Hypoxia inducible factors (HIFs) are central mediators of cellular responses to fluctuations of oxygen, an environmental regulator of stem cell activity. Muscle satellite cells are myogenic stem cells whose quiescence, activation, self-renewal and differentiation are influenced by microenvironment oxygen levels. However, the in vivo roles of HIFs in quiescent satellite cells and activated satellite cells (myoblasts) are poorly understood. Expression analyses indicate that HIF1α and HIF2α are preferentially expressed in pre- and post-differentiation myoblasts, respectively. Interestingly, double knockout of HIF1α and HIF2α (HIF1α/2α dKO) in embryonic myoblasts results in apparently normal muscle development and growth. However, HIF1α/2α dKO in postnatal …

Phytophthora blight is a serious threat to the Midwest vegetable industry, because the oomycete pathogen responsible for this disease, Phytophthora capsici, has a wide host range, can spread quickly in fields, and produces resilient oospores that can survive in soil for years. Phytophthora capsici has become resistant to commonly used fungicides and resistant crop varieties are rare. Amending soil with complex organic substrates has potential to improve soil quality and suppress soil-borne pathogens including P. capsici. Aflatoxin contamination in feed and feed ingredients is of high worldwide prevalence, posing serious risks to the livestock industry. A series of studies were conducted to better understand the comprehensive impact of aflatoxicosis and its interrelationships with dietary modifications in broiler chicks and ducks. Results of these studies emphasize the many factors that the severity of aflatoxicosis depends on, including animal species, age, presence of mycotoxin adsorbent, and concentration of major dietary nutrients. This dissertation empirically examines subsidies, aquaculture technology adoption, and welfare implications of fish farming households in Ghana and Kenya. It is divided into three separate chapters, each addressing a specific empirical issue related to aquaculture in the countries.

The second chapter of this dissertation applies the lognormal double hurdle model to a cross-section of fish farms to analyze commercial demand for improved feed, and whether the government feed subsidy program influences private demand for the feed. Consumers are increasingly concerned about the social and environmental impacts of the foods they purchase. Both choice experiments and best-worst scaling (BWS) methodology have been used to elicit consumer willingness to pay (WTP) for food attributes and the relative importance of food attributes to consumers. The dissertation consists of four essays. Chapter 2 presents a formal law review article exploring the laws associated with wild game harvest, consumption, and inspections. African leafy vegetables (ALVs) are a diverse set of crops grown across sub-Saharan Africa. They have been a staple of traditional diets and contain many critical micronutrients but their importance has been largely ignored by researchers and policymakers at the expense of imported crops. Availability, accessibility, and utilization of ALVs are limited by factors across the supply chain. This research investigated seed systems, drought tolerance, hydroponic production, market barriers, consumer preference, and the impact of gender as related to ALVs. Incremental gains in grain yield of maize hybrids over the decades are the consequence of genotype, environment and management interactions. Historically, genetic improvements in newer hybrids have included longer active grain filling periods (achieved by advancing silking and extending functional stay green in maize leaves); stronger source and sink during grain filling; enhanced tolerance to higher density; and canopy architecture changes. Chronic non-healing wounds (e.g., diabetic foot ulcers and bed sores) impact over 6.5 million Americans per year, costs in excess of $25 billion to treat on an annual basis, and are on the rise due to increasing levels of obesity and diabetes compounded by an aging population. The overall goal of this project was to reduce the aflatoxin level in the final coproduct of ethanol bioprocessing--DDGS. This was pursued by examining the effects of reduction of aflatoxin in the incoming corn prior to bioprocessing, the degradation of aflatoxin in the intermediate products, namely DWG and CDS, during processing, and aflatoxin degradation in the DDGS. Segregation techniques (size screening and density sorting) and detoxification methods (conventional and microwave heating, food additives, and high voltage atmospheric cold plasma) were evaluated for their effectiveness in aflatoxin reduction.

Probiotics are live microorganisms which may confer health benefits on the host when administered in appropriate amounts. Numerous studies have shown that probiotics improve bone health in humans and rodents with less information available on the skeletal health of avians given probiotics. The objective of this study was to determine the effects of probiotic supplementation on bone health of egg-laying and meat-type chickens. Dietary supplementation of a multi-species based probiotic reduced the percentage of shell-less eggs beginning at 4 wk following treatment and increased tibial and femoral bone mineral density in egg-laying hens at 7 wk post-treatment. Seriously mentally ill patients who are taking second-generation antipsychotics have a high risk of metabolic complications, including obesity, diabetes mellitus type II, hypertension, and hyperlipidemia. Guidelines to screen for metabolic syndrome were established by the American Diabetes Association, American Psychiatric Association, American Association of Clinical Endocrinologists, and North American Association for the Study of Obesity (Clark, 2004). Compliance with implementing the guidelines to screen and monitor for metabolic syndrome vary from regular monitoring to little or none. If the world population were to adhere to recommended levels of physical activity, health outcomes would improve. To that end, clinical practices need to consider exercise interventions to improve patient self-efficacy to adhere to recommended physical activity guidelines. Purpose: To evaluate the performance of lesion volumetry in hepatic CT as a function of various imaging acquisition parameters.

Methods: An anthropomorphic abdominal phantom with removable liver inserts was designed for this study. Two liver inserts, each containing 19 synthetic lesions with varying diameter (6–40 mm), shape, contrast (10–65 HU), and both homogenous and mixed-density were designed to have background and lesion CT values corresponding to arterial and portal-venous phase imaging, respectively. Metabolic syndrome is a condition in which the components – central adiposity, insulin resistance, atherogenic dyslipidemia, and elevated blood pressure - confer increased cardiovascular morbidity and mortality. A pilot clinical practice improvement project was developed and implemented using shared medical appointments to address cardiovascular disease risk in adult patients at a rural health care clinic on the southern Kenai Peninsula, Alaska who met the diagnostic criteria for metabolic syndrome. Statistically significant improvement in self-reported minutes of exercise was demonstrated for the nominal group of participants. Background: Enzyme-treated wheat bran (ETWB) contains a fermentable dietary fiber previously shown to decrease liver triglycerides (TGs) and modify the gut microbiome in mice. It is not clear which mechanisms explain how ETWB feeding affects hepatic metabolism, but factors (i.e., xenometabolites) associated with specific microbes may be involved. Background: High-amylose-maize resistant starch type 2 (HAMRS2) is a fermentable dietary fiber known to alter the gut milieu, including the gut microbiota, which may explain the reported effects of resistant starch to ameliorate obesityassociated metabolic dysfunction. Objective: Our working hypothesis was that HAMRS2-induced microbiome changes alter gut-derived signals (i.e., xenometabolites) reaching the liver via the portal circulation, in turn altering liver metabolism by regulating gene expression and other pathways. Trophic cascades involve powerful feeding interactions that can alter the flow of energy and the abundance of species in an ecosystem. In western NY streams, the negative impacts of a new benthic invertivore, the round goby (Neogobius melanostomus) is altering benthic community structure and leaf litter decomposition. Streams with round gobies have a reduced abundance of shredders and slower leaf decay than streams without gobies. However, crayfish, as shredders of leaf litter, may be large or aggressive enough to avoid predation by round gobies.
