes into contact with humans. A proposed study to measure antibiotic resistance when eliminating antibiotic feed will provide a hypothesis …

Disruption Of Type 3 Adenylyl Cyclase Expression In The Hypothalamus Leads To Obesity, Hong Cao, Xuanmao Chen, Yimei Yang, Daniel R. Storm
Molecular, Cellular & Biomedical Sciences
Evidence from human studies and transgenic mice lacking the type 3 adenylyl cyclase (AC3) indicates that AC3 plays a role in the regulation of body weight. It is unknown in which brain region AC3 exerts such an effect. We examined the role of AC3 in the hypothalamus for body weight control using a floxed AC3 mouse strain. Here, we report that AC3 flox/flox mice became obese after the administration of AAV-CRE-GFP into the hypothalamus. Both male and female AC3 floxed mice showed heavier body weight than AAV-GFP injected control mice. Furthermore, mice with selective ablation of AC3 expression in the …

Deepening Knowledge On Cho Cells Metabolism Using Multiple Tracer Substrates, Ana Teixeira, Manuel Carrondo
Deepening Knowledge On Cho Cells Metabolism Using Multiple Tracer Substrates, Ana Teixeira, Manuel Carrondo
Cell Culture Engineering XV
Optimization of culture conditions through trial-and-error has led to much of the past progress on animal cell culture. Today, a renewed account of the underlying biologies through a “systems” view is providing a deeper knowledge of cellular physiology and laying the basis to reengineer desirable states of further increased culture performance. In this work, we took a comprehensive view of cellular metabolism by extensively profiling extracellular and intracellular metabolomes after 13C‑label supplementation in parallel labelling cultures of [1,2-13C]Glucose, [U-13C,15N]Asn, [U-13C,15N]Ser and [1-13C]Pyruvate. This integrative approach was used …

Cell Culture Engineering XV
The Eli Lilly biologics manufacturing facility in Kinsale, Ireland has been operational since 2010 with a 100% cell culture contamination control success rate. The presentation will review the holistic approach to facility protection from adventitious agents that underpins this success including:
- The risk assessment approach to points of entry and management via detectability and control measures
- The approach to personnel training that considers human factors, increased vigilance and event simulations following strategies that are used in chemical synthesis process safety
The presentation will then focus on control of adventitious virus. The talk will briefly comment on the early warning measures …
Adapting An In-Licensed/Acquired Cell Culture Process To Platform Conditions, Raghunath Shivappa
Adapting An In-Licensed/Acquired Cell Culture Process To Platform Conditions, Raghunath Shivappa
Cell Culture Engineering XV
The use of platform materials and processes for new products help streamline process development, enhance predictability and efficiency during manufacturing, and simplify the raw materials supply chain. Although common, adaptation of cell expression systems can require a significant amount of process development work to achieve desired product yield, quality, and comparability. Here, we present a progressive development approach for evaluating platform media and feeds for an acquired cell culture process.
A CHO-DG 44 cell line expressing a novel antibody product was procured through acquisition. The original non-platform media and fed-batch process yielded titers of approximately 3 g/L. A study was …
Impact Of Poloxamer 188 Variability On Biologics Manufacturing: Mitigations And Causal Investigation, Salim Charaniya
Impact Of Poloxamer 188 Variability On Biologics Manufacturing: Mitigations And Causal Investigation, Salim Charaniya
Cell Culture Engineering XV
Poloxamer 188 (P188), a water-soluble, synthetic copolymer with hydrophilic as well as lipophilic properties, is widely used as a cell protectant for bubble-induced shear during cultivation of suspension-adapted mammalian cells for large-scale production of protein-based therapeutics. Poor cell growth and lower product yield has been recently observed at commercial scales in more than one Chinese hamster ovary-based manufacturing process within Roche network when specific lots of P188 were used. An investigation is being conducted to unearth the root cause of “poor-performing” P188 lots and to identify means to mitigate the product supply risks. P188 raw material is currently sourced from …
Development And Application Of Glycosyltransferases For In Vitro Glycoengineering, Alfred Engel
Development And Application Of Glycosyltransferases For In Vitro Glycoengineering, Alfred Engel
Cell Culture Engineering XV
Glycosylation is an important posttranslational modification of proteins influencing protein folding, stability and regulation of the biological activity, e.g. IgG featuring terminal sialic acids were shown to suppress inflammatory response (ADCC) and increase serum half-life. Fully galactosylated structures seem to improve binding to the complement system (CDC).
Despite all efforts in cell-line engineering and process optimization to increase glycosylation homogeneity, today therapeutic proteins still show a heterogeneous glycosylation pattern, with large variations between bioprocesses and even from batch to batch. The use of glycosyltransferases for enzymatic synthesis of well-defined glycan structures will become an essential tool – at least in …
Comparison Of Commercial Cho Cell Media Formulations Using Material-Oriented Recurrent Spectral Libraries, Kelly Telu
Comparison Of Commercial Cho Cell Media Formulations Using Material-Oriented Recurrent Spectral Libraries, Kelly Telu
Cell Culture Engineering XV
Chinese hamster ovary (CHO) cells are commonly used for the production of biological therapeutics. Metabolic profiles of media components can be used to monitor process variability and look for markers that discriminate between batches of product. Currently, there exists no database of CHO media components or a method for the comparison of commercial media formulations. We are creating material-oriented libraries of CHO media components that include LC-MS/MS and GC-MS data. The libraries represent all metabolites that can be detected by LC-MS/MS and GC-MS and consist of recurrent spectra that cover all known fragmentation conditions and precursors. Recurrent spectra occur repeatedly …
Incidence And Potential Implications Of Methylglyoxal In Industrial Cell Culture Revisited, Frank Chaplen
Incidence And Potential Implications Of Methylglyoxal In Industrial Cell Culture Revisited, Frank Chaplen
Cell Culture Engineering XV
Methylglyoxal is a toxic by-product of glycolysis and amino acid metabolism in mammalian systems. The major route for methylglyoxal detoxification is the glyoxalase pathway, which consists of the enzymes glyoxalase I (GLO1) and glyoxalase II (GLO2). A required co-factor for the glyoxalase pathway is reduced glutathione. Evidence suggests that GLOI and methylglyoxal have important roles to play in the signal pathways associated with oxidative stress and necrotic cell death1. Previous work has demonstrated that growth conditions found in industrial cell culture have marked effects on endogenous methylglyoxal levels in Chinese hamster ovary (CHO)2. Furthermore, decreased levels …

