Fanconi anemia (FA) is a genetic disease characterized by bone marrow failure and increased cancer risk. The FA proteins function primarily in DNA interstrand crosslink (ICL) repair. Here, we have examined the role of the PTEN phosphatase in this process. We have established that PTEN-deficient cells, like FA cells, exhibit increased cytotoxicity, chromosome structural aberrations, and error-prone mutagenic DNA repair following exposure to ICL-inducing agents. The increased ICL sensitivity of PTEN-deficient cells is caused, in part, by elevated PLK1 kinase-mediated phosphorylation of FANCM, constitutive FANCM polyubiquitination and degradation, and the consequent inefficient assembly of the FA core complex, FANCD2, and …

Most physiological processes are sensitive to pH (i.e., hydrogen ion activity) and regulation of pH within tissues and body fluids represents a fundamental homeostatic process in living organisms. At the organismal level, acid-base balance is achieved by fast respiratory modulations of the partial pressure of CO2 in combination with slower transepithelial ion movements to accommodate excretion and retention of bicarbonate and protons in response to acid-base disturbances. At the cellular and subcellular levels, acid-base balance is achieved by ion exchange.

Isoforms of the Fcγ receptor III (FcγRIII or CD16) are cell surface receptors for the Fc portion of IgG and important regulators of humoral immune responses. Different ligand binding kinetics of FcγRIII isoforms are obtained in three dimensions by surface plasmon resonance and in two dimensions by a micropipette adhesion frequency assay. We show that the anchor structure of CD16 isoforms isolated from the cell membrane affects their binding affinities in a ligand-specific manner. Changing the receptor anchor structure from full to partial to none decreases the ligand binding affinity for human IgG1 (hIgG1) but increases it for murine IgG2a …

It has been hypothesized that the nicotinic acetylcholine receptors (nAChRs) play important roles in nicotine dependence (ND) and influence the number of cigarettes smoked per day (CPD) in smokers. We compiled the associations between nicotinic cholinergic receptor genes (CHRNs) and ND/CPD that were replicated across different studies, reviewed the expression of these risk genes in human/mouse brains, and verified their expression using independent samples of both human and mouse brains. The potential functions of the replicated risk variants were examined using cis-eQTL analysis or predicted using a series of bioinformatics analyses. We found replicated and significant associations for ND/CPD at …

Simulation Of Value Stream Mapping And Discrete Optimization Of Energy Consumption In Modular Construction, Md Mukul Chowdhury
Theses and Dissertations
With the increased practice of modularization and prefabrication, the construction industry gained the benefits of quality management, improved completion time, reduced site disruption and vehicular traffic, and improved overall safety and security. Whereas industrialized construction methods, such as modular and manufactured buildings, have evolved over decades, core techniques used in prefabrication plants vary only slightly from those employed in traditional site-built construction. With a focus on energy and cost efficient modular construction, this research presents the development of a simulation, measurement and optimization system for energy consumption in the manufacturing process of modular construction. The system is based on Lean …

CML is effectively treated with tyrosine kinase inhibitors (TKIs). However, the efficacy of these drugs is confined to the chronic phase of the disease and development of resistance to TKIs remains a pressing issue. The anti-inflammatory COX2 inhibitor celecoxib has been utilized as anti-tumour drug due to its anti-proliferative activity. However, its effects in hematological malignancies, in particular CML, have not been investigated yet. Thus, we tested biological effects and mechanisms of action of celecoxib in Philadelphia-positive (Ph+) CML and ALL cells. We show here that celecoxib suppresses the growth of Ph+ cell lines by increasing G1-phase and apoptotic cells and …

School of Psychology
PurposeThere is a need for improved measurement of motivation for diabetes self-care. The Elaborated Intrusion Theory of Desire offers a coherent framework for understanding and identifying the cognitive-affective events that constitute the subjective experience of motivation and may therefore inform the development of such an instrument. Recent research has shown the resultant Motivation Thought Frequency scale (MTF) to have a stable factor structure (Intensity, Incentives Imagery, Self-Efficacy Imagery, Availability) when applied to physical activity, excessive snacking or alcohol use in the general population. The current study aimed to confirm the four-factor structure of the MTF for glucose testing, physical activity …

