Agrobacterium is widely considered, when suitably modified, to be the most effective vector for gene transfer into plant cells. For a long time, many cereals crops (monocotyledonous plants) were recalcitrant species to genetic modification, mainly as a result of their recalcitrance to in-vitro regeneration and their resistance to Agrobacterium infection. The aquaculture sector is currently seeking alternative feed ingredients to ensure future socioeconomic and environmental sustainability; microalgae sources are seen to be a promising option. Two nutrition trials were conducted in order to assess substituting plant based protein sources in diets for juvenile Nile tilapia (Oreochromis niloticus), with a range of inclusion levels of two proprietary defatted microalgae samples (Nannochloropsis sp. and Chlorella sp.). The experimental algal ingredients utilised were by-products sourced from a pilot-scale biofuel operation. There is currently worldwide pressure to establish Marine Protected Area (MPA) networks which are self-sustaining and will persistently protect habitats and species. In order for MPA networks to be effective, the species targeted for conservation must be able to disperse between protected areas and maintain a gene-flow necessary for population sustainability and persistence. Quantification of free fatty acids (FFA) in dairy products by gas chromatography flame ion'^ation detection is an important requirement for qua'ity, research, nutritional, authenticity and legislative purposes. Common procedures are typically variants of either the fatty acid methyl esterification (FAME) or direct on-column injection methods developed more than two decades ago. Much work has been done to study strategies of structured weight loss programs and examine their success. Pre-harvest sprouting (PHS) of wheat (Triticum aestivum L.) is a condition characterized by the early germination of spikes during moist environmental conditions. PHS lowers yield, degrades the quality of grain and thus limits the profits of wheat producers groups. During this investigation, proteomics studies of PHS-resistant and PHS-susceptible wheat embryos were conducted at different imbibition time periods via a cutting-edge technology called iTRAQ. To examine the relationship between baseline, post, and follow-up accelerometer derived physical activity (PA) in 9-10 year old children participating in iCook 4-H, a 16-week, 5- state, randomized control family centered childhood obesity prevention program. This family focus intervention was designed to promote PA through lifestyle modifications. Between group differences in minutes-per-day and intensity of PA were determined on accelerometer compliant participants. All subjects wore an accelerometer (Actigraph GT3X+) for 7 days at baseline, post, and follow-up intervention assessment. Recent concern regarding bobcat (Lynx rufus) population status has prompted researchers and managers to gather additional information about bobcats in South Dakota. From 2012–2015, we assessed population dynamics of bobcats occupying the west-central region of South Dakota. Our objectives were to: 1) estimate annual survival rates; 2) determine cause-specific mortality; 3) estimate a population size for the western prairie region of South Dakota; 4) estimate home range size of individually marked bobcats; 5) evaluate reproductive status; and 6) build a population model. Late Quaternary Paleoceanographic Evolution Of The Aegean Sea: Planktonic Foraminifera And Stable Isotopes, Ekrem Bursi̇n İşler, Ali̇ Engi̇n Aksu, Richard Nicholas Hiscott

Aspects of the paleoclimatic and paleoceanographic evolution of the Aegean Sea since ~130 ka are revealed by quantitative variations in planktonic faunal assemblages, the Δ18O and Δ13C isotopic composition of benthic and planktonic foraminifera, and Mg/Ca ratios in planktonic foraminifera extracted from five 6-10-m-long piston cores. Independent sea surface temperature (SST) estimates obtained using planktonic foraminiferal transfer functions and the Mg/Ca ratios show excellent agreement, with r2 correlation coefficients of 0.92-0.95. Planktonic foraminiferal assemblages are similar, through time, across several deep basins, suggesting that major changes must have occurred in near synchroneity across the Aegean Sea. Purpose: The purpose of this study was to collect and analyze data on the workload of students and student-athletes to determine if there is a significant difference in the workloads of student-athletes compared with non-athletes, including undergraduate and graduate students. It was hypothesized that student-athletes would spend more time in athletic activities, but would sacrifice time in other areas. The purpose of this study is to determine influencers and barriers to healthful dietary intake in rural youth in context of the bioecological model. Four focus groups as part of Ignite, a tri-state, five-year, community based partipatory research (CBPR) intervention were conducted with 6th to 8th grade