Inquiry Of Lipid Membranes Interacting With Functional Peptides And Polyphenol Drug Molecules, Chian Sing Ho
Inquiry Of Lipid Membranes Interacting With Functional Peptides And Polyphenol Drug Molecules, Chian Sing Ho
USF Tampa Graduate Theses and Dissertations
Cellular membranes are important targets for many membrane-active peptides and drug compounds. Here we are interested in deciphering how lipid membranes are perturbed by several membrane-active molecules, including the transmembrane domain of the influenza M2 protein (M2TM), aggregates formed by a synthetic polyglutamine peptide, and three polyphenol compounds (i.e., tamoxifen, genistein, and verapamil). We employ phase-separated ternary lipid model membranes in the form of giant unilamellar vesicles (GUVs) to simulate raft-like structures that have been proposed to govern many important processes in plasma membranes (e.g., intracellular singling and trafficking). Specifically, we use fluorescent microscopy to interrogate how those membrane additives …

We previously identified a second-messenger-regulated signaling system in the environmental bacterium Pseudomonas fluorescens which controls biofilm formation in response to levels of environmental inorganic phosphate. This system contains the transmembrane cyclic di-GMP (c-di-GMP) receptor LapD and the periplasmic protease LapG. LapD regulates LapG and controls the ability of this protease to process a large cell surface adhesin protein, LapA. While LapDG orthologs can be identified in diverse

Staphylococcus aureus is a formidable human pathogen that uses secreted cytolytic factors to injure immune cells and promote infection of its host. Of these proteins, the bicomponent family of pore-forming leukocidins play critical roles in S. aureus pathogenesis. The regulatory mechanisms governing the expression of these toxins are incompletely defined. In this work, we performed a screen to identify transcriptional regulators involved in leukocidin expression in S. aureus strain USA300. We discovered that a metabolic sensor-regulator, RpiRc, is a potent and selective repressor of two leukocidins, LukED and LukSF-PV. Whole-genome transcriptomics, S. aureus exoprotein proteomics, and metabolomic analyses revealed that …

Novel Partitivirus Infection Of Bat White-Nose Syndrome (Wns) Fungal Pathogen Pseudogymnoascus Destructans Links Eurasian And North American Isolates, Ping Ren, Sunanda S. Rajkumar
KIP Articles
Bat White-nose Syndrome (WNS) fungus Pseudogymnoascus destructans had caused mass mortality in the North American bats. A single clone of the pathogen (Hap_1) was likely introduced in the United States while Eurasian population comprised of several haplotypes. The origin and spread of P. destructans remain enigmatic due in part to a lack of precise population markers. We searched for P. destructans mycoviruses as they are highly host-specific, and their spread could provide a window on the origin of the host fungus. We discovered a P. destructans bipartite virus PdPV-1 with two double-stranded RNA (dsRNA) segments - LS (1,683 bp) and …

Objective: Regulatory T cells (Tregs) contribute to HIV-1 disease progression by impairing antiviral immunity; however, the precise mechanisms responsible for the development of Tregs in the setting of HIV-1 infection are incompletely understood.
Design: In this study, we provide evidence that HIV-induced expansion of monocytic myeloid-derived suppressor cells (M-MDSCs) promote the differentiation of Foxp3+ Tregs.
Methods: We measured MDSC induction and cytokine expression by flow cytometry and analyzed their functions by coculturing experiments.
Results: We observed a dramatic increase in M-MDSC frequencies in the peripheral blood of HIV-1 seropositive (HIV-1+) individuals, even in those on antiretroviral therapy with undetectable viremia, …

The lengthy duration of multidrug therapy needed to cure tuberculosis (TB) poses significant challenges for global control of the disease. Moreover, chronic inflammation associated with TB leads to pulmonary damage that can remain even after successful cure. Thus, there is a great need for the development of effective shorter drug regimens to improve clinical outcome and strengthen TB control. Host-directed therapy (HDT) is emerging as a novel adjunctive strategy to enhance the efficacy and shorten the duration of TB treatment. Previously, we showed that the administration of CC-3052, a phosphodiesterase-4 inhibitor (PDE4i), reduced the host inflammatory response during Mycobacterium tuberculosis …