Accelerated Bioprocess Characterization By Data Enrichment In Scale-Down Models, Viktor Konakovsky, Graham Mccreath, Jarka Glassey
Cell Culture Engineering XV
Scale-down models are often used for the definition of operating ranges in process development and validation and therefore data exploitation is an important task. Usually, the experimental scientists have very tight timelines and consequentially may often only perform the required routine of cleaning and sorting the data without more sophisticated analyses, even though it might improve data quality. This problem is more exacerbated by the rise of fully automated, miniaturized high-throughput equipment in up- and downstream process development where data processing automation is not just optional but a must. However, cleaned, sorted and time-aligned data alone do not guarantee a …

Sustainable Development: Challange, Policy, Solution, Karl W. Knotoff
Sustainable Development: Challange, Policy, Solution, Karl W. Knotoff
Student Theses 2015-Present
The conflict between our economic markets and environmental needs is going to be the biggest problem in implementing sustainable trends in our society as we proceed toward the sustainable development goals. Although it may seem intuitive and logical that sustainable development goals are not only good but a complete necessity, the implementation and application of sustainable innovations is seen differently by people. For instance, people living in poverty or low income areas see sustainability as a beacon of hope to promote social inclusion, equality and a healthier living environment through cleaner air and water solutions. Creating jobs to implement these …

Moving From A Bioreactor Scale-Up/Scale-Down Approach To A More Holistic Operational Design Space View, Gene Schaefer, Rick Yeager, Ted Deloggio
Cell Culture Engineering XV
Scale-up of bioreactor processes for biologic products has been an area of focus in the industry for over 50 years and the results have generally been successful through the application of a number of specific correlations across scales for mass transfer, mixing, etc. However, the ability to visualize and understand the bioreactor design space in terms of these parameters has generally been limited as has any general understanding of how close a given process comes to certain sensitivity limits at various scales. As part of a project which involved establishing scale-down models at two different sites with long-term objectives of …
An Addition Of Lithium Chloride Improves The Transient Gene Expression Yield In Cho Cells, Che Kim, Tae Kwang, Gyun Lee
An Addition Of Lithium Chloride Improves The Transient Gene Expression Yield In Cho Cells, Che Kim, Tae Kwang, Gyun Lee
Cell Culture Engineering XV
As hundreds of new recombinant therapeutic proteins enter into development each year, a cost-effective transient gene expression (TGE) system is required to assess these proteins at an early phase as a more rapid alternative to a stable gene expression system. While several strategies, such as genetic manipulation and environmental modification, have been attempted in TGE systems to improve the overall yield in Chinese hamster ovary (CHO) cells, chemical supplementation is often employed in industrial processes owing to its simplicity and effectiveness. Lithium chloride (LiCl), which is known to induce G2/M-phase cell cycle arrest, was found to be an attractive chemical …