adolescents from an economically disadvantaged community of rural South Dakota. Arbuscular mycorrhizal fungi (AMF) are obligate symbionts that form a mutualistic relationship with approximately 80% of terrestrial plant species. These obligate symbionts have a generally beneficial effect on their host such as increased nutrient acquisition, better tolerance to biotic and abiotic stresses, and the improvement of soil qualities. A mutant wheat (Triticum aestivum L.) genotype can constitutively produce hypersensitive-like flecks in the absence of any pathogen. The lesions of the mutant are thought to be the result of Reactive Oxygen Species (ROS), which are similarly implicated in hypersensitive reactions to fungal pathogens. Allopurinol can suppress ROS. Consequently, allopurinol was applied to a wheat lesion mutant to test the hypothesis that the same ROS are involved, regardless of the presence of the pathogen. ‘Ning7840’, a lesion mutant and ‘Alsen’, a related genotype that does not express the mutant phenotype were treated with different concentrations of allopurinol under controlled …

This research has been structured into 3 major parts. The first part evaluates a centrifugal evaporator to concentrate micellar casein concentrate (MCC). The second part evaluates a wiped film evaporator (WFE) to concentrate MCC. The third part evaluates heat stability of the high concentration MCC produced by WFE. In part 1, the primary objective was to achieve >25% total solids (TS) without a loss of water removal rate associated with high solids materials. The overall objectives of the six studies were to increase the feed efficiency in lactating dairy cows. The first study evaluated the efficacy of different water sources and treatment systems on ruminal parameters using an in vitro gas production system. Outcomes of the study suggested that the source and nutrient quality of water can affect rate of ruminal fermentation. A previous study revealed a quantitative trait locus (QTL) in maize on the short arm of chromosome 5 (5S) for endosperm amylose content. In that original study, both low and high amylose parental lines, H99ae and GEMS-0067 respectively, were homozygous recessive for amylose extender 1 (ae1) and the polymorphism responsible for this QTL was additive (semi-dominant). Located within the QTL interval is starch branching enzyme 1 (sbe1), which makes it a candidate gene. The objective of this study was to determine if the inclusion of a blend of essential oils and cobalt lactate (EOC) in the diet of lactating dairy cows could result in improved nutrient utilization and lactational performance in addition to assessing the application of EOC to control feed mycotoxins and spoilage. The trial was conducted in two experimental periods (Phase 1 (P1) and Phase 2 (P2)) on a commercial dairy in southwest Minnesota equipped with two robotic milking units. Increased spring rainfall and higher temperatures when combined with changing landuses and extensive tile drainage installation have contributed to the development of sodic and saline/sodic soils in the Northern Great Plains. The objectives of this dissertation were: 1) determine the impact of surface chemical treatments and cover crop on crop yields and soil remediation; 2) determine and describe soil spatial variability and develop a model to identify saline-sodic soils; and 3) evaluate cation impact on dispersion of bentonite clay and selected soils. The incorporation of bioprocessed soybean meals (BSBMs) as a protein replacement for marine-derived fish meal (MDFM) has gained momentum with the potential for aquafeed cost reduction, improved growth performance, and reducing exploitation on wild fish stocks. Further, potential health benefits and optimized production performance resulting from bioprocessed feed ingredients have led to research on the nutraceutical benefits of plant-based ingredients. A novel proprietary MBBM has been developed using a microbial conversion of carbohydrates into digestible protein and additional cellular components that have immunogenic potential. Freezing injury, caused by freezing temperatures in the late fall, midwinter, or early spring, can result in significant loss to grape growers. The damage may range from the partial damage of parts of the plants to the total death of the plant, and may vary between years. Freezing tolerance is a multi-genetic, complex quantitative trait that involves many related traits like dormancy induction, growth cessation, acclimation, deacclimation and bud break. Biochar is produced from incomplete pyrolysis of plant biomass. Due to the unique properties of biochar such as a higher number of micropores, surface area and carbon content, and recalcitrant nature of carbon, biochar is hypothesized to improve physical and hydrological properties of soil. This study assesses the impact of biochar on soil organic carbon, physical and hydrological properties of two landscape positions under a corn (Zea mays L.)-soybean (Glycine max L.) rotation.