BACKGROUND: Tibetan wild barley (Hordeum spontaneum L.) has been confirmed to contain elite accessions in tolerance to abiotic stresses, including salinity. However, molecular mechanisms underlying genotypic difference of salt tolerance in wild barley are unknown. RESULTS: In this study, two Tibetan wild barley accessions (XZ26 and XZ169), differing greatly in salt tolerance, were used to determine changes of ionomic, metabolomic and proteomic profiles in the shoots exposed to salt stress at seedling stage. Compared with XZ169, XZ26 showed better shoot growth and less Na accumulation after 7 days treatments. Salt stress caused significant reduction in concentrations of sucrose and metabolites …
A Comparison Of The Immediate Effects Of Muscle Energy Technique And Joint Mobilizations On Posterior Shoulder Tightness In Youth Throwing Athletes, Maddox L. Reed
Theses and Dissertations
Background: Due to the extreme forces on the glenohumeral (GH) joint during the throwing motion, and the high number of repetitions that occur during overhead sports, alterations in range of motion are a common occurrence in overhead athletes, particularly baseball and softball players. The presence of limited GH motion that occurs as a result of posterior shoulder tightness can increase the risk of injury. Despite clinical evidence pointing to the use of joint mobilizations and muscle energy technique (MET) for the treatment of various pathologies, there currently is no data examining comparing the overall effectiveness of joint mobilizations and MET …

(In)Equitable Subrogation: The Federal Circuit’S Irrational And Unworkable Progress Payment Framework In Balboa, Will F. Hawkins
Duke Law Journal
American taxpayers spend more than $100 billion per year on federal construction projects. Yet massive construction delays, huge budget overruns, and unorganized contractors increase the cost of construction for the federal government. Passed in 1935, the Miller Act attempted to protect the federal government in the event that the contractor defaulted or was unable to complete the project. By requiring contractors to enlist third party “sureties” as guarantors on projects, the Miller Act provides the government with the assurance that another party will step in to complete projects if need be. Contractors are typically paid via periodic progress payments, with …
A Validation Of The Efficacy Of Descriptive Instrumental Collective Case Study Research Methodology For Examining Pilot Cognitive Functioning, Clint R. Balog
A Validation Of The Efficacy Of Descriptive Instrumental Collective Case Study Research Methodology For Examining Pilot Cognitive Functioning, Clint R. Balog
International Journal of Aviation, Aeronautics, and Aerospace
The research conducted developed a descriptive understanding of how the cognitive processes of risk assessment, problem solving, and decision making, as well as other supportive processes, are employed by pilots-in-command (PICs) during the experience of extended, extreme, in-flight emergencies. This understanding is then applied to similar dynamic, operational environments. The research also validated the applicability and efficacy of Robert Stake’s 1995 descriptive, instrumental, collective case study methodology as a tool for investigating such phenomenon and developing such an understanding. Specifically, the research details the necessary procedures for employing this methodology successfully, and provides example of those procedures, and their results, …

Stress Coping Strategies In Indian Military Pilots-Preliminary Observations, Catherine Joseph
International Journal of Aviation, Aeronautics, and Aerospace
ABSTRACT
In aviators, stress coping has been linked to flying performance and has a bearing on flight safety and mission effectiveness. The objective of this research was to study coping strategies utilized by Indian military pilots. A sample of 160 military pilots was asked to complete a demographic data sheet and the Stress Coping Checklist. Preliminary findings suggest that, Indian pilots use flexible problem and emotion focussed coping strategies. This issue needs to be taken into account in both occupational and clinical realms of future research investigations on aviators.

© 2016 The Author(s). Background: Cardiovascular diseases were the leading cause of death in women in the United Arab Emirates (UAE) in 2010. The UAE is expected to experience a tripling of heart diseases in the next two decades as risk factors for heart diseases increase. Research shows that first year survival rates of younger women suffering from a heart attack are lower than in men. Women present with a wider range of symptoms for heart diseases than men; non-recognition of atypical symptoms may explain the delay in seeking treatment and poor prognosis following heart diseases in women. Biochars have shown a great potential to treat stormwater runoff contaminated with heavy metals due to their favorable physical and chemical characteristics. Biochar materials were produced from pyrolysis of oak tree and wood at 400C and 450C respectively, and their Zn adsorption behavior from aqueous solutions were evaluated to assess their applicability as a filter media for stormwater treatment. Two adsorption isotherm models, Freundlich and Langmuir, were used to fit the batch-scale experimental data. The kinetics of Zn adsorption was investigated under two contrasting physical condition (stagnant vs. agitated). The adsorption isotherm was better fitted with the Langmuir model (R2 …

CD99 is a cell surface molecule that has emerged as a novel target for Ewing sarcoma (EWS), an aggressive pediatric bone cancer. This report provides the first evidence of methuosis in EWS, a non-apoptotic form of cell death induced by an antibody directed against the CD99 molecule. Upon mAb triggering, CD99 induces an IGF-1R/RAS/Rac1 complex, which is internalized into RAB5-positive endocytic vacuoles. This complex is then dissociated, with the IGF-1R recycling to the cell membrane while CD99 and RAS/Rac1 are sorted into immature LAMP-1-positive vacuoles, whose excessive accumulation provokes methuosis. This process, which is not detected in CD99-expressing normal mesenchymal …
