Perspectives On Transdermal Electroporation, Kevin B. Ita
Perspectives On Transdermal Electroporation, Kevin B. Ita
Faculty Publications & Research of the TUC College of Pharmacy
Transdermal drug delivery offers several advantages, including avoidance of erratic absorption, absence of gastric irritation, painlessness, noninvasiveness, as well as improvement in patient compliance. With this mode of drug administration, there is no pre-systemic metabolism and it is possible to increase drug bioavailability and half-life. However, only a few molecules can be delivered across the skin in therapeutic quantities. This is because of the hindrance provided by the stratum corneum. Several techniques have been developed and used over the last few decades for transdermal drug delivery enhancement. These include sonophoresis, iontophoresis, microneedles, and electroporation. Electroporation, which refers to the temporary …

Pseudomonas aeruginosa is a leading cause of human morbidity and mortality that often targets epithelial surfaces. Host immunocompromise, or the presence of indwelling medical devices, including contact lenses, can predispose to infection. While medical devices are known to accumulate bacterial biofilms, it is not well understood why resistant epithelial surfaces become susceptible to P. aeruginosa. Many bacteria, including P. aeruginosa, release outer membrane vesicles (OMVs) in response to stress that can fuse with host cells to alter their function. Here, we tested the hypothesis that mucosal fluid can trigger OMV release to compromise an epithelial barrier. This was tested using …

Recombinant subunit vaccines are an efficient strategy to meet the demands of a possible influenza pandemic, because of rapid and scalable production. However, vaccines made from recombinant Hemagglutinin (HA) subunit protein are often of low potency, requiring repeated boosting to generate a sustained immune response. Previously, we demonstrated improved immunogenicity of a plant-made H1 Hemagglutinin (HA) vaccine by chemical conjugation to the surface of the Tobacco Mosaic Virus (TMV) which is non infectious in mammals. Antigen coated TMV is taken up by mammalian dendritic cells and is a highly effective antigen carrier for subunit protein vaccines. In this work, we …

The aim of this project was to examine the effect of microneedle rollers on the percutaneous penetration of tiagabine hydrochloride and carbamazepine across porcine skin in vitro. Liquid chromatography-mass spectrometric analysis was carried out using an Agilent 1200 Series HPLC system coupled to an Agilent G1969A TOF-MS system. Transdermal flux values of the drugs were determined from the steady-state portion of the cumulative amount versus time curves. Following twelve hours of microneedle roller application, there was a 6.74-fold increase in the percutaneous penetration of tiagabine hydrochloride (86.42 ± 25.66 µg/cm2/h) compared to passive delivery (12.83 ± 6.30 µg/cm …

Isavuconazole In The Treatment Of Invasive Aspergillosis And Mucormycosis Infections, Monica A. Donnelley, Elizabeth S. Zhu, George R. Thompson Iii
Faculty Publications & Research of the TUC College of Pharmacy
We have a limited arsenal with which to treat invasive fungal infections caused by Aspergillus and Mucorales. The morbidity and mortality for both pathogens remains high. A triazole antifungal, isavuconazole, was recently granted approval by the US Food and Drug Administration and the European Medicines Agency for the treatment of invasive aspergillosis and mucormycosis. A randomized double-blind comparison trial for the treatment of invasive aspergillosis found isavuconazole noninferior to voriconazole. A separate, open-label study evaluating the efficacy of isavuconazole in the treatment of mucormycosis found comparable response rates to amphotericin B and posaconazole treated historical controls. The prodrug isavuconazonium sulfate …
Counterfeit Industry And The Link To Terrorism, Holly Barbara Holt
Counterfeit Industry And The Link To Terrorism, Holly Barbara Holt
Apparel Design and Hospitality Management
The purpose of this research study is to explore whether consumers would be complicit in the purchase of counterfeit goods once becoming aware of the counterfeit industry being linked to terrorism. Counterfeit goods are defined as identical copies of authentic products and they are produced without the permission of the registered owner (Carpenter & Lear, 2011). Almost any product can be counterfeited from clothing, shoes, jewelry, handbags and even medicines. Counterfeit products are sold at a fraction of the cost of the authentic product. This study identifies the ‘why’ to consumer complicity to purchase the counterfeit items. There are legalities …

Mangrove Enhancement As A Climate Change Adaptation Strategy In The Republic Of The Marshall Islands (Rmi): Potential Ecosystem Service Shifts Following Colonization, Andrew S. Harwood
Environmental Science and Management Professional Master's Project Reports
The ability of Small Island Developing States (SIDS) and Pacific Island communities to adapt to the increasing sea-levels, storm surge, saltwater intrusion, and other threats posed by global climate change is critical to the survival of many of these local communities as well as their cultures (LivingIslands.org). One of the world’s most striking examples of a nation at great risk is the Republic of the Marshall Islands (RMI) in the central Pacific Ocean. Due to a combination of factors, including the country’s extremely low elevation above sea-level (an average of 2 meters) and lack of adequate structures to provide safety …

Environmental Science and Management Professional Master's Project Reports
In this project, I analyzed the status and trends of water quality data describing fecal bacteria (E. coli) and DO in the Amazon and Coyote Creek watersheds of the Southern Willamette Valley, Oregon. I also examined TMDL implementation plans produced by DMAs, determined if and how implementation activities corresponded to changes in water quality, compared management and planning of DMAs in the watersheds, discussed aspects of the current TMDL, and compared state regulations and standards in other states. I concluded by making management recommendations to better facilitate future status and trend analysis.

In 1999 the City of Portland (City) began to require that stormwater management facilities (SMF) be built when private property is newly developed or redeveloped (City Code Chapter 17.38). Proper maintenance and upkeep of SMFs is essential to ensuring they function appropriately. The City's Maintenance Inspection Program (MIP) is tasked with inspecting stormwater management facilities on private properties in order to ensure that they are being properly operated and maintained and to meet provisions of the City's NPDES Municipal Separated Storm Sewer System (MS4) permit.
Greenroofs are one type of SMF that are installed to satisfy this requirement. Understanding the …

Elevated indoor CO2 levels are indicative of insufficient ventilation in occupied spaces and correlate with elevated concentrations of pollutants of indoor origin. Adverse health and well-being outcomes associated with elevated indoor CO2 levels are based on CO2 as a proxy, although some emerging evidence suggests CO2 itself may impact human cognition. Using portable monitors, we conducted an exposure study with 16 subjects in Singapore to understand the levels, dynamics and influencing factors of personal exposure to CO2. Participants carried a CO2 monitor continuously for 7-day periods recording their exposure levels at 1-min intervals. …

It is important to characterize the temporal trajectories of disease-related biomarkers in order to monitor progression and identify potential points of intervention. These are especially important for neurodegenerative diseases, as therapeutic intervention is most likely to be effective in the preclinical disease stages prior to significant neuronal damage. Neuroimaging allows for the measurement of structural, functional, and metabolic integrity of the brain at the level of voxels, whose volumes are on the order of mm3. These voxelwise measurements provide a rich collection of disease indicators. Longitudinal neuroimaging studies enable the analysis of changes in these voxelwise measures. However, …

Optical coherence tomography (OCT) of the human retina is now becoming established as an important modality for the detection and tracking of various ocular diseases. Voxel based morphometry (VBM) is a long standing neuroimaging analysis technique that allows for the exploration of the regional differences in the brain. There has been limited work done in developing registration based methods for OCT, which has hampered the advancement of VBM analyses in OCT based population studies. Following on from our recent development of an OCT registration method, we explore the potential benefits of VBM analysis in cohorts of healthy controls (HCs) and …