Lay Theories Of Morality In The Lives Of Moral Exemplars, Atilla Cidam
Lay Theories Of Morality In The Lives Of Moral Exemplars, Atilla Cidam
Doctoral Dissertations
Psychological research on moral exemplars presents them as near perfect, focusing on their impeccable character and accomplishments. However, just like anybody else, they also have imperfections that are usually unrecognized by psychological research. The aim of this dissertation is to examine the implications of studying moral exemplars as if they are perfect. I will argue that focusing only on the positive qualities of moral exemplars, and ignoring the important ways that failures may have contributed to their development, imply an entity theory of morality where only certain perfect people possess unchangeable moral traits. In Chapter I, I provide an overview …
The Canine Genome: Discoveries, Applications, And Future Potential, Rachael Lander
The Canine Genome: Discoveries, Applications, And Future Potential, Rachael Lander
Honors Theses
A frequent question that educators often encounter is: what is the value of learning? Does knowledge have an inherent value, or should there be an economic benefit? The results of the collaborative efforts to determine the nucleotide sequence of the canine genome were used as a platform to assess these thesis statements. A literature review, practical experience in the laboratory, and interviews with several genome scientists of the contributions of the efforts to determine the genome sequence of the domestic dog, and the biomedical contributions that have been made to both canine and human health demonstrate the inherent value of …

The Mechanosensory Lateral Line System Mediates Activation Of Socially-Relevant Brain Regions During Territorial Interactions, Julie M. Butler, Karen P. Maruska
Faculty Publications
© 2016 Butler and Maruska. Animals use multiple senses during social interactions and must integrate this information in the brain to make context-dependent behavioral decisions. For fishes, the largest group ofvertebrates, the mechanosensory lateral line system provides crucial hydrodynamic information for survival behaviors, but little is known about its function in social communication. Our previous work using the African cichlid fish, Astatotilapia burtoni, provided the first empirical evidence that fish use their lateral line system to detect water movements from conspecifics for mutual assessment and behavioral choices. It is unknown, however, where this socially-relevant mechanosensory information is processed in the …

Mesenchymal Stem Cell Fate: Applying Biomaterials For Control Of Stem Cell Behavior, Hilary J. Anderson, Jugal Kishore Sahoo, Rein V. Ulijn, Matthew J. Dalby
Publications and Research
The materials pipeline for biomaterials and tissue engineering applications is under continuous development. Specifically, there is great interest in the use of designed materials in the stem cell arena as materials can be used to manipulate the cells providing control of behavior. This is important as the ability to “engineer” complexity and subsequent in vitro growth of tissues and organs is a key objective for tissue engineers. This review will describe the nature of the materials strategies, both static and dynamic, and their influence specifically on mesenchymal stem cell fate.

Indeno[1,2-b]fluorenes (IFs), while containing 4n π-electrons, are best described as two aromatic benzene rings fused to a weakly paratropic s-indacene core. In this study, we find that replacement of the outer benzene rings of an IF with benzothiophenes allows the antiaromaticity of the central s-indacene to strongly reassert itself. Herein we report a combined synthetic, computational, structural, and materials study of anti- and syn-indacenodibenzothiophenes (IDBTs). We have developed an efficient and scalable synthesis for preparation of a series of aryl- and ethynyl-substituted IDBTs. NICS-XY scans and ACID calculations reveal an increasingly antiaromatic core from …

Postembryonic Polarity Modification In The Acoel Convolutriloba Longifissura, Eric Young, James M. Sikes Ph.D.
Undergraduate Honors Theses
Metazoans establish the bilateral body plan early in embryogenesis by patterning orthogonal body axes with polarity that is unaltered during the lifetime of most animals. While some organisms re-establish/modify body axes during regeneration and asexual reproduction, the acoel flatworm Convolutriloba longifissura is unusual in its ability to modify left-right (L-R) axis polarity during longitudinal fission. We have developed C. longifissura as a model for studying the mechanisms of L-R polarity modification during postembryonic development. Regeneration experiments have elucidated the temporal dynamics of midline re-specification, suggesting that parallel L-R axes replace the pre-existing midline prior to longitudinal fission. We have characterized …

Foraging Choices Of Vampire Bats In Diverse Landscapes: Potential Implications For Land‐Use Change And Disease Transmission, Daniel G. Streicker, Jacob E. Allgeier
KIP Articles
In Latin America, the common vampire bat Desmodus rotundus is the primary reservoir of rabies, a zoonotic virus that kills thousands of livestock annually and causes sporadic and lethal human rabies outbreaks. The proliferation of livestock provides an abundant food resource for this obligate blood‐feeding species that could alter its foraging behaviour and rabies transmission, but poor understanding of the dietary plasticity of vampire bats limits understanding of how livestock influences rabies risk. We analysed individual‐ and population‐level foraging behaviour by applying δ 13 C and δ 15 N stable isotope analysis to hair samples from 183 vampire bats captured …