Vaccination As A Tool To Reduce Antimicrobial Resistance Worldwide, Bernard Vallat, Elisabeth Erlacher
Vaccine Technology VI
Antimicrobial resistance is a global human and animal health concern that is influenced by the non-appropriate use of antimicrobial agents in both human and veterinary medicine, as well as in the plant sector. To combat antimicrobial resistance, the World Organisation for Animal health (OIE) has developed science-based intergovernmental standards and guidelines covering terrestrial animals and aquaculture.
The OIE also contributed to the development of the WHO Global Action Plan on Antimicrobial Resistance, adopted in 2015 by the World Health Assembly. The 180 Member Countries of the OIE expressed their support to this plan of action through a Resolution, …

How To Deliver New Vaccines Under Very Short Timelines: The Zapi Project, Jean-Christophe Audonnet
Vaccine Technology VI
The ZAPI project is the first One Health project in the European Innovative Medicine Initiative (IMI) program. Its objective is to define and validate unique methodologies that can be applied when new zoonotic infectious diseases will occur in Europe or other regions of the world. The project is supporting an entirely new approach for “anticipating the unexpected”, and is defining new ways to alleviate the technical constraints which are typically encountered with the classical vaccine development processes.
The ZAPI project involves a total of 20 partners, from academic, biotech, and industry origin. One important feature of ZAPI is that it …

Background: Gay, bisexual, and other men who have sex with men (GBMSM) accounted for 67% of new US human immunodeficiency virus (HIV) infections in 2012; however, less than 40% of HIV-positive GBMSM are virally suppressed. Preventing transmission from virally unsuppressed men who have condomless anal sex (CAS) with serodiscordant partners is a public health imperative. New HIV infections in GBMSM are attributed in part to online access to sex partners; therefore, low-cost eHealth interventions are a unique opportunity to reach men where they meet partners.
Objective: To describe the protocol of a randomized controlled trial evaluating whether video-based messaging delivered …

Background: Taeniasis and cysticercosis are major causes of seizures and epilepsy. Infection by the causative parasite Taenia solium requires transmission between humans and pigs. The disease is considered to be eradicable, but data on attempts at regional elimination are lacking. We conducted a three-phase control program in Tumbes, Peru, to determine whether regional elimination would be feasible.
Methods: We systematically tested and compared elimination strategies to show the feasibility of interrupting the transmission of T. solium infection in a region of highly endemic disease in Peru. In phase 1, we assessed the effectiveness and feasibility of six intervention …

Temperature-Dependent Pathogenicity Of Grouper Iridovirus Of Taiwan (Tgiv), Hsin-I Liu, Ying-Chu Lin, Pinwen Peter Chiou, Hsin-Yiu Chou
Journal of Marine Science and Technology–Taiwan
Grouper iridovirus of Taiwan (TGIV) is one of the most devastating infectious pathogens of grouper fry in Taiwan. As environmental temperature often plays an important role in the outbreak of diseases, we assayed the impact of different temperatures (18, 25 and 32℃) on TGIV infection both in vitro and in vivo. Cytopathic effect (CPE) characterized as cell rounding and lysis were observed in TGIV-infected grouper SB (swim bladder) cells at 25℃ and 32℃, but was absent at 18℃. With temperature rising to 32℃, the presence of pycnotic nuclei and chromatin margination became prominent in the infected cells, indicating an apoptotic …

We describe a cohort of 50 elderly subjects, age at least 70 years. We present gender-specific findings in T lymphocyte markers and soluble immune mediators. We show the correlation between cytomegalovirus infection status with CD56dim NK cell responses to a variety of stimuli and with CD56bright/CD56dim NK cell ratio. We also present the correlation of retinol binding protein (RBP)−4 plasma levels with NK cell responses and we explore the relationship between gender and adiponectin, 25(OH)D (vitamin D), and RBP4 in affecting CD56dim NK cell responses. These data are discussed in Al-Attar et al. (2016)
